Analysis of Pectolinarin Content and Antioxidant activity in Cirsium setidens Nakai by Cultivars (산지별 고려엉겅퀴의 Pectolinarin 함량 및 항산화 활성)

  • This study was performed to provide basic data of Cirsium setidens Nakai by cultivars that will be applied for development of functional foods and ingredients. We assessed pectiolinarin content, total flavonoids content and antioxidant effects (DPPH radical scavenging activity and ORAC assay) of C. setidens Nakai. Our results showed that the pectolinarin and total flavonoids contents of C. setidens Nakai by cultivars ranged from $3.95{\pm}0.05$ to $7.29{\pm}0.07mg/g$ and from $40.42{\pm}0.91$ to $76.70{\pm}2.24mg$ pectolinarin equivalent (PNE)/g, respectively. Among C. setidens Nakai by cultivars, the pectolinarin content was highest in GW-D extract. Futhermore, the DPPH radical scavenging activity of C. setidens Nakai ranged from 31.25 to 81.93%, respectively. The oxygen radical absorbance capacity (ORAC) value was highest in GW-D and GW-E extracts (514.49 and $501.73{\mu}M\;TE/g$, respectively). These results suggest that C. setidens Nakai extract could be considered as a good sources of natural antioxidants and functional food ingredients.

Antioxidative Effects of Silymarin and Silybin Purified from Silybum marianum on Lipid Peroxidation (엉겅퀴로부터 분리 정제한 Silymarin 및 Silybin의 지질 과산화에 대한 항산화 효과)

  • This study was undertaken to evaluate as antioxidant activity against lipid peroxidation. Silymarin and silybin extracted from Silybum marianum were successively purified wit solvent fractionation by silica gel column chromatography. These isoflavonoid inhibited superoxide anion production in the xanthine oxidase system. In the rat liver microsomes, silymarin or silybin rapidly inhibited lipid peroxidation which was initiated enzymatically by reduced nicotinamide adenine dinucleotide phosphate(NADPH) or non-enzymatically by ascorbic acid or Fenton's reagent (H2O2+Fe2+). Mitochondrial lipid peroxidation was also inhibited by silymarin and silybin. silymarin and silybin inhibited on terminating radical chain reaction during lipid peroxidation in the enzymatic system of microsomes or in the linoleic acid hydroperoxide induced peroxidation system.

Effect of Fermented Cirsium japonicum Extract on Testosterone Deficiency Syndrome (엉겅퀴 발효 추출물을 통한 남성 갱년기 증상 개선 효과)

  • As men get older, total testosterone levels decline gradually, and concentrations of free and bioavailable testosterone decline sharply with each decade beyond their 30s. Andropause or testosterone deficiency syndrome (TDS) is defined as a decrease in sexual satisfaction or decline in general well-being accompanied by low levels of testosterone in older men. This male climacteric is characterized by nervousness, reduced potency, decreased libido, irritability, fatigue, depression, memory problems, sleep disturbances, and hot flushes. Cirsium japonicum (CJ) is used as a traditional medicine for hemorrhage, blood congestion, and inflammation in Korea. However, there is no report on the efficacy of CJ treatment for TDS. In this study, we observed the mitigating effect of CJ extract (CE) and fermented CJ extract (FCE) on symptoms of TDS. In elderly male rats, total and testosterone levels, hind limbs muscles, forced swimming time, and total and motile sperm counts significantly increased after daily intake of CE and FCE for 6 weeks. In contrast, sex hormone binding globulin, retroperitoneal fat, total serum cholesterol, and triglyceride levels were significantly reduced in CE and FCE groups. However, there was no difference in prostate specific antigen, aspartate aminotransferase, and alanine aminotransferase levels among all groups, which means CE and FCE did not have putative adverse effects. In a cell experiment, we also observed that CE and FCE enhanced expression of genes related to testosterone biosynthesis but reduced genes involved in testosterone conversion. On the whole, these positive effects on TDS were greater in FCE compared to CE. Thus, these results suggest the potential of FCE as a promising natural product for recovering testosterone levels and alleviating TDS.

Optimization of Extraction Process for Antioxidant from Persimmon Leaf and Thistle Using Response Surface Methodology (반응표면분석법을 이용한 감잎과 엉겅퀴로부터 항산화성분의 추출공정 최적화)

  • In this study, we extracted an antioxidant from natural products which are known to have a high antioxidant content and also optimized the extraction process by applying a response surface methodology (RSM). In addition, by measuring the total flavonoids and total polyphenols of the active ingredient extracted persimmon leaf and thistle, the functionality of the active ingredient was evaluated. Both ultrapure water and alcohol were used as extraction solvents and the ratio of ultrapure and alcohol, amount of sample, extraction time, extraction temperature were set as independence variables. Also, the yield, total polyphenols, and flavonoids was set as the response. Optimal extraction conditions were as follows; for persimmon leaf, the extraction time = 3.1 h, ratio of alcohol/ultrapure = 63.4 vol%, and temperature = $54.6^{\circ}C$ while for thistle the extraction time = 2.9 h, ratio of alcohol/ultrapure = 40.7 vol%, and temperature = $68.4^{\circ}C$. Also, the response were as follows; for persimmon leaf, the yield = 27.7%, total polyphenols = 33.2 mg GAE/g, and total flavonoids = 47.8 mg QE/mg dw, whereas for thistle the yield = 27.0%, total polyphenols = 17.9 mg GAE/g, and total flavonoids = 28.8 mg QE/mg dw at the optimal conditions. The overall satisfaction level was 71.7%.

Development of Beverages Drinks Using Mountain Edible Herbs (산채류를 이용한 음료 개발에 관한 연구)

  • Beverages using mountain edible herbs(MEH) were formulated by determining optimum ratio of juices of MEH to several other ingredients. Fermented beverages were made by mixing juices(1 volume) fermented with Lactobacillus helverticus with syrup(3 volumes), followed by homogenization and flavoring . The general analysis and quality change of the beverages during storage were Peformed. The analysis of the beverages showed that the pH of Synurus deltoides and Cirsium sctidens were 3.8 and 3.7, titratable acidities were 0.50 and 0.49, optical densities were 1.201 and 1.119, respectively, The pH and color were not significantly changed when Synurus deltoides and Cirsium sctidens were stored at room temperature and 37$^{\circ}C$ for 6 months. The pH of Synurs deltoides beverage ranged 3.95~3.96 and pH of Cirsium sctidens was 3.83~3.95 at room temperature and 3.87~3.98 at 37$^{\circ}C$, respectively. The analysis of fermented beverages showed that pHs of Synurus deltoides and Cirsium sctidens were 3.65 and 3.70, titratable acidities were 0.57 and 0.60. Solids-non-fat were 3.2 and 3.1, and total counts of lactic acid bacteria were 2.5$\times$10$^{8}$ and 4.0$\times$10$^{8}$ , respectively. The changes of pH and titratable acidities stored at 4$^{\circ}C$ for 15 days were 3.39~3.56 and 0.61~0.81 for Synurus deltoides and 3.48~3.67 and 0.60~0.78 for Cirsium sctidens, respectively.

Anti-obesity Activities of Cirsium setidens Nakai Ethanolic Extract (고려엉겅퀴 주정 추출물을 함유하는 임상시험제품의 항비만 활성 평가)

  • The purpose of this study was to evaluate anti-obesity activity of Cirsium setidens Nakai test material (CNTM) in 3T3-L1 adipocytes and obese C57BL/6J mice fed with a high-fat diet using various obesity-related in vitro experiments. During adipocyte differentiation, CNTM significantly inhibited lipid accumulation and ROS production compared to controls. To evaluate whether CNTM could exert glycerol release effects on mature 3T3-L1 adipocytes, we treated cells with various concentrations of CNTM for 1 h. Treatment of mature adipocytes with $160-320{\mu}g/mL$ of CNTM increased the release of glycerol, but not in a significant dose-dependent manner. Anti-adipogenic and anti-lipogenic effects of CNTM seemed to be mediated by the inhibition of $PPAR{\gamma}$ and $C/EBP{\alpha}$. Moreover, CNTM stimulated fatty acid oxidation in an AMPK-dependent manner. CNTM-treated groups of C57BL/6J mice showed reduced body weights and adipose tissue weight with improving serum lipid profiles and adiponectin protein expression in obese C57BL/6J mice fed with a high-fat diet. These results suggest that CNTM might have anti-obesity effect on adipogenesis and lipid metabolism in vitro and in vivo. This presents the possibility of developing a treatment for obesity using nontoxic natural resources.

From the roots of this plant which were used for arthritis, neuralgia and carbuncular abscess as folkloric remedies, linarin, mp $255{\sim}257^{\circ}$, was isolated and identified by chemical reactions and spectral data with authentic samples.
