BACKGROUND: RNA interference (RNAi) eliminates or decreases gene expression by disrupting the target mRNA or by interfering with translation. Recently, RNAi technique was applied to generate new crop traits which provide protection against pests. To establish the environmental risk assessment protocol of RNAi LMO in lab scale, we developed dsRNA expression system using E. coli and tested acute oral toxicity assay to honey. METHOD AND RESULTS: The dsRNA expression vector, L4440, was chosen and cloned 240 bp of Snf7 and GFP gene fragment. To develop the maximum dsRNA induction condition in E. coli, we tested induction time, temperature and IPTG concentration in media. To estimate the risk assessment of dsRNA to honey bee, it has been selected and cultured with dsRNA supplement for 48 hours according to OECD guideline. As a result, the optimum condition of dsRNA induction was $37^{\circ}C$, 4 hours and 0.4 mM IPTG concentration and the difference between Snf7 and GFP dsRNA molecules from E. coli was not significant in survival and behavior to honey bee. Furthermore, blast search results indicated that effective match of predicted dsRNA fragments were not existed in honey bee genome. CONCLUSION: In this study, we developed and tested the acute oral toxicity of dsRNA using E. coli expression system to honey bee.

특정 제품이 가진 "감성 이미지"란 그 제품의기능과 함께 제품의 감각적 요소 그리고 사용자가 제품에 대해 가진 개념적인 사고 과정의 산물이다. 감성이미지의 국제 비교 연구는 사용자가 특정 제품에 대해 가지고 있는 이미지를 국가나 문화의 맥락 속에서 상호 비교 평가하여, 각 국가나 사회집단을 특팅지울 수 있는 문화적 감성 요소를 추출하고자 하는 것이다. 즉, 각 국가의 생활문화 속에서 사용자들이 제품에 대해 가지는 감성이미지를 계량적으로 규명하고 비교 평가하며, 이 정보를 기초로 각국의 문화와 정서에 적합한 제품 디자인의 모형을 제시하는데 본 연구의 목적이 있다. 구체적인 연구방법은 우선, 자동차 제품을 중심으로 '제품 이미지'를 나타내는 '언어 이미지'스케일을 각 문화 집단 별로 구성한다. 이 제품 이미지에 대비하여, 감성적 요소로 선정된 표준 색채 스케일을 중심으로 나타난 집단의 '이미지'를 상호 비교 평가한다. 이 경우, 각 국가별 집단에서 특징적으로 나타나는 제품의 주요 특성이 감성 이미지로 표현되는 색채 이미지에 어떻게 반영되는가를 상호 비교한다. 이 연구에서는 특정 지역의 사람들이 제품에 적용하는 감성이미지는 색채나 기타 감성 요소에 대한 선호의 문제가 아니라 그 집단의 사람들이 그 제품을 통해 표현하고자 하는 사회적 표상의 반영이라는 것을 보여준다. 따라서, 제품의 이미지가 각 국가별로 공유되는 감성 이미지로 변환되기 위해서는 동일한 의미나 상징성을 지닌 감성 요소를 활용하여 그 제품의 이미지를 포장할 수 있는 방법을 적용하는것이다. 본 연구에서는 각 문화 집단들이 특정 제품에 대해 가지고 있는 사회적 표상(social representation) 체계를 색채스케일에서 나타난 감성 이미지를 통해 구체화하는 것이다.로서는 방전효율의 저하가 없는 양호한 성능을 보였으며, SC의 시범 작동시험을 실차(소나타 1800cc)에 장착하여 수행한 결과 20회 이상의 연속시동에서도 아무런 문제점 없이 잘 동작하였다.되는 데이타를 입력한후 마우스로 원하는 작업의 메뉴를 선택하면 된다. 방법을 타액과 혈청내 testosterone 농도 측정에 응용하여 RIA의 결과와 비교하여 본 바 상관관계가 타액에서 r=0.969, 혈청에서 r=0.990으로 두 결과가 잘 일치하였다. 본 실험에서 측정된 한국인 여성의 타액내 testosterone농도는 107.7$\pm$12.0 pmol/l이었고, 남성의 타액내 농도는 274.2$\pm$22.1 pmol/l이었다. 이상의 결과로 보아 본 연구에서 정립된 EIA 방법은 RIA를 대신하여 소규모의 실험실에서도 활용할 수 있을 것으로 사려된다.또한 상실기 이후 배아에서 합성되며, 발생시기에 따라 그 영향이 다르고 팽창과 부화에 관여하는 것으로 사료된다. 더욱이, 조선의 ${\ulcorner}$구성교육${\lrcorner}$이 조선총독부의 관리하에서 실행되었다는 것을, 당시의 사범학교를 중심으로 한 교육조직을 기술한 문헌에 의해 규명시켰다.nd of letter design which represents -natural objects and was popular at the time of Yukjo Dynasty, and there are some documents of that period left both in Japan and Korea. To develop transgenic orchardgrass (Dactylis glomerata L.) resistant to high temperature, a thermptolerance gene, DgP23, was introduced into orchardgrass using Agrobacterium - mediated transformation method. PCR and Southern blot analyses using genomic DNA showed specific DNA band on agarose gel and hybridization signal on X- ray film in transgenic orchardgrass harboring the recombinant DgP23 gene, but not in the wild type and empty vector control plants. RT-PCR and Southern blot analyses using total RNA also showed specific DNA band and hybridization signal. Transgenic orchardgrass did not showed ny morphological aberration both in the green house and field cultivation. Thermotolerance of transgenic plants was not detected in laboratory test. but may detected in field test.
It has been well known that the alkali-aggregate reaction between the aggregates and cement paste is one of the reasons of a concrete siructre expansion. Because of a serious demage on the concrete stnicture from the expansion, in many countries, the safety of the materials is checked in laboratory by mortar-bar test and the upper limit of expansion in length is 0.1%. The prescriptions are presented in the ASTM C227 and 490 of U.S. which has been international currency and in the KS Handbook F2503, F2546 and L5107 of Korea published by Korean Standards Association. Both of the prescriptions are almost same in their contents. Actually, in the process of preparing and measuring the mortar-bar according to the prescription mentioned above, it seems that there are no problems for its own purpose but a few points are found to be improved upon the methods to increase the accuracy for laboratory work as follows. 1. The prescription of blending ratio(aggregate, cement and water) should be noted by volume not by weight. 2. It is unreasonable to measure the initial length of mortar bars after 24$\pm$2 hours at once regardless the kind of aggregates. 3. It may bring about errors in calculating the expansion ratio under the condition of the denominator value fixed as 254mm. 4. The measuring methods of specific gravity are selected according to the purposes but the pure specific gravity displays the highest accuracy among them.
Experiments have been carried out in barge enclosure near Bukhangang area and arificially eutrophicated pond. Applied plants are three terrestrial plants (Big cone pine, Pine needles and Ginkgo) and one aquatic plant (Water chestnut). In the experiment pond, applied plants are big cone pine, pine needles and ginkgo at the concentration of 0.5 g/l and water chestnut of 0.3 g/l. And it used pine needles at the concentration of 0.3 g/l in barge enclosure. The dominant species was Microcystis aeruginosa. Chlorophyll a concentration decreased to the extent of average 65% as compared with the beginning of experiment by the input of big cone pine and pine needles in artificially eutrophicated pond. But it was decreased to the extent of average 34% by the input of water chestnut. Especially, pine needles significantly inhibited the growth of algae from the early days in barge enclosure, it was declined at a rate of average 74%. It was proved that pine needles was the most inhibitory to the growth of algae at the treatment with $0.3{\sim}0.5\;g/l$, Ginkgo and water chestnut are needed more study. We purified that the dissolved phenolic compounds from the various plants can affect algal growth.
Seed-propagation of Solomon's seal (Polygonatum odoratum) takes 2 years to shorten the period for becoming a practical method. The experiment was done to establish a proper method of breaking the epicotyl dormancy in bulk seed-propagation. Seedlings with a bulbil were treated with $GA_3$ every 2 days for 4 or 8 days and chilling treatments at $3^{\circ}C$ were enforced for 4, 6, 8 or 12 weeks. Emergence- and growth-related characteristics were examined immediately after the treatments, 3 and 6 weeks later. Rate of cotyledonary sheath rupture immediately after $GA_3$ treatment was greater in its 8-day treatment than in 4-day although its effect disappeared later. However, any epicotyl treated with $GA_3$ solution did not elongate so that new seedlings disemerged over the bed soil. That resulted from not breaking the epicotyl dormancy since $GA_3$ did not rupture all of the cotyledonary sheath formed with several sheets and consequently, the solution did not reach it. The $GA_3$ treatment for bulk seed-propagation, therefore, was impractical method. On the contrary, the chilling treatment was able to be applied to the seed-propagation because of getting the cotyledonary sheath rupture and the epicotyl elongation. Seedling emergence and its growth after chilling treatment were influenced by chilling period although required at least over 6-week treatment for satisfactory results.
A lab-scale vertical flow subsurface (VFS) wetland composed of three parallel columns with polypropylene synthetic fiber as main substrate was operated. Piggery stormwater diluted from swine excreta was fed to the wetland on the basis of three different hydraulic regimes or hydraulic retention time (HRT) of 2, 4, and 8 days with daily internal recirculation. Then, monitoring of common water quality parameters was carried out. Unexpectedly, an increase of effluent COD concentration accompanying the appearance of foams was observed during a distinct period in the wetland with HRT 2, 4, and 8 days, successively. Subsequently, a series of experiments was conducted to investigate the origin of the foams. Foams and the increase of COD concentration were found to be induced by the release of organic matter from the synthetic polypropylene fiber which was fed with piggery stormwater. Meanwhile, nitrogen removal was found to be enhanced during a period which overlapped the distinct foaming period signifying that foaming played two important functions in biological nitrogen removal. Foams which form rapidly and then burst easily could hold up and then release oxygen for nitrification. Foams which contain organic surfactants could serve as carbon sources for denitrification as well. Hence, nitrogen removal was enhanced during the foaming stage. After that, COD concentration decreased slowly to a level prior to the foaming stage, and nitrogen removal efficiency declined as well.

An HPLC analysis method was developed for standard determination of marmesin as a functional health material in Broussonetia kazinoki extract. HPLC was performed on a $C_{18}$ Kromasil column ($4.6{\times}250mm$, $5{\mu}m$) with a gradient elution of 0.1% (v/v) trifluoroacetic acid and acetonitrile at a flow rate of 1.0 mL/min at $30^{\circ}C$. The analyte was detected at 330 nm. The HPLC method was validated in accordance with International Conference on Harmonization guidelines for analytical procedures with respect to specificity, precision, accuracy, and linearity. The limit of detection and quantitation were 6.2 and $18.6{\mu}g/mL$, respectively. Calibration curves showed good linearity ($r^2$>0.9999), and the precision of analysis was satisfactory (less than 0.3%). Recoveries of quantified compound ranged from 100.35 to 101.18%. This result indicates that the established HPLC method is very useful for the determination of marker compounds in B. kazinoki extracts.
This study investigated the optimum depth for the application of bioremediation in contaminated coastal sediment using a lab scale column experiment. The biostimulants were placed in the top surface of the sediment facing seawater, 3cm, 6cm and 10cm of the depth from the surface, respectibely. During the experiment, the changes of organic pollutants and heavy metal fractions in the sediment were monitored in 1 month and 3month time intervals. The organic pollutants found during various analysis such as chemical oxygen demand, total solids and volatile solids, significantly reduced when the depth of the biostimulant was 3cm or less. In contrast, at a depth of over 6cm, the reduction of organic pollutants decreased, and the results were similar to the control. Heavy metals fractions in the sediment also changed with the depth of the biostimulants. The exchangeable fraction of the metals was quite reduced at the sediment surface in the column, but the organic bound and residual fractions considerably increased at a depth of 3cm. Based on this study, the optimum biostimulants depth for in-situ bioremediation of contaminant coastal sediment is 3cm from the sediment surface.
Congestion control is a key task in reliable multicast along with error control. However, existing tree-based congestion control schemes such as MTCP and TRAMCC are designed for one-to-many reliable multicast and have some drawbacks when they are used for many-to-many reliable multicast. We propose an efficient congestion control mechanism, TMRCC, for tree-based many-to-many reliable multicast protocols. The proposed scheme is based on the congestion windowing mechanism and a rate controller is used in addition. The feedback for error recovery is exploited for congestion control as well to minimize the overhead at the receivers. The ACK timer and the NACK timers are set dynamically reflecting the network condition changes. The rate regulation algorithm in the proposed scheme is designed to help the flows sharing the same link to achieve the fair share quickly The performance of the proposed scheme is evaluated using ns-2. The simulation results show that the proposed scheme outperforms TRAMCC in terms of intra- session fairness and shows good level of responsiveness, TCP-friendliness, and scalability. In addition, we implemented the proposed scheme by integrating with GAM that is one of many-to-many reliable multicast protocols and evaluated the performance in a laboratory-wide testbed.
