Effects of Acidic Treatments for Anthocyanin and Proanthocyanidin Extraction on Black Bean (Glycine max Merrill.) (검정콩 안토시아닌 및 프로안토시아니딘 추출에 대한 산 처리의 영향)

  • This study was performed to investigate the effects of various acidic treatment for anthocyanin and proanthocyanidin extraction from black bean. Extracts were prepared with 80% methanol solutions containing HCl, acetic, formic, phosphoric, and citric acids of different concentrations (0.1, 0.2, 0.3, 0.5, and 0.7%). Total anthocyanin content ranged from 0.74 mg/g in 0.7% citric acid to 1.74 mg/g in 0.3% HCl, depending on acid type and concentration. The major anthocyanins were delphinidin-3-glucoside (D3G), cyanidin-3-glucoside (C3G), and petunidin-3-glucoside (Pt3G). The highest C3G content was 1.12 mg/g in 0.3% HCl. Proanthocyanidin content ranged from 2.01 mg/g in 0.5% acetic acid to 5.29 mg/g in 0.3% HCl. Thus, acidic condition is a significant factor affecting anthocyanin and proanthocyanidin extraction from black bean, and the optimum extraction conditions were determined as 80% methanol containing 0.3% HCl.

Anthocyanin Composition and Characteristics of 'Gaeryangmeoru', 'Kyoho', and 'Hongisul' Grape Varieties ('개량머루', '거봉' 및 '홍이슬' 품종의 anthocyanin 조성과 특징)

  • This study was investigated the anthocyanin composition of 'Gaeryangmeoru', 'Kyoho', and 'Hongisul' grapes cultivated in Korea using ultra-performance liquid chromatography (UPLC) coupled to a mass spectrometer (MS) equipped with an ESI (electrospray ionization) source. 'Gaeryangmeoru' is a dark-blue grape used for winemaking that can reach its coloring in unfavorable weather. The 'Kyoho' and 'Hongisul' varieties are hybrid grapes that feature black and pink skin, respectively. The anthocyanins extracted from the peels of grapes were analyzed using UPLC-ESI-MS/MS. Twenty-five anthocyanins were identified in the 'Gaeryangmeoru' and 'Kyoho' varieties, and 21 were identified in the 'Hongisul' variety. Eight, 14 and five predominant anthocyanins were identified in 'Gaeryangmeoru', 'Kyoho' and 'Hongisul' grape respectively. In all three varieties, mono-glucosides were 2.3-5.9 times more abundant than di-glucoside. Malvidin was the predominant anthocyanidin in 'Gaeryangmeoru' (44.1%) and 'Kyoho' (56.5%), but cyanidin (96.8%) was in 'Hongisul'. The acylated anthocyanins in 'Gaeryangmeoru' (2.0%) were rare, whereas acylated anthocyanins with p-coumaric acid were predominant in 'Kyoho' (40.9%) and 'Hongisul' (70.7%). In particular, cyanidin feruloyl glucoside was found only in the 'Hongisul' cultivar and considered to be useful as a criterion for identification of the variety. As a result, the grape varieties were demonstrated to have variety-specific anthocyanin characteristics, enabling classification based on anthocyanin composition in terms of anthocyanidins, glucosylation and acylation. The taxonomical application of anthocyanin composition confirmed the possibility that 'Gaeryangmeoru' originated from Vitis amurensis or its hybrids, and the 'Hongisul' grape was distinguished from other grapes by cyanidin feruloyl glucoside.

Effect of Sorbitol and Salicylic Acid on Quality and Functional Food Contents of Tomato Fruit (Solanum lycopersicum) (솔비톨 및 Salicylic Acid 처리가 토마토 과실의 품질 및 기능성 물질 함량에 미치는 영향)

  • In this study, we aimed to understand the effect of sorbitol and salicylic acid (SA) on quality and functional food contents of tomato. Sole or combined application of sorbitol and SA in different concentrations (0.5 mM and 1.0 mM) greatly improved the characteristics of tomato, such as red color tone, soluble solid content, hardness and weight as well as increasing the functional food contents such as of lycopene, anthocyanins and vitamins in comparison with the control. The cyanidin content increased approximately 52-61% and delphinidin contents also increased 20-24% and 39-41% in tomato plants treated with SA alone and sorbitol with SA, respectively. Furthermore, the sole or combined application of sorbitol and SA also increased the vitamin $B_1$ and C contents. In particular, vitamin C content was increased 1.2-fold by treatment with sorbitol alone and was increased 2-fold by SA treatment compared to the control. In the sorbitol with SA treatments, vitamin C content was approximately 4-fold increased relative to control plants.

Establishment of Pre-Harvest Residue Limits of Clothianidin and Thiacloprid in Ginseng (인삼 중 Clothianidin 및 Thiacloprid의 생산단계 농약잔류허용기준 설정)

  • The residue patterns of clothianidin and thiacloprid, insecticides registered in the ginseng, were investigated to predict pre-harvest residues limits (PHRL). Pesticides were treated under Korea GAP (Good Agricultural Practices) with the recommended dose (single dose) and twice of recommended dose (double dose). Samples were collected 11 times over 42 days (each 0, 2, 5, 8, 12, 16, 20, 24, 28, 33, 42 days after treatment). Residues of clothinidin and thiacloprid were analyzed by UPLC/TQD. Biological half-life of clothinidin in single dose and double dose were 14.6 days and 10.2 days and that of thiacloprid were also 9.7 days and 11.2 days, respectively. The PHRL of ginseng on 10 days before harvest was 0.3 mg/kg in clothianidin and 0.18 mg/kg in thiacloprid.

Quality characteristics of semi-dried persimmons soaked in different concentration of sugar solution (당액침지에 따른 반건시 가공품의 품질특성)

  • This study was conducted to investigate the quality and characteristics of semi-dried persimmons soaked in different concentrations of sugar solution and stored at $7^{\circ}C$ for 20 days. The L value and chroma value were significantly higher in S5 and S10 compared to the other concentrations of sugar solution. The ${\Delta}E$ and browning degree were increased according to the increase in concentration of sugar solution. Total sugar, reducing sugar, and sugar free contents were higher in the control (semi-dried persimmon) than those in S0, but they increased according to the increase in concentration of sugar solution. Polyphenol oxidase and peroxidase activities were decreased according to the increase in sugar solution concentration, which were highest in S0 among other semi-dried persimmons soaked in sugar solutions. Total ascorbic acid content was highest in S10 (12.29 mg/g), followed by S0 (2.54 mg/g), S5 (7.76 mg/g), S15 (6.05 mg/g), and S20 (5.05 mg/g). Total polyphenols, flavonoids and proanthocyanidins contents were the highest in S10 compared to other semi-dried persimmons soaked in sugar solutions. Furthermore, the same tendency was observed with DPPH radical scavenging ability. These results showed that 10% sugar solution could be applied to semi-dried persimmons in order to achieve high quality, nutritional value, and browning inhibition.

Autopoietic Machinery and the Emergence of Third-Order Cybernetics (자기생산 기계 시스템과 3차 사이버네틱스의 등장)

  • First-order cybernetics during the 1940s and 1950s aimed for control of an observed system, while second-order cybernetics during the mid-1970s aspired to address the mechanism of an observing system. The former pursues an objective, subjectless, approach to a system, whereas the latter prefers a subjective, personal approach to a system. Second-order observation must be noted since a human observer is a living system that has its unique cognition. Maturana and Varela place the autopoiesis of this biological system at the core of second-order cybernetics. They contend that an autpoietic system maintains, transforms and produces itself. Technoscientific recreation of biological autopoiesis opens up to a new step in cybernetics: what I describe as third-order cybernetics. The formation of technoscientific autopoiesis overlaps with the Fourth Industrial Revolution or what Erik Brynjolfsson and Andrew McAfee call the Second Machine Age. It leads to a radical shift from human centrism to posthumanity whereby humanity is mechanized, and machinery is biologized. In two versions of the novel Demon Seed, American novelist Dean Koontz explores the significance of technoscientific autopoiesis. The 1973 version dramatizes two kinds of observers: the technophobic human observer and the technology-friendly machine observer Proteus. As the story concludes, the former dominates the latter with the result that an anthropocentric position still works. The 1997 version, however, reveals the victory of the techno-friendly narrator Proteus over the anthropocentric narrator. Losing his narrational position, the technophobic human narrator of the story disappears. In the 1997 version, Proteus becomes the subject of desire in luring divorcee Susan. He longs to flaunt his male egomaniac. His achievement of male identity is a sign of technological autopoiesis characteristic of third-order cybernetics. To display self-producing capabilities integral to the autonomy of machinery, Koontz's novel demonstrates that Proteus manipulates Susan's egg to produce a human-machine mixture. Koontz's demon child, problematically enough, implicates the future of eugenics in an era of technological autopoiesis. Proteus creates a crossbreed of humanity and machinery to engineer a perfect body and mind. He fixes incurable or intractable diseases through genetic modifications. Proteus transfers a vast amount of digital information to his offspring's brain, which enables the demon child to achieve state-of-the-art intelligence. His technological editing of human genes and consciousness leads to digital standardization through unanimous spread of the best qualities of humanity. He gathers distinguished human genes and mental status much like collecting luxury brands. Accordingly, Proteus's child-making project ultimately moves towards technologically-controlled eugenics. Pointedly, it disturbs the classical ideal of liberal humanism celebrating a human being as the master of his or her nature.

Multi User-Authentication System using One Time-Pseudo Random Number and Personal DNA STR Information in RFID Smart Card (RFID 스마트카드내 DNA STR Information과 일회용 의사난수를 사용한 다중 사용자 인증시스템)

  • Thia paper suggests a milti user-authentication system comprises that DNA biometric informatiom, owner's RFID(Radio Frequency Identification) smartcard of hardware token, and PKI digital signqture of software. This system improved items proposed in [1] as follows : this mechanism provides one RFID smartcard instead of two user-authentication smartcard(the biometric registered seal card and the DNA personal ID card), and solbers user information exposure as RFID of low proce when the card is lost. In addition, this can be perfect multi user-autentication system to enable identification even in cases such as identical twins, the DNA collected from the blood of patient who has undergone a medical procedure involving blood replacement and the DNA of the blood donor, mutation in the DNA base of cancer cells and other cells. Therefore, the proposed system is applied to terminal log-on with RFID smart card that stores accurate digital DNA biometric information instead of present biometric user-authentication system with the card is lost, which doesn't expose any personal DNA information. The security of PKI digital signature private key can be improved because secure pseudo random number generator can generate infinite one-time pseudo randon number corresponding to a user ID to keep private key of PKI digital signature securely whenever authenticated users access a system. Un addition, this user-authentication system can be used in credit card, resident card, passport, etc. acceletating the use of biometric RFID smart' card. The security of proposed system is shown by statistical anaysis.

Flavonoid Metabolic Engineering for Modification of Flower Color in Chrysanthemum (국화 꽃색 변경을 위한 플라보노이드 대사공학)

  • In ornamental crops, the color and shape of flowers are one of the important traits. Generally, flower colors are determined by accumulating pigments such as carotenoids, flavonoids, and betalains. Among them, flavonoids are responsible for broad ranges of colors. Chrysanthemums are one of the most popular ornamental crops in the world, and there have been many efforts to change their flower color. In chrysanthemum flowers, cyanidin-based anthocyanin confers pink or red color, whereas terpenoid-based carotenoids are mainly responsible for yellow and green colors. However, blue colored chrysanthemums do not occur in nature. To date, there have been attempts to obtain blue or violet-colored chrysanthemum flowers through the introduction of a novel gene for accumulating delphinidin-based anthocyanins, while other studies have reported changing endogenous metabolites through the reconstruction of flavonoid biosynthesis. Since various transcription factors are involved in the regulation of flavonoid biosynthesis, it is important to understand not only the structural genes, but also the transcription factors required for the modification of flavonoid-based flower color. Therefore, in this paper, we describe the flavonoid biosynthetic pathway and its regulation, and review previous studies on the change in flower color through modification of flavonoid biosynthesis. This effort could be an important milestone in successfully achieving the modification of chrysanthemum flower color by means of plant biotechnology.

Interaction with Polyphenols and Antibiotics (폴리페놀 화합물과 항생제의 상호작용)

  • Polyphenols are secondary metabolites produced by higher plants and have been used as antiallergic, anticancer, antihypertensive, antiinflammatory, antimicrobial and antioxidant agents. They are generally divided into flavonoids and non-flavonoids. The antimicrobial activity of flavonoids are stronger than that of non-flavonoids. The skeleton structures of flavonoids possessing antimicrobial activity are chalcone, flavan-3-ol (catechin), flavanone, flavone, flavonol and proanthocyanidin. The flavonols are shown antibacterial activity against several gram-positive bacteria (Actinomyces naeslundii, Lactobacillus acidophilus and Staphylococcus aureus) and gram-negative bacteria (Fusobacterium nucleatum, Porphyromonas gingivalis, Prevotella melaninogenica and Prevotella oralis). Among of non-flavonoids, caffeic acids, ferulic acids and gallic acids showed antimicrobial activity against gram-positive (Listeria monocytogenes and S. aureus) and gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa). These are found to be more efficient against the E. coli, L. monocytogenes, P. aeruginosa and S. aureus than antibiotics such as gentamicin and streptomycin. The kaempferol and quercetin showed synergistic effect with ciprofloxacin and rifampicin against S. aureus and methicillin resistant S. aureus (MRSA). Epigallocatechin gallate (EGCG) acts synergistically with various ${\beta}-lactam$ antibiotics against MRSA. In particular, the epicatechin, epigallocatechin (EGC), EGCG and gallocatechin gallate from Korean green tea has antibacterial activity against MRSA clinical isolates and the combination of tea polyphenols and oxacillin was synergistic for all the clinical MRSA isolates.

Biological activities in Aronia melanocarpa depending on drying methods (건조방법에 따른 아로니아의 생리활성)

  • To investigate biological activities in Aronia melanocarpa various drying methods were employed such as vacuum freeze drying, hot air drying and cold air drying. DPPH radical scavenging activity and ABTS radical scavenging activity of vacuum freeze dried Aronia melanocarpa was higher than hot and cold air dried Aronia melanocarpa. Vacuum freeze drying method showed the greatest contents of total phenol (15.34 g GAE/100 g), flavonoid (3.10 g GE/100 g) and tannin (2.46 g TE/100 g). Total anthocyanin content decreased to 163.52 mg C3G/100 g and 50.15 mg C3G/100 g for hot and cold air drying, respectively. Vacuum freeze-dried method increased the total anthocyanin content (743.09 mg C3G/100 g) when compared with fresh Aronia melanocarpa (163.52 mg C3G/100 g). Total proanthocyanidin content of vacuum freeze dried Aronia melanocarpa has increased to 6.21 g CE/100 g more than eight times compared with fresh Aronia melanocarpa (0.71 g CE/100 g). Chlorogenic acid and neochlorogenic acid content of vacuum freeze dried Aronia melanocarpa were higher than hot air dried and cold air dried Aronia melanocarpa, increasing about three times compared with fresh Aronia melanocarpa. These results suggested that vacuum freeze drying is optimal drying method to enhance biological activities in Aronia melanocarpa.
