Evaluation for Application of IOM Sampler for Agricultural Farmer's Inhalation Exposure to Kresoxim-methyl and Fenthion (농작업자의 Kresoxim-methyl과 fenthion에 대한 호흡노출량 측정을 위한 IOM 채집기의 효율성 평가)

  • An IOM sampler equipped with glass fiber filter has been recently utilized instead of solid adsorbent, which was used to measure the inhalation exposure of agricultural operator to pesticides. The aim of this study is to validate the efficacy of an IOM sampler by measuring the trapping efficiency and breakthrough using kresoxim-methyl water-dispersible granule and fenthion emulsifiable concentrate. On LC-MS/ MS, minimum detection level was 12.5 pg and method limit of detection was 5.0 ng/mL. Good linearity ($R^2$ > 0.999) for matrix matched standards was obtained. Recoveries of pesticides from glass fiber filter were 102-109% (kresoxim-methyl) and 97-104% (fenthion) while those from XAD-2 resin were 94-98% (kresoxim methyl) and 93-100% (fenthion). Trapping efficiency test was performed with personal air pumps and IOM sampler (glass fiber filter) connected with solid adsorbent (XAD-2 resin) with two types of formulation (solid and liquid) which were diluted by standard rate and sprayed to IOM sampler. Those pesticides were trapped only in glass fiber filter without any breakthrough to solid adsorbent. After spiking of pesticides to glass fiber filter, breakthrough test was carried out with IOM sampler (glass fiber filter) which was connected with solid adsorbent. As a results, 87-101% of kresoxim-methyl and 96-105% of fenthion remained in spiked glass fiber filter, however, no pesticides were detected in second glass fiber filter and solid adsorbent. In conclusion, IOM sampler which equipped with glass fiber filter can be applied widely for pesticide inhalation exposure study since it has good trapping efficiency and adsorption capacity, regardless of the solid or liquid formulation.

STUDIES ON THE CYSTINE COMPONENT IN THE SERICULTURAL PROTEINS OF BOMBYX MORI L. (가잠사단백질의 각과정에서의 Cystine 성분에 대한 연구)

  • The purpose of this treatise is to prove the presence of cystine in silk fiber through wide sampling throughout all the sericultural processes of Bombyx mori.; also to show that disulfide cross linkages exist in the silk fiber. The conclusions reached were as follows: 1. Crystalline cystine was obtained from silk fibroin using Folin's Method. 2. Analytical data showing the cystine content of silk fiber and its related materials were obtained using Sullvan's Method as follows: Material Percent Cystine A. Mulberry leaf protein 0.175 B. Silkworm egg 0.33 C. Silkworm Body, matured, fat extracted, without silk gland 0.41 D. Silk gland, matured 1.23 E. Silkworm feces none F. Silkworm pupa, fat extracted 0.30 G. Silkworm moth, fat extracted 0.60 H. Raw Silk 0.22 I. Fibroin 0.175 J. Sericin 0.30 3. The presence of cystine in the silkworm was substantiated the existence of 0.175 % methionine in mulberry leaves and 0.12% methionine in the silk gland. 4. Part of the sulfhydryl compounds in the silk gland is believed to transfer to serine and methionine, with the former being secreted into the liquid silk finally as silk fiber and the latter used for nutritive purposes in the growing of silk gland tissue. 5. The cystine content is variable by mulberry species, silkworm species, sex, breeding process, and other culturing environments. 6. Hybrid silkworms require more nutritive amino acids for effective growth than the original parents, and secrete less of them as silk fiber. 7. From such an observation, the amino acid composition of silk fiber is believed to be fairly flexible. Cystine if included in the amorphous part of the fiber, especially in sericin. 8. The result from enriching the silkworm diet with pure cystine or wool cystine did not result in any advantage, therefore it is believed that the natural cystine and methionine contents in the mulberry leafaregoodenoughforsilkwormnutrition. 9. The disulfide cross linkage in silk fiber was verified by using the Harris Method. Contraction took place following the treatment of the fiber with various salts and acids. Comparisons were made with wool fiber. 10. During these experiments, the fibrious structure of silk fiber and the net-globular liquid form were photographed microscopically. It is believed that the globules of liquid silk are net-formed by the inter attraction of the OH ion of the globular peptide and the H ion of water as shown by the hair cracking behavior of the film. The net-globular protein precipitation from the mulberry protein solution showed that mulberry is a proper diet for the formation of fibrous protein in the silk fiber. 11. The significance of the presence of cystine in silk fiber as emphasized in this paper should result in modification of the general conception that cystine is absent from this fiber.   • In this study, effective gene extraction methods were compared to identify raw materials of processed meat products through molecular biological methods. Species specific primers were used to identify ingredients of processed foods and, as a sample, 13 kinds of processed meat products including beef, pork and chicken. According to the type of sample, 13 kinds of samples were classified into liquid type, source type and powder type. The samples were pre-treated (centrifugation) and (or) performed Whole Gene Amplification (WGA) kit for amplification of the extracted DNA. As a result, it was possible to identify the raw material of products through the centrifugation of sample 1 ml for liquid type of processed meat products. For source type of products after gene extraction, it was required to perform WGA for the identification of ingredients. For powder type products did not required any further pre-treatment and WGA. In this study, it was an opportunity to confirm the possibility of identification of raw material from the gene extraction of processed meat products and this method could be used to examine the authenticity of raw material of products.

Total Sugar and Artificial Sweetener Contents of Health Functional Foods in Seoul (서울지역 유통 건강기능식품의 당 및 인공감미료 함량)

  • This study was carried out to investigate and evaluate total sugar and artificial sweetener contents in health functional foods. In this study, HPLC with evaporative light scattering detector (ELSD) and HPLC-UV were used to determine the contents of total sugar and artificial sweetener in health functional foods. Sixty-six chewable products and sixty red ginseng products were collected from markets in Seoul. The average content of 126 samples per daily intake portion was 1.96 g ranging from not-detected (N.D.) to 12.61 g. The mean total sugar content per serving of chewable product was 1.26 g and N.D. to 10.39 g. The average amount of total sugar per daily intake of ginseng and red ginseng was 2.70 g and N.D. to 12.61 g. The average amount of sugar per daily intake of chewable products was 2.10 g for children, 1.43 g for nutrients, and 0.35 g for functional raw material. For children's products, the content of sugar per serving was ranged from 1.03 g to 5.33 g, from N.D. to 10.39 g for nutrients and from N.D. to 2.61 g for functional raw materials. The average content of sugar per daily intake of ginseng and red ginseng product was 4.25 g in liquid product, 1.51 g in concentrate product and 1.49 g in powder product. The contents of sugar per the daily intake of the liquid product ranged from N.D. to 10.80 g, from 0.01 g to 12.61 g for the concentrated product, and from 0.06 g to 5.64 g for the powdered product. Analysis of artificial sweeteners showed that artificial sweeteners were detected in three cases. No artificial sweeteners were detected in ginseng and red ginseng products. Two of the chewable products and one of the functional raw materials were detected. The detected artificial sweeteners were aspartame, 3.09 g/kg in nutrients and 1.09 g/kg in functional raw material.

Vitamin D analysis in the Korean total diet study and UV/sun light irradiated mushrooms (한국형 총식이조사 및 UV/태양광 조사 버섯에서의 비타민 D 분석)

  • This study was conducted to evaluate vitamin D intake of Koreans in a total diet study (TDS) and to determine the effect of irradiation on vitamin D synthesis in mushrooms. For analysis, sample were saponified and extracted with hexane, and vitamin D was quantified by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Based on the validation results, the recovery of the National Institute of Standards and Technology (NIST) standard reference sample (SRM) 1849a was 96.7% and the z-score of -1.6 was obtained by the Food Analysis Performance Assessment Scheme (FAPAS) proficiency test (PT) 21115. Vitamin D2 was not detected in any samples, and the highest level of vitamin D3 was detected in mackerel and anchovies ranging from 24.2 to 120.2 ㎍/kg. The mean daily intake of vitamin D was 0.99 ㎍/day, as estimated from the vitamin D contents of the analyzed foods and their corresponding intake. The adequate intake (AI) of vitamin D based on the Dietary reference intakes for Koreans provided by the Ministry of Health and Welfare is 5-15 ㎍/day for Koreans aged 6 to 75 years. Compared with this AI, vitamin D intake of Koreans estimated in this study was inadequate. For that, the increased vitamin D content in ultraviolet (UV)/sun light irradiated mushrooms warrants further research to increase vitamin D intake of Koreans through diet.

Isolation of endosulfan degrading bacteria and their degradation characteristics (유기염소계 농약 endosulfan을 분해하는 미생물의 분리 및 분해 특성)

  • A bacterium, which was named to be Bacillus sp. E64-2, capable of degrading endosulfan was isolated from the environmental sample using enrichment culture technique. The Bacillus sp. E64-2 was able to degrade 99% of 10 mg/L endosulfan in the culture media within 7 days at $30^{\circ}C$. Endosulfan diol was the only intermediate by the endosulfan degrading bacterial culture and the pH value of the culture media was significantly increased to pH 8.4 from pH 7.0 after 7 days of incubation. When the endosulfan and the crude extract of the strain were incubated, endosulfan diol was a major metabolite. Both the enzymatic reaction and the pH-increasing effect contribute to the degradation of endosulfan by the bacterial culture.

Effects of Mixed Scutellaria baicalensis Extracts as Natural Preservative on Efficacy and Storage of Lactic Acid-Fermented Garlic Extract (천연보존료 복합 황금추출물이 유산균발효 마늘추출물의 저장성 및 기능성에 미치는 영향)

  • The natural preservative "complex Scutellaria baicalensis extract (BHC)" contains Scutellaria baicalensis, Glycyrrhiza uralensis (liquorice), Zizyphus jujube (jujube), and Astragalus propinquus (milk vetch root). BHC has been used as a natural preservative for more than 10 years to increase storage duration and quality of food with strong antibacterial activity. BHC has been added into functional foods as a subsidiary ingredient. However, no studies have been performed to test whether or not BHC affects the activity of main functional ingredients. In this study, we tested whether or not BHC has any effect on the hepatoprotective activity of lactic acid-fermented garlic extract (LAFGE) when formulated in a clinical test supplement. $H_2O_2-induced$ oxidative damage in HepG2 cells was not attenuated by BHC, indicating that BHC had no influence on the protective effect of LAFGE against oxidative damage. Furthermore, BHC had no effect on the hepatoprotective effect of LAFGE against acetaminophen-induced acute liver injury in rats, as indicated by no changes in alanine transaminase and aspartate transaminase levels. In conclusion, BHC, formulated in the clinical test supplement with LAFGE, had no effect on hepatoprotective activity, indicating BHC could be considered as a suitable natural preservative for liquefied functional food materials.

Preparation of Water-Soluble Solid Forms Containing Ursolic Acid Using Supercritical Fluid (초임계유체를 이용한 우르솔산 함유 고체 가용화 제제의 제조)

  • In this work ursolic acid (UA), a poorly water-soluble compound, was inclusion complexed with 2-hydroxypropyl-$\beta$-cyclodextrin (HP-$\beta$-CD) by various methods such as kneading, solvent evaporation and two types of supercritical fluid processes. The solubility and characteristics of these UA/HP-$\beta$-CD complexes were investigated by scanning electron microscopy, x-ray diffraction and HPLC. The water solubilities of the two complexes obtained from solvent evaporation and ASES processes were observed to increase up to 6$\sim$240 folds and 12$\sim$56 folds, respectively, compared with that of unprocessed UA. The stability of UA/HP-$\beta$-CD complex samples in cosmetic formulations was examined at various temperatures for one month. The UA/HP-$\beta$-CD complex prepared by solvent evaporation was found to be most stable among all the cosmetic formulations tested in our experiments.

A Study on the Synthesis of Potassium Hexatitanate Whisker by the Slow Cooling Calcination Process (서냉 소성법에 의한 육티탄산칼륨 Whisker의 합성에 관한 연구)

  • Fibrous potassium hexatitanate whisker with the size of $ID=0.5{\sim}1{\mu}m$ and length=$100{\sim}1000{\mu}m$ (aspect ratio=100~1000) was produced through the reaction between titanium dioxide and potassium carbonate using the slow-cooling calcination followed by water leaching treatment. The optimum condition for the production of fibrous potassium titanate was calcination temperature of $1100^{\circ}C$ for 5hrs, $TiO_2$ mole ratlo to $K_2CO_3$ of 4.5 and slow-cooling rate of $0.5^{\circ}C/min$ to $860^{\circ}C$. Fibrous crystal are grown by the association between the solid potassium titanate and liquid phase during the slow-cooling process. The Proper water leaching condition for removing of K component was leaching time of 10hrs in boiling water. Pressurizing of the mixture of $K_2CO_3$ and $TiO_2$ to be calcinated became effective on the growth of fibrous crystal.

MCC 입자의 표면화학적 특성에 따른 부유부상 효과

  • 부유부상 공정은 현재 신문고지와 사무용 고지의 탈묵을 통한 신문용지 및 화장지 제조 에 널리 사용되고 있는 고지재활용을 위한 핵심공정이다. 하지만 이들 공정은 갈수록 열악 해지는 국내 고지원료의 품질변화에 따라서 잉크 및 토너의 분리효율이 저하되고 있어 생산 되는 탈묵 펄프의 품질저하, 리젝트 발생량의 증대, 폐수 처리공정의 부하 증가 등 다양한 문제점을 발생시키고 있어 이에 대한 대책의 수립이 시급히 요청되고 있다. 현재와 같이 다양한 형태의 잉크 및 인쇄방식으로 인쇄된 많은 종류의 재활용지가 원료 로 투입되고 있는 경우 탈묵공정의 효율 향상을 위해서는 부유부상 공정에 대한 좀 더 체계 적이고 논리적인 접근이 필요하다. 이러한 문제점을 극복하기 위해 먼저 고지 재활용 공정 의 핵심 단위공정인 부유부상 공정에 관련된 복잡한 문제점을 단순화하여 고상 및 액상의 표면화학적 특성을 평가하고, 이에 따른 부유부상 공정의 효율을 분석하였다. 본 연구에서는 부유부상 공정을 기초과학적 측면에서 구명하기 위해 마이크로 크리스탈린 셀룰로오스(Microcrystalline cellulose: MCC)를 모델 물질로 사용하였고, 친수성 의 표면 특 성을 나타내는 MCC의 표면 특성을 바꾸기 위하여 AKD(alkyl ketene dimer)로 처리비율을 달리하여 사이징 처리하였다. 부유부상 실험에 사용된 MCC는 친수성을 띠는 것과 소수성 을 부여한 것을 구별하고 그 비율을 백색도를 통해 측정하기 위하여 자체적으로 친수성을 가지는 MCC는 세척 견뢰도가 높은 검은색 염료로 염색하였다. 준비된 친수성과 소수성 M MCC의 혼합비율 별로 패드를 작성하여 백색도를 측정함으로써 검량선을 작성하였다. 또한 부유부상 시간에 따른 제거효율을 알아보기 위하여 부유부상 시간별로 각각의 리젝트율과 수율올 측정하고, 리젝트 시료로부터 패드를 제조하여 백색도를 측정하였다. 실험 결과 소수성 MCC의 소수화 정도에 따라서 리젝트율이 증가하였으며, 이를 통해 표 면에 소수성을 띠는 입자는 소수성이 강할수록 부유부상공정에서 제거율이 증가한다는 것을 확인하였다. 부유부상 처리한 MCC로 패드를 제조한 뒤 백색도를 비교한 결과에서도 이를 확인하였다. 한편 리젝트로 함께 제거된 MCC 내에 존재하는 친수성 MCC의 양은 극히 미 세하였다. 또한 부유부상를 실시하는 초기에 상당량의 리젝트가 발생함을 확인하였는데, 이 는 전체 부유부상을 통해 제거되는 양의 45-68%였다. 한편 부유부상이 진행됨에 따라서 리젝트 양의 증가폭이 둔화되는 경향을 나타내었다. 이러한 경향은 MCC의 소수성이 강할 수록 더욱 뚜렷하게 나타났다. 이를 통해 계 내로부터 소수성인 물질이 급속하게 제거됨을 알 수 있었으며 필요 이상의 부유부상 처리는 잉크제거 효율을 높일 수는 있으나 소모되는 시간에 비하여 비효과적임을 알 수 있었다.
