The Components of the Sap from Gorosoe(Acer mono Max.) and Sugar Maple(Pseudo-sieboldianum Kom.) (고로쇠나무 및 당단풍나무 수액의 성분조성)

  • Even though the saps have been consumed as beverage in Korea for a very long period of time, a little research has been conducted on the chemical composition. We determined mineral, free sugar, composition amino acids and nucleotides in the sap of Gorosoe(Acer mono Max.) and Sugar Maple(Pseudo-sieboldianum Kom.) collected from Sancheong, Kurye and Hamyang. The contents of solid, crude protein and ash were 1.1~3.3%, 0.03~0.12% and 0.02~0.05% in the sap of Gorosoe and Sugar Maple, respectively. The detectable nucleotides were CMP, UMP, IMP, GMP, AMP and hypoxanthine. Free sugar that we determined in the sap of Gorosoe and Sugar Maple were sucrose, fructose and glucose, but maltose was not detected. The chief component of saccharides in the sap was sugar and ranged from 7.1 to 36.6g/L. Especially, the sucrose contents in the sap of Gorosoe and Sugar Maple from Hamyang were higher 3.7 and 2.2 times than those from the other samples. The prominent minerals in the sap were calcium and potassium. The calcium concentration was ranged from 99.0~153.3mg/L in the sap of Gorosoe and was ranged from 21.2~32.4mg/L in that of sugar maple. The sap of Gorosoe and Sugar Maple were composed of 18 and 15 kinds of amino acid respectively, and the total contents of amino acid were in the range of 0.7~29.3mg%. The major amino acids in the sap of Gorosoe were taurine, serine, glutamic acid and aspartic acid in decreasing order. Taurine contents in the sap of Gorosoe and Sugar Maple was found to be the dominant amino acid.

Effects of UV Sterilization on Quality of Acer mono Sap (자외선 살균이 고로쇠 수액의 품질에 미치는 영향)

  • An ultraviolet (UV) sterilization system was developed to decrease the number of microorganisms in filtered Acer mono sap (FAS). The Acer mono sap (AS) was passed through 18 strips of PTA fluoroplastic tubing with 30 UV lamps (total 1,170 W). During passage, the AS was effectively exposed to the UV rays. The total bacteria, coliform group and fungi were sterilized at a flow rate ranging from 852 to 1,358 mL/min and a UV power higher than 156 W. Although the sensory score of the UV-sterilized AS was significantly lower than that of the untreated AS, the sensory score was better under the condition of 390 W and 852~1,358 mL/min than under the other conditions examined. ANOVA did not reveal a significant difference in pH, total acidity, sugar content and color characteristic under all conditions tested (p<0.05). Considering the overall attributes of the AS quality, the optimum sterilization condition was determined to be 390 W and 852~1,358 mL/min.

Effect of shelf-life extension of birch sap using high pressure processing (초고압 기술이 자작나무 수액의 저장성 향상에 미치는 영향)

  • Effects of high pressure processing on physicochemical and microorganisms properties in birch sap were investigated using variable high pressure processing conditions. The viable cell counts of untreated birch sap was 4.0 log CFU, whereas high pressure processed sap were not detected. In birch sap was treated with 450 to 550 MPa, microorganisms were not detected during storage period, and physicochemical properties as well as color were slightly changed. The more processing time and pressure, its quality variations were more stable and then its optimum processing condition was determined with 120 sec at 550 MPa. The microorganisms and physicochemical properties of treated birch sap were investigated during storage at $5^{\circ}C$ and $10^{\circ}C$ for 45 and 28 days. Changes of physicochemical properties of treated birch sap were smaller than those of the untreated, but viable cell count were not detected during storage period. As for pH, $^{\circ}Brix$, and turbidity result of birch sap, quality shelf life of control and treatment were 4 and 24 days, respectively. Especially, ${\Delta}E$ value of instrumental color was untreated birch sap 4 days similar with the high pressure processed it for 28 days. These results indicated that the high pressure processing can be used as an effective method to improve the shelf life of birch sap.

The Antimicrobial Activity of Fermented Extracts from Korean Dendropanax morbifera (국내 황칠나무 발효 추출물의 항균력 평가)

  • We investigated the fermentation conditions for extracts of leave/branches and sap from Korean Dendropanax morbifera (D. morbifera) using Lactobacillus plantarum (L. plantarum) ilchiwhangchil 1785 and L. plantarum ilchiwhangchil 2020. Log growth phase cultured L. plantarum ilchiwhangchil 1785 and L. plantarum ilchiwhangchil 2020 were used for fermentation. The pH and growth of the microorganisms in broth were monitored during the fermentation period. The results revealed that the optimum fermentation conditions for 20 wt% of leave/branches extracts and 1 wt% of sap extract was 2 days incubation at $37^{\circ}C$. The minimum inhibitory concentration (MIC) method and a disk diffusion assay were used to evaluate the antimicrobial activity of the fermented extracts of the leave/branches and sap against Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa. The antimicrobial activity increased in all three strains grown on the medium containing fermented extracts of the leave/branches and sap as compared with that of the strains grown on medium containing non fermented extracts. Furthermore, the antimicrobial activity increased in proportion to the contents of the fermented extracts. Our data suggest that fermented extracts of leave/branches and sap of D. morbifera have applications as natural bio functional materials, such as preservatives, cosmetic materials, and natural packaging materials.

Physicochemical Characteristics and Nutritional Components of Goroshoe (Acer mono Max.) Sap with Collection Periods (고로쇠 수액의 유출시기별 이화학적 특성과 영양성분)

  • This study evaluated the physicochemical characteristics and nutritional components of Goroshoe (Ace rmono Max.) sap related with collection periods. The changes of browning index, turbidity, pH, total acidity, organic acids, free sugar, crude protein, crude ash, and minerals were investigated. Browning index and turbidity were slightly increased, while pH and total acidity were not changed as collection periods increased. Citric acid decreased slightly from 0.081 to 0.045 mg/mL at middle collection periods. Malic acid also decreased from 0.494 to 0.416 mg/mL at middle collection periods but then afterward increased to 0.674 mg/mL as collection periods increased. Sucrose decreased from 1.759 to 1.000%, while fructose increased from 0.023 to 0.044% as collection periods increased. Crude protein increased remarkably from 0.80 to 17.49 mg% as collection periods increased. Crude ash decreased slightly from 0.04 to 0.03% at middle collection periods, and then increased afterward to 0.06% as collection periods increased. Potassium increased from 13.43 to 49.75 mg/L as collection periods increased. These results indicate that the useful components of Goroshoe sap increased with increasing collection periods, but turbidity could be reduced.

Storage-life and Palatability Extension of Betula platyphylla Sap Using Lactic Acid Bacteria Fermentation (유산균 발효를 이용한 자작나무 수액의 저장성 및 기호성 증진 기술)

  • In this study, a new method for extending storage-life and palatability of Betula platyphylla sap by applying lactic acid bacteria fermentation was developed. The fluids of saps were filtered through 0.22 ${\mu}m$ membrane filter and each fermented by 8 different lactic acid bacteria which are Lactobacillus acidophilus, Lactobacillus brevis, Leuconostoc mesenteroides, Leuconostoc lactis, Lactococcus lactis, Pediocossus pentosaceus, Pediococcus dextrinicus, Streptococcus thermophilus. All the tested lactic acid bacteria except P. dextrinicus grew fast up to $10^6{\sim}10^7cfu/mL$ levels and lowered pH down to about pH 4 levels in 48 hours in both saps. The produced organic acids and lowered pH level inhibited the growth of spoilage microorganisms almost completely for 2 weeks during storage at room temperature. Addition of xylitol in the saps before fermentation accelerated the growth of lactic acid bacteria and increased the sweetness and overall taste of final product. The filtration process did not affect the mineral compositions of Betula platyphylla saps. Also the compositions and amounts of minerals showed very minor differences before and after fermentation in Betula platyphylla saps inoculated with L. acidophilus. By applying lactic acid fermentation to extend storage-life of tree saps instead of heat treatment, it was possible to keep natural minerals in active forms without any modifications.

Stability of Aminophylline, Ceftriaxone Sodium and Ampicillin/Sulbactam in Total Parenteral Nutrient Solution (고영양수액제 중 아미노필린, 세프트리악손 및 암피실린/설박탐의 안정성에 관한 연구)

  • Aminophylline, ceftriaxone 및 ampicillin/sulbactam (Unasyn)을 미숙아용 고영양수액제에 직접 첨가하거나 Y-site로 투여하는 경우의 안정성에 관해 조사하였다. Aminophylline 주사액 (25mg/mL) $300{\mu}l$와 ceftriaxone sodium (37.5 mg/mL) 2mL를 각각 고영양수액제 직접 첨가하였다. 또한 Y-site에서의 안정성 조사를 위해 ceftriaxone sodium (37.5 mg/mL)을 고영양수액제에 각각 1:1 및 1:2 부피비가 되도록 혼합하였고 Unasyn (25mg/mL)은 고영양수액제와 1:1의 부피비로 혼합하였다. 이상과 같이 조제한 혼합액을 $25^{\circ}C$$4^{\circ}C$에 보관하여 aminophylline은 48시간 동안 그리고 항생제들은 24시간 동안의 경시변화를 HPLC를 이용하여 분석하였다. Aminophylline은 위 보존조건에서 48시간동안 안정하였다(변화율 <$10{\%}$). Ceftriaxone sodium을 고영양수액제에 직접 첨가한 경우 ceftriaxone의 잔존률은 $25^{\circ}C$에서 4시간째에 $90.5{\pm}1.8{\%}$이었고 $4^{\circ}C$에는 $95.1{\pm}1.4{\%}$로 측정되었다. Y-site에서의 안정성과 관련하여 ceftriaxone sodium을 고영양수액제와 1:1로 혼합한 경우 양 보존조건에서 ceftriaxone은 24시간 동안 안정하였으나 1:2로 혼합한 경우에는 $4^{\circ}C$ 보관 시에만 안정하였고 $25^{\circ}C$에서는 24시간째에 약 $14{\%}$ 정도 분해되는 것으로 나타났다. 한편, Unasyn의 경우 ampicillin은 24시간째에 $4^{\circ}C$ 보관 시에만 안정하였고 $25^{\circ}C$에서는 24시간째에 약 $14{\%}$정도 분해되는 것으로 나타났다. 한편, Unasyn의 경우 ampicillin은 24시간째에 $4^{\circ}C$에서는 안정하였으나 $25^{\circ}C$에서는 $30{\%}$까지 감소되는 것으로 분석되었고 sulbactam은 24시간째에 온도와 관계없이 안정한 것으로 나타났다. Ceftriaxone sodium을 TPN과 Y-site 혼합 후 1-2시간 이내에 침천이 형성되었고 Unasyn의 경우에는 12시간째에 침천이 형성되었다. 혼합액의 pH나 색상은 연구기간 동안 일정하였다. 이러한 연구결과에 기초할 때, aminophylline은 고영양수액제와 혼합가능하고 ceftriaxone과 Unasyn은 고양양수액제와 혼합시 최소 1시간 동안은 안정한 것으로 평가되었다.

Chemical Composition, Nutritional Value, and Saponin Content in the Spring Sap of Acer mono (고로쇠나무 수액(樹液)의 화학적(化學的) 성분(成分), 영양가치(營養價置)와 사포닌 함유(含有) 여부(與否)에 관(關)한 연구(硏究))

  • This study was conducted to analyze the chemical composition, nutritional contents, and saponin in the xylem sap of Acer mono Max. From Feb 25 to Mar 4, 1994, spring sap was collected by making holes with 1.7cm diameter on the trunk of trees in Mt. Baekwoon and Mt. Jiri. Sugars were quantified by HPIC, amino acids by amino acid analyzer, saponins by TLC, HPLC, and $^1H$ & $^{13}C$ NMR. Major component of the sap was sucrose at a concentration range of 0.68 to 2.01%. Following minor components were found: glucose at 0.03-0.11%, and fructose at 0.01-0.03% as sugars, lipid at 0.03%, threonine at 0.152%, lysine at 0.038%, arginine at 0.068% as amino acids, ash at 0.1%, Ca at 175ppm, Fe at 2ppm, P at 19ppm, K at 16ppm, Na at 31ppm as minerals, vitamin $B_1$ at 0.6ppm, vit. $B_2$ at 0.1ppm, and vit. C at 19ppm. A trace of phenolic compounds was found by TLC, while saponin commonly reported in high-quality ginseng roots was not found in maple sap. It is concluded that sap of Acer mono contains a good variety of natural compounds such as sugars, amino acids, Ca, Fe, and vitamins to serve as an excellent source of very natural and health-promoting drink.

Effect of Triglyceride Chain Length on the Stability of Total Nutrient Admixtures (고영양수액제의 안정성에 대한 트리글리세라이드의 측쇄 길이의 영향)

  • Background :긴 측쇄의 트리글리세라이드(long chain triglyceride, LCT)는 대두 또는 홍화씨로부터 제조된 지방유제의 주요 성분이다. 중 측쇄 트리글리세라이드(medium chain triglyceride, MCT)는 LCT와 다른 물리적 성질을 가지고 있다. MCT의 지방산은 LCT의 지방산 보다 작은 분자량을 가지기 때문에 물에 더 잘 녹으며 다른 경로를 통하여 더 빨리 대사된다. Objectives & Methods :본 연구에서는 지방산의 길이에 따른 고영양수액제(total nutrient admixture, TNA)의 안정성을 비교하기 위하여 서로 다른 LCT와 MCT 조성을 가지는 4 가지 TNA 처방의 안정성을 실온 및 냉장 조건에서 측정하였다. Results :그 결과 MCT와 LCT의 혼합 조성의 TNA는 LCT 단독 조성의 TNA 와 유사한 안정성을 나타내었다. MCT와 LCT 혼합 TNA는 LCT 단독 TNA에 비하여 작은 입자크기를 보였지만 외관, 산도, 과산화수소가, 삼투농도, 아연 농도, 니코틴아미드 및 리보플라빈 농도에서는 의미 있는 차이를 보이지 않았다. Conclusion : 따라서 MCT와 LCT의 혼합 TNA는 LCT 단독 조성 TNA에 상응하는 안정성을 가지고 LCT단독 TNA을 대체제로 사용할 수 있다. 결론적으로, TNA의 2가지 타입 모두 3일 동안 실온과 냉장에서 안정하고, MCT와 LCT의 혼합 TNA는 LCT 단독 조성 TNA보다 생리화학적으로 더 안정하므로 환자들에게 좀 더 효과적인 에너지원이 될 것으로 전망된다.

Silk Degumming by Electrolyzed Alkaline Water (전해 알칼리수에 의한 견의 정련)

  • This studies were carried out to develop the new silk degumming method by elelectrolyzed alkaline water on the silk degumming process. Using to this method, it could be collect pure sericin from the degumming water, because the degumming water by elelectrolyzed alkaline water was not contaminated by chemicals including soap. The range of elelectrolyzed alkaline water was pH 11.5 to 11.7 and maintained the first value for 8 days under the cool and closed conditions. The degumming ratio of silk was higher in elelectrolyzed alkaline water(pH 11.5~11.7)than that of soap and alkaline bath. When the pH value of elelectrolyzed alkaline water was adjusted at pH 11.0, the degumming ratio of silk was similar to that of soap and alkaline degumming. After degumming the pH value of degumming water decreased largely in the elelectrolyzed alkaline water compared to that of soap and alkaline bath. The tenacity and elongation of degummed silk by elelectrolyzed alkaline water was almost same those of soap and alkaline degumming for 90 min.
