Protein Hydrolysis with Formic Acid and Analysis of Amino Acid Using Butylthiocarbamyl - trimethylsilyl (BTC - TMS) Derivatives by Gas Chromatography

  • The protein hydrolysis with 6 M formic acid containing 0.3% tryptamine was a superior method for amino acid analysis of standard amino acid and protein than 6 M HCI containing 0.3% tryptamine. The recoveries of standard amino acid after acid hydrolysis were more accurate in the 6 M formic acid hydrolysis than 6 M HCI hydrolysis, especially recovery of tryptophan showed higher values of 1.5 times than that of 6 M HCI hydrolysis. The results of analysis on the standard protein, bovine serum albumin, showed very similar values compared to the sequence analysis reported in the literature for the 6 M formic acid hydrolysis than 6 M HCI hydrolysis, especially in the tryptophan recovery as standard amino acid recovery. Butylthiocarbamyl - trimethylsilyl (BTC - TMS) derivatives of 22 standard amino acids were successfully resolved DB-17 capillary column. Excellent reproducibility of standard amino acid recovery and composition of bovine serum albumin were obtained with BTC-TMS derivatives.

  • A kinetic model for evaluating effects of surfactant on the biodegradation of HOC(hazardous organic chemicals) in soil-slurry systems was developed. The model includes the partition of HOC and surfactant, the dissolved-, micellar-, and sorbed-phase biodegradation, the enhanced solubilization of HOC by surfactant addition, and the mass transfer of HOC. Phenanthrene as HOC and Trition X-100, Tergitol NP-10, Igepal CA-720, and Brij 30 were used in the model simulations. The biodegradation rate was increased even with a small micellera-phase bioavailability. The biodegradation was not greatly enhanced due to decreased aqueous HOC concentration by increasing surfactant dose in both cases with and without micellar-phase bioavailability. The effect of sorbed-phase biodegradation on total biodegradation rate was not highly important compared to aqueous- and micellar-phase biodegradation. The model can be applied for surfactant screening and optimal design of surfactant-based soil bioremediation process.

Biodegradation of Pyrene in Marine Environment (해양환경에서 Pyrene의 생분해)

  • The biodegradation of recalcitrant polycyclic aromatic hydrocarbon, pyrene was investigated in microcosm simulating the beach sand and seawater. The natural biodegradation rates of pyrene were between 30-2,200 ng/g(ml)/day in beach sand and seawater when the pyrenc loading rates were 100- 1,000 ppm at 5-$20^{\circ}C$. The effects of the inoculum size, pyrene concentralion, incubation temperature and surfactant addition were investigated in fertilized (Inipol EAP 22) samples. Generally the biodegradation in beach sand was higher than that in seawater. A mixed inoculum (Pseudomonus, Acinetobacter, Moruxella) showed the 3,120 nglglday of biodegradation rate in beach sand with 200 ppm pyrene, which was 7.8 times higher than the natural biodegradation rate. The highest transformation rate, 4,860 ng/g/day was obtained in the bioaugmented beach sand (1,000 ppm pyrene). The glucose and surfactant addition to enhance the removal have negatively influenced on the biodegradation of pyrene. In case ol surfactants, CMC (critical micell concentration) might bc the control factor for the biodegradation.

  • Copyrolysis of biomass/plastic mixture was carried out from room temperature to $600^{\circ}C$ with varing the heating rates of 10, 20, and $30^{\circ}C/min$ using a thermogravimetric analyzer. Waste wood chip (WWC) and linear low density polyethylene (LLDPE) were selected as a biomass and plastic, respectively. Individual pyrolysis temperature ranges were $430{\sim}550^{\circ}C$ and $230{\sim}600^{\circ}C$ for LLDPE and WWC, respectively. For the copyrolysis of WWC and LLDPE, the decomposition temperature range of WWC was not varied, while the decomposition temperature range of LLDEP was increased to a higher temperature. The results imply that the interaction might occur between LLDEP and WWC during copyolyis of LLDPE and WWC.

  • The changes of nitrogen utilization ratio (NUR), Protein solution ratio (PSR) and free sugar contents during the amino acid soysauce manufacturing process by a low hydrochloric acid, temperature were investigated. On hydrolysis by 6%-HCI (3 liquor rate of defatted soybean weight, 3LR) at $85^{\circ}C$, NUR and PSR were 74.51%, 56.49% at 65 hours. At the same time free sugars were detected glucose, galactose, arabinose, fructose, xylose. on hydrolysis at $95^{\circ}C$, NUR and PSR were 77.72%, 64.04% at 50 hours, and 5 free sugars of the above statement were detected at 5 hours. Remarkable decreases in the levels of free sugars, only glucose were observed after 80 hours of the hydrolysis. On hydrolysis by 12%-HCI(3LR) at $95^{\circ}C$, NUR and IRA were 88.41%, 69.47% at 50 hours, free sugar were nearly disappeared after 20-35hours. On hydrolysis, galactose's disappearence rate was faster than glucose's.

  • Picosecond time-correlated single photon counting system and time-resolved luminescence spectrometer were constructed, employing a mode-locked picosecond laser, fast electronics, and microchannel plate tube. It has been shown that the instrument response function critically depends on laser pulse shape, timing jitter and walk of the electronics, and characteristics of detector and amplifier. Correcting time dispersion in the optical system, the best instrument response function obtained appears to be 25 ps, which made it possible to measure the luminescence lifetime with less than 10 ps resolution in the picosecond to microsecond range. range.

  • In this study, two methods to evaluate the disassemblability of the parts are proposed, which are constructed from the concepts of ease-of-disassembly and disassembly time. A method to evaluate the disassemblability of the products is consequently developed using the concept of minimum parts. The methods are applied to evaluate the disassemblability of computer cases. The results show the conclusions that the new model has less disassemblability than the old one. These methods are expected to be used for the evaluation of the possibility of remanufacturing and recycling of a part.

배추 및 무의 Glucosinolate 분석과 가공처리중 항암물질의 형성

  • 십자화과 채소에는 여러종류의 glucosinolate가 존재하며 이들 중 특히 indolw형 glucosinolate는 여러가지 요인에 의해 분해되어 항암효과를 나타내는 물질들을 생성한다고 보고 되고 있다. 이에 다라 우리나라 농가에서 대량 생산되고 있는 십자화과 채소인 배추 및 무에서 항암전구물질인 indole glucosinolate를 포함한 glucosinolate를 분석 및 동정하고 가공처리조건에 따라 항암물질인 indoleacetonitrile과 indolemethanol의 함량을 측정하였다. 1. GC 및 GC-MS를 이용하여 배추, 무의 품종별 ; 이들 씨에서 4-8종류의 glucosinolate를 동정하였다. 2. Thymol에 의한 총 glucosinolate의 함량은 배추 및 무에서는 복돌이 배추와 왕관무가 3.21, 2.0$\nu$mole/g으로 가장 많았고 배추씨, 무씨에서는 각각 6.38, 4.07$\nu$mole/g이었다. 3. 배추 및 무의 저장중 총 glucosinolate함량은 저장 기간이 지남에 따라 점차 감소하는 경향이였다. 4. 열처리 시간에 따른 배추와 무의 indoleacetonitrile함량은 열처리 시간이 지남에 따라 증가하며 40분 열처리시 최대치를 나타내었다. 5. indolemethanol이 생성될때 방출되는 배추, 무, 배추씨 및 무씨의 Thiocyanate ion의 함량은 열처리 시간이 지남에 따라 증가하며 40분 열처리시 최대치를 나타내었으며 자가분해에 따른 함량도 분해시간이 경과함에 따라 증가하며 30분경과시는 최대치를 나타내었고 이는 40분 열처리시보다 많은 함량이였다.함량이였다.

  • This study was carried out to determine the effects of media compositions on cell growth and casein hydrolysis for cell production in order to add Micrococcus sp. LL3 as a potential agent for industrial application for the shortening of ripening period. Monosaccharides like glucose, mannose and fructose were mare excellent as carbon source, but arabinose and xylose markedly inhibited cell growth and caseinolysis. Among the organic nutrients, yeast extract was more effective for cell growth and for caseolysis. However, inorganic nitrogen sources were less effective than organic sources. Urea inhibited cell growth severely. Cell growth and caseinolysis were rather increased a little in the broth containing 1% NaCl, and the organism tolerated and grew in relatively high concentrations of NaCl up to 9%. Addition of vitamin did not affect cell growth and caseolysis in level of $0.1\;{\mu}g/ml$ concentration. Cell growth and caseinolysis were stimulated by addition of glutamic acid and $MgSO_4$ with concentration of 0.2% and 0.05% respectively.
