The Antibacterial Effects of Oriental Medicinal Herbs on Bacteria Isolated from Contaminated Beverages (오염된 음료에서 분리된 세균에 대한 한약재의 항균성)

  • The use of synthetic additives for preservation and enhancement of the market quality of food products has been emerging as a societal issue in terms of safety as well as changes in consumption patterns. Various research related to natural additives is being conducted to address these issues. This study examined the antibacterial effects of 79 types of medicinal herbs used as oriental remedies on bacteria isolated from beverages of damaged marketable quality. The antibacterial effects of methanol extracts on 13 Bacillus sp. and three Paenibacillus sp. were evaluated. We found that 43 of the herbal medicines analyzed had antibacterial effects on the isolated bacteria. Of those, eight were selected, and their antibacterial effects were further examined using water, ethanol, methanol, and ethyl acetate as solvents. The results revealed that Prunus mume, Rhus javanica, and Coptis japonica had excellent antibacterial effects against the isolated bacteria. In particular, they exerted greater antibacterial effects when water and ethanol were used as solvents. This result indicates the possibility of developing natural additives using these substances. Since P. mume in particular, has not been sufficiently studied compared to other herbal medicines, it presents an opportunity for additional investigation and the possibility for development as a new product in the future.

Seed Disinfectant Effect of Pleurotus ostreatus (Heuktari) Extract on Fusarium fujikuroi Nirenberg (흑타리버섯 추출물의 벼 키다리병에 대한 종자소독 효과)

  • This study was carried out to investigate antimicrobial activities of water extracted Pleurotus ostreatus var Heuktari (PO), and their application to F. fujikuroi Nirenberg growth inhibition material. Various organic solvents (chloroform, hexane, ethyl acetate, ethanol) were tested to investigate the antimicrobial activities of Pleurotus ostreatus var Heuktari against F. fujikuroi Nirenberg. Chloroform, hexane, ethyl acetate and hot water extracts had no antimicrobial activities, but butanol extract showed 2.5% strong activities in order of F. fujikuroi Nirenberg disk diffusion test. Then we observe Antifungal activity using green house. Bed soil and mushroom extract mixed at 10%, 7.5%, 5%, 2.5%, 1% total bed soil mass. Screening of Antifungal activity was tested two periode 18day and 25day. The Antifungal activity rate of each period and extract density was 7.5% mixed bedsoil has investigated 80% Antifungal activity rate. The result indicates that the Pleurotus ostreatus var Heuktari extract using F. fujikuroi Nirenberg registance, which shows the development of organic agriculture seed disinfectant.

Commercial ${\gamma}$-Oryzanol Inhibits the Formation of C-7 Oxidized Cholesterol Derivatives (OCDs) in an Aqueous Model System during Cholesterol Autoxidation (수용성 모델시스템 내에서의 상업적 ${\gamma}$-Oryzanol의 C-7 산화 콜레스테롤 유도체 생성 저해효과)

  • The inhibition of cholesterol autoxidation by commercial ${\gamma}$-Oryzanol (0, 50, 100, and 300 ppm) was studied in an aqueous model system for 20 h at pH 5.5 and $80^{\circ}C$. The inhibition effectiveness of the commercial ${\gamma}$-Oryzanol was followed by the retention of cholesterol and the formation of C-7 oxidized cholesterol derivatives (OCDs). Changes in the amount of ${\gamma}$-Oryzanol in the aqueous system were determined during cholesterol autoxidation. A method to detect the levels of 7-ketocholesterol, $7{\alpha}$-hydroxycholesterol and $7{\beta}$-hydroxycholesterol in an aqueous model system with ${\gamma}$-Oryzanol was developed by using the hexane-ethyl acetate extraction system and high-performance liquid chromatography. Results showed that the levels of C-7 OCDs in an aqueous dispersion containing 300 ppm of ${\gamma}$-Oryzanol were not significantly (p>0.05) increased, when compared to other treatments (0, 50, and 100 ppm), during the accelerated cholesterol oxidation.

Anti-mycobacterial Effects of the Extract of Humulus japonicus (환삼덩굴(Humulus japonicus) 추출물의 항결핵 효과)

  • The present study aimed to evaluate the in vitro antimycobacterial effects of hop plant, Humulus japonicus. Methanol extract of H. japonicus (MeOH extract) showed strong direct bactericidal effects against Mycobacterium tuberculosis in vitro. Furthermore, the MeOH extract significantly inhibited M. tuberculosis growth in human macrophages. When five fractions obtained from MeOH extract were examined using the same methods, the hexane and ethyl acetate fractions showed bactericidal effects against M. tuberculosis in vitro, whereas the butanol and water fractions inhibited M. tuberculosis growth in macrophages. Because H. japonicus extract exhibited antimycobacterial activity against both free M. tuberculosis in culture medium and intracellular M. tuberculosis in human macrophages, this plant might be a good candidate for development of a new anti-tuberculosis drug.

Establishment of Analytical Method for Pencycuron in Representative Agricultural Commodities by High-Performance Liquid Chromatography (대표 농산물 중 살균제 Pencycuron의 HPLC 정밀 잔류분석법 개발)

  • The single residue analytical method was developed for determining fungicide pencycuron residues in various agricultural commodities with high-performance liquid chromatography (HPLC). Pencycuron residue was extracted with acetone from representative crops such as Korean cabbage, apple, brown rice and green pepper. After ethyl acetate/n-hexane partition and subsequent clean-up with silica gel chromatography, pencycuron residue was quantified by reversed phase HPLC with UV detection at 240 nm. The suspected residue of pencycuron was confirmed using selected-ion monitoring (SIM) LC/mass spectrometry (MS). Instrumental limit of quantitation (ILOQ) and method LOQ (MLOQ) were set at 2 ng and 0.02 mg/kg, respectively. Overall recoveries of pencycuron from different crop samples fortified at three levels (MLOQ, 10MLOQ, 100MLOQ) were 72~108%. This proposed method could be useful as official analytical method for quantification of pencycuron residues in agricultural commodities.

Effects of Pine Needle Ethyl Acetate Fraction on Acctylcholine(ACh) and Its Related Enzymes in Brain of Rats (뇌 조직의 아세틸콜린 및 그 관련효소에 미치는 솔잎(Pine Needle) 에틸아세테이트획분의 영향)

  • This study was designed to investigate the effects of ethyl acetate(EtOAc) fraction of pine (Pinus densiflora Sieb et Zucc) needle on cholesterol and lipofuscin(LF) accumulations, acetylcholine(ACh) and its related enzyme activities such as choline acetyltransferase(ChAT) and acetylcholinesterase(AChE), and monoamine oxidase-B(MAO-B) activity, which destroyed the catecholamine related neurotransmitters in brain membranes of Sprague- Dawley (SD) rats. Male SD rats were fed basic diets (control group) and experimental diets (EtOAc-25, EtOAc-50 and EtOAc-100) for 45 days. Cholesterol accumulations in mitocholndria and microsomes were significantly inhibited (11.8-12.1% and 9.6-13.0%, respectvely) in EtOAc-50 and EtOAc-100 groups. ACh levels and ChAT activities were significantly increased about 10% in membranes of EtOAc-100 group compared with control group. AChE activities were significantly increased about 8 -12% in membranes of EtOAc-50 and EtOAc-100 groups compared with control group. MAO-B activities were significantly inhibited about 10% in membrane of EtOAc-l00 group compared with control group. These results suggest that ethyl acetate fraction of pine needle may play an effective role in inhibiting cholesterol and improving a membrane fluidity, and learning and memory impairments. Moisturizing and Anti-oxidation Effect of Astragalus membranaceus Root Extract (황기추출물의 보습 및 항산화 효과)

  • We investigated the effect of moisturizing and anti-oxidation of Astragalus membranaceus (Astragali Radix) root extract with respective to growing districts and extract methods for the purpose of development of cosmetic ingredients. Astragalii Radix was collected in Jecheon, Jeongseon, Taebaek, and Yeongju in Korea and China as growing districts. Formononetin was determinated by HPLC method as one of the various active agents in Astragalus membranaceus root extract. The 75% ethanol extract demonstrated to be more effective than $H_2O$-extracted one for a scavenging activities to DPPH radicals and reactive oxygens. The 75% ethanol extract showed $IC_{50}$ (50% scavenging concentration) of 2.162 mg/mL and 2.981 mg/mL in case of free radical scavenging activity test and reactive oxygen scavenging activity test, respectively Especially, free radical scavenging activity of isoflavonoids isolated from ethylacetate fraction was similar to scavenging activity of genistein. Astragalus membranaceus root extract of Jecheon district by sonicating extraction method was more effective in skin hydration compared with others.

Biological activity of supercritical extraction residue 60% ethanolic extracts from Ulmus davidiana (느릅나무 초임계 추출박 60% 주정추출물의 생리활성)

  • Ulmus davidiana supercritical fluid residue EtOH extracts(USCFR) and ethyl acetate solvent fraction (USCFREA) of supercritical extraction foil were investigated in order to examine the recycling of supercritical extraction foil in the process of studying Ulmus davidiana branch supercritical extract. Experiments were performed for the determination of total phenol content. The $IC_{50}$ value(ppm) of DPPH radical scavenging activity and ABTS radical scavenging activity was $7.42{\pm}0.09$, $7.50{\pm}0.05$, $22.94{\pm}0.09$, $6.43{\pm}0.10$, and USCFREA, respectively, as compared with the positive control (vitamin C) with values $17.80{\pm}0.14$ and $5.34{\pm}0.06$, respectively. The antioxidative activities of USCFR and USCFREA were confirmed to be superior to the positive control group. In anti-allergic activity studies, both USCFR and USCFREA showed concentration-dependentanti-allergic activity, and USCFREA showed strong anti-allergic activity even at very low concentrations. Thetotal phenolic contents (ugEG, ugGA; ppm) of USCFR were $134.17{\pm}0.13$, $132.02{\pm}0.24$ and USCFREA were $154.77{\pm}1.05$ and $153.18{\pm}1.10$, respectively. Based on the above results and strong antioxidant activity, USCFR and USCFREA hold the potential to be considered as basic research materials for the development of therapeutic supplements based medicines or functional cosmetics related to chronic inflammatory skin immunity diseases.

Antioxidant Activity and Inhibitory Effect on Oxidative DNA Damage of Ethyl Acetate Fractions Extracted from Cone of Red Pine (Pinus densiflora) (솔방울 에틸아세테이트 분획물의 항산화 및 산화적 DNA 손상 억제 활성)

  • Antioxidant activity and inhibitory effect on oxidative DNA damage of ethyl acetate fractions extracted from Cone of Red Pine (Pinus densiflora) were investigated to find utilization of Cone, by-product of Red Pine, thrown out after berry shatter, as a new natural plant resource. Cone from P. densiflora was extracted with methanol (MeOH) and separated to petroleum ether, ethyl acetate and water fraction. Among them, ethyl acetate fraction was used. The antioxidant activity was conducted by the 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical, 2, 2'-Azino-bis (3-ethylbenzothiazoline-6 sulfonic acid) diammonium salt (ABTS) radical scavenging assay, Fe2+ chelating assay and reducing power assay. The inhibitory effect on oxidative DNA damage was determined by DNA cleavage assay using φX-174 RF I plasmid. The results of DPPH and ABTS radical scavenging activity at 200 ㎍/㎖ of extracts were 86.50% and 95.80% respectively, which were similar figures compared with L-ascorbic acid as control. Fe2+ chelating activity was 77.96% and reducing power was 0.77 at 200 ㎍/㎖. Total phenolic component was 27.29±0.3 ㎎/g and Vitamin C content was 1.84±0.1 ㎎/g. Also ethyl acetate fraction from Cone has inhibitory effect, using φX-174 RF I plasmid on DNA cleavage assay. In conclusion, Cone, by-product of P. densiflora, showed high antioxidant activity and inhibitory effect on oxidative DNA damage. Therefore this study suggests Cone, useless by-product, can be developed as a new natural plant resource with lots of utilization such as an effective antioxidant, natural medicine, food, cosmetics and so on.

Effects of Extracts from Sarcocarp, Peels, and Seeds of Avocado on Osteoblast Differentiation and Osteoclast Formation (아보카도 과육, 과피 및 씨 추출물이 조골세포 분화 및 파골세포 형성에 미치는 영향)

  • Avocado (Persea americana Mill., Family Lauraceae) is an important subtropical crop in the Americas where it has been cultivated for several thousand years. To investigate the bioactivities of avocado, which acts on bone formation, we prepared methanol extracts from the sarcocarp, peels, and seeds of avocado. The methanol extracts of peels and seeds showed higher bone-forming activity than avocado sarcocarp extracts accompanied by MC3T3-E1 osteoblast proliferation and alkaline phosphatase (ALP) activity. Additionally, the extracts of sarcocarp and peel from avocado also decreased tartrate-resistant acid phosphatase (TRAP) activity against differentiation of osteoclasts, derived from mouse bone marrow macrophages. The hexane fraction from avocado peels showed strong bone-forming activity accompanied by osteoblast proliferation and ALP activity (170.7${\pm}$8.4%), and the ethyl acetate fraction from avocado peel decreased TRAP activity (5.2${\pm}$0.3%) and differentiated osteoclasts at 50 ${\mu}g$/mL. Therefore, avocado is expected to be a natural source for developing medicinal agents to prevent bone-related diseases, such as osteoporosis, by increasing osteoblast differentiation and reducing osteoclast activity.
