Effects of Conjugated Linoleic acid(CLA) on the Growth of Tumor Cells and the Production of Interleukin-1 and Interleukin-2 (Conjugated Linoleic Acid(CLA)의 암세포 증식 억제효과 및 Interleukin-1과 Interleukin-2의 생성에 미치는 영향)

  • Conjugated dienoic derivatives of linoleic acid(CLA) are a series of positional and gemotric isomers of linoleic acid which are found naturally in food, mainly dietary products and breef. We studied the effects of CLA on the growth of tumor cells and the production of interleukin-1(IL-1) and interleukin-2(IL-2). CLA treatment markedly inhibited the growth of Yac-1 cells and sarcoma-180 cells by 99 and 82% to that of control, respectively, after four days of incubation at 37$^{\circ}C$. To elucidate the immunological mechanism of antitumor activity of CLA, spleen cells of Balb/c mouse were exposed to 31, 63, 125, 250 $\mu\textrm{g}$ of CLA per ml for 24 hrs at 37$^{\circ}C$. The culture supernatants of CLA-exposed spleen cells reduced the production of IL-1 and IL-2 in all of the test conditions. These results indicate that the anticarcino-genic effect of CLA was mediated by the other actions rather than the production of the Il-1 or IL-2. We suggest that CLA might have an antiinflammatory effect in part due to its inhibitory action on the production of IL-1.

Antiproliferative Effect of the Salviae miltiorrhizae Radix Extracts on the Cancer Cell Lines (단삼 추출물이 암세포주에 미치는 세포증식 억제 효과)

  • Objectives : The purpose of this study was to identify antiproliferative effects of Salviae miltiorrhizae Radix(SM) extracts against cancer cell lines. Methods : We used 2 kinds of cancer cell lines such as colon cancer cells(HT-29), human oral epitheloid carcinoma cells(KB). MTT assay was performed to examine the efficacy of SM extracts on the cytostaticity of cancer cells in proportion to time and doses. Apoptosis was evaluated by DNA laddering and DAPI nuclei staining. Results : The MTT absorbances against HT-29 and KB of SM extracts were significantly decresed. DNA ladders could be identified in KB of SM extracts. The morphological change were observed and number of cells were decreased by SM extracts. Conclusions : SM extracts is considered to be effective to induce apoptosis and inhibit cancer cell proliferation.

The Antimutagenic Activity and the Growth Inhibition Effect of Cancer Cells on Methanol Extracts from Small Water Dropwort (돌미나리 메탄올 추출물의 항돌연변이 작용과 암세포증식 억제효과)

  • The study was carried out to evaluate the antimutagenic and anticancer effects of small water dropwort. The methanol extracts from small water dropwort significantly reduced the mutagenicity induced by aflatoxin $B_1\;(AFB_1)$ and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in Salmonella typhimutium TA 100. Also, the methanol extracts inhibited the growth of AZ-521 human gastric cancer cells and HT-29 colon cancer cells. The chloroform fraction from methanol extracts of small water dropwort inhibited $40\;to\;80\%$ of the mutagenicity by $AFB_1$ in Sal. typhimurium TA 100 by the addition of 2.5 to $10\%$. To separate active compounds, the chloroform fraction was subjected to column chromatography on a silica gel and separated into five fractions. Among the five fractions, fraction 4 showed the highest antimutagenic effect against $AFB_1$ and an anticancer effect in the HT-29 colon cancer cell. As the result of the analysis in GC-MS, 1-napthalene carbonitrile, 5,6,7,8-tetrahydrol and benzene, 1,1'-(1,4-pentadiene-1,5-diyl) bis-,(E,E) were identified potentially from fraction 4.

Effects of Purple Kohlrabi (Brassica oleracea var. gongylodes) Flesh and Peel Ethanol Extracts on the Antioxidant Activity and Antiproliferation of Human Cancer Cells (자색 콜라비 가식부와 껍질 에탄올 추출물의 항산화 활성 및 암세포 증식 억제효과)

  • This study examines the effects of purple Kohlrabi fresh and peel ethanol extracts on the antioxidative activity and antiproliferation of human cancer cells (Hep G2 human liver, HCT-116 human colon, and A549 human lung cancer cells.) The total flavonoid and anthocyanin content of purple Kohlrabi ethanol extracts were much greater in the peel than in the flesh. The DPPH radical scavenging activity and antioxidative index of purple Kohlrabi peel extracts were similar to those of the BHA and the BHT. Antiproliferation effects of purple Kohlrabi peel extracts on human cancer cells (Hep G2, HCT-116, and A549) strengthened in a dose-dependent manner. In particular, the antiproliferation activity of purple Kohlrabi peel extracts exceeded 40% in colon cancer cells. These results indicate that the purple Kohlrabi peel may contain bioactive compounds such as flavonoids as well as anthocyanin and that these compounds may facilitate cancer prevention.

Effect of the Hexane Extract of Saussurea lappa on the Growth of HT-29 Human Colon Cancer Cells (목향 헥산추출물이 대장암세포인 HT-29 세포의 증식에 미치는 영향)

  • In Asia Saussurea lappa (SL) has been used as a traditional herbal medicine to treat abdominal pain and tenesmus. Recently, in vitro cell culture studies have shown that SL has anti-ulcer, anti-inflammatory, and anti-tumor properties. To explore its potential chemopreventive and chemotherapeutic effects in colon cancer, we examined whether the hexane extract of SL (HESL) could inhibit the growth of HT-29 human colon cancer cells, and investigated the mechanisms for this effect. The cells were cultured with various concentrations (0-5 ${\mu}g/mL$) of HESL. The results indicated that HESL markedly decreased the numbers of viable HT-29 cells; whereas at the concentration of 5 ${\mu}g/mL$, HESL slightly decreased the viable cell numbers of CCD 1108Sk human skin normal fibroblasts at 72 hr. HESL substantially increased the numbers of cells in the sub G1 phase, and dose-dependently increased apoptotic cell numbers. Western blot analysis of the total cell lysates revealed that HESL increased Bax protein levels, but did not affect Bcl-2 levels. HESL induced the cleavage of poly (ADP-ribose) polymerase and caspases 8, 9, 7, and 3. This study demonstrated that HESL inhibits cell growth and induces apoptosis in HT-29 cells, which may be mediated by its ability to increase Bax levels and activate the caspase pathway. These findings may lead to the development of new therapeutic strategies for colon cancer treatment.

Apoptotic Effect of Extract from Artemisia annua Linné by Akt/mTOR/GSK-3β Signal Pathway in Hep3B Human Hepatoma Cells (Hep3B 간암세포에서 개똥쑥추출물로부터 Akt-mTOR-GSK3β 신호경로에 의한 apoptosis 효과)

  • Extracts from Artemisia annua Linné (AAE) have been known to possess various functions, including anti-bacterial, anti-virus, and anti-oxidant effects. However, the mechanism of those effects of AAE is not well-known. The aim of this study was to analyze the inhibitory effects of AAE on cell proliferation of the human hepatoma cell line (Hep3B) and to examine its effects on apoptosis. Activation by phosphorylation of Akt is cell proliferation through the phosphorylation of TSC2, mTOR, and GSK-3β. We suggested that AAE may exert cancer cell apoptosis through Akt/mTOR/GSK-3β signal pathways and mitochondria-mediated apoptotic proteins. For this, we examined the effects of extracts of AAE on cell proliferation according to treatment concentration. Treatment with AAE not only reduced cell viability, but also resulted in the induced release of lactate dehydrogenase (LDH). These results were determined with a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and a lactate dehydrogenase (LDH) assay. Furthermore, we determined the effects of apoptosis through Hoechst 33342 staining, annexinⅤ-propidium iodide (PI) staining, 5,5′, 6,6′-tetrachloro-1,1′,3,3′-tetraethyl-imidacarbocyanine iodide (JC-1) staining, and Western blotting. Our study showed that the treatment of liver cancer cells with AAE resulted in the inhibition of Akt, TSC2, GSK-3β-phosphorylated, Bcl-2, and pro-caspase 3 and the activation of Bim, Bax, Bak, and cleaved PARP expressions. These results indicate that AAE induced apoptosis by means of a mitochondrial event through the regulate of Akt/mTOR/GSK-3β signaling pathways.

Analysis of the effects of δ-Aminolevulinic acid on the proliferation and apoptosis of mammalian cells (포유류 세포주에서 δ-Aminolevulinic acid (ALA)의 세포증식과 사멸에 미치는 영향분석)

  • ${\delta}$-Aminolevulinic acid (ALA) is a compound which is widely present in the biosphere and plays an important role in the living body as an intermediate of the tetrapyrrole compound biosynthesis pathway that leads to heme in mammals and chlorophyll in plants. ALA is of interest as a biodegradable mediator, a growth regulator, a precursor of heme proteins, and an effective agent used in therapy of cancer. It has been recently reported that ALA is commonly used in dermatology, due to good effects of skin therapy. Although for the last few decades a substantial amount of research has been focused on the elucidation of the mechanism of ALA and the improvement of its therapeutic activity, it's effect on the cell functions and growth was not cleared. Here, we identified that ALA treatment could attenuate cell proliferation of HEK293T and HaCaT cells. In addition, ALA treatement could induce apoptosis of HeLa cells. These results suggest that apoptosis induced by ALA treatment might be responsible for inhibition of cell proliferation. These results propose the possibility of the improved therapeutic strategy making ALA one of the effective drugs used in human cancers.

Anti-proliferative Effects of Traditional Korean Doenjang across Different Aging Periods on Cancer Cell Lines (숙성기간으로 구분된 전통된장의 암세포 증식억제 효과)

  • Doenjang is a major fermented soy-based food in Korea. Recent investigations have shown that fermented soybean foods have immunity-enhancing, anti-cancer, anti-obesity and anti-diabetic effects. Several studies also have reported that genistein and daidzein, which are easily absorbed in the body are produced in larger quantities in aged doenjang. The purpose of this study was to evaluate the variations in the anti-cancer effects of commercialized doenjang as it ages. Four groups were formed for this study according to aging periods of doenjang, namely short (under 5 years, S group), mid (under 10 years, M group), long (under 15 years, L group) and very long (over 15 years, E group). The anti-cancer effects of doenjang were determined by cell cytotoxicity assays in A549, YAC-1, and HepG2 cancer cell lines. Also, NK cell activity and splenocyte proliferation were assayed for cancer immunotherapy. The quantities of phenolic compounds in doenjang at different ages were also measured. The results showed that the anti-cancer effects increased in the S and M groups for all three cancer cell lines. Interestingly, similar to this result, splenocyte proliferation and NK activity were also the highest in the S and M groups. In contrast, the E group showed significantly reduced splenocyte proliferation. The quantity of phenolic compounds was similar to that of the anti-cancer results. Collectively, these results suggest that the fermentation period of doenjang plays a very important role in determining its anti-cancer effects.

Antiproliferative Effects of Caesalpinia sappan Extract on Human Epithelial Cell Line HaCaT and Cancer Cell Lines (인체 상피세포주와 암세포주에 대한 소목 추출물의 증식억제 효과)

  • Caesalpinia sappan L. (Leguminosae) is an oriental medicinal herb distributed in China and Taiwan, and its heartwood has been traditionally used as an analgesic, a therapy for thrombosis or tumor. This study was to investigate the proliferation and inhibition effects of Caesalpinia sappan extracts against human epithelial cell and cancer cell lines. The methanol extract of dried C. sappan heartwood was evaporated (KS-6), and then sequentially extracted by hexane (KS-01), chloroform (KS-02), ethyl acetate (KS-03), n-butanol (KS-04), and water (KS-05). After 48 hr of exposure, these fractions at a concentration of $50{\mu}g/ml$ significantly increased, and reduced cell proliferation in both human normal epithelial and cancer cell lines. The ethyl acetate fraction (KS-03) among organic solvent fractions was 120.2% of the most proliferation activity ($50{\mu}g/ml$) against HaCaT human epithelial cell. However, fractions from chloroform, butanolic and methanolic extract had 7.2, 28.7 and 20.8% of antiproliferative effect on HaCaT cell, respectively. In cell proliferation effects of C. sappan extract on HeLa, SiHa and C33A human cervical cancer cells, chloroform fraction (KS-2) was the most antiproliferative activity, its antiproliferative rate (dosage, $50{\mu}g/ml$) relative to control was 25.8, 12.2 and 17.4% for SiHa, HeLa and C33A, respectively. The results indicated that the six extract fractions could induce cell cycle stimulate or arrest in some way. Finally, further investigation is needed to assess the molecular mechanisms mediated anticancer activities of this plant.

Gamma Irradiation Induces a Caspase-dependent Apoptotic Mechanism in Human Prostate Cancer PC-3 Cells (인간 남성호르몬 비의존형 전립선 PC-3 암세포에서 감마선의 Caspase-의존성 세포자멸사 유도 효과)

  • Prostate cancer is the most predominant cancer in men and related death rate increases every year. Till date, there is no effective therapy for androgen independent prostate cancer. To investigate the mechanism for cell growth inhibition or apoptosis in human androgen independent prostate cancer PC-3 cells after gamma irradiation. The aim of this study was to examine the potential of gamma irradiation to induce apoptosis in PC-3 cells and to assess the mechanism of gamma irradiation-induced apoptosis. Five different assays were employed in this study: cell proliferation assay, morphological assessments of apoptotic cells, DNA fragmentation analysis, quantification of apoptosis by annexin V (AV) and propidium iodide (PI) staning, and western blot analysis. Cell viability was inversely related to radiation dose. DAPI-positive cells were detected 48 hr after 40 Gy radiation exposure. And nuclear morphological changes of cells were observed by gamma irradiation. DNA ladder patterns in the cells exposed to gamma-radiation were appeared at 24 hr. Also, gamma irradiation induces apoptosis of PC-3 cells via Caspase3, Bax and PARP-dependent fashion.
