Phylogenetic and Morphological Comparison between Thamnaconus septentrionalis and T. modestus Collected in Southwest Seashore (서남해에서 채집된 말쥐치 (Thamnaconus modestus)와 유사종 (T. septentrionalis)의 형태 및 계통유전학적 비교)

  • Thamnaconus modestus, distributed in the Northwest Pacific, has high economic value and is used in various seafood. In this study, the morphological and genetic characteristics of T. modestus and T. septentrionalis were compared and analyzed. We observed the external and internal morphology of T. modestus, sketched skeletal elements, and analyzed phylogenetic evolutionary relationships using the cytochrome c oxidase subunit I (COI) gene on mitochondrial DNA compared to T. septentrionalis. The T. modestus observed in this study had blackish-brown patterns irregularly scattered on the gray-brown body, and the fins were blue-green. Genetic analysis results based on the COI sequences of T. modestus showed seven types of base sequence variation; however, the homology was more than 98.8%. In addition, as a result of comparison of the COI nucleotide sequences and phylogenetic analysis in Tetraodontiformes, two T. septentrionalis sequences (JN813099, MW485059) were similar to T. modestus with 99% homology, and the other two T. septentrionalis sequences (EF607583, KP267619) were similar to those of species belonging to another genus Thamnaconus with 95% homology with T. modestus. It was not easy to classify the species based on morphological characteristics, and phylogenetic analysis between T. modestus and T. septentrionalis confirmed the difference in classification. These results provide the external and internal morphology of T. modestus and will be used as important information for the taxonomic study of T. modestus and T. septentrionalis.

A Study on the Induction of Infertility of Largemouth Bass (Micropterus salmoides) by CRISPR/Cas9 System (CRISPR/Cas9 System을 활용한 배스의 불임 유도에 대한 연구)

  • A largemouth bass (Micropterus salmoides) is an ecosystem disturbance fish species at the highest rank in the aquatic ecosystem, causing a serious imbalance in freshwater ecosystems. Although various attempts have been made to eradicate and control largemouth bass, no effective measures were found. Therefore, it is necessary to find an approach to maximize the effective population reduction based on the unique characteristics of largemouth bass. This study used the transcriptome analysis to derive 182,887 unigene contigs and select 12 types of final target sequences for applying the CRISPR/Cas9 system in the genes of IZUMO1 and Zona pellucida sperm-binding protein, which are proteins involved in sperm-egg recognition. After synthesizing 12 types of sgRNA capable of recognizing each target sequence, 12 types of Cas9-sgRNA ribonucleoprotein (RNP) complexes to be used in subsequent studies were prepared. This study searched the protein-coding gene of sperm-egg through the Next Generation Sequencing (NGS) and edited genes through the CRISPR/Cas9 system to induce infertile individuals that produced reproductive cells but could not form fertilized eggs. Through such a series of processes, it successfully established a composition development process for largemouth bass. It is judged that this study contributed to securing the valuable basic data for follow-up studies to verify its effect for the management of ecological disturbances without affecting the habitat of other endemic species in the same water system with the largemouth bass.

Changes in hematological parameters and plasma components of olive flounder, Paralichthys olivaceus exposed to acute microplastics (넙치, Paralichthys olivaceus의 미세플라스틱 급성 노출에 따른 혈액성상 및 혈장성분의 변화)

  • Juvenile olive flounder, Paralichthys olivaceus (mean weight 66.7±7.1 g; mean length 19.2±0.9 cm) in a bio-floc environment were exposed to microplastic (PE: polyethylene, size 40-48 ㎛) at 0, 4, 20, 100, 500 and 2,500 mg L-1 for 96 hours. No P. olivaceus deaths were observed following microplastic exposure. In the plasma components, calcium was significantly decreased whereas there was no significant change with magnesium following microplastic exposure. Glucose was significantly decreased with over 100mg L-1 at 48 hours and 20mg L-1 at 96 hours. Cholesterol was significantly decreased with over 20mg L-1 after 48 hours, whereas there was no significant change in the total protein content. In enzymatic plasma components, the AST(Aspartate aminotransferase) was significant decreased by microplastic exposure. The results of this study indicate that acute exposure to microplastic induces blood physiological changes in P. olivaceus.

Ichthyofauna and Community Structure of Fish in Chuncheon and Andong Reservoirs (춘천호와 안동호의 어류상과 군집 특성)

  • Ichthyofauna and community structure of fish were studied for two fisheries resource reserves, Chuncheon reservoir and Andong reservoir from 2017 to 2018. Fish were sampled 8 times at each reservoir. In Chuncheon reservoir, total number of fish sampled was 1,197 representing 34 species and 13 families. Dominant species and sub-dominant species were Lepomis macrochirus (23.2%) and Zacco platypus (17.4%), respectively. In Andong reservoir, total number of fish sampled was 1,393 representing 24 species and 9 families. Dominant species and sub-dominant species were L. macrochirus (15.7%) and Carassius carassius (15.2%), respectively. The diversity index of two sampling site increased compared to previous investigation. The evenness index was not changed much in Andong reservoir but increased in Chuncheon reservoir. Ichthyofauna and community structure of fish at sampling sites in both reservoirs maintained stable except high proportion of exotics species with needed to be reduced by consistent effort. For better fish population in these communities, however, various efforts such as seed stocking and installment of artificial spawning ground should be provided to these reservoirs.

Optimal Management of Mackerel in Korea: A Maximum Entropy Approach (최대 엔트로피 기법을 이용한 한국 연근해 고등어 최적 관리에 관한 연구)

  • Mackerel is one of the most widely consumed aquatic products in Korea. Concerns about the depletion of stocks have also arisen as the catch has decreased. The primary purpose of this study is to estimate the mackerel stock and derive the optimal level of catch in Korea. We apply a generalized maximum entropy econometric method to estimate the mackerel growth function, which does not require the steady state assumption. We incorporate a bootstrapping approach to derive the significance levels of parameter estimates. We found that the average ratio of catch to the estimated total stock was less than 30% before the 1990s but exceeded 40% in the 1990s. After 2000, it dropped back to about 36%. This finding indicates that mackerel may have been over-fished in the 1990s, but the government regulations introduced in the 2000s alleviated over-fishing problems. Nevertheless, our dynamic optimization analysis suggests that the total allowable catch may need to be carefully controlled to achieve socially optimal management of resources.

Analysis of the Antioxidant Properties of 2,2-diphenyl-1 Picrylhydroazyl, Hydroxyl Radicals, and Nitric Oxide in Alaska Pollock Roe, with or without Natural Fermented Seasoned (알래스카 명란의 DPPH, OH, NO의 항산화 특성 분석)

  • Alaska pollock (Gadus chalcogrammus) is a marine fish species belonging to the family Gadidae. In this study, whether the Alaska Polloack Roe have antioxidant properties, 2,2-diphenyl-1 picrylhydroazyl (DPPH), hydroxyl radical (OH) reducing activity, and nitric oxide radical (NO) scavenging activity were evaluated in distilled water extract (DWE) and ethanol extract (ETE) of raw Alaska pollock roe, Gochujang Pollock roe, and fermented seasoned Pollock roe. The DPPH scavenging activity of the DWE with Gochujang Pollock roe was 71.9% at a concentration of 1.0 mg/ml and that of the ETE was 73.7% at the same concentration. The DPPH scavenging activity of the DWE with fermented seasoned Pollock roe was 78.0% at 1.0 mg/ml, whereas that of ETE was 78.4% at the same concentration. The $IC_{50}$ values of the DWE and ETE of raw Pollock roe for DPPH were $11.65{\mu}g/ml$ and $11.47{\mu}g/ml$, respectively. The OH scavenging activities of raw Pollock roe, Gochujang Pollock roe, and fermented seasoned Pollock roe ethanolic extracts at a concentration of 1.0 mg/ml were 70.9%, 79.0, and 80.6%, respectively. The $IC_{50}$ values of the DWE and EWE of raw Pollock roe for NO were $11.45{\mu}g/ml$ and $11.41{\mu}g/ml$, respectively. The DPPH, OH, and NO scavenging abilities in DWEs and ETEs of Gochujang and fermented seasoned Pollock roe were higher than those of instant (no Gochujang or season treatment) treatment Pollock roe. Both the Gochujang and fermented seasoned Pollock roes have natural radical scavenging ability and may be useful potential antioxidant food supplements.

Antioxidant Properties (ABTS, FRAP, Total Phenolic Content) of Alaska and Gochujang Pollock Roes and Fermented Pollock Roe Seasoning (고추장과 발효액이 첨가된 알래스카 산 프리미엄 명란의 ABTS, FRAP, total phenolic acid의 항산화 특성 분석)

  • The Alaska Pollock (Gadus chalcogrammus) is distributed in an arc across the North Pacific Ocean. Distilled water extracts (DWE) and ethanol extracts (ETE) of 1.0 mg/ml concentrations of raw Alaska Pollock roe, premium Gochujang Pollock roe, and premium fermented Pollock roe seasoning were evaluated for estimated 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS), ferric reducing antioxidant potential (FRAP), and total phenolic content. The ABTS scavenging activity of the raw Alaska roe DWE and ETE were evaluated at 50.1% and 53.1%, respectively. The ABTS scavenging activity of the Gochujang roe DWE was 68.7% and of the ETE was 70.4%; for the fermented seasoning it was 71.3% and 71.6% for the DWE and ETE, respectively. The ABTS $EC_{50}$ values of the raw roe DWE and ETE were 12.49 ug/ml and 12.21 ug/ml, respectively. The FRAP $EC_{50}$ values of the Gochujang roe DWE and ETE were 10.67 ug/ml and 10.56 ug/ml, respectively, and the $EC_{50}$ values for total phenolic content for the fermented seasoning DWE and ETE were 10.45 ug/ml and 10.31 ug/ml, respectively. When Gallic was acid used as a control, the relative total phenolic content scavenging activity in each ETE was 52.0% (raw Alaska roe), 61.1% (Gochujang roe), and 63.6% (fermented seasoning). In the present study, higher ABTS, FRAP, and total phenolic content were observed in the Gochujang Pollock roe and fermented roe seasoning than in the Alaska Pollock roe.

Ecological Characteristics of Phoxinus phoxinus (Cyprinidae) at the Spring Water in Eocheon Stream, Korea (어천 용천수에 서식하는 연준모치 Phoxinus phoxinus (Cyprinidae)의 생태 특성)

  • We investigated the ecological characteristics of Phoxinus phoxinus in the spring water in Eocheon stream from January to December 2016. The water temperature was $9.7{\sim}14.3^{\circ}C$ and remained below $15^{\circ}C$ throughout the year. The fishes cohabiting with minnow were Rhynchocypris kumgangensis (70.14%), Cottus koreanus (13.63%), Rhynchocypris oxycephalus (3.16%), Oncorhynchus mykiss (0.97%), Zacco koreanus (0.49%), and Iksookimia koreensis (0.24%). The sex ratio of female to male was 1:0.86. The frequency analysis of total length indicated that the fishes with the total length of 30 - 50 mm were one year old, those of 50 - 65 mm were two years old, those of 65 - 75 mm were three years old, and those of 75 mm or more were four years old. The sexually mature fishes were 50 mm or longer for female and 60 mm or longer for male. The spawning season was from April to July, and the water temperature was $12.3-14.3^{\circ}C$ during the period. The prosperous spawning season was from June to July, and the water temperature was $13.8-4.3^{\circ}C$ during the period. The average number of eggs in the ovaries was 508 per matured female, and the matured eggs were yellowish and spherical with a mean diameter of $1.58{\pm}0.13mm$. The length-weight correlation of P. phoxinus was $BW=0.000007TL^{3.09}$ with the constant a as 0.000007 and the parameter b as 3.09. The condition factor (K) was 0.99 (0.76 - 1.32) in average.

Analysis of Fish Community according to Habitat in the Woraksan National Park, Korea (월악산국립공원의 서식지에 따른 어류군집 분석)

  • This study was conducted to analyze the current status of fish fauna and characteristics of the fish community according to the habitat of Woraksan National Park, Korea. The spatially balanced sampling selected 20 stations from major streams of Woraksan National Park, and three surveys were conducted in each season. The physical environments of the habitat were mostly mountain streams (Aa), with large stones and gravels scattered over the stream. The average altitude of the habitat was 304.4 m, and the average depth of water was 40.3 cm, being less than 1 m in most cases, and the river water level was distributed from 3rd to 5th streams. The principal component analysis of the physical environmental factors by habitat showed that the substrate properties differed according to the altitude. The survey identified a total of 2,183 individuals in 16 species belonging to 7 families. The dominant species was Zacco koreanus(86.2%), and the subdominant species was Rhynchocypris oxycephalus(3.8%). Pseudopungtungia tenuicorpa, classified as the endangered wildlife II, was the first endangered legally protected species found in this survey. Analysis of the rank abundance curve model in the fish community showed the Zipf model at 9 out of 20 points, the Lognormal model in 3 points, and the Preemption model in 4 points. The remaining 4 habitats showed only one species and were not analyzed. The canonical correspondence analysis of 20 stations and fish species was performed to understand the characteristics of the fish community according to environmental factors. The fish communities were divided according to differences in habitat environment by the altitude.

The effect of starvation on the characteristics of morphometric measurement and body-sectioned shape in Far Eastern catfish (Silurus asotus) (메기(Silurus asotus) 기아 시 외부형태 및 체 절단면 변화)

  • A 210-day experiment was conducted to examine the effects of starvation on survival, external morphometric traits, and sectioned morphometric traits in Far Eastern catfish(Silurus asotus). The survival rate decreased to 92.2±0.47% in the fed group and to 74.4±2.59% in the starved group. Eight dimensions of Ls, DALPO/Ls, DADOP/Ls, ED/Ls, CH/Ls, BDAA/Ls, BDMA/Ls, and BWAA/Ls showed significant differences (p<0.05), and 16 other dimensions of DALAD/Ls, DPDPL/Ls, HPLAA/Ls, HALAV/Ls, HALOP/Ls, DAUPO/Ls, DADAV/Ls, DADAA/Ls, Max BL/Ls, Man BL/Ls, LD/Ls, HWOP/Ls, DAUF/HL, DAUS/HL, IW/HL, and DAUE/HL were not significantly different among the initial, fed, and starved groups (p>0.05). The fed group showed higher total height (THX), wider width (WX), and larger area (AX) than those of the starved group in each body section. This research provides information on the morphometric traits and body sections of starved Far Eastern catfish. These results can be used as a basic guide to indicate nutritional status and assist in aquaculture methods for Far Eastern catfish. Moreover, the methods used in this study were considered helpful because the survey analysis method can be used to investigate information on the nutritional status of other fish species.
