Effects of Flower of Pueraria lobata on Lipid Peroxidation and Activities of Alcohol Metabolic Enzymes in Alcohol-treated Rats (갈화가 에탄올을 투여한 흰쥐의 지질과산화와 알코올 대사효소의 활성도에 미치는 영향)

  • This study was designed to investigate the effect of flower of Pueraria lobata on liped peroxidation and activities of alcohol metabolic enzymes in alcohol-treated rats. Male Spra gue-Dawley rats were given 25% ethanol (Alcohol), 25% ethanol and 5 mg tectorigenin/kg B.W.(Alc.-Tec), 25% ethanol and 5mg kaikasaponin III/kg B.W. (Alc-Kai). The contents of serum total lipid, triglyceride and phospholipid were increased by ethanol treatment and were lower in the Alc.-Tec and Alc.-Kai group than in the Alcohol group. Decreased serum HDL-cholesterol by alcohol treatment was recovered by tectorigenin and kaikasaponin III. Microsomal cytochrome P-450, aniline hydroxylase and aminopyrine N-demethylase activities were increased by ethanol and were lower in the Alc. Tec and Alc.-Kai group than in the Alcohol group. Activity of hepatic alcohol dehydrogenase was increased by ethanol and was higher in the Alc.-Tec and Alc.-Kai group than in the Alcohol group. Microsomal ethanol oxidizing system activity was higher in Alc.-Tec group than in the other group. No significant difference was found in catalase activity among treatment groups. These data indicate that tectorigenin and kaikasaponin III were effected alcohol metabolic enzyme system and the liver damage associated with chronic ethanol consumption.

  • Cellulosic alcohol fermentation has recently gained more attention in the production of ethanol, butanol, and 2,3-butanediol. However, it was revealed that the process had several hurdles, such as, an expensive cost for biomass decomposition to yield fermentable sugars and a production of byproduct lignin. As an alternative for the process through biomass saccharification, the alcohol production through syngas from biomass has been studied. In this study, we reviewed acetogen and its central metabolic pathway, Wood-Ljungdahl route, capable of utilizing syngas. Furthermore, the metabolic engineering strategies of acetogen for bio-alcohol production from syngas was also reviewed with a brief perspective.

Prevalence of the Nonalcoholic Fatty Liver Disease in Obese Children (소아 비만증에서 비알코올성 지방간염의 유병률)

  • Purpose : Obesity is, along with metabolic syndrome, closely related with nonalcoholic fatty liver disease. This study tried to evaluate the prevalence of nonalcoholic liver disease in obese children and verify the factors associated with the disease. Methods : Two hundred and seventy nine children who showed a body mass index of 95 percentile over the baseline in health examinations of surrounding schools were evaluated. Questionnaires, body measurements, blood examinations, and ultrasonographic measurements of abdominal fat were examined. Results : Out of 279 children enrolled for the study, 27 children were found to possess nonalcoholic liver disease(9.7%). Among those found to be positive for nonalcoholic liver disease, it's prevalence increased to 15.2%(22 out of 144 children) among children with severe obesity. Factors known to be involved with metabolic syndrome, namely waist/hip circumference ratio and thickness of abdominal fat, were found to be closely related to nonalcoholic fatty liver as well. Conclusion : The prevalence of nonalcoholic fatty liver in obese children was 9.7%, with higher incidence observable in severer obesity. Factors responsible for metabolic syndrome were closely associated with nonalcoholic fatty liver disease, and the level of insulin resistance, which is an useful index in both diseases, can be utilized in evaluation of the effect of treatment and control of risk factors.

Activities of Antioxidation and Alcohol Dehydrogenase Inhibition of Methanol Extracts from Some Medicinal Herbs (약용식물 추출물에 대한 항산화성과 알코올 탈수소효소 저해성 연구)

  • The activities of antioxidation and alcohol dehydrogenase in hibitionin methanol extracts of thirty two medical herbs were tested using the method of DPPH activity, nitrite scavenging effect and alcohol dehydrogenase assay in vitro. In DPPH method, Eugenia caryophyllata, Thea sinensis, Paeonia suffruticosa, Alnus japonica showed over 90 % of free radical scavenging activities. The nitrite scavenging ability appeared Zanthoxylum bungeanum, Alnus japonica, Thea sinensis, Hovenia dulcis(cortex) and Illicium verum showed the high value. In connection with in vivo alcohol metabolism, thirteen medicinal herbs were screened for inhibition. As a reasult, we found significant inhibition of ADH by methanolic extracts of Glycyrrhiza uralensis, Pueraria thunbergiana(radix), Alnus japonica. These results indicate that the antioxidative effect was strongly related with alcohol dehydrogenase inhibitor; Thea sinensis and Alnus japonica.

Effects of Cudrania Tricuspidata Root Extract (CTE) on Ethanol-Induced Hangover via Modulating Alcohol Metabolizing Enzyme Activities and Blood Gas Levels in Rats (꾸지뽕나무 뿌리 추출물의 알코올 대사 효소 활성 및 혈액 산성화 기전 조절을 통한 숙취해소 효과)

  • To investigate the anti-hangover effects of Cudrania tricuspidata root extract (CTE), the blood alcohol metabolism and blood gas imbalance of CTE in rats treated with 10 ml/kg alcohol were examined. CTE (500 mg/kg and 750 mg/kg) was administrated after 30 minutes of alcohol consumption (10 ml/kg). Blood collection was implemented from the tails of the animals after 1, 3, and 5 hours post alcohol consumption. The Condition drink (a commercial anti-hangover beverage) was used as a positive control. Single administration by the oral route was performed. The consumption of CTE (500 mg/kg and 750 mg/kg) decreased the serum alcohol concentration by increasing and maintaining both the alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) enzyme activity levels in the blood and liver. In addition, CTE led to recovery from the imbalances in the blood gas levels, including carbon dioxide ($CO_2$) and changes in pH, bicarbonate ($HCO_3{^-}$) and lactic acid levels due to alcohol ingestion. In conclusion, CTE exerted a more pronounced anti-hangover effect than a commercial anti-hangover drink. Therefore, CTE can be a novel and safe anti-hangover natural product agent for the prevention or treatment of symptoms caused by excessive alcohol consumption.

Effect of Monascus Pigment Extract on the Alcohol Metabolism in Rats (흰쥐에 있어서 홍국 색소 추출물이 알코올대사에 미치는 영향)

  • To investigate the alcohol metabolizing system in liver of rats drunken 10% ethanol with Monascus pigment extract (MPE), Sprague-Dawley male rats weighing about 250 g have been drunken 10% ethanol containing 1, 2.5 and 5% Monascus pigment extract for a month. Three groups of rats drunken 10% ethanol with MPE gained somewhat less body weight than normal group, but the changes of body weight was not significantly different among the former groups. All groups drunken MPE supplemented alcohol had no remarkable changes in liver function on the basis of liver weight/body weight, the serum levels of alanine aminotransferase and xanthine oxidase activity. 10% alcohol drunken animals (control group) showed significantly increased activity of hepatic alcohol dehydrogenase (ADH) by 87% compared with normal group and the animals drunken 1%, 2.5% and 5% MPE showed respectively 34%, 29% and 21% increased activity of hepatic ADH, whileas Km value of ADH in 1, 2.5 and 5% MPE group decreased by 40%, 30% and 19% respectively compared with the control, but Vmax showed no significant changes among MPE groups. In case of aldehyde dehydrogenase (ALDH), 1% MPE group showed significantly increased activity by 32% and 2.5% or 5% MPE group showed increasing tendency compared with control, and Km value in three experimental groups declined by 27% and no particular changes were found among those. Furthermore, Vmax value in 1, 2.5 and 5% MEP group increased by 88,56 and 22% respectively with the control. In the aspect of the area under the curve of a ethanol concentration versus time (AUC) profile obtained after administration of 10% alcohol with 1 or 5% MPE, the decreasing rate of AUC to the control was 18% in 1% MPE treated rats whereas 10% in 5% MPE group.
