Studies on the Preparation of Radioactive iodine Labelled Concanavalin-A, Lectin Extracted from Korean Native Plant “Banha”, and Their Conjugation Products and the Hemagglutination Tests of These Labelled Compounds in Vitro.

  • Concanavalin-A, lectin extracted from Korean native plant “Banha”(Pine-llia Termata) and their conjugation products with tyrosine and 5-iodo-6-aminouracil were labelled by radioactive iodiae-125, and these labelled compounds were subjected to a hemagglutination test in vitro. The labelling procedures were presented with respect to labelling yield and the trends on the conjugation reaction of lectin with various amino acids were discussed. The preliminary results on the hemagglutination tests against cancer tissues were presented and the potential clinical applications of these labelled compounds were discussed.

Analysis of Nutritional Composition and Phenolic Compound in Propolis Collected from Falseacacia and Chestnut Tree in Korea (국내산 아까시나무와 밤나무 유래 propolis의 영양성분 및 페놀성 화합물 분석)

  • Nutritional composition and phenolic compounds of raw propolis collected from falseacacia (Robinia pseudoacacia L.) and chestnut tree (Castanea crenata), and their 70% ethanol extracts of propolis (EEP) were analyzed. Propolis had high crude lipid content, but no significant differences in general compositions in terms of collection area and plant origins. Mineral contents varied greatly depending on the plant origins, with falseacacia propolis showing the highest mineral content. Sixteen amino acids were analyzed, among which aspartic acid content was the highest at $328.4{\sim}410.6\;mg%$ and methionine the lowest at $0{\sim}21.1\;mg%$. Extraction yield for EEP was relatively high at $64.2{\sim}81.9%$, and total polyphenol and flavonoid contents were $13.9{\sim}23.7$ and $8.6{\sim}10.8%$, respectively. HPTLC and HPLC analysis on the phenolic compounds revealed the overall chromatographic patterns were almost equal, showing similar polyphenol compositions between the propolis. About 16 peaks were identified by HPLC analysis, among which 6 peaks of p-hydroxy benzoic acid, caffeic acid, ferulic acid, benzoic acid, cinnamic acid, and chrysin were identified.

Effect of Various Fruit-loads on Nitrogen Partitioning, Accumulation, and Remobilization in Young Trees of 'Fuyu' Persimmon (착과 정도가'부유'단감 유목의 질소화합물 분배와 축적 및 재이용에 미치는 영향)

  • This study was conducted to determine the distribution of nitrogenous compounds to various tree parts and the extent of reserve accumulation in persimmon (Diospyros kaki) under various fruit-loads. This study also ascertained the proportion of storage nitrogen made available for the new growth the following year. On June 15, the fruit-load was adjusted to a leaf-fruit (L/F) ratio of 10, 20, and 30, and some trees were completely defruited. Between June 15 and November 11, the increase of total amino acids were greater with a high L/F ratio. The amino acids increased in the root were negligible at the 10-L/F ratio. Of the total amino acids increased during this period, the proportion distributed to the root was 64% in the 20-L/F, 18.5% in the 30-L/F, and 81% in the defruited trees, and the distribution to the fruits was 81% in 10-L/F, 12% in 20-L/F, and 35% in the 30-L/F trees. Leaf amino acids decreased in the 10-L/F trees. Total proteins increased in autumn were greater as the L/F ratio was higher. Total proteins were in the fruits the most, and the distribution to the permanent parts was decreased as the L/F ratio was decreased. At the L/F ratio of 30, 59% of the total proteins increased in the autumn was distributed to the fruits and 40% to the root. Leaf proteins decreased at 10 and 20 L/F ratios. During the new growth from April 10 to June 10 the following year, amino acids decreased in the old wood and 1-yr-old shoot, whereas proteins decreased only in the 1-year-old shoots. Amino acids and protein decreased by 540 mg and 610 mg, respectively, in the roots of the defruited trees. Total amino acid and proteins in the newly-grown parts were the most at 730 mg and 1290 mg, respectively, when defruited the previous year. They were the least at the 10-L/F ratio, being 120 mg and 400 mg, respectively.

  • Pyrazoloazines are extremely useful in agriculture and medicine. The main objective of this article is to synthesize some new pyrazoloazines. Ethyl 3-amino-1H-pyrazole-4-carboxylate undergoes diazotization, couples with activated methylene compounds and cyclizes to form pyrazolo[5,1-c][1,2,4]tri-azine derivatives. The title compound also reacts with $\alpha$-substituted cinnamonitriles to produce pyrazolo[1,5-a]pyrimidine derivatives. Structures of newly synthesized compounds are established via chemical and spectral methods.

Compositions of Opuntia ficus-indica (손바닥 선인장의 성분 특성)

  • Attempts were made to determine the compositions of Opuntia ficus-indica var. saboten for the utilization as food materials. The major components of Opuntia ficus-indica and aloe in proximate compositions were nitrogen free extract. Total mineral contents of stem, fruit and seed of Opuntia ficus-indica were 9400.8, 6151.2 and 1096.8 mg%, respectively, and their major minerals were Ca, P and Mg. The major free amino acids of fruit were tyrosine, proline and arginine, those of stem were glycine and arginine and that of seed was glutamic acid. The major amino acid of fruit was glutamic acid, those of stem were glycine and arginine and those of seed were glutmic acid and arginine. Vitamin C contents of fruit and stem were 163.8 and 71.2 mg% but not presented in seed. Vitamin A was also not presented in fruit, stem and seed. Contents of total polyphenols and flavonoids were changed by extraction solvent and temperature. Total polyphenols of fruit were higher than those of stem and seed. Total flavonoids of fruit were similar to those of stem.

Stem Girding Increases Seed Production and Nitrogenous Compounds in Larix leptolepis (환상박피 처리에 의한 일본잎갈나무의 착과유도 효과와 질소 화합물 함량의 증가)

  • Japanese larch (Larix leptolepis) has been extensively planted in Korea as a reforestation species but their supply has been a major bottleneck due to sporadic natural seed production. In this study, stem girdling was applied to 32-year-old grafted Japanese larches, resulting in significantly enhanced seed production compared to the controls. Stem girdling induced about 4 times higher cone production than that of controls in the Japanese larch seed orchard. Time-dependent metabolic alterations after girdling were investigated by stable isotope ratio mass spectrometer, HPLC, and GC-MS analysis. In girdled trees, the contents of total nitrogen, sucrose, and total free amino acids were significantly higher than the non-girdled trees at the flowering differentiation season (from July to August). Moreover, the numbers of female strobilus per tree were positively correlated with the contents of both total nitrogen (r=0.765, p<0.01) and total amino acids (r=0.802, p<0.01) in the bark being collected at the flowering differentiation time (August 20). Interestingly, the levels of various individual amino acids at the flowering differentiation times, such as aspartic acid, glutamic acid, glycine, serine, and cysteine, were also significantly correlated (p<0.05) with the numbers of strobilus, suggesting that those amino acids might be involved in the induction of female strobilus formation of Japanese larches.

Analysis of the General Components of Syneilesis palmata Maxim (우산나물(Syneilesis palmata)의 성분 분석)

  • We measured the levels of soluble protein, sugar, free amino acids, minerals, total polyphenols, and flavonoid compounds in Syneilesis palmata as part of a study on nutritional and functional materials for development of valuable foods. The content of soluble protein in the aerial and root portions were 210.36 mg% and 870.42 mg%, respectively. The amount of reducing sugar was 848.12 mg% (aerial parts) and 1,420.91 mg% (roots), and that of free sugar was 14.85 mg% in aerial parts and 355.00 mg% in roots. The free amino acid level in aerial parts was 1,613.10 mg% and that in roots was 3,282.96 mg%. The mineral content of aerial parts was 3,531.53 mg% and that of roots was 1,878.34 mg%. The K, Ca, and Mg levels were greater than those of other minerals, with K comprising more than 75% of the total mineral content. The levels of polyphenol and flavonoid compounds in aerial parts were 1,920.00 mg% and 843.95 mg%, respectively, and those in roots were 487.56 mg% and 91.07 mg%, respectively.

Analysis on the Components of the Vitex rotundifolia Fruit and Stem (순비기나무(Vitex rotundifolia) 열매와 줄기의 함유성분 분석)

  • This study was conducted to analyze components of the fruit and stem of Vitex rotundifolia as part of a study on the nutritional and functional materials, and the development of a functional food. General components of V. rotundifolia fruit are moisture 12.92%, carbohydrate 78.67%, crude protein 3.22%, crude fat 1.73% and 3.46% crude ash. V. rotundifolia stem was moisture 11.30%, carbohydrate 80.87%, crude protein 4.78%, crude fat 0.64 % and 2.41% crude ash. The content of V. rotundifolia reducing sugar was 646.07 mg% (fruit) and 1,547.97 mg% (stem). The total amount of free sugar was fruit 5.66 mg% and stem 90.79 mg%. The content of soluble protein was 3,268.12 mg% in fruit and 4,927.55 mg% in stem. The polyphenol compound content was 608.06 mg% and 808.06 mg%, respectively. Total amount of hydrolyzed amino acid of V. rotundifolia fruit and stem were 3,095.75 mg% and 2,135.84 mg%, while that of free amino acid of fruit and stem were 79.99 mg%, 81.20 mg%, among which cysteine (2,010.82 mg%) was the highest in the V. rotundifolia fruit. In the results of mineral analysis, the content of K was the highest in fruit (2,184.00 mg%) and stem (1,469.20 mg%).

  • 근년 들어 수요가 급증하고 있는 수산물 액젓류는 다량의 식염 (20% 이상)을 함유하고 있으므로 저장성이 비교적 양호하고 부패되기 어려운 식품이나 시중에 유통되고 있는 일부 시판 액젓류는 비교적 단기간에 갈변 또는 흑변이 일어나고 있다. 이러한 현상은 액젓 내에 함유되어 있는 성분 중 아미노기를 가지는 물질 즉, 암모니아, 아민, 아미노산, 펩티드 및 단백질과 지방산화에 의해서 생성된 카보닐 화합물 또는 Bacillus속과 같은 내열성 세균과 곰팡이 등이 잔존하여 이 같은 현상을 일으키는 것으로 알려져 있다 (Chung and Toyomizy, 1976; Oh, 1996).   • 효소는 일반적으로 건전한 생체에서는 원형질에서 충분히 생성되어진다. 또한 미생물의 배양조건을 달리하던가 씨앗의 발아 시기와 같이 생체가 특수한 상황일 때에는 급격히 증가하기도 한다. 이러한 효소는 주로 단백질로 구성되어 있으며, 그 대부분의 아미노산이 peptide 결합을 하고 있는 고분자 화합물로서 효소의 종류에 따라 특정한 반응에만 특이적으로 작용하는 기질특이성을 가지고 있다. 효소를 이용한 건강식품으로는 체내 과산화지질의 분해를 촉진하는 SOD(superoxide dismutase), 활성 산소나 과산화수소 등의 체외 배출을 돕는 글루타치온 생성효소를 이용한 제품 외에 야채의 미생물 발효를 통하여 미생물 자체나 효소를 이용하는 과채발효 음료 등이 있지만, 여기에서는 특별히 미생물 유래의 효소를 이용한 분말형 효소 제품을 소개하고자 한다.
