Regulation of Arabidopsis Circadian Clock by De-Etiolated 1 (DET1) Possibly via Histone 3 Acetylation (H3Ac) (히스톤 3 아세틸화(H3Ac)를 통한 De-Etiolated 1 (DET1)의 애기장대 생체시계 조절)

  • The circadian clock is a self-sustaining 24-hour timekeeper that allows organisms to anticipate daily-changing environmental time cues. Circadian clock genes are regulated by a transcriptional-translational feedback loop. In Arabidopsis, LATE ELONGATED HYPOCOTYL (LHY) and CIRCADIAN CLOCK-ASSOCIATED 1 (CCA1) transcripts are highly expressed in the morning. Translated LHY and CCA1 proteins repress the expression of the TIMING OF CAB EXPRESSION 1 (TOC1) transcripts, which peaks in the evening. The TOC1 protein elevates the expression of the LHY and CCA1 transcripts, forming a negative feedback loop that is believed to constitute the oscillatory mechanism of the clock. In mammals, the transcription factor protein CLOCK, which is a central component of the circadian clock, was reported to have an intrinsic histone acetyltransferase (HAT) activity, suggesting that histone acetylation is important for core clock mechanisms. However, little is known about the components necessary for the histone acetylation of the Arabidopsis clock-related genes. Here, I report that DET1 (De-Etiolated1) functions as a negative regulator of a key component of the Arabidopsis circadian clock gene LHY in constant dark phases (DD) and is required for the down-regulation of LHY expression through the acetylation of histone 3 (H3Ac). However, the HATs directly responsible for the acetylation of H3 within LHY chromatin need to be identified, and a link connecting the HATs and DET1 protein is still absent.

Immunostimulating and Anticancer Activities of Hot Water Extract from Capsosiphon fulvescens (매생이 열수추출물의 면역 및 항암 활성)

  • To investigate the immunostimulating and anticancer activities from hot water extract of Capsosiphon fulvescens, tumor cell toxicity, sarcoma-180 growth inhibition activity, complement system-activity, intestinal immune system and oral toxicity were performed. The extract of Capsosiphon fulvescens was prepared by hot water and precipitated by using ethanol. Partially purified extract (CFE) was obtained after dialysis and ultrafiltration. The polysaccharide compositions consisted of xylose(19.1%), fucose(15.3%), mannose(4.2%) and galactose(7.9%). The tumor cell toxicity of CFE slightly showed at high concentrations of 10-30 ${\mu}g/ml$, but inhibition ratio against mouse solid tumor was more increased for CFE of 40.1-59.4% than the control. Blood leukocyte counts increased to a maximum of 83% including liver, spleen and thymic of mouse. Immunoglobulin A binding amounts showed a high level of CFE of $2,454{\pm}113.8-2,670{\pm}133.1{\mu}g/mg$ in comparison with the control of $2,092{\pm}123.0{\mu}g/mg$. Acute toxicity of CFE was not detected at the concentration of 2,000 mg/kg in normal mouse.

An Experimental Study on the Effectiveness of Microwave Hyperthermia Combined with Radiation on the Small and Large Intestine in rats (흰쥐의 장조직에 X-선 조사와 마이크로파 온열요법의 효과에 관한 실험적 연구)

  • The synergistic effect of combining radiation therapy and hyperthermia kills significantly more cells than using either modality alone. The reason for enhanced cell killing from the combined treatment is that the two modalities are complementary. For histopathological exmination, 102 rats were divided into 4 groups as hyperthermia, radiation, hyperthermia combined with radiation and normal control groups. The effect of prior irradiation (6-15 Gy of X-ray) on the response of small and large bowel of rats to $40^{\circ}C-44^{\circ}C$ (for 30 minutes) microwave (2450 MHz) hyperthermia was investigated. The musculature of the small and large intestine remained intact and the circumference of the histological sections were not significantly altered by the heated at $43^{\circ}C$ for 30 minutes. Thermal enhancement ratios of normal tissue is 1.0 Thermal enhancement ratio was not increased in combination therapy by evaluation of histopathologic changes in small and large intestine.

  • Purpose: Optical imaging is providing great advance and improvement in genetic and molecular imaging of animals and humans. Optical imaging system consists of optical imaging devices, which carry out major function for monitoring, tracing, and imaging in most of molecular in-vivo researches. In bio-luminescent imaging, small animals containing luciferase gene locally irradiate light, and emitted photons transmitted through skin of the small animals are imaged by using a high sensitive charged coupled device (CCD) camera. In this paper, we introduced optical imaging system for the image acquisition of bio-luminescent signals emitted from small animals. Materials and Methods: In the system, Nikon lens and four LED light sources were mounted at the inside of a dark box. A cooled CCD camera equipped with a control module was used. Results: We tested the performance of the optical imaging system using effendorf tube and light emitting bacteria which injected intravenously into CT26 tumor bearing nude mouse. The performance of implemented optical imaging system for bio-luminescence imaging was demonstrated and the feasibility of the system in small animal imaging application was proved. Conclusion: We anticipate this system could be a useful tool for the molecular imaging of small animals adaptable for various experimental conditions in future.

Ecosystem Risk Assessment Using the Indicator Species (지표종을 이용한 생태계 위해성평가)

  • Risk assessment by living indicator species provides the information about the ecosystem disturbance, disapperance of symbiosis and change of living group. In the initial stage of this kind research, the degree of contamination was reported using the level of simple number, but simple number may not represent the risk itself which can be casued in the living organisms. Risk assessment using various indicator species overcomes these limitations and can be expanded to the DNA level. In many developed counties, the government has supervised the researches about the indicator species for the monitoring and its application to ecosystem restoration. Several living indicator species found in the vicinity of the abandoned Au mines such as fern, earthworm, bacteria, rhizosphere-rhizoplane, salamamdor and DNA change of these species are described in this paper.

Effect of Environmental Factors on Appressorium Formation of Colletotrichum truncatum (환경조건(環境條件)이 대두(大豆) 탄저병균(炭疽病菌)(Colletotrichum truncatum)의 부착기형성(附着器形成)에 미치는 영향(影響))

  • The results investigated of the effect of light, pH, and nutrients in vitro and the levels of fertilizer(P,K and Ca) in vivo in order to find out the effect of environmental factors in forming an appressorium of Colletotrichum truncatum were as follows; The ratio of appressorium formation promoted more in a light treatment than in a dark treatment according to investigation after three and six hours passed away. But after the period, the ratio of appressorium formation promoted highly in both treatments. Reasonable pH of appressorium formation was pH 6-pH 8. The ratio of appressorium formation was the most favorable at none nutrient level in every treatment of C-source and N-source, and suppressed in a high concentration of C-source and N-source. But according as the time passed away, the ratio of appressorium formation promoted in C-source and N-source. When their quantities were increased in P and K treatments, the ratio of appressorium formation was somewhat suppressed but it was highly suppressed in the gradual order of none fertilizer level < standard fertilizer level < doubled fertilizer level in Ca treatment according to investigation after the first three hours passed away. But after the period, the ratio of appressorium formation promoted highly regardless of the level of fertilizer.

  • In this paper, requirements of efficient group key creation in multiple hierarchy structure environment with clear distinction of hierarchical roles within organizations are explained and the method of creating a group key that satisfies such requirements is proposed. The proposed method creates the group key through logical sum operation of hierarchy identifier created using uni-directional hash chain and group identifier randomly created according to the access right. The problem of excessive possession of key information by upper group users in the existing static group key creation technique was resolved. At the same time, lower group users were prevented from deducing key information of upper group users. In addition, as a result of comparative analysis performed with an experiment on existing super group key creation technique and multiple hierarchy group key method, the proposed method was found to be equivalent or superior to existing method in terms of various items including the total number of keys created, the number of keys possessed by users, the number of keys used for encoding and decoding of information, and expandability of keys.

Influence of Chilling Stress on Photosynthetic and Physiological Reponses of Cucumber (Cucumis sativus L.) Seedlings (오이묘에 냉온 스트레스가 광합성 및 생리반응에 미치는 영향)

  • This study were conducted to investigate the influence of chilling on photosynthetic rate, root activity, contents of total sugars and fatty acids of cucumber seedlings grown in a greenhouse. Even though photosynthetic activity of seedlings exposed to $0^{\circ}C$ for 5 hours was little or insignificantly influenced, it was reduced by 52.8% and 67.7% in seedlings exposed to the same temperature for an extended 10 and 24 hours, respectively. Photosynthetic rate decreased significantly when seedlings were illuminated, as compared to continuously held under darkness, during 15 hours of chilling treatment at 3$^{\circ}C$. Recovery of photosynthetic ability was also retarded by illumination during a recovery period after chilling treatment. Root activity, as measured by the oxidation power of $\alpha$-naphtylamine, was significantly reduced by chilling treatment at 0 to 6$^{\circ}C$, but amount of bleeding xylem sap collected at 40 days after chilling treatment was not significantly different among treatments. Total sugar content increased by 12 and 23% as compared to the control in seedlings chilled for 24 hours, respectively, at 3$^{\circ}C$. Contents of unsaturated linolenic and oleic acids increased, while content of saturated palmitic acid decreased with chilling treatment.

  • The physicochemical characterstics of El Salvadoran Coffea arabica cv. Bourbon coffee extracts under various roasting conditions were investigated. Green beans were roasted under four different conditions (Light-medium, Medium, Moderately dark, and Very dark). The coffee extracts were prepared by using the espresso or drip methods. As the roasting degree increased, the coffee bean moisture content decreased and the ash content increased. The lightness and yellowness of the beans and coffee extracts decreased along with the increasing roasting degree. In the drip coffee, the reducing sugar content decreased and the pH value increased along with the increasing roasting degree. Both in the espresso and drip coffee, total organic acid and chlorogenic acid contents decreased, while the caffeine content increased along with the increasing roasting degree. Therefore, it is suggested that the roasting degree affects the physicochemical characteristics of coffee extracts.

Investigation of Axially Loaded Jacked Pile Behavior by Pile Load Test (말뚝재하시험을 통한 압입강관말뚝의 연직지지거동 분석)

  • Jacked pile that involves the use of hydraulic jacks to press the piles into the ground is free from noise and vibration, and is possibly installed within a limited construction area. Thus, as an alternative to conventional pile driving methods, pile jacking could become widely accepted for the construction projects in urban area (e.g., reconstruction or remodeling construction projects). Great concern has arisen over the prediction of axially loaded jacked pile behavior. Against this background, a series of pile load tests were hence conducted on a jacked steel pipe pile installed in weathered zone (i.e., weathered soil and weathered rock). From the test results, base resistance and shaft resistance for each test condition were evaluated and compared with the values predicted by the previous driven pile resistance assessment method. Test results showed that the previous driven pile resistance assessment method highly underestimated both the base and shaft resistances of a jacked pile; differences were more obviously observed with the shaft resistance. The reason for this discrepancy is that a driven pile normally experiences a larger number of loading/unloading cycles during installation, and therefore shows significantly degraded stiffness of surrounding soil. Based on the results of the pile load tests, particular attention was given to the modification of the previous driven pile resistance assessment method for investigating the axially loaded jacked pile behavior.
