Effects of Elevated CO2 Concentration and Increased Temperature on the Growth of Gastrodia elata Blume, Parasitic Medicinal Plant (CO2 농도와 온도 변화가 기생 약초인 천마의 생육반응에 미치는 영향)

  • Gastrodia elata (Orchidaceae) is a perennial parasitic plant that has a unique lifestyle of being in a symbiotic interaction with fungi. The underground root of Gastrodia is used for medicinal treatment to cure high blood pressure, stroke, leukemia, headaches, and especially neurasthenia. This study is intended to investigate the effect of the global warming on the ecological responses of Gastrodia. We cultivated the small tuber of Gastrodia with the oak wood lot and mulberry fungus under control (ambient $CO_2$ concentration + ambient temperature, ACAT), temperature treatment (ambient $CO_2$ concentration+elevated temperature, ACET), and $CO_2$+temperature treatment (elevated $CO_2$ concentration+elevated temperature, ECET). The elevated $CO_2$ concentration was about twice in the ambient air while the elevated temperature was about $2^{\circ}C$ higher than the control group. And then we observed the growth and production of reproductive organs and the underground root. The observation showed that the number of flower stalk was highest at ACET and lowest at ECET. The flower stalk was longest at ACET and shortest at ECET. The inflorescence was longest at ACAT and shortest at ECET. The seed capsule was heaviest at ACET and lightest at ECET. The aboveground biomass was highest at ACET and lowest at ECET. The number of rhizomes was highest at ACET and lowest at ECET. The total rhizome biomass was highest at ACET and lowest at ECET. The average rhizome biomass was highest at ACET and lowest at ECET. The results showed that the growth of Gastrodia increased because of more active growth of Gastrodia elata when only the temperature increased and decreased when both $CO_2$ concentration and temperature increased, indicating the poor growth of Gastrodia elata under the global warming condition. Therefore, the Gastrodia elata plantation should be maintained at the temperature of $20-25^{\circ}C$ and not be exposed to a high $CO_2$ concentration.

Effect of Achyranthis Radix and Drynariae Rhizoma Extracts on Antioxidant Activity and Antioxidant Enzymes (우슬과 골쇄보의 추출물이 항산화 활성 및 항산화 효소 대사에 미치는 효과)

  • In vitro and in vivo experiments using Achyranthis radix and Drynariae rhizoma extracts were conducted. Antioxidant properties were analyzed and the effects on bone, glucose and lipid metabolism were investigated. Drynariae rhizoma (64.67%) obtained higher DPPH radical scavenging activity compared to Achyranthis radix (19.03%). Similar results were obtained in the reducing power. No differences were observed on the ABTS radical scavenging ability and SOD. In contrast, Achyranthis radix (77.60%) has higher chelating ability compared to Drynariae rhizoma (46.21%). In vivo experiments revealed higher plasma TBARS in OVX-DR than in OVX-AR. Opposite result was seen in erythrocyte TBARS. Hepatic, nephritic and erythrocyte enzymes were considered for the antioxidant enzyme activities. GSH-Px and PON of hepatic enzymes were higher in OVX-AR. While the CAT and GR were higher in OVX-DR. SOD, GSH-Px, GR and PON of nephritic enzymes of OVX-DR were higher compared to OVX-AR. Almost similar values were obtained in CAT using both extracts. The OVX treated rats obtained higher CAT and GR in the erythrocyte enzymes compared to SHAM. The SOD of erythrocyte enzymes in OVX-DR was higher compared to OVX-AR. On the other hand, the GSH-Px was higher in OVX-AR.

Photoprotection and Anti-inflammatory Effects of Chinese Medical Plants (약용식물추출물의 광보호 효과와 항염증 효과 연구)

  • Chronic exposure to solar radiation, particularly ultraviolet (UV) light, causes a variety of adverse reactions on human skin, such as sunburn, photoaging and photocarcinogenesis. Free radicals and reactive oxygen species (ROS) caused by UV exposure or other environmental facts play critical roles in cellular damage. And, repeated-UV irradiation activated the expression of the matrix metalloproteinase (MMP) and induced skin irritation. Therefore, the development of effective and safe photoprotectants that can reduce and improve the skin damage has been required. The purpose of this study was to investigate the photo-protective effect of several chinese medical plants (Juniperus chinensis) on the UV -induced skin cell damages. We tested free radical and superoxide scavenging effect in vitro. Fluorometric assays of the proteolytic activities of MMP-1 (collagenase) were performed using fluorescent collagen substrates. UVA induced MMP-1 synthesis and activity were analyzed by enzyme-linked immunosorbent assay (ELISA) and gelatin-based zymography in skin fibroblasts. We also examined anti-inflammatory effects by the determination test of proinflammatory cytokine, interleukin 6 in HaCaT keratinocytes. Expression of prostaglandin E$_2$ (PGE$_2$) after UVB irradiation was measured by competitive enzyme immunoassay(EIA) using PGE$_2$ monoclonal antibody. In the human skin we tested anti-irritation effect on the SLS-induced damage skin after appling the extract containing emulsion. We found that Juniperus chinensis extract had potent radical scavenging effect by 98% at 100$\mu\textrm{g}$/mL. The extract of Juniperus chinensis showed strong inhibitory effect on MMP-1 activities by 97% at 100 $\mu\textrm{g}$/mL and suppressed the UVA induced expression of MMP-1 by 79% at 25$\mu\textrm{g}$/mL. This extract also showed strong inhibition on MMP-2 activity in UVA irradiated fibroblast by zymography. In the test of proinflammatory cytokines of human keratinocytes Juniperus chinensis extract decreased expression of interleukin 6 about 30%. The amount of PGE$_2$ by HaCaT keratinocytes was significantly increased at the doses of above 10 mJ/$\textrm{cm}^2$ of UVB (p < 0.05). At the concentrations of 3.2-25$\mu\textrm{g}$/mL of this extract, the production of PGE$_2$ by HaCaT keratinocytes (24 h after 10mJ/$\textrm{cm}^2$ UVB irradiation) was significantly inhibited in culture supernatants (p < 0.05). In SLS-induced skin irritation model in vivo, we found to reduce skin erythema and improve barrier recovery after appling Juniperus chinensis extract containing emulsion when compared to irritated non-treated and placebo-treated skin. Our results suggest that Juniperus chinensis extract can be effectively used for the prevention of UV and SLS-induced adverse skin reactions and applied as anti-aging and anti-irritation cosmetics.

Effects of Nitrogen Fertilization on the Yield and Effective Components of Chrysanthemum boreale M. (질소시비가 산국의 수량과 유효성분에 미치는 영향)

  • Chrysanthemum boreale M. (hereafter, C. boreale M.), a perennial flower, has been historically used as a natural medicine in Korea. With increasing concerns for health-improving foods, the demand for C. boreale M. has become higher than ever. Howevr, the amount of wild C. boreale M. collected from mountainous areas is not enough to cover all demands. The cultivation system and fertilization strategy are required to meet increasing demand on C. boreale M. with a good quality. We investigated the effects of nitrogen application on plant growth and effective components of C. boreale M. to suggest optimum rate of nitrogen fertilization. C. boreale M. was cultivated in a pot scale (1/2000a scale), and nitrogen applied with rate of 0(N0), 50(N50), 100(N100), 150(N150), 200(N200), and $250(N250)kg\;ha^{-1}$. Phosphate and potassium were applied at the same level ($P_2O_5-K_2O=80-80kg\;ha^{-1}$) in all treatments. Maximum yield achieved in 246 and $226kg\;ha^{-1}$ N treatment on the whole plant and the flower part, a valuable part as a herbal medicine, respectively. Proline was the most abundant amino acid in the flower of C boreal M. and the contents of amino acids increased with increasing nitrogen application rate in flower. Nitrogen recovery efficiency was high more than 41% in all nitrogen treatments and increased to 61.8% in nitrogen N100 treatment. From the nitrogen content, the high nitrogen uptake, the low residue of mineral N and the reasonably good apparent fertilizer recovery, it can be inferred that C. boreale M. made efficient use of the available nitrogen. In flower, contents of Cumambrin A. which is a sesquiterpene compound and has the effect of blood-pressure reduction, decreased with increasing nitrogen application. However, the amount of Cumambrin A in flower increased as nitrogen rate increased, because of increasing flower yield. Conclusively, nitrogen fertilization could increase yields and enhance quality. The optimum nitrogen application rate might be on the range of $225{\sim}250kg\;ha^{-1}$ in a mountainous soil.

  • In the present study, the vegetation was classified using the phytosociological method and canonical-correlation analysis (CCA) was implemented to analyze correlation between community structure and environmental factors in the natural habitats of forest byproducts, especially medicinal plants, such as Cudrania tricuspidata, Sorbus commixta, and Hovenia dulcis, in 2021-2022 to provide primary ecological data to establish environmental conditions for wild vegetable cultivation. A total of 11 plots in five regions, 8 plots in three regions, and 17 plots in 5 regions were selected for the natural habitats of C. tricuspidata in southern Korea, S. commixta in high mountains, and H. dulcis in valleys of central Korea, respectively. The importance value in each community was respectively analyzed as follows, in C. tricuspidata community, the importance value of C. tricuspidata (61.10) was the highest, followed by Celtis sinensis, Pinus thunbergii, Neolitsea aciculata, Styrax japonica, Carpinus coreana, Quercus serrata, and Q. acutissima. In Sorbus commixta community, Q. mongolica (57.21) was the highest, followed by, S. commixta (42.58), Betula ermani, Tilia amurensis, A. pseudosieboldianum, A. tschonoskii var. rubripes, Cornus controversa, Magnolia sieboldii, and Taxus cuspidata. In H. dulcis community, H. dulcis (64.58) was the highest, followed by Zelkova serrata, Cornus controversa, A. mono, Q. serrata, C. cordata, and Juglans mandshurica. As the result of the analysis on DBH of the major species having the high importance value, in C. tricuspidata community, C. tricuspidata, C. sinensis, Neolitsea aciculata, and C. coreana show the density of normal distribution, so the dominant status of these species is likely to continue. In S. commixta community, S. commixta show the density of reverse J-shaped curve, so the dominant status of these species is likely to be stable, and Q. mongolica, B. ermani and T. amurensis, show the density of normal distribution, so the dominant status of these species is likely to continue. In H. dulcis community, C. cordata, and J. mandshurica show the density of reverse J-shaped curve, so the dominant status of these species is likely to be stable, and H. dulcis, Z. serrata, C. controversa and A. mono had a formality distribution, suggesting a continuous domination of these species over the other species for the time being. The results of CCA ordination analysis using 11 environmental factors and 30 communities of three taxa classified by TWINSPAN analysis revealed that the altitude showed the strongest correlation with the vegetation. C. tricuspidata community was distributed on the moderate and gentle northeastern slope at low altitude with the highest pH, C.E.C, Ca2+, and Mg2 and various P2O5, whereas S. commixta community was distributed on the steep slope at high altitude with the highest O.M and T-N and lower P2O5, Ca2+, Mg2+, C.E.C and pH, which is the opposite tendency of the environment of C. tricuspidata community. H. dulcis community was distributed on the gentle northern slope at lower altitude with an average pH, O.M, T-N, Ca2+, Mg2+, and C.E.C, except higher P2O5.

Use of Auxins for Rhizome Propagation and Shading Effect on Growth of Asarum sieboldii Mio. (세신(細辛)의 근경삽(根莖揷)번식에 있어서 오옥신의 이용과 차광재배(遮光栽培))

  • This study was carried out to investigate the auxin(IAA, IBA, NAA) treatment and the effect of shading rate in Asarum sieboldii. The results obtained were summerized as follows: By the soaking treatment of auxins to the cutted rhizome enhanced root growth and plant weight. By the increment of shading rate, plant growth was much better compare to the control. Leaf fallen times appeared about 20 days more earlier at plain area then the alpain area. Root yield was much higher by the treatment of shading then the conventional cultivation so it seem to be the useful for large scale cultivations of A. sieboldii.

Antioxidant Activity and in vitro for Anticancer Effects of Manufactured Fermented Mulberry Leaf Tea (뽕잎발효차 제조에 따른 항산화 및 in vitro 항암활성)

  • This study was conducted to compare and analyze the qualitative property of MLT (mulberry leaf tea) and FMLT (fermented mulberry leaf tea) based on the antioxidant, anticancer activities of various extracts. When the antioxidant activity of MLT and FMLT extracts was evaluated, the electron donating activity was found to increase proportionally as the concentration of each extract increased. In addition, the extract of FMLT showed a higher electron donating activity than that of MLT. Furthermore, the ethanol extracts showed the highest electron donating ability. When the SOD activity was evaluated, it was also found to increase proportionally with the concentration of each extract. Furthermore, the SOD activity of the ethanol extract group was the highest, whereas the SOD like activities of both MLT and FMLT were similar. When nitrite decomposition was evaluated for each pH, the highest value was observed at pH 1.2. Finally, the nitrite deleting ability was the highest for the ethanol extracts. When each extract of MLT and FMLT was analyzed in vitro for anticancer effects, they were found to decrease the number of cancer cells proportionally as the concentration of extract increased for both HeLa cells and MCF-7 cells. Furthermore, FMLT was found to exert a greater inhibition of cancer cells than MLT. Among the extract groups, the ethanol extract induced the greatest inhibition of the development of cancer cells, and these effects were greater against MCF-7 cells than HeLa cells.

Effect of Diameter and Length of Root on Yield in Rehmannia glutionosa Libosch (지황(地黃) 종근(種根)의 굵기와 길이가 수양(收量)에 미치는 영향(影響))

  • This experiment was carried out to clarify the effect of root diameter and length on yield in Rehmannja glutionosa on experimental fields of Chungbuk Provincial Rural Development Ad­ministration. Emergance date was faster Sand 10 days with root diameter 3mm and 9mm than that of root diameter 6mm. Emergance ratio was high in the order of 6mm >7mm > 9mm with root diameter, and bolting ratio was increased in the order of 9mm > 6mm > 3mm with root diameter. Leaf length had no differences betwere root diameter and length, and leaf width had same tendency. Content of inorganic matter such as T - N, $P_2O_5,\;K_2O,\; C_aO,\;M_gO$ was high measuring date was delayed, and higher in root than leaf. Sugar content was higher in November than in October and had no differences with root diameter, but was more de­creased as the root length was longer. Yield was increased 9 % with root diameter 9mm. Comparing with root length, yield was higer 5% with 6cm than that of 1, 172kg/10a with 3cm, and decreased 25% with 9cm. And the profitable root was considered root diameter 8mm and root length 6cm.

Increment of Germanium Contents in Angelica keiskei Koidz. and Panax ginseng G.A. Meyer by In Vitro Propagation (명일엽(明日葉)(신선초(神仙草)) 및 인삼(人蔘)의 기내배양(器內培養)을 통한 Germanium 함량(含量) 증대(增大))

  • This study was carried out to find optimum concentration of germanium compounds and pH of medium on the induction and growth of callus from A. keiskei and P. ginseng and to intend to increase Ge. absorption by calli while those calli were subculturing on MS medium. Callus from a. keiskei was rarely induced under light condition. Under dark condition, callus in­duction from A. keiskei was good up to 5ppm, retarded at 50ppm of $GeO_2$, or C. E. Ge. O., and rarely done at 100 ppm of $GeO_2$ but was somewhat well at 100 ppm of C. E. Ge. O. The induction and growth of callus was good in order of pH 5. 7 > pH 5. 4 > pH 6. 0 Under light condition, the growth of callus induced from P. ginseng was poor at $1{\sim}10\;ppm$ of $GeO_2$, or C. E. Ge. O., but shooting from callus occurred frequently. Under dark condtion, the growth of callus from A. keiskei was good up to 5 ppm of $GeO_2$, or C. E. Ge. O. and was rarely done at 50 ppm of $GeO_2$, but was somewhat well even at 100 ppm of C. E. Ge. O. Shooting from callus occurred frequently in a. keiskei, especially at pH 5.7. The growth of callus from P. ginseng was poor at 10 ppm of $GeO_2$, or 50 ppm of C. E. Ge. O. Under dark condition, the amount of Ge absorption by callus induced from A. keiskei was much high­er than that from P. ginseng. The amount of Ge. absorption by callus treated with $GeO_2$, was higher than that treated with C. E. Ge. O.

Effect of Fermented (Oenanthe stolonifera DC) Extract on the Activity of Enzymes Related to Liver Function of Alcohol-administered Rats and Mice (밭미나리 발효액이 알콜투여 흰쥐의 간기능관련효소활성에 미치는 영향)

  • It has long been recognized that dropwort contains specific funtional subtances for protecting human liver and preventing and curing its diseases, and thus it has been widely utilized in traditional folk remedy. In the present study. fresh of fermented extract of dropwort shoots grown on dryland and fresh extract of those grown on flooded fie1d were fed to the rats suffering from acute, subacute or chronical toxication induced by alcohol administration, and their affects were investigated. Administration of alcohol to rats and mice for 2 days at 5ml of 30% EtOH/kg/day raised total cholesterol and total glyceride which were, however, great1y supressed when alcohol was administered to the laboratory animals previously fed on fresh or fermented extract of dryland dropwort, or fresh extract of flooded field-grown dropwort for 20 days, without significant differences among the extracts. The levels of alanine aminotransferase and aspartate aminotransferase which were raised by alcohol adminstration were also lowered by feeding dropwort extract, among which that of fermented dryland-grown one was more effective than the other two. Chronic alcohol toxication was induced to rats by administering 10% alcohol for 10 months and fermented dropwort extract or tap water was fed to the rats for 5 days. The rats fed on fermented dropwort extract were lower in total cholesterol by 40% and in tota1 glyceride by 60% than the control. Alanine aminotransferase and aspartate aminotransferase in the rats fed on fermented dropwort extract were decreased by 87.2% and 91.7%, respectively, compared to the control, and the rats recovered almost to normal. Activity of alkaline phosphatase, catalase, superoxide dismutase or glutathione peroxidase changed greatly by alcohol administration in the rats suffering from chronic as well as acute toxication. The extract of fermented dry land dropwort significantly lowed the activity of those enzymes, especially, alkaline phosphatase and superoxide dismutase. The present results suggesting the possible medicinal effect of fermented dropwort extract to liver diseases.
