Effects of Thermal and Salinity Stress on Expression of FK506BP in the Red Seabream (Pagrus major) (참돔(Pagrus major)에서 온도 및 염분 스트레스가 FK506BP 발현에 미치는 영향)

  • FK506BP (FK506 binding protein 12) is a small peptide with a single FK506BP domain. It is involved in suppression of immune response, oxidative stress and inflammation. The purpose of this study was to investigate the gene expression of FK506BP in red seabream (Pagrus major) exposure to low water temperature (8℃, 33 psu) and low salinity (20℃, 10 psu). Results showed that, the expression of FK506BP was significantly increased in the experiment groups, such as low water temperature (8℃, 33 psu), and low salinity (20℃, 10 psu). These results suggest that FK506BP was played roles in biomarker gene on the environmental stress such as water temperature and salinity.

Protective Effect of Plantago asiatica L. Extract Against Ferric Nitrilotriacetate (Fe-NTA) Induced Renal Oxidative Stress in Wistar Rats (차전초 추출물을 투여한 랫드에서의 Fe-NTA 유발 산화스트레스에 대한 신장보호 효과)

  • Plantago asiatica L. (PA), which is widely distributed in Korea, Japan and China, has traditionally been used as a popular folk medicine for the treatment of liver diseases. A variety of activities of PA was reported, that is hepatoprotective, anti-inflammatory, anti-glycation and anti-oxidant effect. Ferric nitrilotriacetate (Fe-NTA) is a potent nephrotoxic agent and has been reported to induce renal proximal tubular necrosis. In the present study, pre-treatment with PA extract (PAE) in Wistar rat followed by Fe-NTA i.p. treatment (13.5 mg Fe/kg body weight) was performed to detect the renal protective effect of PAE. Only Fe-NTA treated group showed increases in the level of serum blood urea nitrogen (BUN) and serum creatinine (Cr), and renal tissue malondialdehyde (MDA), product of lipid peroxidation. Moreover, the level of biomarkers indicate the antioxidants status, reduced glutathione (GSH), glutathione-S-transferase (GST) and glutathione reductase (GR) were decreased. However, PAE pre-treated group showed decreases in the levels of serum BUN, serum Cr and renal tissue MDA in concentration dependent manner and increases in the level of GSH, GST and GR. These results are significantly different (p < 0.05) to the other groups. Our data suggest that PAE may be used as an chemopreventive material against Fe-NTA-mediated renal oxidative stress.

Effect of Tremella fuciformis Berk on Anti-stress activities during Long-Term and Short-Term in Mice (마우스에서 흰목이버섯 (Tremella fuciformis Berk)의 장.단기적 항스트레스 효과)

  • The objective of this study was to evaluate the protective effect of aqueous extracts from Tremella fuciformis Berk(Tf AE) against stress during long-term and short-term in ICR mice. All the animals were randomly divided into two groups which had been bred for 5 months that were treated by immobilization stress for 8 weeks (total 7 months breeding, equivalent to human beings aged 20) with or without Tf AE, and one out of two groups was continuously bred until they become 18 months old (equivalent to human beings aged 60) without Tf AE. Afterwards, the changes of serum and hepatic metabolites were investigated on the basis of the index of stress-related in vivo oxidative damage. As a result, it was found that stress increases serum triglyceride (TG) and aspartate aminotransferase (AST) and decreases serum HDL-cholesterol in the long-term (total 18 months breeding) and short-term (total 7 months breeding). In addition, stress concerned the decrease of total antioxidant status (TAS) and superoxide dismutase (SOD) as well as the increase of malondialdehyde (MDA) in liver. On the other hand, Tf AE-fed groups reversed all these biochemical indices. These results suggest that stress in one's youth causes negative results in TG, HDL-cholesterol, LDL-cholesterol, AST, TAS, SOD and MDA measured in one's senescent. The administration of Tf AE in the stressed mice decreases serum TG and AST that are increased by stress, and exerts influence on the increase of serum HDL-cholesterol. Also Tf AE recovered the values of liver TAS, SOD and MDA in the stressed mice. In conclusion, Tf AE represented protective effect in the stressed mice to some degree.

Protective Effect of Functional Perilla frutescens Hot-water Extract Against tert-butyl hydroperoxide-Induced Liver Oxidative Damage in Rats (랫드에서의 t-BHP 유발 산화스트레스에 대한 기능성 들깻잎 열수 추출물의 간 보호 효과)

  • Perilla frutescens usually dieted in East Asian country such as Korea and Japan. Antioxidant, antiinflammatory and anticancer activities of perilla leaves have been founded. In previous study, we confirmed that caffeic acid, major compound of perilla, was accumulation by sucrose aqueous solution and thus antioxidant effect of perilla was enhanced. In this study, we investigated the protective effect of functional perilla leaves extract (PLE) against tert-butyl hydroperoxide(t-BHP) induced-oxidative hepatotoxicity. The pretreatment with PLE (250, 500 and 1000 mg/kg b.w.) for 5 days before a single dose of t-BHP (i.p.; 0.5 mmol/kg) significantly lowered the serum levels of aspartate aminotransferase, alanine aminotransferase and lactate dehydrogenase dose-dependently. And we confirmed that the indicators of oxidative stress were remarkably reduced in the liver, such as the glutathione contents and malondialdehyde, marker of lipid peroxidation. Pathological histology of the rat livers tissues showed that PLE reduced the hepatocyte degeneration and neutrophilic infiltration of liver induced by t-BHP. These results suggest that functional perilla frutescens has the protective effect of liver against t-BHP-induced oxidative hepatic stress in rats.

  • Evaporative Demand Drought Index (EDDI)는 미국해양대기관리처 (National Oceanic and Atmospheric Administration, NOAA)에서 2016년에 개발한 가뭄지표로, 기존의 가뭄지표가 주로 강수량과 기온에 초점을 두고 가뭄을 판단하는 반면 토양의 수분 스트레스의 신호를 바탕으로 증발산수요의 상대적인 변화를 계산하여 가뭄지표에 대한 조기 경보를 제공한다. EDDI는 강수량을 이용한 기존의 가뭄지수와 달리 증발/산 요구량 (evaporative demand)에 초점을 맞춰 보다 짧은 시간의 척도와 공간 분포 및 시계열 결과의 도출로 잠재적 가뭄 예보에 활용할 수 있어 가뭄의 조기 경보 및 가뭄 모니터링 도구로 사용할 수 있다. 현재 NOAA에서는 EDDI Map Archive(https://www.esrl.noaa.gov/psd/eddi/)를 활용하여 1980년부터 현재까지 1-week부터 12-months 시간척도의 미국 전역의 EDDI 지도를 제공하고 있으며, 짧은 기간의 급속하게 발생하는(rapid-onset) Flash drought의 조기경보지표로 활용되고 있다. 본 연구에서는 최근 3년간 우리나라에 발생한 극심한 가뭄 사상을 대상으로 EDDI의 적용함으로서 시공간적 가뭄 특성을 파악하고자 한다.

Evaluation on the stress using HRV according to elapsed time of MRI noise (HRV를 이용한 자기공명영상 소음의 시간 변화에 따른 스트레스 평가)

  • The noise of MRI shooting is 100dB loud and has an intensive psychological and physiological influences on the human body. ECG signals were measured by experimental methods, while wearing earplugs for 15 minutes in the stable state. Then the ECG signals were measured for 30 minutes while listening to about 100dB of sound in a MRI equipment. In this study, the heart rate variability of men and women was analyzed according to the MRI noise stress level through the frequency analysis. As the MRI noise level is about 100dB, HRV analysis resulted in an imbalance between the sympathetic and parasympathetic. During the period from the resting state up to 10 minutes, the maximum stress state was shown. This study will encourage MRI workers to take interests in hearing protection for the patient and to make objective indicators about MRI noises.

Anti-stress Effect during Long and Short-Term of Vitamin E in Mice (마우스에서 vitamin E의 장.단기적 항스트레스 효과)

Effects of Rosa multiflora Yoghurt on Sociopsychological Stress (사회 . 심리적 스트레스에 미치는 장미(Rosa multiflora) 요구르트의 영향)

  • The effects of anti-stress rose (Rosa mutiflora) fruit extract yoghurts (RFEY-1.0, RFEY-3.0, RFEY-5.0 containing with 1.0, 3.0 and 5.0% of rose fruit extract) were tested for the anti-stress effects. ICR male mice (20$\pm$2 g) were fed with basic experimental diets and given free access to water containing these ingredients for 18 days. Psychological stress and sociopsychological stress exposed by foot-shock for 1 hour (10 sec duration at intervals of 120 sec) every day for 3 days. RFEY-1.0, RFEY-3.0, RFEY-5.0 groups in the sociopsychological stress resulted in a significant decrease of 11.7%, 16.0% and 24.7% in plasma corticosterone (CS) secretion compared with psychological stress (control group). Noradrenaline (NA) secretions in the brain were significantly increased 15.6%, 25.0% and 40.8%, respectively, in RFEY-1.0, RFEY-3.0, RFEY-5.0 groups compared with control group. MHPG -SO$_4$ (3-methoxy-4-hydroxy-phenylethyleneglycol sulfate) levels in the brain resulted in a marked decreases of 17.0%, 25.3% and 28.4%, respectively, in RFEY-1.0, RFEY-3.0, RFEY-5.0 groups compared with control group. NA/MHPG-SO$_4$ ratios in the brain of RFEY-1.0, RFEY-3.0, RFEY-5.0 groups resulted in a significantly increase of 39.5%, 67.3% and 96.3%, respectively, compared with control group. These results suggest that rose fruit extract yoghurt may be tried to apply for human consumption such as sociopsychological stress.

Multiple-biometric Attributes of Biomarkers and Bioindicators for Evaluations of Aquatic Environment in an Urban Stream Ecosystem and the Multimetric Eco-Model (도심하천 생태계의 수환경 평가를 위한 생지표 바이오마커 및 바이오인디케이터 메트릭 속성 및 다변수 생태 모형)

  • The objectives of the study were to evaluate the aquatic environment of an urban stream using various ecological parameters of biological biomarkers, physical habitat quality and chemical water quality and to develop a "Multimetric Eco-Model" ($M_m$-E Model) for the ecosystem evaluations. For the applications of the $M_m$-E model, three zones including the control zone ($C_Z$) of headwaters, transition zone ($T_Z$) of mid-stream and the impacted zone ($I_Z$) of downstream were designated and analyzed the seasonal variations of the model values. The biomarkers of DNA, based on the comet assay approach of single-cell gel electrophoresis (SCGE), were analyzed using the blood samples of Zacco platypus as a target species, and the parameters were used tail moment, tail DNA(%) and tail length (${\mu}m$) in the bioassay. The damages of DNA were evident in the impacted zone, but not in the control zone. The condition factor ($C_F$) as key indicators of the population evaluation indicator was analyzed along with the weight-length relation and individual abnormality. The four metrics of Qualitative Habitat Evaluation Index (QHEI) were added for the evaluations of physical habitat. In addition, the parameters of chemical water quality were used as eutrophic indicators of nitrogen (N) and phosphorus (P), chemical oxygen demand (COD) and conductivity. Overall, our results suggested that attributes of biomarkers and bioindicators in the impacted zone ($I_Z$) had sensitive response largely to the chemical stress (eutrophic indicators) and also partially to physical habitat quality, compared to the those in the control zone.

Influence of metabolizable energy on blood biomarkers in duck under heat stress (대사에너지가 열 스트레스에 노출된 오리의 혈액 생체지표에 미치는 영향)

  • The objective of this study was to determine the influence of dietary metabolic energy (ME) on blood parameters in duck under heat stress. A total of 240 meat ducks Cherry valley (Anas platyrhynchos) were assigned into four treatment groups with a randomized block design for 42 days. The four treatments were: ME 2900 kcal/kg, ME 3000 kcal/kg, ME 3100 kcal/kg, and ME 3200 kcal/kg. Blood lipid profiles was higher in ME 2900 but lower in ME 3100 and ME 3200 than that of ME 3000 (p < 0.05). Blood aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels were higher in ME 3100 and ME 3200 compared those in ME 3000 (p < 0.05). The blood red cell and platelet profiles were increased in ME 3100 and ME 3200, but reduced in ME 2900 compared to those in ME 3000 (p < 0.05). Among blood electrolytes, chloride ($Cl^-$) concentration was decreased in ME 2900 compared to that in ME 3000. Blood gas $PCO_2$ was reduced in ME 2900 compared to that in ME 3000 (p < 0.05). Blood immunoglobulin (IgG) level was reduced in ME 2900 compared to that in ME 3000 (p < 0.05). Level of stress hormone, corticosterone was increased in ME 2900, but decreased in ME 3100 and ME 3200 compared to that in ME 3000 (p < 0.05).
