Analysis of the behavior of microorganisms isolated from the medium during cultivation of Agaricus bisporus (button mushroom) (양송이 재배 중 배지에서 분리한 미생물의 상호작용 분석)

  • This experiment investigates the characteristics of microorganisms isolated from a medium during cultivation process and reveals the relationship between these microorganisms and the growth of Agaricus bisporus. The domestically grown strains of Agaricus bisporus displayed a higher inhibition growth rate against microorganisms isolated from straw, chicken manure, and medium than imported strains. As for inhibition of mycelial growth among mushroom cultivars of the microorganisms separated by each fermentation step from the mushroom medium, the domestic cultivar, 'Saedo,' grew more vigorously among other cultivars. As the fermentation progressed, it was confirmed that inhibitation of microorganisms against Agaricus bisporus was weakened. A total of 21 strains of microorganisms that promote mushroom growth were isolated in the 4th turning process, and the microorganisms isolated from the mushroom medium affect the growth and as yield of the mushroom through secretory substances.

Damages and Developmental Characteristics of Fungus Gnat, Lycoriella ingenua (Diptera: Sciaridae) in Button Mushroom Cultivation (양송이버섯 재배에서의 긴수염버섯파리(Lycoriella ingenua)의 발달과 피해)

  • Mushroom sciarid fly Lycoriella ingenua (Dufour), syn. L. mali (Fitch) is one of the most common fly pests affecting the mushroom cultivation in Korea. This study was carried out to investigate the development at different temperatures and damages of L. ingenua in A. bisporus mushroom cultivation. Rearing of mushroom flies were carried out on mycelial culture in Petri dishes. The development of L. ingenua from egg to adult at the temperature of 16, 20, 24, $28^{\circ}C$ were 35.2, 25.8, 23.5, and 22.2 days, respectively. Adult flies invade mushroom farms and oviposit in freshly spawned compost. Damages are mainly caused by the larvae of 1-7 mm, which feed on growing mycelium and also developing stipes and caps of mushrooms. Adult flies spoil the appearance of the fruiting bodies, spread various fungal and bacterial disease, and transmit mites. The damages by mushroom flies increased without pest control causing yield loss of up to 27%.

Characteristic evaluation of collected strains of Agaricus spp. based on ITS rDNA sequence (ITS 계통분석을 이용한 주름버섯류 수집균주의 특성평가)

  • Phylogenic relationships and morphological characteristics were classified and investigated among the 233 collected strains of Agaricus spp. The 38 strains were differently identified as different characteristic group using analysis of ITS regions in rDNA. A. bitorquis was showed close relationship in groupA whereas A. campestris was in groupC as different characteristic group among with A. bisporus. There was no phylogentic difference with strains collected by country and different pileus colored Agaricus bisporus. Also the strains were cultivated twice to investigate morphological characteristics of fruiting bodies. The characteristics and yield of collected strains were compared with molecular varieties and seasons by the cultivation. In this result, A. campestirs showed good yield and quality in terms of hardness off-white mushroom was more harder than other number of A. bisporus. Also earliness and color of pileus was influenced by external environment all conditions.

Evaluation of Traits of Button Mushroom Agaricus bisporus (양송이버섯 수집균주의 다양한 형질특성평가)

  • White button mushroom (Agaricus bisporus) is the most widely consumed mushroom in the world, and domestic consumption is increasing owing to expansion of the westernized diet in recent years. Before 2000, most Korean mushroom farmers cultivated foreign varieties, but recently, Korean breeders are developing hybrid strains using molecular breeding tools. To produce a better mushroom cultivar, we evaluated some traits of button mushroom Agaricus bisporus strains. Mycelial growth rate at different culture temperatures was investigated by 25>20>15>30>$10^{\circ}C$. Recently developed domestic varieties had superior mycelial growth rate compared to the traditionally grown species. In particular, the ASI1338 strain showed excellent mycelial growth rate at different temperatures. Further, we confirmed that ASI1007, ASI1085, ASI1310, ASI1339 were strongly resistant to bacterial brown blotch, whereas ASI1053, ASI1103, ASI1140, ASI1146, ASI1177, ASI1183, ASI1195, ASI1321, ASI1331, ASI1336 were relatively weakly resistant. The average number of days for 16.5, and that for harvest was 5.7; the average yield was 142.7 g/2-kg bag. Mycelial growth rates at 10, 15, 20, $25^{\circ}C$ were similar, but that at $30^{\circ}C$ was lower. The mushroom yield was highly correlated with the number of days for pinheading and harvest.

Composition of Organic Acid and Fatty Acid in Pleurotus ostreatus, Lentinus edodes and Agaricus bisporus (느타리, 표고와 양송이버섯의 유기산 및 지방산 조성)

  • Composition of organic acids and fatty acids in three edible mushrooms, Pleurotus ostreatus, Lentinus edodes and Agaricus bisporus were analyzed by gas-liquid chromatography and compositional differences of these components depending on species, sizes and portions (pileus and stipe) were investigated. Lactic, oxalic. fumaric. citric and pyroglutamic acid were identified in each of the three mushrooms and the major organic acids were malic, citric and pyroglutamic acid, respectively. The total organic acid contents were in the range of 2.4-4.0% in Pleurotus ostreatus, 1.7-3.6% in Lentinus edodes and 1.9-3.1% in Agaricus bisporus. Especially oxalic and pyroglutamic acid in Pleurotus ostreatus, malic and citric acid in Lentinus edodes and fumaric acid in Agaricus bisporus were higher in pileus than stipe. The total organic acid contents decreased as the size of pileus developed and especially the decrease of citric acid were marked. On the other hand, the major fatty acids in three mushrooms were linoleic, palmitic and oleic acid. But fatty arid compositions were not significantly different depending on species, sizes and portions.

Characteristics of Mummy Disease on Agaricus bisporus and A. blazei in Korea (양송이와 신령버섯 미이라병 (Mummy Disease))

  • Mummy disease has been observed for a long time in the button mushroom, Agaricus bisporus farms in Korea, and severe mummy disease occurred on “Shinryung” mushroom, A. blazei recently. Typical symptoms of mummy disease were observed on the mushroom-cultivation beds infected ; tilted caps of mushrooms, browning and lignified internal tissue of stipe, overdevelopment of mycelium around the base of the stipe, and mummified mushrooms. Electron micrographs prepared from internal tissue of stripe of the diseased mushrooms showed that many bacterial cells present inside hyphal cells of the diseased mushroom, which is one of the characteristics of mummy disease reported previously, Survey in Buyo, Chungnam showed that mummy disease occurred at 55% and 83% frequency on bottom mushroom (brown strain) and “Shinryung” mushroom cultivation during year 2000. It indicates that mummy disease is one of the major diseases for the mushrooms cultivation.

Contents of Free-Sugars & Free-Sugaralcohols in Pleurotus ostreatus, Lentinus edods & Agaricus bisporus (느타리버섯, 표고버섯 및 양송이의 유리당(遊離糖)과 당(糖)알코올 조성)

  • Compositions of free-sugars and sugar-alcohols in three cultivated mushrooms, Pleurotus ostreatus, Lentinus edodes and Agaricus bisporus were analyzed by gas-liquid chromatography and compositional differences of these components depending on species, sizes of fruiting body and portions(pileus and stipe) were investigated. Glucose, fructose, trehalose, glycerol, arabitol and mannitol were identified in each of the three mushrooms. The major components were mannitol and arabitol in Lentinus edodes, mannitol in Agaricus bisporus, trehalose and mannitol in Pleurotus ostreatus. The total free-sugars and sugar-alcohols content were in the range of 12.7-18.9% in Lentinus edodes, 9.2-16.1% in Agaricus bisporus and 8.2-10.6% in Pleurotus ostreatus. Total contents of these components in Pleurotus ostreatus and Agaricus bisporus were higher in pileus than those in stipe and it was decreased as pileus size development in all of three edible mushrooms.

Analysis of trends in brown button mushroom consumption for raising awareness (갈색양송이 인지도 제고를 위한 소비 성향 분석)

  • Cultivation of brown mushrooms, rather than that of white variants is preferred by Korean mushroom farmers, as the former are resistant to diseases. However, brown mushrooms were cultivated only in selective eco-friendly agricultural farms due to lack of consumer awareness. After providing information about brown mushrooms to respondents through a 1-minute video clip, a survey was conducted on social network service (SNS) to assess recognition and preference for brown mushrooms. A food evaluation was then conducted among 200 people randomly selected from the survey respondents. Most respondents (83%) had not encountered brown button mushrooms previously, and 98% of the respondents were willing to buy these mushrooms because they were "curious about its taste" (44%). In the food evaluation, 32% of the respondents found the brown button mushrooms to be delicious, 28% reported a good flavor, and 31% described a good texture. In addition, we confirmed that 95% of respondents were interested in purchasing brown mushrooms after sampling. Therefore, in the present study, we evaluated public perception, preference, and taste of brown button mushrooms, and confirmed that availability of information on nutrition and benefits s of mushroom consumption could induce consumers to buy brown button mushrooms.

Selection and appropriate culture conditions of antagonistic bacterium Bacillus altitudinis HC7 against button mushroom cobweb disease caused by Cladobotryum mycophilum (양송이버섯 솜털곰팡이병균(Cladobotryum mycophilum)에 대한 길항미생물 Bacillus altitudinis HC7의 선발 및 적정 배양조건)

  • This study was conducted to selection and investigate appropriate conditions for mass production of antagonistic microbes to control cobweb disease caused by Cladobotryum mycophilum. A grampositive bacterium was isolated from spent substrate of Agaricus bisporus and showed significant antagonistic activity against Cladobotryum mycophilum. The bacterium was identified as Bacillus altitudinis. based on the cultural, biochemical and physiological characteristics, and 16S rRNA sequence. The isolate is saprophytic, but not parasitic nor pathogenic to cultivated mushroom whereas it showed strong inhibitory effects against C. mycophilum cells in vitro. The control efficacy of B. altitudinis HC7 against cobweb disease of C. mycophilum was up to 78.2% on Agaricus bisporus. The suppressive bacterium may be useful for the development of biocontrol system. To define the appropriate conditions for the mass production of the Bacillus altitudinis HC7, we have investigated appropriate culture conditions and effects of various nutrient source on the bacterial growth. The appropriate initial pH and temperature were determined as pH 6.0 and 30℃, respectively. The appropriate concentration of medium elements for the growth of pathogen inhibitor bacterium(Bacillus altitudinis HC7) was determined as follows: 3.0% soluble startch, 10% soytone, 1.0% (NH4)2HPO4, 1.0 mmol KCl, and 0.5% L-asparagine.
