Differences of Essential Oil Content in Valeriana fauriei var. dasycarpa Hara, V. officinalis var lalfolia Miq and V. wallichii DC (광릉쥐오줌풀, 넓은잎쥐오줌풀, 네팔산쥐오줌풀의 정유성분(精油成分)차이)

  • In this study the essential oil and pharmacologically active components of different valerian species (Valeriana fauriei var. dasycarpa Hara and V. officinalis L. from Korea and V. wallichii DC from Nepal) were analyzed to search for native plant resources that can be used for medicinal herb and tobacco flavoring. The oil contents in dry roots of V. fauriei, V. officinalis and V. walhchii were 1. 71%, 0. 82% and 0. 83 %, respectively. The composition of the essential oil among different valerian species was compared. In V. fauriei 47 compounds were identified, while 21 and 25 compounds were identified in V. officinalis and V. wallichii. The major compounds in the oil of V. fauriei were bornyl acetate (33.8%) and camphene (10.8%), cedrol (4. 1 %), -pinene (3. 2%) and unidentified sesquiterpene alcohol (3. 0%). The major compounds were borneol (62. 5%) and ${\beta}-sesquiphelandrene$(6. 8%) and spathulenol (2. 1%) in V. officinalis, and borneol (74. 3%) and ${\alpha}-elemene$ (2.7%) and -sesquiphellandrene (2. 3%) in V. wallichii. Among the components known as major pharmacologically active compounds in European or Japanese valerian roots, valeranone, valerenal and -kessyl acetate was detected in a small amount in V. fauriei, but kessoglycol diacetate was not detected in V. faudei and V. officinalis Among the valepotrate compounds, major pharmacologically active components in V. wallichii, valtrate was detected in a small amount in V. fauriei and V. officinalis., and detected 1. 42% in V. wallichii. Didrovaltrate was also detected in the three valerian species tested, but acevaltrate was not detected except V. wallichii. On the other hand, antioxidative activity was slightly higher in V. fauriei than those of V. officinalis.

Occurrence of Earthworm and Effect of Plant Extracts on Earthworm in Golf Courses (골프장에서 지렁이의 발생과 식물체 추출물이 지렁이에 미치는 영향)

  • Occurrence of earthworms for turfgrass growing seasons and effect of earthworms by some plant extracts and plant oils in golf courses were investigated at the three locations golf courses in Korea 2005 to 2006. There were no differences in monthly occurrence of earthworm except Anseong Benest Golf Club, namely, Anseong Benest Golf Club was highly occurred in July in 2006. Density of earthworm was not different between fairway and rough in Dongrae Benest Golf Club (opened in 1971) while density was higher in rough than fairway in Anseong Benest Golf Club (opened in 1999) and Gapyeong Benest Golf Club (opened in 2000). In case of Gapyeong Benest Golf Club, earthworm density was higher in old courses than recently opened courses. Effect of plant extracts such as mustard oil, wintergreen oil, Quisqualis indica, Daphne genkwa, Pharbitis nil, Zingiber officinale, Xanthium strumarium, and Camellia sinensis on earthworm, mustard oil, wintergreen oil and tea saponin were highly toxic to earthworm, Eisenia andrei. In the pot experiment, 100% of Eisenia andrei was dead at the treatment of 500-fold of aquatic solution of tea saponin (12.4% a.i.).

A Study on the Characteristics of Natural Preservative Agent-treated Fabrics for Textile Cultural Properties Preservation (섬유문화재 보존에 활용하기 위한 천연보존제로 처리한 직물의 특성연구)

  • Natural preservative agents are generally made of antibiotic substances that are extracted from plants. They are used mainly to keep in an original good state food, natural cosmetic goods and medicines which are likely to get rotten. The purpose of this study is to investigate whether natural preservative agents can be applied to fabrics for the preservation of textile cultural properties. For the purpose, this researcher experimented with a certain natural preservative agent which was developed to preserve natural cosmetic goods. The study found that when treated with the foresaid natural preservative agent whose ratio to water was 1%, fabrics showed little changes in color and tensile strength, almost neutral pH, high antibiosis and anti-fungus and high resistance to Bacillus cereus that is much detrimental to silk fabrics. As a part of the study, a preliminary test on the possibility of using natural preservative agents to preserve textile cultural properties, found that when kept covered up with the 1% natural preservative agent-treated fabrics for 72 hours, excavated textile relics showed a dramatic decrease in microbial growth.

  • The 1-methylcyclopropene (1-MCP) is the gaseous ethylene action inhibitor, which delay senescence and wilting by blocking the binding of its receptors. We investigated the effects of 1-MCP with different treated duration (0h (Control), 1h, and 4h) in cut flowers of Eustoma grandiflorum 'Voyage' and Cymbidium 'Lapine Hat'. The relative water uptake increased by 14.5% than control (9.8%) and the flower diameter maintained high from 3 to 11 days in 1-MCP treatment for 4h in E. grandiflorum 'Voyage'. The relative fresh weight was averagely high in 1-MCP treatment for 4h, however, the vase life was not significantly different in both cut flowers. Since the effects of 1-MCP treatment was differently responded by plant species and cultivars, interacted studies of duration and levels of 1-MCP are needed for the pronounced effects of postharvest quality.

  • 들깨(Perilla frutescens L.)는 꿀풀과 1년생 초본으로 한국, 중국 등의 동부 아시아가 원산지로 식용, 약용 및 공업용 등 다양한 용도로 이용되고 있다 들깨는 우리나라에서 오랜시간 재배해 왔으나 체계적인 연구기간이 짧아, 다른 유료작물에 비해 육성품종이 적고 유전자원의 체계적인 수집평가가 이루어지지 않았다. 특히 외국에서는 들깨와 근연종을 이용하여 색소 및 정유성분에 대한 연구를 하는데 비해 우리나라에서는 주로 식용으로만 연구가 진행되고 있다. 들깨 육종에 있어서 중요한 것은 유전자원 확보와 이를 정리 보존하여 우량한 육종재료를 효율적으로 이용하는 것이다. 본 연구에서는 국내에서 수집한 들깨 44자원의 형태적 특성을 조사하여 품종 육성을 위한 기초자료를 제공하고자 하였다. 본 연구에 사용된 들깨 자원은 경북 수집 10자원으로 가장 많았으며 전북 8자원, 경남 7자원, 강원과 경기에서 각각 6자원, 충남 3자원, 전남과 충북에서 각각 2자원이었다. 화색은 들깨 44자원 모두 흰색이었으며 줄기색은 연녹색이 29자원, 녹색 9자원, 자주색 4자원으로 조사되었다. 잎의 앞면은 녹색 27자원, 연녹색 10자원, 진녹색 7자원이었으며 뒷면색은 녹색 26자원, 연녹색 18자원으로 조사되었다. 잎의 모양은 장타원형 21자원, 피침형 15자원, 심장형 7자원이었으며 잎의 모용은 23자원이 적음, 14자원이 보통, 6자원이 많게 조사되었다. 잎의 길이는 35자원이 15.1cm이상으로 조사되었고 잎넓이는 11.1cm이상인 자원이 37자원으로 조사되었다. 종피색은 갈색이 28자원, 회색이 6자원, 암갈색이 3자원 순으로 조사되었다. 본 실험에서 조사된 들깨 유전자원의 형태적 특성은 들깨 선발에 있어 기초 정보로 활용 가능할 것이며 추가적으로 농업형질, 기능성 물질 분석등의 들깨 육종 프로그램을 위한 다양한 평가 및 선발이 필요할 것이다.

Analysis of growth pattern, gene expression and flavonoid contents under LED light wavelength in Lettuce (Lactuca sativa L.) (상추에서 LED광질에 따른 플라보노이드 생합성 관련 유전자들의 발현 및 이차대사 산물의 성분 분석)

  • We analyzed the effects of various LED light treatments (red 655 nm, blue 456 nm, white and mixed light) on growth pattern, gene expression and flavonoid contents in lettuce leaf. Plants treated with mixed light (red+blue+white) showed better growth performance than those treated with single LED and fluorescent lamp (FL). Expression analysis of the eight genes involved in flavonoid biosynthesis in plants treated with LED light was examined. Results showed that red lettuce grown under mixed light showed high expression levels of LsC4H, LsF3H and LsDRF genes. Morever, the same treatment plants possessed higher content of gallic acid, chlorogenic acid and quercetin contents than those in plants exposed to single light. However, the highest total anthocyanin content was identified in plants treated with red+blue light and the lowest content was identified in plants exposed to white fluorescent lamp and single LED light condition. Thus, this study indicates that the ratio of blue to red LEDs is important for the morphology, growth, and phenolic compounds with anthocyanin properties in the two lettuce cultivars tested.

Agronomic Characteristics and Aromatic Compositions of Korean Wild Codonopsis lanceolata Collections Cultivated in Field (한국 야생더덕 수집종의 노지 재배시 생육 특성과 향기성분 조성)

  • The native ecological environment and aromatic constituents of Korean wild Codonopsis lanceolata and one Japanese strain were investigated to find Codonopsis lanceolata strains showing high aromatics, and to know regional differences among these strains. The results were as follows : There were no remarkable differences among the Korean wild C. lanceolata strains in ecological environments. Recovery yield of essential oils was highest in Togyusan strain with 0.009%. Difference in protein band patterns among these strains was not recognized, and peroxidase and esterase pattern changes were appeared in different collected regions at the leaf and root tissues. Major aromatic constituents were 11 kinds of aliphatic alcohols such as trans-2-hexenal, 1-hexanol, cis-3-hexanol, and trans-2-hexanol. And Togyusan strain, Sobaeksan strain, and Kayasan strain have the highest aliphatic alcohols of plant essential oils. In particular, BHT(butylated hydroxytoluene), one of the antioxidants, was detected in Chirisan strains.

  • An ideal anti-bacterial medication for oral infection requires selective effect on pathogens causing dental caries and periodontal disease but not on normal flora. In addition, it should be less toxic for human and even for environment. This study was to seek such a natural anti-bacterial medication and thus anti-bacterial effect of Hamamelis virginiana was evaluated. Many recent researches on the anti-bacterial effect of natural plant extract and essential oil have reported that natural products can be used as medication for prevention and restrainment of dental caries, halitosis and periodontitis. It has been also reported that Hamamelis virginiana has anti-bacterial effect on Porphyromonas gingivalis, Fusobacterium nucleatum, Capnocytophaga gingivalis, Veilonella parvula, Eikenella corrodens, Peprostreptococcus micros, and Actinomyces odontolyticus. This study evaluated anti-bacterial effect of Hamamelis virginiana on Streptoccoccus mutans, Haemophylus actinomycetemcomitans, and Klebsiella pneumoniae to expand its anti-bacterial effect on other important oral pathogens and eventually to develop its oral care products or apply to clinical purpose. In this study, anti-bacterial tests for antibiotic disk susceptibility, minimal inhibitory concentration and minimal bactericidal concentration were performed to evaluate anti-bacterial effect of Hamamelis virginiana against Streptoccoccus mutans, Haemophylus actinomycetemcomitans, and Klebsiella pneumoniae. The results showed that Hamamelis virginiana has anti-bacterial effect on all pathogen strains tested in this study and furthermore Hamamelis virginiana possesses bactericidal effect other than bacteriostatic effect on Streptoccoccus mutans, Haemophylus actinomycetemcomitans, Klebsiella pneumoniae. This study indicates that a natural anti-bacterial medication for oral diseases can be developed using Hamamelis virginiana.

Distribution of Bacterial Decomposers in Lake Khuvsgul, Mongolia (몽골 훕스굴 호수 수층에서 유기물질 분해세균의 분포)

  • To understand the ecological function of heterotrophic bacterial community in water column of large freshwater lakes in the permafrost zone, we investigated the structure and function of bacterial community in Lake Khuvsgul, Mongolia. Species composition of overall bacterial community was analyzed by denaturing gradient gel electrophoresis (DGGE) of 16S rRNA gene fragments, and bacteria that can be cultured at 10oC were isolated and characterized. Based on the depth profile of environmental parameters, thermocline and chemocline were recognized at the 5~10 m zone of the water column. The stratified DGGE profile indicated that the discontinuity of water properties might influence the structure of bacterial community: band profiles in the 0~5 m zone were diverse with large change by depth, but the profile was relatively stable at the $\geq$10 m zone, with predominance of the band identified as Acidovorax facilis. Bacterial cultures were screened for protease, cellulase, amylase and lipase activity, and 23 isolates were selected for high activity of the hydrolytic enzymes. The isolates were identified based on their 16S rRNA gene sequences. In the surface water (zero meter depth), Acidovorax defluvii and Sphingobacterium faecium with high cellulase activity were present. Flavobacterium succinicans, Mycoplana bullata and A. facilis were stably predominant isolates at 2 m, 5 m, and $\geq$10 m depths, respectively. F. succinicans isolates showed high protease activity while M. bullata isolates showed moderate levels of protease and celluase activity. A. facilis isolates showed either cellulase or lipase activity, exclusively to each other. According to the profile of growth rates of the isolates in the temperature range of $0\sim42^{\circ}C$, the surface-zone (0~5 m) isolates were facultative psychrophiles while isolates from $\geq$10 m depth were typical mesophiles. This stratification is believed to be due to stratified availability of organic materials to the bacterial decomposers. In the water column below the chemoline, the environment is extremely oligotrophic so that the trait of rapid growth in low temperature might not be demanded by deep-lake decomposers. The stratified distribution of community composition and decomposer activity in Lake Khuvsgul implies that ecological functions of bacterial community in lakes of cold region are sharply divided by water column stratification.

Bioactive Constituents and Utilities of Artemisia sp. as Medicinal Herb and Foodstuff (쑥(艾)의 생리활성 물질과 이용)

  • Mugwort (Artemisia sp.) abounds on hedgebank, waysides and grassy places in most part of Europe, Asia and Northern America. This herb has long been associated with witch-craft and magic as a protective charm. Mugwort has been a value as a foodstuff of relieving famine. Proximate composition of mugwort is similar to green vegetables, but it has high level in calcium, potassium and vitamin A content. For medicinal uses, the leaves are harvested twice in spring and autumn before the plant comes into flower and are dried for later uses. Aqueous or organic solvents extracts often have physiologically active constituents. Some extracts of mugwort include cineol, thujone, borneol, camphor, caryophyllene, coumarin, cubebene, pinene, linalool, absinthin etc. The dried leaves have traditionally been used as an antihelmintic, anti-inflammatory, antispasmodic, antitumor, hepatic stimulant, chologogue, emmenagogue, febrifuge, stomachic, tonic etc. Mugwort has been used the dried or green leaves as a flavoring and coloring agents for tea, cake. pastry, bread, noodle, alcoholic liquor, soap and hygienic band.
