Estimation of Heritabilities and Genetic Gains for Height and Diameter Growth of Korean White Pine Open-Pollinated Progeny Stands (잣나무 풍매차대검정림(風媒次代檢定林)의 수고(樹高) 및 직경생장(直徑生長)의 유전력(遺傳力)과 개량효과(改良效果)에 관한 연구(硏究))

  • This study was carried out to analyze the genetic variation and to estimate both heritabilities and genetic gains for height and diameter growth of 20-year-old open-pollinated progenies of Korean white pine in three different sites. For there, analysis of variance for both height and diameter growth was conducted to see if there exist significant differences among families, sites, blocks, and their interactions exist or not and to analyze the variance components for each factor. Mean height and diameter at Gapyeong site were 7.65m and 11.92cm, respectively. they were 7.42m and 11.35cm at Gwangiu site, 6.13m and 8.41cm at Youngdong site, and 7.12m and 10.68cm for the overall sites. The family No. 20 showed the most excellent growth of 7.99m in mean height and 12.14cm in mean diameter for all sites surveyed. The estimates of heritabilities for individual-tree and family were, 0.35~0.73 and 0.65~0.83 for height growth, and 0.12~0.40 and 0.46~0.75 for diameter growth, respectively. For the combined data from all the sites, the estimates of individual tree heritability were 0.60 for height and 0.20 for diameter, and those of family were 0.91 for height and 0.77 for diameter. Heritability estimates varied with testing sites, and those for height were higher than those for diameter in all sites. Given equal intensity of selection, combined selection method provided the most efficient genetic gains for both height and diameter growth.

Clipping of T2 Sympathetic Chain Block for Essential Hyperhidrosis (다한증 환자에서의 Clipping에 의한 T2 Sympathetic Chain Block의 효과)

  • Background: A definitive cure for an essential hyperhidrosis can be obtained by an upper thoracic sympathectomy. However, this is offset by the occurrence of a compensatory hyper hidrosis as a side effect and it is irreversible. We performed a thoracoscopic sympathetic chain block using an endoscopic clip in order to avoid the compensatory hyperhidrosis. Material and Method: From Aug. 1998 to Nov. 1998, 42 cases of thoracoscopic clipping of the T2 sympathetic chain were performed. The sympathetic chain was clipped using an endoscopic clip instead of cutting. Result: Bilateral procedure took less than 40 minutes and occasionally necessitated one night in the hospital. There were no mortality nor life- threatening complications. Horners syndrome occurred in two cases. At the end of postoperative follow-up(median 3 months), 95.0% of the patients were satisfied with the results. Compensatory sweating occurred in 31 cases(77.5%) where nine of those cases were classified as either embarrassing(6 cases-15.0%) or disabling(3 cases-7.5%). Conclusion: Endoscopic thoracic T2 sympathetic chain block using endoscopic clipping is an efficient, safe and minimally invasive surgical method for the treatment of palmar and craniofacial hyperhidrosis and the results were similar to those underwent T2 sympathicotomy. We recommend that patients receive endoscopic sympathetic chain block in summer.

Change in Protein Composition of Filefish Muscle during Post-Mortem Lapse (말쥐치육의 사후경과에 따른 단백질조성의 변화)

  • Protein compositions of filefish (Navoden modestus) skeletal muscle and their changes in postmortem with reference to freshness kept at $0^{\circ}C$ were investigated. The muscle protein was approximately composed of $31\%\;sarcoplasmic,\;55\%$ myofibrillar, $10\%$residual intracellular, and $4\%$stroma protein. The sarcoplasmic and myofibrillar protein decreased while the residual intracellular protein increased with the decline of freshness during post-mortem lapse. In the analysis of electrophoretograms and its densitograms, the myofibrillar protein resembled to other fishes in protein composition: $70\%$ actin and myosin, $20\%$ regulatory proteins, and $10\%$ unknown proteins. And most of the residual intracellular protein was estimated as myofibrillar protein. Troponin T, troponin C and myosin light chain 2 of the myofibrillar protein constituents were decreased during storage. Amino acid composition of the protein from the at-death muscla was similar to those of other fishes except that tryptophan and sulfur-containing amino acids were scant. Proline and cysteine were remarkably decreased whereas leucine, isoleucine and phenylalanine were slightly increased in the protein from the muscle lapsed of 18 days. In free amino acid composition, alanine, glycine, lysine, and especially taurine were rich in the at-death muscle. The muscle lapsed of 18 days showed an increase of taurine, histidine, valine and methionine, and a decrease of lysine, arginine, aspartic acid, threonine, leucine, and isoleucine.

  • The transfer of isolated nuclei from Agrocybe aegerita mycelia of wild type into Pleurotus florida protoplasts of auxotroph was induced with polyethylene glycol. The type 1 of nuclear transfer products was spontaneous segregants of both parental morphology of colony. Hyphae of A. aegerita type had clamp connections while that of P. florida type lacked. Type 2 was main products of nuclear transfer which formed true clamp connections. Type 3 was clampless products. They all produced primordia and developed basidiocarps similar to Agrocybe aegerita. A comparison of nuclei transfer products was made using isozyme analyses of alcohol dehydrogenase, esterase, lactate dehydrogenase and peroxidase. Isozyme band patterns of some type 2 strains produced a new band. Band patterns from mycelial extracts of the other strains could be characterized by parental bands.

The Folk Plants in Southern Region of Chungcheongbuk-do, Korea (충청북도 남부지역의 민속식물)

  • This study was carried out in order to catalogue the folk plants of 5 counties and cities of Chungcheongbuk-do Province from March to October, 2011. 429 pages of field notes relating to the folk plants were collected from approximately 92 local inhabitants and analyzed subsequently. The identified folk plants were of 173 taxa, composed of 150 species, 1 subspecies, 22 varieties and 129 genera under 56 families. Naturalized plants were included. The use by its usage was: 349 taxa; edible, 68 taxa; medicinal, 2 taxa; dye, 1 taxa; aroma, 4 taxa; spice, 1, taxa; ornamental, 3 taxa; oil, 1 taxa; starch, 18 taxa; others respectively, so the edible use is the highest. The most useful part was the leaf, followed by stem, and whole plant. The consistency comparison between the scientific name and the local name were the highest in the 50's and the lowest in 90's.

Comparison of Muscle Texture between Red Seabreams Cultured by Feeding and Starving (급이 및 비급이 참돔의 조직감 비교)

  • For the effective use of red seabream, the texture between red seabream muscles cultured by feeding and starving were compared. Moisture and crude lipid contents of red seabream muscle cultured by starving (RCS) were 72.7% and 3.7%, respectively, which was 3% higher in the moisture compared to that of red seabream muscle cultured by feeding (RCF), while 3% lower in the crude lipid. The hardness of RCS was $0.93\;kg/cm^2$, which was higher than that ($0.47\;kg/cm^2$) of RCF. No differences in total collagen content, acetic acid-solublized collagen content, its thermal denaturation temperature and SDS-PAGE patterns between RCS and RCF were found. The results suggested that the difference in muscle texture between RCS and RCF was probably due to the difference of lipid content.

  • Interspecific fusion products were obtained from fusion of Ganoderma applanatum and Ganoderma luridum. Frequency of fusion was 0.77-1.38%. Fusion products were selected by the comparision of morphology and color of colony. Fusion Products had chracteristics of two parental strains and generally grew faster than the parents. Some fusants were segregated on GCM. Fusion products were confirmed by mycelial morphology and electrophoretics pattern of esterase isozyme from mycelium. Most of fusion products were lack in clamp connection but those fusion products, that were segregated produced mycelium with and without clamp connection. Some of the fusion products produced fruit body on sawdust medium.

A Study on Paralytic Shellfish Poison of Sea Mussel, Mytilus edulis -Food Poisoning Accident in Gamchun Bay, Pusan, Korea, 1986- (진주담치의 마비성독에 관한 연구 -1986년 부산 감천만 중독사고를 중심으로-)

  • At various times and places all over the world men have become ill and some have died after eating shellfish that were intoxicated with paralytic shellfish poison(PSP) caused by Protogonyaulax spp. In late March, 1986, two persons were dead by ingesting wild sea mussels, Mytilur edulis, grown at bottom of an anchored waste ship to be dismantled at Gamchun Bay, Pusan, Korea. The samples were collected from the bottom of the ship during April $1\~April$ 8 of the year to find the cause of the food poisoning accident. The toxicity was estimated by bioassay with ICR male mouse, while the toxins were extracted and characterized. The toxins were extracted with acidified $80\%$ ethanol. The extract was defatted three times with dichloromethane, treated with activated charcoal, and then purified by chromatography on Bio-Gel P-2 and Bio-Rex 70. The toxic fractions obtained were analysed by cellulose acetate membrane electrophoresis, thin layer chromatography and high performance liquid chromatogaphy. The range and the average of PSP-toxicity of the samples were $132\~295\;MU/g$, 203 MU/g respectively. The amount of PSP was $26.4\~58.9{\mu}g/g$ of whole meat in range and $40.6{\mu}g/g$ in average. The toxicity of the digestive gland of the samples was 9 times higher than that of edible meat (except digestive gland) as $439\~979MU/g$, and it was about $70\%$ in total toxin. The compositional analytical results of the paralytic shellfish toxin, Gonyautoxin $1\~4$ were the major part of the PSP and Saxitoxin and neosaxitoxin were detected as the minor component. It was concluded that the food poisoning accident was caused not by Saxitoxins but by Gonyautoxins.

Comparative Study of Proteolytic Activities of Some Commercial Milk Clotting Enzymes on Bovine Skim Milk (상업적 응유효소의 탈지유에 대한 단백질 분해 작용)

  • Proteolytic activities of some commercial milk clotting enzymes(rennet, trypsin, pepsin, papain W-40, neutrase 1.5 and protease S) in bovine skim milk containing 0.02% $CaCl_2$ were determined by measuring DH(Degree of Hydrolysis), NPN(Non Protein Nitrogen) and by comparing patterns of SDS-PAGE(Sodium Dodecyl Sulphate Polyacrylamide Gel Electrophoresis). The DH of microbial enzymes(neutrase 1.5 and protease S) and trypsin in bovine skim milk were higher than those of pepsin and papain W-40. The amounts of NPN in the milk treated with trypsin and the other animal enzymes(rennet and pepsin) showed the highest and lowest degrees of proteolysis, respectively. SDS-PAGE showed that trypsin and protease S hydrolyzed $\alpha$-lactalbumin and papain W-40 hydrolyzed $\beta$-lactoglobulin slightly, while neutrase 1.5 hydrolyzed both $\alpha$-lactalbumin and $\beta$-lactoglobulin after treating for 90 min. Trypsin and protease S easily hydrolyzed ${\alpha}_s$-casein and $\beta$-casein, which were not hydrolyzed by rennet. Papain W-40 hydrolyzed $\kappa$-casein more than rennet as shown in SDS-PAGE. Based on the results of the experiments, the DH and NPN of trypsin, neutrase 1.5 and protease S were shown to be higher than those of the other enzymes. The SDS-PAGE patterns of papain W-40 and neutrase 1.5 were similar with that of rennet.

Hydrolysis Characteristics of Goat Milk $\beta-Casein$ by Enzyme and Angiotensin Converting Enzyme Inhibition Effects of Hydrolysate (산야유 $\beta-Casein$의 효소 가수분해 특성과 가수분해물의 Angiotensin Converting Enzyme 저해 효과)

  • This study was carried out to understand hyrolytic characteristics of $\beta-casein$ by enzyme in goat milk and to measure the inhibition effect of the ACE of the hydrolysate. In order to conduct the experiment, $\beta-casein$ of goat milk was separated using Mono S HR 5/5, a cation exchange column. The separated $\beta-casein$ was treated with trypsin of animal hydrolysis enzymes, in an effort to verify the characteristics of hydrolysis. The inhibition activity of ACE was measured and the results are as follows. By analyzing the hydrolysate separated from the trypsin-processed $\beta-casein$ of goat milk, the inhibition effect of the ACE was measured trypsin-hydrolyzed $\beta-casein$ demonstrated a $25.36\pm0.79\%$ of inhibition effect and the $IC_{50}$ of the hydrolysate from the trypsin-processed $\beta-casein$ reached $308.7\pm2.77({\mu}g/mL)$.
