양식 넙치에서 연쇄구균 혼합백신 접종 후 항체가 변화 조사 (Development of antibodies after combination Streptococcus iniae and Streptococcus parauberis of commercial vaccine injection in olive flounder (Paralichthys olivaceus) in farm)

  • Streptococcosis, caused by Streptococcus iniae and Streptococcus parauberis is an important bacterial disease that affects in olive flounder in Korea. In Korea, multivalent bacterial vaccines are used to prevent streptococcal diseases in aquaculture. In this study, commercial vaccines containing formalin-inactivated bacterial cells of S. iniae and S. parauberis were administered at six fish farms and one unvaccinated fish farm were designated for investigation (Wando; 4 sites and Jeju; 3 sites). Blood was collected from vaccinated and unvaccinated olive flounders, and titers of antibodies against S. iniae and S. parauberis in serum were analyzed using ELISA. After a one shot vaccination in the farms at Jeju (farm A) and Wando (farm D), the proportion of individuals with specific antibodies against S. parauberis OD values of 0.4 or higher was 60% and 53.5%, respectively. But after booster vaccination, the proportion of individuals with serum OD values of 0.4 or higher was higher substantially increased to 96.6% (farm A) and 100% (farm D). The levels of S. parauberis specific antibodies of olive flounder were increased after vaccination in three fish farms (farm D, E, and F), but not S. iniae specific antibodies.

3년간(1998-2000년) A군 연구균 감염의 혈청학적 형분류와 역학적 특징 (Serological Analysis and Epidemiologic Characteristics of Group A Streptococci in Seoul(1998-2000))

  • 목 적: A군 연쇄구균의 세포벽은 M단백과 T단백으로 구성되어 있고, M단백의 항원성은 현재 90여 가지로 분류되고 있으며 균의 독성과 관련이 있고 질병의 병인론을 연구하는데 도움이 된다. 반면에 T단백은 항원성 분류는 연쇄구균의 역학적 상황을 이해하는데 유효하다고 알려져 있다. 이에 저자들은 지난 3년 동안 서울시내 5개 지역 병의원의 외래 및 입원환자를 대상으로 분리된 A군 연구균의 T단백 분류를 통해 역학적 동태를 알고자 하였다. 방 법: 1998년부터 2000년까지 서울 시내(남부, 북동부, 북부, 북서부, 북부)를 5개 지역으로 분류하여 외래 및 입원환자를 대상으로 분리된 A군 연구균의 T단백의 혈청형 분석을 시행하여 연도별 A군 연구균의 혈청학적 형 분류와 침투성질환에서 분류된 연구균의 혈청학적 형 분류의 특성을 알아보았다. 결 과 :1998년에 5개 지역에서 시행한 92례의 아동에서 A군 연구균을 분리하여 T12, T4, Non-typed(NT)가 72.2%를 차지하였고, 이중 성홍열은 7례가 있었으며 T12와 T4가 각각 3, 4례를 차지하였으며 경부임파선염 1례(T12), 편도농양 2례(T8, NT)이었다. 1999년에는 41례의 아동에서 시행하였으며 T12, T4, T1가 68%의 분포를 보였으며 성홍열은 5례 중 T12, T4가 3례를 차지하였고 폐렴 2례(T4, T1)와 경부임파선염 1례(T8/25)를 나타내었다. 2000년도는 서울 중부를 제외한 지역에서 시행한 83례의 아동에서 T12, T4, T1이 전체의 63.9%를 보였고 성홍열은 3례(T12, T4, T5 각각 1례)와 편도농양 T12 1례와 폐렴 NT 1례, 패혈증 T1 1례를 나타내었다. 결 론: A군 연구균의 T단백의 혈청학적 분석은 서울 내에서의 지역적 특정 분포는 없으나 T12가 감소하는 연도별 차이를 보였다. 성홍열이나 침투성 질환에서 특정 T단백의 분포도를 보이지는 않았다. 이에 T단백의 혈청학적 분석이 질병의 진단과 치료에 도움이 될 것으로 사료되며 앞으로 주기적으로 더 많은 아동을 대상으로 하는 역학적 검사가 이루어 져야할 것이다.

혈액성상(血液性狀)에 따르는 양식어(養殖魚)의 건강진단(健康診斷) (Health Assessment by Hematological Studies and Blood chemistries in cultured carps)

  • 양식어류(養殖魚類)의 건강평가기준(健康評價基準)을 위한 기초자료(基礎資料)를 확보하고자 저자(著者)등은 1988년(年) 6월(月) 부터 1989년(年) 5월(月)까지 이스라엘잉어를 이용한 일련의 혈액조사(血液調査)를 실시하여 다음과 같은 결과(結果)를 얻었다. 1. 건강(健康)한 이스라엘잉어의 혈액상(血液像)은 Hct $32{\pm}3.4%$, Hb $8.3{\pm}0.9g/dl$, RBCs $132{\times}10^4{\pm}27.6{\times}10^4mm^3$, 총단백(總蛋白) $2.7{\pm}0.7g/dl$, GOT $143{\pm}19.5units$, GPT $50{\pm}14.2units$, 총혈당 $70{\pm}12.6mg/dl$, BUN $7.5{\pm}1.9mg/dl$ 등이었다. 2. 온수의 변화(變化)에 따른 혈액상(血液像)은 $23^{\circ}C$를 대조군(對照群)으로하여 $5^{\circ}C$ 하강(下降)시($18^{\circ}C$) Hct 및 RBCs수(數)가 감소(減少)하였으며, $5^{\circ}C$ 상승(上昇)시($28^{\circ}C$) Hct, RBCs수(數) 및 glucose량(量)이 증가(增加)하였다. 3. 용존산소량(溶存酸素量)의 변화(變化)에 따른 혈액상(血液像)은 4.5ppm을 대조군으로 하여 용존산소 감소(減少)시(3ppm) Hct 및 RBCs수가 증가(增加) 하였으며 용존산소 증가(增加)시(7ppm) Hct 및 RBCs수가 감소 하였다. 4. 연쇄구균(連鎖球菌) $10^6cells$/fish를 근육(筋肉)에 접종(接種)한 경우 염증성(炎症性) 반응(反應)과 함께 Hct, RBCs, glucose, BUN치(値)가 현저(顯著)히 감소(減少) 하였으며 이들은 6 일(日)만에 회복 되었다. 5. 연쇄구균 $10^4cells$/fish를 복강내(腹腔內)에 접종한 경우 역시 염증성 반응과 함께 Hct, RBCs, BUN치(値)가 감소 되었으나, 이 경우 폐사(斃死)된 예(例)는 없었다. 6. Edwardsiella tarda균 $1.2{\times}10^8cells$/fish를 근육(筋肉)에 접종한 경우 Hct 및 RBCs치(値)는 감소하였고, TCHO, GOT, GPT, BUN치(値)는 증가 하였으며, 격심(激甚)한 염증성(炎症性) 반응(反應)과 함께 접종 3~4일(日) 내(內)에 접종어(接種魚)의 60%가 폐사(斃死)하였다.

치면세균막에서 분리한 뮤탄스 연쇄상구균 및 Streptococcus anginosus의 수종 항생제에 대한 감수성 조사 (ANTIBIOTIC SUSCEPTIBILITY IN MUTANS STREPTOCOCCI AND STREPTOCOCCUS ANGINOSUS ISOLATED FROM DENTAL PLAQUE)

  • The aim of this study was to investigate the susceptibility of mutans streptococci (S. mutans and S. sobrinus) and Streptococcus anginosus, for seven antibiotics, penicillin G, amoxicillin, ciprofloxacin, cefuroxime, erythromycin, bacitracin, and vancomycin. The minimum inhibitory concentration (MIC) of seven antibiotics against 3 species (type strains) of mutans streptococci and S. anginosus, 10 strains (wild type) of S. mutans, 7 strains (wild type) of S. sobrinus, and 11 strains (wild type) of S. anginosus, were measured by broth dilution method. All of the type strains of mutans streptococci and S. anginosus had the same susceptibility for penicillin G, amoxicillin, cefuroxime and bacitracin. Type strain of S. anginosus was sensitive in ciprofloxacin, but those of mutans streptococci were not. All of the clinical isolates of mutans streptococci and S. anginosus had the same susceptibility for the seven antibiotics. Our data reveal that mutans streptococci and S. anginosus have similar antibiotic-resistant character. In addition. these results may offer the basic data to verify the antibiotic-resistant mechanism of mutans streptococci and S. anginosus.

Streptococcus iniae의 유전학적 다양성과 RAPD fingerprint profile의 비교 (Phylogenetic Diversity and Comparison of RAPD Fingerprint Profile of Streptococcus iniae)

  • 2004년부터 2005년까지 제주도에서 전형적인 연쇄구균증상을 나타내는 양식넙치로부터 Streptococcus sp.를 분리하였다. 생화학적 성상 조사와 multiplex PCR assay법을 통해 S. iniae 94 균주를 동정하였다. RAPD fingerprint profile을 조사해 본 결과 A, B, C-type 총 3가지의 genotype을 확인하였으며, 동일종에 대해 외국에서 보고된 선행 연구결과와 비교하였을 경우, 지리적으로 다른 유전적 다형현상을 나타내는 것으로 추정되었다. 근래에 제주도에 우점하는 genotype은 A-type이었고, RAPD fingerprint profile은 약 2,000 bp, 1,300 bp, 1,000 bp, 850 bp, 550 bp의 밴드 양상을 나타내었다. S. iniae의 serotype 동정에 유효한 것으로 보고되어진 P14 random primer는 유전적 다형현상에 의해 제주도에서 분리된 S. iniae의 serotype 동정에 적합하지 않은 것으로 판단되었다.

치아우식경험과 Streptococcus mutans의 관련성 (Relationship between Streptococcus mutans levels and dental caries experience)

  • Objectives : The aim of the present study was to measure the level of Streptococcus mutans(S. mutans), the major causative agent in dental caries, and to examine the relationship between S. mutans level and dental caries experience. Methods : The present study was carried out in 41 university students. S.mutans plaque was detected in the saliva using Dentocult$^{(R)}$-SM and was measured in absorbance. The data were analyzed in the relevance for number of dental caries experience. Results : The group of S. mutans value has progressive caries, which was higher than that of non-suffering caries(p<0.05). More caries experience showed higher value of S. mutans(p>0.05). According to the relationship with DMFT, level of S. mutans and absorbance, correlation with the level of salivary S. mutans is shown statistically highly significant (p<0.01). The level of S. mutans in saliva and plaque and absorbance is statistically significant(p<0.05). Conclusions : The levels of S. mutans in both group of high caries experience and progressive caries was highly detected, continuous oral health management should be carried out to prevent the possible dental caries.

연쇄상구균의 약제내성과 전이성 R-plasmid의 특성 (Multiple Drug Resistance and Transferable R-plasmid of Streptococci Isolated from Diseased Olive Flounder)

  • MICs of 8 chemotherapeutic agents against forty streptococcal isolates were determined. These strains were isolated from diseased olive flounder, Paralichthys olivaceus, and showed 2-5 multiple drug resistance against different antibacterial agents including ampicillin, doxycycline (DOXY), erythromycin, florfenicol, flumequine, norfloxacin, oxolinic acid, and oxytetracycline (OTC). In conjugation experiment, we found transferable R plasmids carrying OTC and DOXY resistance determinant in 3 drug resistance strains analyzed. Six out of 40 isolates showed positive signal in colony hybridization with the R plasmid DNA (pST9) as a probe. It suggests that other types R plasmid different from pST9 is also involving in multiple drug resistance of streptococci isolated from olive flounder.

연쇄상구균의 표현형적 특성과 RAPD profiles 비교 (Comparison of RAPD Profiles and Phenotypical Characters of Streptococcal Strains)

  • Streptococcal infection is one of the most serious disease of cultured olive flounder, Paralychthys olivaceus in Korea and caused by more than one species. However, there has been considerable confusions about the taxonomic position of the fish pathogenic streptococci. In this study, We performed the randomly amplified polymorphic DNA(RAPD) pattern analysis to evaluate the possible classification in 8 streptococci isolated from diseased olive flounder and reference strains based on their DNA structure. RAPD PCR with DNA solution prepared by simple boiling and 10-mer random primer was appeared to be a good tool for discrimination of different streptococcal strains. Phenotypical characters by simple biological test and API 20 Strep corresponded well to the specific profiles of RAPD in streptococcal isolates of this study. Therefore, the RAPD profile was considered as one of differential characters to discriminate the streptococcal isolates from diseased olive flounder.

Streptococcus mutans에 대한 티트리의 항균 효과 (Antibacterial effect of tea tree on Streptococcus mutans)

  • Objectives: The purpose of this study is to investigate the antibacterial effect of Streptococcus mutans of tea tree ingredient. Methods: The experimental groups were each given with different concentrations (30 or 50 vol%) of tea tree prepared in saline solution. The control group applied only saline solution. The tea tree coating of the specimen were examined under a scanning electron microscope. For the antibacterial activity test of the tea tree, the contact angle of the tea tree- coated specimen's surface was analyzed. The antibacterial effect against Streptococcus mutans was determined by counting the colony forming units (CFU). The statical statics were evaluated by using one-way ANOVA and paired t-test. Results: The tea tree treated group of hydrophilic more than non treated group. Antibacterial experiments demonstrated that tee tree solution was effective against Streptococcus mutans. However there was no significant difference in depending solution concentration groups. Conclusions: The antimicrobial activity of the tea tree containing solution showed its potential for use as coating for denture and medical materials.

Jaborandi extract와 Sorbitol이 구강미생물 증식에 미치는 영향 (Antimicrobial activity of jaborandi extract and sorbitol to oral microbes)

  • Objectives : The aim of the study is to investigate the antimicrobial activity of jaborandi and sorbitol to various oral microbes. Methods : Jaborandi leaves contain pilocarpine. The conditions for extraction were optimized on the basis of substances, temperatures and pHs. Total pilocarpine content after extraction was analyzed by HPLC. The herbal antimicrobial activity of jaborandi and sorbitol were evaluated for oral microbes containing ATCC 25175 S. mutans, ATCC 13419 S. salivarius, ATCC 6249 S. mitis, ATCC 33398 S. equi, ATCC 29213 S. aureus, ATCC 14053 C. albicans. Results : The optimum conditions for highest yielding extraction were pilocarpine content after boiling at $100^{\circ}C$ for 1 hour at pH 3. The level of total pilocarpine content was analyzed at 833 mg/kg by HPLC. The most effective antimicrobial activity was obtained by combination of pilocarpine and sorbitol rather than pilocarpine, menthol and sorbitol, respectively. Conclusions : This results supported the preventive oral health care using safe and convenient jaborandi herb.
