유산균 발효에 의한 십전대보탕의 발효 전 후 성분 변화 연구 (Analysis of Constituents in Sipjundaebo-tangs Fermented by Lactic Acid Bacteria)

  • 십전대보탕의 발효 전 후 성분 변화를 분석하기 위해 8종의 유산균을 이용하여 발효한 십전대보탕과 발효하지 않은 십전대보탕을 비교 분석하여 발효 후 증가한 3종의 성분과 감소한 2종의 성분을 규명하여 각 유산 균주에 대하여 5종 성분의 정량 분석을 실시하였다. 증가한 3종의 성분은 각종 크로마토그래픽 기술을 이용하여 분리 정제 후 NMR, MS 등의 분광학적 분석기법을 이용하여 구조를 규명하였으며, cinnamyl alcohol (1), liquiritigenin (3) 및 nodakenetin (5)으로 규명되었다. 발효 후 감소한 성분은 표준품과 HPLC 비교분석(retention time, DAD spectrum)을 통하여 liquiritin (2) 및 nodakenin (4)으로 구조를 규명하였다. 8종의 유산 균주로 발효한 십전대보탕의 성분 변화를 분석한 결과 L. plantarum KFRI 144 발효물이 다른 유산균 발효물에 비하여 cinnamyl alcohol (1), liquiritigenin (3) 및 nodakenetin (5) 성분이 대체적으로 가장 높게 증가하였으며, liquiritin (2) 및 nodakenin (4) 성분이 가장 큰 비율로 감소하여 8종의 유산 균주 중 발효 효과가 가장 우수하였다.

창출 성분의 장관면역 자극을 통한 골수세포 증식활성 (Bone Marrow Cell Proliferation Activity through Intestinal Immune System by the Components of Atractylodes lancea DC.)

  • 십전대보탕의 10가지 구성 천연물을 열수추출하여 장관면역 활성을 비교 검토한 결과, 창출(Atractylodes lancea DC., ALR)과 인삼(Panax ginseng C.A. Meyer, PG)에서는 높은 활성을 나타내었으나 약한 활성을 갖는 황기(Astragalus membranacues, ASR)와 당귀(Angelica aculiloba Kitagawa, AR)를 제외하고는 어떤 생약에서도 거의 활성을 갖지 않았다. 한편 이러한 활성을 가진 생약(ALR, PG와 ASR) 중에서도 특히 ALR만이 종류, 생산지 및 육종조건에 관계없이 항상 일정하게 높은 활성을 보여주어 창출의 열수추출물 획분(ALR-0)이 십전대보탕의 장관면역 활성에 중요하게 관여하는 시료로 선정되었다. ALR-0 획분은 다시 용매별로 분획되어 메탄올-가용성 획분(ALR-1), 메탄올-불용성/에탄올-가용성 획분(ALR-2) 및 조다당 획분(ALR-3)으로 조제되었으며 이들의 활성을 측정한 결과, 단지 ALR-3 획분만이 Peyer's patch를 통한 골수세포의 증식을 활성화시키는 장관면역 활성을 시료농도에 의존적으로 나타내었으며 그 외의 획분에는 활성이 거의 없었다. 한편 ALR-3 획분을 $NaIO_4,\;NaClO_2$ 및 pronase로 처리한 후 활성을 측정한 결과, 모든 처리군에서 활성이 감소되었으며 특히 periodate 산화에 의해 심한 활성감소를 보였다. 이러한 결과를 토대로 십전대보탕의 장관면역 활성은 10종의 생약 중에서도 특히 창출이 중요하게 활성에 관여하는 것으로 보이며 다양한 용매별 분획을 통해 저분자보다는 에탄올에 침전된 고분자가 주요 함유물질인 조다당 획분이 장관면역 활성에 관여하는 것으로 추정되었다. 또한 periodate 산화에 의해 활성이 크게 감소되는 것으로 보아 주로 다당류가 창출의 장관면역에 중요한 활성물질인 것으로 생각된다.

십전대보탕가미방(十全大補湯加味方)의 면역조절효과(免疫調節效果)에 관한 실험적(實驗的) 연구(硏究) (Immunomodulatory effects of Kamishipjundaebotang(KSDBT))

  • This study was done to evaluate the immunomodulatory effects of KSDBT. KSDBT significantly enhanced the lymphocyte proliferation through 3H thymidine uptake and also increased T and T helper cell. It upregulated IL-2, IL-12(p35, p40) and INF-${\gamma}$. NO and carbon clearance were significantly increased by KSDBT. These results indicate KSDBT exert antitumor activity by immunomodulation of cytokines.

십전대보탕(十全大補湯)과 가미십전대보탕(加味十全大補湯)이 뇌허혈 흰쥐의 뇌혈류역학에 미치는 실험적 영향 (Experimental Effects of Sibjeondaebo-tang and Gamy-Sibjeondaebo-tang on Cerebral Hemodynamics in Cerebral Ischemia Rats)

  • This Study was designed to investigate the effects of Sibjeondaebo-tang (SDT) and Gamy-Sibjeondaebo-tang (GST, Sibjeondaebo-tang adding Cervi Pantotrichum Cornu) on the improvement in regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats, and in the rats with cerebral ischemia induced by middle cerebral artery occlusion, and further to determine the mechanisms. And, It was to investigate the effects of the SDT and GST with the change of histologic examination through the BDNF in the hippocampus CA1. In changes of cerebral hemodynamics, SDT and GST significantly increased rCBF in a dose-dependent manner but decreased MABP in normal rats. In mechanism of cerebral hemodynamics, Increase of GST-induced rCBF was significantly inhibited by pretreatment with methylene blue (0.01 mg/kg, i.p.), an inhibitor of guanylate cyclase, and Decrease of GST-induced MABP was significantly increased by pretreatment with methylene. These results suggested that the action of GST was mediated by guantlate cyclase pathway. In cerebral ischemics, the rCBF was stably improved by SDT (10 mg/kg, i.p.) significantly and stably increased by GST (10 mg/kg, i.p.) during the period of cerebral reperfusion, which contrast with the findings of rapid and marked increase in Control group. These results suggested that GST had anti-ischemic action in cerebral ischemic state. In histological examination through TTC stain, Sample A group and Sample B group decreased discoloration in the cortical part at $28^{th}$ day after MCAO induction. In immunohistochemistric response of BDNF, Sample A group and Sample B group increased respondent effect at $28^{th}$ day after MCAO induction. These results suggest that GST can Increase rCBF in normal state, as well as improve the stability of rCBF in cerebral ischemic state. Furthermore, methylene blue inhibitor study suggested the mechanism of blood flow enhancement by GST may be mediated by guanylate cyclase pathway.

십전대보탕(十全大補湯) 와송(瓦松) 및 십전대보탕가와송(十全大補湯加瓦松)의 항암효과(抗癌效果)와 면역반응(免疫反應)에 관(關)한 연구(硏究)

  • In order to research the effects of Shipjundaebotang(SDT) and Orostachys Herba(OH) on anti-tumor and immune response, the author performed this experimental study. Experimental groups are divided into five groups, which are solid extract of Orostachys Herba by water(OHW), solid extract of Orostachys Herba by ethanol(OHE), solid extract of Shipjundaebotang (SDT), solid extract of Shipjundaebotang added by solid extract of Orostachys Herba by water(SDT+OHW) and solid extract of Shipjundaebotang added by solid extract of Orostachys Herba by ethanol (SDT+OHE). In these experimental studies, extension of survival days for anti-tumor effect was observed, and de layed type hypersensitivity and rosette forming cell for cell-mediated immune response, hemagglutinin titers and hemolysin titers for humeral immune response, spleenic natural killer cell activity and carbon clearance (K-index) in vitro were measured with mice. The result were summerized as follows: I. SDT, SDT+OHW and SDT+OHE treated groups were significantly recognized to extend the survival days of tumor bearing mice as compared with the control group. 2. Delayed type hypersensitivity was significantly increased in SDT, SDT+OHW and SDT+OHE treated groups as compared with control group. 3. Hemagglutinin titer was increasred in all sample groups as compared with control group, but not significantly. 4. Hemolysin titers was significantly increased in SDT, SDT+OHW and SDT+OHE treated groups as compared with control group, and SDT+OHE treated group showed the increasing effect with significance as compared with the other sample groups. 5. For the effect of roselle forming cell quantitation, SDT, SDT+OHW and SDT+OHE treated groups showed the increasing effect with significance as compared with control group. 6. Natural killer cell activity was significantly increased in SDT+OHW as compared with comrol group, but the other groups, except OHW and SDT+OHW treated groups, revealed the increasing effect as compared with control group, but the significance was not admitted. 7. For the effect of K-index(Carbon clearance), SDT, SDT+OHW and SDT+OHE treated groups showed the increas ing effect with significance as compared with control group. 8. The study didn't show that Orostachys herba had any significance with survival days, anti-tumor effect and immune re sponse.
