Effect of Ethanol on Prostaglandins Production of Monocytes (에탄올이 단핵구의 Prostaglandins 생산에 미치는 영향)

  • The increase in alcohol consumption level has been noticed in Korea recently. Alcohol appreciably inhibits cell mediated immunity and this may contribute to the high prevalence of serious infection such as pulmonary tuberculosis among alcoholic subjects. The present study was undertaken to examine the effect of ethanol on the cyclooxygenase metabolites of human monocyte in vitro. Monocytes were activated with 800 units of gamma interferon(IFN-${\gamma}$) for 3 days following apply of Ficool-hypaque density gradient and gelatin coated flasks for separation of monocytes. Ethanol with addition of 100mM, 300mM and 600 mM for 30 minutes to 106 monocytes with/without previous IFN-${\gamma}$ treatment caused a dose dependent decrease in the production of thromboxane B2, 6-keto-PGE1$\alpha$ and PGE2 by radioimmunoassay at 6 hours after ethanol treatment. Quite different from the findings after 6 hours there was dose dependent increase in three prostaglandins without IFN-${\gamma}$ treatment after 24 hours of incubation. With previous treatment of IFN-${\gamma}$ reduced productions of three prostaglandins at 24 hours than control is spite of ethanol stimjulation. These findings show that IFN-${\gamma}$ can inhibit alcohol induced derangement of arachidonic acid metabolism of monocytes.

  • We provide the reader with a brief introduction to the neurobiology of neuropeptides. Several comprehensive reviews of the distribution and neurochemical, neurophysiological, neuropharmacological and behavioral effects of the major neuropeptides have recently appeared. In reviews of the large number of neuropeptides in brain and their occurance in brain regions thought to be involved in the pathogenesis of major psychiatric disorders, investigators have sought to determine whether alternations in neuropeptide systems are associated with schizophrenia, mood disorders, anxiety disorders, alcoholism and neurodegenerative disease. There is no longer any doubt that neuropeptide-containing neurons are altered in several neuropsychiatric disorders. One of the factors that has hindered neuropeptide research to a considerable extent is the lack of pharmacological agents that specifically alter the synaptic availability of neuropeptides. With the exception of naloxone and naltrexone, the opiate-receptor antagonists, there are few available neuropeptide- receptor antagonists. Two independent classes of neuropeptide-receptor antagonists has been expected to be clinically useful. Naltrexone, a potent ${\mu}$-receptor antagonist, has been used successfully to reduce the need for alcohol consumption. And cholecycstokinin antagonists are now in development as a new class of anxiolytics, which would be expected to be free from tolerance and physical dependence and lack of sedation. In this review, we deal with these two kinds of neuropeptide system, the opioid system and cholesystokinins in the brain. The role of opioid systems in the reinforcement after alcohol consumtion and that of cholesystokinins in the pathogenesis of anxiety will be discussed briefly. As we know, the future for neuropeptides in psychiatry remains bright indeed.

Production of cheese containing alcohol metabolism using Lactobacillus with fig enzyme (무화과(Fig) 효소를 첨가한 유산균을 이용하여 알코올 대사활성 함유 치즈의 제조)

  • In this study, we evaluated the alcohol degradation ability of fig enzyme in the production of cheese using Lactobacillus kitasatonis, Lactobacillus amylophillus, and Leuconostoc mesenteroides sub. The strains were highly resistant to ethanol, acid, and bile acid. When 10% of fig enzyme was added, the alcohol dehydrogenase and aldehyde dehydrogenase activities in each strain were approximately 170, 270, and 190% higher, respectively, than in samples without fig enzyme. The addition of 10% of fig enzyme to produce cheese with the L. amylophillus strain showed an approximately 250% increase in alcohol dehydrogenase and aldehyde dehydrogenase degradation. In conclusion, when fig enzyme was added to produce cheese using L. amylophillus, high alcohol degradation ability was observed. The applicability of fig enzyme addition was confirmed for the production of functional food.

Effect of Tenebrio molitor Larvae Fermented with Saccharomyces cerevisiae M1 (KACC 93023) on Non-alcoholic Fatty Liver-induced Rats (Saccharomyces cerevisiae M1 (KACC 93023)으로 발효한 갈색 거저리 유충이 비 알코올성 지방간 유발 흰 쥐에 미치는 영향)

  • In this study, Tenebrio molitor (brown mealworm) larvae powder was fermented with Saccharomyces cerevisiae M1 (KACC 93023). The fermented and non-fermented powders were subsequently administered to rats with induced non-alcoholic fatty liver disease to compare the degree of improvement. The rats exhibited abnormal lipid metabolism due to orotic acid, and the group that were fed the fermented larvae powder showed similar weight to normal rats. AST, ALT, ALP, and LDH activities, all known indicators of liver health, and various other lipid-related indicators appeared positive, suggesting improvement. The lipid peroxide and glutathione content in the liver tissue and serum were similar to or better than those of the normal group. Finally, morphological observations of liver tissue using H&E and oil red O stainings revealed that the fermented powder group showed markedly reduced fat along with normal hepatocyte shape and arrangement. To summarize, it was confirmed that dietary brown mealworm larvae improve non-alcoholic fatty liver function, and, when fermented with yeast, most indicators showed positive improvements to almost normal levels. As such, the fermented powder can be considered to improve non-alcoholic fatty liver, suggesting that it may be used as a material for various products in the future.

  • The variations of asparagine and aspartic acid contents among the soybean varieties and in the different parts of soybean sprout after the different durations of storage were investigated. Asparagine and aspartic acid are known to have the detoxifying effects on acetaldehyde, which is highly toxic metabolites in the process of alcohol metabolism in the human body. The contents of asparagine and aspartic acid of beansprout showed continuosuly increasing trends along with the days to cultivation with a great varietal difference, especially in the roots. The duration of seed storage did not affect contents of asparagine content; the longer storage decreased aspartic acid content remarkably.

The Hepatoprotective Effects of Hep G2 Cells and the Alcohol-Metabolizing Enzyme Activities of Lemon-Myrtle (Backhousia citriodora) Leaf Extracts (레몬 머틀 잎 추출물의 Hep G2 세포에서의 간 보호 효과 및 알코올대사 효소활성)

  • Lemon myrtle (Backhousia citriodora), a plant in the Myrtaceae family, is native to the semitropical rain-forests of Queensland and is presumably the most commercialized native spice. In Australian thousands of lemon-myrtle trees are under tillage. This study was carried out to investigate the alcohol metabolism, hepatoprotective effects and antidiabetic, tyrosinase inhibitory activity of hot-water (LMW) and 80% ethanol (LME) extracts from lemon-myrtle leaves. The alpha-glucosidase (${\alpha}$-glucosidase) inhibitory activities of the LMW and LME extracts were 7.66% and 40.29% at 1 mg/ml (p<0.05), respectively. The tyrosinase inhibitory activity of the LME extract was about 38.26 % at 1 mg/ml. The effects the LMW and LME extracts had on alcohol-metabolizing activities were determined by measuring the generation of reduced nicotinamide-adenine dinucleotide (NADH) by acetaldehyde dehydrogenase (ALDH) and alcohol dehydrogenase (ADH). The ADH activities of the LMW and LME extracts significantly increased in a dose-dependent manner and were about 154.40% and 192.03% at 1 mg/ml, respectively (p<0.05). The ALDH activities of the LMW and LME extracts also significantly increased in a dose-dependent manner and were about 151.14% and 192.34% at 1 mg/ml, respectively (p<0.05). At $100{\mu}g/ml$, the LMW and LME extracts showed significant protective effects against tacrine-induced cytotoxicity in Hep G2 cells. The results suggested that Backhousia citriodora leaf extracts have the potential to be significant sources for natural health products.

Cudrania tricuspidata Fruit Extract Ameliorates Free Fatty Acid-induced Lipid Accumulation in HepG2 Cells (유리지방산으로 지방축적을 유도한 HepG2 cells 대한 꾸지뽕 열매 추출물의 개선 효과)

  • Non-alcoholic fatty liver disease (NAFLD) is a chronic liver disease associated with various metabolic syndromes, such as obesity, dyslipidemia, hypertension, and diabetes. Cudrania tricuspidata is a medicinal plant distributed widely in Asia and has been used in clinical practice to treat various diseases. The aim of this study is to determine the lipid-lowering effects of C. tricuspidata fruit extract (CTE) using a cell model induced by free fatty acids (FFAs). HepG2 cells were exposed to 1mM FFAs (palmitic acid:oleic acid = 2:1) for 24 hr to simulate the conditions of NAFLD in vitro. CTE attenuated the increases of lipid accumulation, intracellular triglyceride, and cholesterol content and inhibited 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMG-CoA reductase) activity in the HepG2 cells in a dose-dependent manner. Also, CTE inhibited the protein expression of lipogenesis-related genes, such as sterol regulatory element-binding protein-1/-2 (SREBP-1/-2), fatty acid synthase (FAS), and stearoyl CoA desaturase-1 (SCD-1) in FFAs-induced lipid accumulation in HepG2 cells. In addition, CTE-induced adenosine monophosphate-activated protein kinase (AMPK) phosphorylation in HepG2 cells. These results suggest that CTE attenuates hepatic lipid accumulation by inhibiting lipogenesis through the modulation of the AMPK signaling pathway on FFAs-induced lipogenesis in HepG2 cells and may potentially prevent NAFLD.

A Case Report of Nonalcoholic Steatohepatitis with Metabolic Syndrome (대사증후군을 동반한 비알코올성 지방간염 치험 1례)

  • Objective: The purpose of this case report is to describe the efficacy of treatment with Korean medicine of a patient with nonalcoholic steatohepatitis (NASH) and metabolic syndrome. Methods: The patient was treated with Korean medicine (Saenggangunbi-tang) from April 27, 2016 to October 22, 2016. Results: Post-treatment, the patient's AST and ALT levels were significantly decreased. Triglyceride levels were within the normal range during the treatment. Abdominal ultrasonography revealed a significant improvement in hepatic steatosis. Conclusions: The results suggest that Saenggangunbi-tang could be an effective treatment for NASH with metabolic syndrome.

The Effect of Estrogen Deficiency on Ethanol Metabolism in Ovariectomized Rats (난소절제 랫트에서 에스트로겐 저하가 알코올 대사에 미치는 영향)

  • This study was designed to examine the effect of estrogen deficiency on the metabolism of ethanol in ovariectomized rats. Female rats were assigned to an ovariectomy (OVX) and a sham (SHAM) surgery group. Gain body weight was greater in incresed in OVX group and especially uterus weight significantly decrease depending on the concentration of estrogen after 3 month of ovariectomy. Ethanol at the tolerative dose (6 g/kg) was injected to rats by oral administration to measure the concentration of ethanol in blood. The area under the blood concentration time curve (AUC) was significantly lower in OVX group than SHAM group. The significant decrease in AUC in OVX group indicates that the estrogen deficiency leads to changes of the factors related to ethanol metabolism. Activity of hepatic alcohol dehydrogenase was not significantly influenced by the ovariectomy and also the ethanol elimination rate in vivo was not different. Cytochrome P450 isozymes did not show any changes except CYP 1A1 and 2E1. Level of hepatic glutathione in OVX group was higher after treatment of ethanol. Therefore the reduction of AUC appears not to be directly associated with the difference of ethanol metabolizing enzyme, but to be related with the physical factors like body weight.

Effect of Ganoderma Lucidum on the Liver Function and Lipid Metabolism in Alcohol-Consuming Rats (영지가 알코올 섭취한 흰쥐의 간기능 및 지질대사에 미치는 영향)

  • The effects of Ganoderma Lucidum consumption (GL) on the liver function and lipid metabolic disorder induced by alcohol consuming were studied in rats using GL powder(1%, 2%, 4%) and 10% ethly alcohol. Thirty-five rats(Sprague-Dawley, male) were divided into five groups and fed experimental diets for seven weeks. The concentrations of serum cholesterol, HDL-cholesterol and triglyceride were the highest in the alcohol group, while the levels were apparently reduced by in groups fed GL. The atherogenic indices(AI) also tended to be higher in the alcohol group. Glutamic oxaloacetic transaminase(GOT) activities were significantly lower in body alcohol + 2% GL and alcohol + 4% GL groups, and glutamic pyruvic transaminase(GPT)activities lower only in the alcohol + 2% GL compared with the alcohol group. Alkaline phosphatase(ALP) activities were not significantly different among the groups. The elevated levels of liver cholesterol and triglyceride due to alcohol consumption, were apparently reduced by GL supplementation. On microscopic observation of liver tissues, fat droplets appeared extensively on the liver-lobule in the alcohol group, while fat droplets appeared only on the central vein in the alcohol + 1% GL group. However, in groups administered 2% and 4% GL, fat droplets appeared similar to the alcohol group. The concentrations of fecal bile acid were significantly increased in groups fed GL. These results indicate that GL exerted some beneficial effects on lipid metabolic disorder caused by long-term alcohol consumption. Thus, it seems that GL may be effective in preventing or curing some aspects of alcohol toxicity. More detailed studies are needed in order to determine proper dietary levels of GL for combating alcohol toxicity.
