Antimicrobial Activity of Grape Seed Ethanol Extract (포도종실 에탄올 추출물의 항균 활성)

  • Antimicrobial activities of grape seed ethanol extract and its serial solvent fractions were investigated against various food poisoning microorganisms. The grape seed ethanol extract showed dose dependant antimicrobial activity against Bacillus subtilis ATCC 9372 or Staphylococcus aureus ATCC 6538, whereas had limited effect on Pseudomonas aeruginosa IFO 3080, Salmonella enteritidis IFO 3313 and Escherichia coli ATCC 25922. Ethylacetate and butanol fractions ameng the serial solvent fractions of grape seed ethanol extract contained the catechin at the levels of 35.7 mg/g and 20.2 mg/g, respectively. Nevertheless, the butanol fraction of grape seed ethanol extract showed intense antimicrobial activity compared with the ethylacetate fraction on all microorganisms tested. It was found that the butanol fraction was mainly composed of oligomeric or polymeric polyphenols such as condensed tannins by the fractionation on C l8 cartridge according to the difference in the degree of polymerization. Therefore, it seems that the antimicrobial activity of grape seed ethanol extract is related to the degree of polymerization of proanthocyanidin as well as the total content of flavan-3-ol composing the proanthocyanidin.

Antioxidant Activity of Korea Traditional Fermented Sauces Made with Asparagus (아스파라거스를 이용한 전통장류의 항산화 효과)

  • Asparagus have anti-cancer, antioxidant, and anti-inflammatory properties. Since the high functionality of Korean traditional sauces was discovered, various studies have been conducted using asparagus. In this study, the antioxidant activity of traditional sauces containing asparagus was confirmed. The antioxidant capacity of asparagus was independent of the concentration and showed the highest activity at 70 mg/ml. For instance, concerning Meju containing asparagus, high antioxidant activity was observed in water and ethyl acetate extracts; the total polyphenol content was equivalent to 1.4 mg/g and 0.7 mg/g of tannic acid in the water and ethyl acetate fractions respectively. The DPPH and ABTS radical scavenging activities were 27.6% and 9.04%, respectively, in the water layer, and 10.7% and 52.4%, respectively, in the ethyl acetate layer. Additionally, the antioxidant activity of asparagus-containing traditional sauces was measured using 95% ethanol extracts; soy sauce showed the highest activity, retained in a wide range of concentrations.

Extractives from the needles of Juniperus rigida Siebold et Zucearin and Antioxidant activity (노간주나무(Juniperus rigida Siebold et Zucearini) 잎의 추출성분 및 항산화 활성)

  • The dried neeldes(1.5 kg) of Juniperus rigida were ground, extracted with acetone-H2O(7:3, v/v), concentrated, and fractionated with a series of hexane, CH2Cl2, EtOAc and water on a separatory funnel. Each fraction was freeze dried to give dark-brown powder and EtOAc and water soluble fractions were chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluent. Spectrometric analysis such as NMR and FAB or EI-MS including TLC were performed to characterize the structures of the isolated compounds. The needles of Juniperus rigida contained a large amount of (+)-catechin, quercetin-3-O-α-L-rhamnopyranoside and isoconiferin, in addition to a small amount of umbelliferone and quercetin-3-O-β-D-rutinoside. The antioxidative activities of each fraction and isolated compounds were tested by DPPH radical scavenging method, and EtOAc soluble fraction, (+)-catechin and quercetin-3-O-α-L-rhamnopyranoside were effective.

Antioxidant activities of Citrus junos seed shell extract and fractions cultivated in Korea (국내산 유자씨박 (Citrus junos seed shell) 추출물 및 분획물의 항산화 활성 평가)

  • In the present study, 70% ethanol extract, the ethyl acetate fraction were prepared from citron (Citrus junos)seed and their antioxidative ability was evaluated. The yields of extract and fractions were 5.1 and 0.9% per dried powder, respectively. In the 1,1-Phenyl-2-picrylhydrazyl (DPPH) radical test, free radical scavenging activities ($FSC_{50}$) of 70 % ethanol extract, ethyl acetate fraction were 512.1 and $514.0{\mu}g/mL$, respectively. Evaluation of total antioxidant capacities ($OSC_{50}$) using $Fe^{3+}$-EDTA system. Their $OSC_{50}$ of ethyl acetate fraction were $86.5{\mu}g/mL$. this antioxidant capacities higher than that of 70% ethanol extract. but lower than that of L-ascorbic acid ($1.72{\mu}g/mL$), known as a prominent water soluble antioxidant. The cellular protective effects on the $^1O_2$-induced cellular damage of rabbit erythrocytes were evaluated and the results showed that the extract was lower than (+)-${\alpha}$-tocopherol and low concentration of ethyl acetate fraction was similar to (+)-${\alpha}$-tocopherol. but not at high concentrations of ethtyl acetate fraction. it was able to induce cellular damage at high concentration.

Antioxidative Effects and Component Analysis of Graviola (Annona muricata) Leaf Extract/Fractions (그라비올라(Annona muricata) 잎 추출물 및 분획물의 항산화 활성과 성분 분석)

  • In this study, the antioxidative effects and component analysis of 50% ethanol extract, ethyl acetate fraction and aglycone fraction obtained from Annona muricata leaf were investigated. Free radical scavenging activities were performed by 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay, total antioxidant capacities were estimated using luminol-dependent chemiluminescence assay and $^1O_2$ quenching effects were estimated. Free radical scavenging activities ($FSC_{50}$) of 50% ethanol extract, ethyl acetate fraction and aglycone fraction were 45.6, 29.8 and $18.0{\mu}g/mL$, and total antioxidant capacities ($OSC_{50}$) were 4.4, 1.1 and $2.8{\mu}g/mL$, respectively. As a result of $^1O_2$ quenching experiment, ethyl acetate and aglycone fraction showed similar activities to L-ascorbic acid used as a comparative control. The cellular protective effects of 50% ethanol extract on the $^1O_2-induced$ cellular damage of human erythrocytes were exhibited at concentration-dependent ($5-50{\mu}g/mL$). TLC and HPLC were used to analyse components in the ethyl acetate fraction and aglycone fraction of Annona muricata leaf. In ethyl acetate fraction, rutin (quercetin-3-O-rutinoside), kaempferol-3-O-neohesperidoside, nicotiflorin (kaempferol-3-O-rutinoside), p-coumaric acid were identified. In aglycone fraction, kaempferol was identified. These results suggest that the extracts of Annona muricata leaf have the applicability as antioxidant cosmeceutical ingredients.

Antioxidative Activities and Whitening Effects of Solvent Fraction from Prunus davidiana (Carriere) Franch. Fruit (산복사나무(Prunus davidiana (Carriere) Franch.) 열매 분획 추출물의 항산화 및 미백 효능)

  • In this study, methanol extract and its organic solvent fractions were prepared from Prunus davidiana (Carriere) Franch. fruit. Antioxidative activities, polyphenolic and flavonoid contents, and tyrosinase inhibitory activities were evaluated. Among the fractions, ethyl acetate fraction showed the highest antioxidative activity with $IC_{50}$ values of 5.8 ${\mu}g/mL$ and 88.1 ${\mu}g/mL$ for DPPH radical and nitrite scavenging, respectively, which were comparable to those of ascorbic acid. Further, total flavonoid and polyphenolic contents were highest in the ethyl acetate fraction with $IC_{50}$ values of 244.5 mg/g and 210.2 mg/g, respectively. $IC_{50}$ values for tyrosinase inhibitory activity were 0.310 mg/mL and 0.329 mg/mL for the ethyl acetate and n-butanol fractions, respectively. Based on these results, it is suggested that the ethyl acetate fraction of Prunus davidiana (Carriere) Franch. fruit could be used as a functional material in the food and pharmaceutical industries.

Antioxidative and Antibacterial Activities of Artemisia princeps Pampanini Extracts (사자발쑥 추출물의 항산화 및 항균 활성)

  • In the present study, the antioxidative and antibacterial activities of Artemisia princeps Pampanini (A. princeps Pamp.) extract were investigated. The ethyl acetate fraction of A. princeps Pamp. showed the most prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}=12.27{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of A. princeps Pamp. extract on $Fe^{3+}-EDTA/H_2O_2$ systems were investigated using a luminol-dependent chemiluminescence assay. The ethyl acetate fraction of the extract ($OSC_{50}=0.33{\mu}g/mL$) had a 5 times greater ROS scavenging activity than L-ascorbic acid ($1.50{\mu}g/mL$), known as a water soluble antioxidant. The cellular protective effects of fractions of A. princeps Pamp. on the rose-bengal sensitized photohemolysis of human erythrocytes were examined. The aglycone fraction of extracts suppressed photohemolysis in a concentration dependent manner. The inhibitory effects of A. princeps Pamp. extract on tyrosinase were investigated to assess their whitening efficiency. The ethyl acetate fraction demonstrated a 7 times higher tyrosinase inhibitory effect ($IC_{50}=29.20{\mu}g/mL$) than albutin, known as a whitening agent. The antibacterial activity of ethyl acetate fractions against various normal skin flora were measured. The results showed that the antibacterial activity of the fraction was the highest on Staphylococcus aureus, Bacillus subtilis, and Propionibacterium acnes. Antioxidant substances were isolated and purified from the ethyl acetate fractions. Eupatilin and jaceosidin were identified. These results indicate that the extract/fractions of A. princeps Pamp. can function as antioxidant and/or antibacterial agents for the skin.

A Determination of the Moisturizing Effect and Stability of a Cream Containing Lespedeza cuneata G. Don Extracts (비수리 추출물 함유 크림의 보습 효능 및 안정성 평가)

  • In this study, the skin moisturing effect and stability of cream containing L. cuneata G. Don extract (ethyl acetate fraction) were evaluated. The skin hydrating effect of the cream containing extract was 1020% higher than the placebo cream, and the TWEL of the cream containing extracts was decreased to $7.7g/m^2h$ compared to the control ($10.2g/m^2$) and placebo cream ($8.9g/m^2$). The pH, viscosity, and absorbance were measured under the 4, 25, 37, $45^{\circ}C$ and the sun light during the 12 weeks. The pH change between cream containing extract and placebo cream did not show the significant difference under the 4, 25, 37, $45^{\circ}C$ except for the sun light. Both creams showed high decrease (about 59%) of viscosity at $45^{\circ}C$. However, there was no significant change under other conditions. The absorbance of the cream containing the extract and the placebo cream was decreased similarly at all conditions. This decrease in absorbance was relatively small compared to the decrease of absorbance of the extract in ethanol solution under the sun light (Fig. 7). In addition, any change in color or smell of the cream was not observed during the 12 weeks. Also physical changes as creaming and cohesion were not shown. These results indicate that the cream containing L. cuneata extract has the skin moisturizing effect and is relatively stable. Therefore, it is suggested that the ethyl acetate fraction of L. cuneata extract could be applicable to cosmetics as a new cosmetic material with its antioxidative and antibacterial activities reported previously.

Antimicrobial and Antioxidant Activities of Perilla frutescens var. acuta Extract and Its Fraction and Their Component Analyses (자소엽 추출물의 항균 및 항산화 효과와 성분분석)

  • In this study, antimicrobial and antioxidative activities of Perilla frutescens var. acuta were investigated with 50% ethanol and the ethyl acetate fraction and also the components were analyzed. The minimum inhibitory concentration (MIC) of the ethyl acetate fraction for both Staphylococcus aureus and Pseudomonas aeruginosa were $78{\mu}g/mL$, indicating high antimicrobial effects. The free radical scavenging activity ($FSC_{50}$) and the reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) in $Fe^{3+}-EDTA/H_2O_2$ system values of the ethyl acetate fraction were $25.90{\mu}g/mL$ and $1.40{\mu}g/mL$, respectively. After the cell damage induced by $400mJ/cm^2$ UVB irradiation, the cytoprotective effect of the ethyl acetate fraction of P. frutescens var. acuta showed the concentration dependent manner ranging from 2.0 to $16.0{\mu}g/mL$. The intracellular ROS inhibitory activity in HaCaT cells decreased to 28.6% and 40.7% for the 50% ethanol extract and ethyl acetate fraction, respectively at the concentration of $32{\mu}g/mL$. Components of rosmarinic acid, luteolin, apigenin, caffeic acid and ethyl caffeate were identified in the ethyl acetate fraction. These results suggest that the extract and fraction of P. frutescens var. acuta may be applied to the field of cosmetics as a natural material that protects the skin from an external environment by having antimicrobial and antioxidative activities.

Antioxidant and Anti-inflammatory Effects of Solvent Fractions from the Peel of the Native Jeju Citrus ‘Hongkyool’ and ‘Pyunkyool’ (제주 재래종 감귤 ‘홍귤’, ‘편귤’ 과피 분획물의 항산화, 항염증 효과)

  • This study investigated the anti-oxidative and anti-inflammatory activity of unripe fruit peel solvent fractions of the native jeju citrus ‘Hongkyool’ (Citrus tachibana Tanaka) and ‘Pyunkyool’ (C. tangerina Hort. ex Tanaka). The total polyphenol content and total flavonoid content were highest in the butanol fraction of both ‘Hongkyool’ (534.4 mg/g, 431.8 mg/g) and ‘Pyunkyool’ (342.9 mg/g, 415.7 mg/g). In both cultivars, the butanol fraction showed the strong antioxidant activity by DPPH radical scavenging and ABTS radical scavenging. The DPPH radical scavenging of the butanol fraction from ‘Hongkyool’ and ‘Pyunkyool’ was 89% and 64% at a concentration of 1 mg/ml, respectively. The ABTS radical scavenging of the butanol fraction from ‘Hongkyool’ and ‘Pyunkyool‘ was 94% and 85% at a concentration of 1 mg/ml, respectively. We investigated the effect of the anti-inflammatory activity of the ethyl acetate fraction from ‘Hongkyool’ and ‘Pyunkyool’ on LPS-induced NO production, IL-6, iNOS, and COX-2 protein expression in Raw 264.7 cells. At concentrations of 50 and 100 μg/ml of the ‘Hongkyool’ ethyl acetate fraction, the anti-inflammatory effect was excellent. These results suggest that ethyl acetate and butanol fractions from ‘Hongkyool’ and ‘Pyunkyool’ could be useful functional materials, with anti-oxidative and anti-inflammatory properties.
