Application of Exergy in Aquatic Ecosystems Health Assessment : Experimental Approach and Field Observations (수계 생태계의 건강성 평가 척도로서의 엑서지 적용성에 관한 연구: 실험 및 야외 관찰)

  • The results of field experiments with mesocosms on Lake Baikal, containing natural plankton assemblage, and laboratory experiments with microcosms containing Daphnia magna and Chlorella vulgaris demonstrated decrease of the structural exergy of the communities after the addition of allochtonous compounds peptone, diesel oil, o-diphenol, $CdCl_2$ to mesocosmsassemblage, phenol, $CoCl_2$ and $CuSO_4$ to micro-cosms. Structural exergy changes were more expressed than changes of components biomasses and total biomass of the community. Comparison of exergy content for benthos in cleanand affected by the discharges of Baikalsk Pulp and Paper Combine also showed sufficient docrease of structural exergy in polluted area. It points to the possibility of the use of structural exergy as ecosystem health reflecting parameter.

Preparation and Evaluation of Microcapsule/Emulsions via the Electroatatic Interations of Polysaccharide and Protein (식물 유래 다당류/단백질 기반 마이크로캡슐/에멀젼 제조 및 평가)

  • A novel microcapsule/emulsions for cosmetics was studied. Our present studies demonstrate that the biopolymer-stabilized microemulsion composed of polysaccharide and protein can encapsulate and stabilize remarkably coenzyme-Q10 (Q10). Polysaccharide and protein complex were incorporated in the microcapsule in order to reinforce the physical strength of the microspheres. We compared the long-term stability of the activity of Q10 in biopolymer-stabilized microemulsion. There was no noticeable negative effect on the activity of Q10. Optical microscope (OM) and transmission electron microscope (TEM) showed that microcapsules were spherical and had a smooth surface. Consequently, the polysaccharide/protein emulsion produced in this study may be beneficial in improving the emulsion stability and the protection capability of labile substances.

Insecticidal toxicities of essential oils extracted seven plants against Ricania sp. nymphs and adults (돌발 해충, 갈색날개매미충에 대한 7종 식물체 오일성분의 살충)

  • The insecticidal toxicities of seven essential oils against Ricania sp. nymph and adults, newly recorded pest, were evaluated using leaf-dipping and spray methods. At 72 h exposure time, the $LC_{50}$ values of the essential oils of Artemisia vulgaris, Anethum graveolens, Citrus paradisi, Melaleuca alternifolia, Melaleuca leucadendron, Neroli bigarade and Tagetes erecta were 251.02, 387.31, 286.21, 353.54, 275.64, 424.55, $165.68mg/cm^2$ when using the leaf dipping method against Ricania sp. nymphs, and 278.31, 402.11, 306.11, 379.64, 298.22, 459.56, $183.21mg/cm^2$ when using the spray method against Ricania sp. adults, respectively. These results indicate that the essential oil of the Tagetes erecta can be potentially used as new natural insecticidal agents against Ricania sp. nymph and adults.

Synthesis of Oleic Acid Esters containing Thiocarbamate Group as Anti-wear Additives (티오카바메이트 그룹을 함유하는 올레인산 에스테르계 내마모제의 합성)

  • A series of oleic acid esters containing mercaptobenzothiazole and dialkylenedithiocarbamate, which are potential environmentally friendly lubricating grease additives, were synthesized as 90% yield through several method such as nucleophilic substitution of dialkyl amine and carbon disulfide, reduction reaction, and condensation reaction. The structures of the additives were confirmed by $^1H$-NMR, FT-IR and EA analysis. The additives (1 wt%) were soluble in 100 N BO except C4-DTC-OE and soluble in soybean oil. The tribological properties as lubricating additives in 100 N BO were evaluated using 4-ball tester and the results showed as follows: Bz-thia-OE < C4-DTC-OE < Pyrro-DTC-OE < C8-DTC-OE. Whereas, the 4-ball anti-wear properties were not shown in soybean oil.

  • Biocatalytic modification of natural resources can be used to generate novel compounds with specific properties, such as higher viscosity and reactivity. The production of hydroxy fatty acids (HFAs), originally found in low quantities in plants, is a good example of the biocatalytic modification of natural vegetable oils. HFAs show high potential for application in a wide range of industrial products, including resins, waxes, nylons, plastics, lubricants, cosmetics, and additives in coatings and paintings. In a recent study, Pseudomonas aeruginosa strain PR3 was used to produce 7,10-dihydroxy-8(E)-octadecenoic acid (DOD) from oleic acid. This present study focused primarily on the utilization of three natural nut oils obtained from the Philippines -pili nut oil (PNO), palm oil (PO), and virgin coconut oil (VCO)- to produce DOD by P. aeruginosa strain PR3. Strain PR3 produced DOD from PNO and PO only, with PNO being the more efficient substrate. An optimization study to achieve the maximum DOD yield from PNO revealed the optimal incubation time and medium pH to be 48 h and 8.0, respectively. Among the carbon sources tested, fructose was the most efficiently used, with a maximum DOD production of 130 mg/50 mL culture. Urea was the optimal nitrogen source, with a maximum product yield of 165 mg/50 mL culture. The results from this study demonstrated that PNO could be used as an efficient substrate for DOD production by microbial bioconversion.

Synthesis and Lubricating Properties of Dimer Acid Derivatives Based on Used Vegetable Oil (폐식물유 기반 다이머산 유도체의 합성 및 경유의 윤활성능)

  • Vegetable oil-based dimer acid derivatives were prepared through a two-step procedure and their lubricating properties for diesel fuel were evaluated using high frequency reciprocating ring (HFRR) method to investigate wear scar diameter (WSD). Diels-Alder reaction at an elevated temperature transformed fatty acid to dimer acid, subsequently converted into dimer acid derivatives by esterification with methanol. It should be noted that the derivatives were dissolved well in diesel oil up to 1 wt%. After adding 120 ppm of the derivatives to pure diesel, the WSD significantly decreased to $300{\sim}05{\mu}m$, compared to $552{\mu}m$ of WSD in pure diesel. Dimer acid derivatives having carboxylic acid show superb in lubricating property which does not depend on the alkyl group in the derivatives.

Suppression Effect on Soft-rot by Bacteriocin-producing Avirulent Pectobacterium carotovorum subsp. carotovorum Pcc21-M15 (박테리오신을 분비하는 비병원성 돌연변이주에 의한 무름병 방제 효과)

  • Pectobacterium carotovorum subsp. carotovorum causes soft rot disease in diverse plants. Carocin D is bacteriocin that is produced by Pectobacterium carotovorum subsp. carotovorum Pcc21 strain. Nonpathogenic mutant P. carotovorum subsp. carotovorum Pcc21-M15 strain was obtained by mutagenesis with Tn5 insertion and screened pathogenesity. P. carotovorum subsp. carotovorum Pcc21-M15 and E. coli (pRG3431), carocin D gene-transformed E. coli, produce carocin D against P. carotovorum subsp. carotovorum Pcc3. Pathogenic P. carotovorum subsp. carotovorum Pcc3 and mixture with Pcc21-M15 or E. coli (pRG3431) were treated with lettuces. Pcc21-M15 and E. coli (pRG3431) effectively suppressed the development of soft-rot disease. While symptoms in 90% of Pcc3-treated lettuces were observed after 3 days, only 25% of Pcc3 and Pcc21-M15-treated lettuces were observed to be infected after 6 days. These results suggest that the nonpathogenic strain P. carotovorum subsp. carotovorum. Pcc21-M15 and E. coli (pRG3431) are effective to soft-rot disease suppression.

A Comparative Study of Physiological Activity of Glycyrrhiza uralensis Fischer Stems and Leaves by Processing Methods (감초 경엽(莖葉)의 포제방법에 따른 생리활성 비교)

  • This study investigates the change of chemical components, antioxidant and antimutagenic activity in Glycyrrhiza uralensis Fischer stems and leaves (GU) by the various processing methods, as follows: fresh (GU-1), dried under the shade (GU-2), blanched (GU-3), roasted 3 times (GU-4), roasted 4 times (GU-5). The components have been identified and quantified through the use of gas chromatograph-mass spectrometry (GC-MS). At results, cis-1,3-dimethyl-2-methylenecyclohexaneat at 19.7 min and n-hexadecanoic acid at 21.5 min were detected in five kinds of extract. 3-O-Methyl-D-fructose at 17.8 min was observed in four extracts except in GU-1 and aminopyrazine at 19.8min was observed in four extracts except in GU-4. The total phenolics contents was high in GU-4(3.38 g/100 g). ABTS radical scavenging was high in GU-5($EC_{50}$, 0.57 ug) and DPPH radical scavenging was high in GU-4($EC_{50}$, 2.66 ug). The extracts of GU-3, GU-4, and GU-2 were most potent in anti-mutagenicity activity against 1-NP, Trip-P-1, and Trip-P-2, respectively. GU-3 and GU-4 also showed most potent effect of anti-mutagenicity on 2-AA and AFB1, respectively.

Agrobacterium-mediated transformation produces transgenic oilseed rape with a high-yield trait (아그로박테리움 형질전환법을 이용한 수량증대 유채 식물체 개발)

  • This study includes the transformation of genes such as ORE7, the increase of gene expression, and the use of the bar gene as a selectable marker that shows herbicide resistance with Agrobacterium tumefaciens using hypocotyls from the oilseed rape "Youngsan" cultivar. To establish an Agrobacterium transformation system for the production of oilseed rape with a high-yield trait, infection time and co-cultivation period with Agrobacterium were tested. Therefore, when hypocotyls from the oilseed rape "Youngsan" cultivar were infected with Agrobacterium for 20 min and co-cultivated for 3 days, approximately 32-36 putatively transformed hypocotyls with shoots including roots survived from 100 inoculated hypocotyls after 4 weeks of transformation on a selection medium containing 20 mg/L of phosphinothricin (PPT) as a selectable agent. Additionally, a PCR assay was performed to confirm the insertion of target genes and showed the presence of the ORE7 gene as a high-yielding trait and the bar gene as a selectable marker. Treatment with 0.5% (v/v) Basta solution as a selectable agent for 6 days with leaves from transformed oilseed rape expressed the bar gene. Therefore, this study can contribute to the development of special oilseed rapes containing agriculturally useful traits such as herbicide resistance, drought tolerance, high yielding traits, and high oleic acid content.

Clean Flotation Process to Recycle useful Materials from Fly Ash (비산재로부터 유용성분을 회수하는 청정부유선별공정)

  • All coal ash, generated from coal-fired power plants, is entirely dumped onto a landfill site. As coal ash contains 80% fly ash, a clean floating process was developed in this study to recover useful components from coal ash and to use them as high value-added industrial materials. When the unburned carbon (UC) was recovered from the fly ash, soybean oil, an eco-friendly vegetable oil, was used as collector instead of a non-ionic kerosene collector to prevent the occurrence of odor from the kerosene. After the UC was separated by flotation, particulate ceramic microsphere (CM) was recovered, without generating acidic wastewater, through hydro-cyclone instead of sulfuric acid solution in order to separate ceramic microsphere (CM) and cleaned ash (CA) from the residue. By utilizing soybean oil as a collector, the recovery rate of UC turned high at 85.8% due to the increased adsorption of UC, the high viscosity of soybean oil, and the increase in floating properties caused by the linoleic acid contained in soybean oil. All of the combustible components contained in the recovered UC were carbon components, with the carbon content registering high when soybean oil was used. The recovered UC had many pores with a rough surface; thus, it could be easily ground and then used as an industrial material for its fine particles. The CM and CA recovered by the clean separation process using hydro-cyclone had a spherical shape, and the particles were clearly separated without clumping together. The average diameter (D50) of the particles was 5 ㎛, so it was possible to realize the atomization of CM through a process change.
