  • Experimental studies were conducted to investigate the effect of Sagan-Tang on analgesic, sedative, antipyretic, isolated ileum and blood vessel and so on. The results of this investigation were summarized as follows; Analgesic action by the acetic acid stimulating method in mice were recognized. Prolonging action against the hypnotic duration induced by thiopental-Na was noted in mice. Antipyretic effect in typhoid vaccine febrile rats was recognized. Spontaneous motility of the isolated ileum of mice was suppressed and contractions of the isolated ileum of mice and guinea-pig induced by accetylcholine chloride, barium chloride and histamine were remarkably inhibited. Vaso-diating and hypotensive actions were recognized in rabbits. GOT and GPT activities in the serum of rats damaged by $CCl_4$ and galactosamine were decreased remarkably.

  • Hyangsayangwee-Tang, a combined preparation of crude drugs, which has been used for stomach and intestinal disorder, was examined for anti-spasmodic, anti-ulcerative and anti-emetive effects. Spontaneous motility of isolated ileum was strongly suppressed and inhibitory effects against contraction of isolated ileum induced by acetylcholine and barium chloride were shown in mice. And, contraction of isolated guinea-pig ileum by histamine was inhibited. In rats fundus preparations, Hyangsayangwee-Tang elicited strong relaxation and had antagonist effects against the spasm induced by acetylcholine and barium chloride. A significant inhibitory effect on the intestinal propulsion of barium sulfate in mice was shown. In pylorus-ligated rats, Hyangsangwee-Tang inhibited gastric secretion and showed a strong anti-peptic activity. Protective effects against gastric ulceration induced by pyloric ligation, water-immersion stress, histamine and aspirin were significantly recognized in mice and rats. Hyangsayangwee-Tang decreased cupric sulfate-induced vomitting in frogs.

신원방우황청심원액의 뇌허혈 및 중추신경계에 대한 약효 (Pharmacological Actions of New Wonbang Woohwangchungsimwon Liquid on Cerebral Ischemia and Central Nervous System)

  • In order to investigate pharmacological properties of New Wonbang Woohwangchungsimwon Liquid(NSCL) and Wonbang Woohwangchungsimwon Liquid(SCL), the effects of NSCL and SCL on cerebral ischemia and central nervous system were compared. Cerebral ischemia insult was performed using unilateral carotid artery occlusion in mongolian gerbils. The histological observations showed a preventive effect of NSCL and SCL treatments with ischemia-induced brain damage. The ATP in brain tissue was decreased in vehicle-treated ischemic gerbils. This decrease was prevented by NSCL and SCL treatment. In contrast to what was seen with ATP, lipid peroxide were elevated in vehicle-treated ischemic gerbils. This elevation was inhibited by NSCL and SCL treatments. While NSCL and SCL had no effects on the hexobarbital-induced sleeping time, they showed sedative effect in rotarod and spontaneous activity test. NSCL and SCL prevented the seizures induced by electric shock and strychnine, but the effect of NSCL was less than that of SCL. Furthermore, NSCL and SCL showed anti-stress effect. Our findings suggest that the pharmacological profiles of NSCL on cerebral ischemia and central nervous system are similar to those of SCL.

신원방우황청심원액의 심혈관계에 관한 약효 (Pharmacological Actions of New Wonbang Woohwangchungsimwon Liquid on Cardiovascular System)

  • In order to investigate the pharmacological properties of New Wonbang Woohwangchungsimwon Liquid (NSCL), effects of Wonbang Woohwangchungsimwon Liquid (SCL) and NSCL were compared. In isolated rat aorta, NSCL and SCL showed the relaxation of blood vessels in maximum contractile response to phenylephrine (10$^{-6}$ M) regardless to intact endothelium or denuded rings of the rat aorta. Furthermore, the presences of the inhibitor of NO synthase and guanylate cyclase did not affect the relaxing effect of NSCL and SCL. NSCL and SCL inhibited the vascular contractions induced by acetylcholine, prostaglandin endoperoxide or peroxide in a dose-dependent manner. In conscious spontaneously hypertensive rats (SHRs), NSCL and SCL significantly decreased heart rate. NSCL and SCL, at high doses, had a negative inotropic effect that was a decrease of left ventricular developed pressure and (-dp/dt)/(+dp/dt) in the isolated perfused rat hearts, and also decreased the contractile force and heart rate in the isolated rat right atria. In excised guinea-pig papillary muscle, NSCL and SCL had no effects on parameters of action potential such as resting membrane potential, action potential amplitude, APD$_{90}$ and V$_{max}$ at low doses, whereas inhibited the cardiac contractility at high doses. These results suggested that NSCL and SCL have weak cardiovascular effects with relaxation of blood vessels and decrease of heart rate, and that this effect is no significant differences between cardiovascular effects of two preparations.s.

  • 민간에서 주로 사용되는 약용식물들 중 간독성의 발현 저해 및 간기능 보호 효과가 우수한 약물을 찾아 임상적인 약효를 밝혀보고자 하는 실험과정으로 탄소화합물의 불완전 연소 및 열분해에 의해 생성되는 간장해 물질인 benzo(a)pyrene으로 유도한 rats의 간독성 발현에 미치는 자근 수침액의 예방 및 치료 효과를 실험한 결과 자근 수침액의 투여는 B(a)P 투여로 현저하게 증가된 혈청 및 간장의 AST, ALT, LDH, ALP 활성을 유의성 있게 감소시켰고 B(a)P 투여로 증가한 혈청 total cholesterol 및 phospholipid 함량 역시 감소시킬 수 있었으나 그 효과는 현저하지 않았다. 즉, 자근 수침액의 투여는 B(a)P에 의한 간독성 발현을 유의성 있게 감소시켰으며 그 효과는 전처리가 후처리에 비해 우수하였다.

  • This study was performed in order to investigate the effect of renal denervation on diuretic action of UK 14, 304, $\alpha$$_2$-Adrenergic Agonist, administered into the vein and the carotid artery in dog. The diuretic action of UK 14, 304 administered into the vein or the carotid artery was reversed to the antidiuretic action by renal denervation, this time, the decrease of N $a^{+}$excretion amounts in urine ( $E_{Na}$ ) and the increase of N $a^{+}$ reabsorption rates in renal tubule ( $R_{Na}$ ) were exhibited. This results suggest that central diuretic action of UK 14, 304 is mediated by renal nerves and the antidiuretic action of UK 14, 304 in denervation kidney is caused by the increase of N $a^{+}$reabsorption rates ( $R_{Na}$ ) in renal tubules in dog.n dog.

비소세포폐암 환자에 있어서 Epidermal Growth Factor Receptor Tyrosine Kinase Inhibitors의 약효 및 rash 발생과 관련한 인자에 대한 연구 (Factors associated with effectiveness of and rash occurrence by Epidermal Growth Factor Receptor Tyrosine Kinase Inhibitors in patients with non-small cell lung cancer)

  • Purpose: Currently lung cancer ranks second in cancer for incidence rate and is a disease that ranks first for a death rate by cancerous growth because it is already advanced at the time of diagnosis. The purpose of this paper was to analyze the factors that affect the effectiveness of and rash occurrence by Epidermal Growth Factor Receptor Tyrosine Kinase Inhibitor (EGFR TKI) in patients with non-small cell lung cancer. Methods: A retrospective chart review of 100 patients, who took EGFR TKI (erlotinib, gefitinib) among patients who were diagnosed with non-small cell lung cancer in a Hospital in Korea between May 2005 and February 2008, was conducted. The drug effectiveness was evaluated by Response Evaluation Criteria In Solid Tumor. Results: EGFR mutation was the only factor associated with drug response (complete response and partial response). When stable disease was added to drug response as the evaluation parameter, ECOG and rash as well as EGFR mutation were found to be important factors. Survival, however, was not affected by EGFR mutation. The factors influenced on survival were older age (${\geq}65$), low ECOG ($1{\sim}2$), adenocarcinoma and rash. In the case of rash, group with EGFR mutation or low ECOG showed significantly higher chance of occurrence. There was no significant difference in rash occurrence between gefitinib and erlotinib groups. Conclusions: Based on the results, EGFR mutation positive and low ECOG ($1{\sim}2$) were significantly important factors for both effectiveness of EGFR TKI and rash occurrence. Also, rash itself was found to be an independently significant factor for the disease control and survival. Therefore, while administering EGFR TKI, patients who have the factors associated with rash occurrence should be closely monitored for effective and safe drug therapy.

단기 육묘재배 당귀근의 부위별 성분연구 (Study on the Constituents of Angelica gigas root cultivated by Short-time Growing seedling)

  • 참당귀(當歸) 추대억제(抽臺抑制) 재배법(栽培法)을 개발하기 위하여 종자를 조기에 파종, 육묘하여 본포에 이식한 후 당년가을에 수확한 뿌리의 부위별(部位別) 주성분(主成分)을 관행재배한 당귀와 비교하므로서 1년생 당귀의 추대억제효과와 약재로서의 가치를 평가하고저 하였다. 1. 관행(慣行) 재배(栽培)한 2년생 당귀의 뿌리내 decursin 과 decursionol angelate의 함량을 합한 평균치는 6.47% 임에 비하여, 단기육묘(短期育苗)하여 수확한 1년생 뿌리 내에서의 함량은 4.82%로 낮은 경향이었다. 2. 약효성분(藥效成分)의 부위별(部位別) 함량은 세근(지근)과 피충부가 높은 반면 중심주에서는 낮은 경향이었다. 3. Decursin및 decursinol angelate의 구성비(構成比)는 1년생에서는 10 : 7.5, 2년생에서는 10 : 6의 비율로 2년생에서 는 decursinol angelate 함량이 감소하는 경향이었다. 또한 2년생에서는 부위에 관계없이 두성분의 구성비가 60% 정도로 안정적인데 반해, 1년생에서는 세근과 지근에서 decursional angelate 구성비가 높은 경향이었다. 4. 조기(早期) 육묘재배(育苗栽培)한 1년생 당귀의 생약적(生藥的) 가치(價値)는 일정 면적(面積)에서의 수량(收量)이나 또는 단순한 성분함(成分含) 양면(量面)에서 무리가 없을 것으로 사료되지만, 주성분인 decursin의 구조이성체인 decursinol angelate와의 구성비(構成比)에 따른 효능상의 문제는 추후 검토가 요망된다.

  • 시호(柴胡)의 주(主) 약효성분(藥效成分)인 saikosaponin a 및 c의 함량(含量)에 미치는 영향(影響)을 조사(調査)하기 위하여 산지에서 채취(採取)한 1년근(年根) 중(中)의 saikosaponin a, c의 함량(含量)과 재배지(栽培地) 토양(土壤)의 이화학성(理化學性)과의 관계(關係)를 규명하였다. 시호근(柴胡根) 중(中)의 saikosaponin a 및 c의 함량(含量)은 각각(各各) 0.141~0.479%와 0.042~0.094%의 범위에 속(屬)하였다. Saikosaponin a의 함량(含量)은 표토(表土)의 전질소함량(全窒素含量)과 부(負)의 상관(相關)이 인정(認定)되었다. Saikosaponin a 및 c의 함량(含量)은 시호근(柴胡根) 중(中)의 Fe의 함량(含量)과 공(共)히 부(負)의 상관(相關)이 인정(認定)되었다.

수도 수묘대의 조류방제에 관한 연구 (Studies on Algae Control in Wetted Nursery of Rice)

  • 수도 수묘형 육묘에 있어서 종자침하에 의한 부패, 발아지연, 종자의 부유에 의한 종자가 한쪽으로 몰리는 등의 해를 야기시켜 발아율이나 성묘율율 저하시키는 조류나 괴불방제를 시도하여 수묘대에서 파종전후 5일에 Cuperic Sulfate, Lime Sulfur, Swep 80wp. Dithane A-40, Dichlone 50wp를 처리하여 다음과 같은 결과를 얻었다. 1. 조류 및 괴불방제의 효과가 뚜렷한 약제는 Dichlon 50.wp, Dithane A-40, Swep 80 wp 였으나 Dithane A-40, Swep 80wp는 약해등으로 성묘율이 낮었다. 2. Cuperic Sulfate는 조류 및 괴블에 대한 약제효과는 인정되었으나 약제처리후의 파종시 발아억제 및 종자침하현상이 심하였다. 3. 파종후 25일묘 조과결과 Dichlone 50wp를 제외한 기타 약제처리구는 Check구에 비하여 지하부의 근수, 지상부의 생체중 및 건물중비율이 낮았고 초장은 약제처리구가 전반적으로 낮은 편이었다. 4. Dichlone 50wp는 조류 및 괴블 방제효과 과 뚜렷하여 종자발아 및 성묘비율을 높일 수 있었으며 약효의 지속기간은 처리후 4일 정도임이 인정되었다.
