Synthesis of Hydroxyapatite Using a Cationic Surfactant (양이온성 계면활성제를 이용한 수산화인회석 합성)

  • Hydroxyapatite (HAP) containing hexadecyltrimethylammonium chloride (CTAC) as a cationic surfactant was prepared by a precipitation method. X-ray diffraction (XRD), transmission electron microscopy (TEM) and micropore physisorption analyzer were used for characterizing the crystal phase, morphology and specific surface area of HAP and CTAC-HAP. After thermal treatment, the specific surface area of both pure HAP and CTAC-HAP were reduced. The sharp rod morphology of CTAC-HAP was changed into a round shape with a smaller aspect ratio after the heat treatment. The morphological change by thermal treatment was also observed in pure HAP. Therefore, the morphological change and decrease of the specific surface area suggested that pores from the removal of CTAC during thermal treatment were not retained.

THE INFLUENCE OF pH AND LACTIC ACID CONCENTRATION ON THE FORMATION OF ARTIFICIAL ROOT CARIES IN ACID BUFFER SOLUTION (산 완충용액의 pH 및 유산의 농도가 인공치근우식의 형성에 미치는 영향)

  • The purpose of this study is to compare and to evaluate the effect of pH and lactic acid concentration on the progression of artificial root caries lesion using polarizing microscope, and to evaluate the morphological changes of hydroxyapatite crystals of the demineralized area and to investigate the process of demineralization using scanning electron microscope. Artificial root caries lesion was created by dividing specimens into 3 pH groups (pH 4.3, 5.0, 5.5), and each pH group was divided into 3 lactic acid concentration groups (25 mM, 50 mM, 100 mM). Each group was immersed in acid buffer solution for 5 days and examined. The results were as follows : 1. Under polarized microscope, the depth of lesion was more effected by the lactic acid concentration rather than the pH. 2. Under scanning electron microscope, dissolution of hydroxyapatite crystals were increased as the lactic acid concentration increased and the pH decreased. 3. Demineralized hydroxyapatite crystals showed peripheral dissolution and decreased size and number within cluster of hydroxyapatite crystals and widening of intercluster and intercrystal spaces as the pH decreased and the lactic acid concentration increased. 4. Under scanning electron microscope evaluation of the surface zone, clusters of hydroxyapatite crystals were dissolved, and dissolution and reattachment of crystals on the surface of collagen fibrils were observed as the lactic acid concentration increased. 5. Under scanning electron microscope, demineralizatlon of dentin occurred not only independently but also with remineralization simultaneously. In conclusion, the study showed that pH and lactic acid concentration influenced the rate of progression of the lesion in artificial root caries. Demineralization process was progressed from the surface of the cluster of hydroxyapatite crystals and the morphology of hydroxyapatite crystals changed from round or elliptical shape into irregular shape as time elapsed.

Effect of remineralization and inhibition to demineralization after fluoride gel or hydroxyapatite paste application on stripped enamel (치간 삭제 후 불소 또는 수산화인회석 도포 시 재광화 및 탈회억제에 관한 연구)

  • Objective: The aim of this study was to evaluate the effect of remineralization and inhibition to demineralization after fluoride gel (acidulated phosphate fluoride, APF) or hydroxyapatite (HAp) paste application on interdentally stripped teeth. Methods: After interdental stripping, 1.23% APF or 5%, 10% HAp paste were applied for 7 days for remineralization. Afterwards, teeth were exposed to lactate carbopol buffer solution for demineralization. Scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDS) were used to compare change in surface contents and crystal structures after remineralization, and then after demineralization. Results: EDS analysis indicated that calcium (p < 0.001) and phosphate (p < 0.01) contents were increased after 10% HAp paste application on stripped enamel, calcium (p < 0.05) and phosphate (p < 0.01) contents were increased after 5% HAp paste application, and fluoride (p < 0.01) contents were increased after 1.23% APF application. SEM image showed that enamel surfaces became smoother and crystal structures became small and compact after APF or HAp application. After demineralization, calcium (p < 0.05) and phosphate (p < 0.05) contents remained increased on the enamel remineralized with 10% HAp paste, and phosphate (p < 0.05) contents remained increased on the enamel remineralized with 5% HAp paste. After demineralization, surfaces looked less destroyed in the enamel remineralized beforehand than those of the control, and small pores between crystal structures, formed by remineralization were remained. Conclusions: Hydroxyapatite paste and fluoride gel were helpful to remineralize and inhibit deminerlization on stripped enamel.

  • Effect of the roasted barley tea in commercial markets on the adherence to the saliva-coated hydroxyapatite(HA) beads and the cell surface hydrophobicity of Streptococcus mutans and Streptococcus sobrinus as cariogenic microorganism was examined in vitro. Adherence activity and hydrophobicity in bacteria tested in all the barley tea samples decreased and the values were different according to the type of tea and the type of treatment. The inhibition of bacterial adsorption to HA beads suggest that barley tea active molecules as catechins and melanoidins may adsorb to a host surface, preventing the tooth receptor from interacting with any bacterial adhesions. The obtained results showed that the barley tea may inhibit bacterial adherence, the first step of the pathogenesis of dental caries in which these microorganism are involved.

불완전 결정성 아파타이트 박막의 특성 및 응용

  • 뼈 결정과 같이 결정도가 낮은 탄산 인회석 결정의 화학조성 및 구조는 순수 수산화인회석 과 물리적 및 화학적 성질에 있어서 매우 다르다. 극미세 결정으로 결정도가 매우 낮은 불완전 결정이며, 결정 표면은 매우 반응성이 높은 이온으로 구성되어 있어 유기분자나 세포와 활발히 반응할 수 있다. 이와 같은 뼈 결정의 특성을 고형 기질 표면에 불완전 결정도 인회석 박막을 형성시켜 생체재료에 재현하였다. 이 박막에는 뼈세포들이 잘 부착하였는데 특히 골모세포의 부착, 증식, 분화를 촉진하여 장차 이와 같은 뼈 탄산 인회석의 특성을 갖는 인회석을 생체이식 재료로 이용하므로써 우리 몸의 생리에 좀 더 가까운 인공 매식체를 개발할 수 있을 것으로 기대된다.

Changes in surface content and crystal structure after fluoride gel or hydroxyapatite paste application on stripped enamel (치간 삭제 후 불소 및 수산화인회석 도포 시 법랑질 표면의 함량 및 결정구조 변화에 관한 연구)

  • Objective: The aim of this study was to evaluate the remineralization of interdentally stripped teeth after fluoride gel or hydroxyapatite paste application. Methods: After interdental stripping, 1.23% fluoride gel or 10% hydroxyapatite paste was applied three times a day, with a duration of four minutes, for a week. Scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDS) were used to compare the change of surface contents and crystal structures before and after the application of fluoride gel or hydroxyapatite paste. Results: EDS analysis indicated that calcium contents were increased after 10% HAp paste application on stripped enamel (p < 0.01). SEM view showed that enamel surfaces in groups of 1.23% APF gel or 10% HAp paste application were smoother than those of control group, which was regarded as filling of the pore structure. And pores between crystal structures in groups treated with 1.23%, APF gel or 10% HAp paste were smaller than those of control group. Conclusions: Application of APF or HAp on stripped enamel could positively influence on the surface contents or crystal structure.

  • HAP란 Hydroxylapatite의 준말이며 우리말로는 수산화인회석으로도 불린다. 본 실험에서는 다양한 농도의 염기조건(NaOH $10^{-3}$, $10^{-2}$, $10^{-1}$, 1, 10, 30 M)에서 서로 다른 형태의 HAP를 수열합성법을 통해 합성하였다. XRD (X-ray powder diffraction) 로 관찰한 결과 NaOH 농도 $10^{-1}M$ 이상에서부터 HAP가 합성됨을 확인하였다. Transmission and scaning electron microscopy 를 이용하여 HAP의 모양과 표면을 관찰해본 결과, NaOH의 농도가 진해 질수록 육각기둥의 형태에서 사각형으로 변화하였다. 6개의 각각의 HAP의 표면에 Pd (Palladium)을 도입하고 그 양을 정량화 하였다. 합성된 Pd-HAP를 C-C coupling reaction에 이종상 촉매로 사용하였다.

The effect of different crystallization temperature of the hydroxyapatite coating produced by ion beam-assisted deposition on anodizing-treated titanium disks on human osteosarcoma cells (양극산화처리된 티타늄 표면에 이온빔보조증착방식을 이용한 수산화인회석 코팅시 소결온도의 차이가 조골세포에 미치는 영향)

  • Purpose: The aim of this study was to study the effect of hydroxyapatite (HA) coating crystallinity on the proliferation and differentiation of human osteosarcoma cells. Materials and methods: Surface roughness of the titanium disks increased by anodizing treatment and then HA was coated using ion beam-assisted deposition (IBAD). HA coating was crystallized by heat-treated at different temperature ($100^{\circ}C$, $300^{\circ}C$, $500^{\circ}C$, $800^{\circ}C$). According to the temperature, disks were divided into four groups (HA100, HA300, HA500, HA800). With the temperature, crystallinity of the HA coating was different. Anodized disks were used as control group. The physical properties of the disk surface were evaluated by surface roughness tests, XRD tests and SEM. The effect of the crystallinity of HA coating on HOS cells was studied in proliferation and differentiation. HOS cells were cultured on the disks and evaluated after 1, 3, 5, and 7 days. Growth and differentiation kinetics were subsequently investigated by evaluating cell proliferation and alkaline phosphatase activity. Results: Regardless of the heat-treated temperature, there is no difference on the surface roughness. Crystallinity of the HA was appeared in the groups of HA500, HA800. HOS cells proliferation, ALP activity were higher in HA500 and HA800 group than HA100 and HA300. Conclusion: Within the results of this limited study, heat treatment at $500^{\circ}C$ of HA coating produced by IBAD has shown greater effect on proliferation and differentiation of HOS cells. It is considered that further in vivo study will be necessary.

  • Enamel layer consists of carbonated hydroxyapatite of 85%, water of 12% and protein of 3% and lipid and tentin consists of carbonated hydroxyapatite of 47%, water of 20% and protein of 33% and lipid so if heat is increased in teeth and then teeth color is discolor. In case high temperature is applied in teeth, teeth color changed quality is measured about change of time for minimize teeth change of color. Spectroscopic light intensity special quality by change of time rise at specification temperature.

THE CHANGE OF THE CONFIGURATION OF HYDROXYAPATITE CRYSTALS IN ENAMEL BY CHANGES OF PH AND DEGREE OF SATURATION OF LACTIC ACID BUFFER SOLUTION (유산 완충용액의 pH 및 포화도 변화에 따른 법랑질 내 수산화인회석 결정 형태의 변화)

  • Since it was reported that incipient enamel caries can be recovered, previous studies have quantitatively evaluated that enamel artificial caries have been, remineralized with fluoride showing simultaneously the increase of width of surface layer and the decrease of width of the body of legion. There is, however, little report which showed that remineralization could occur without fluoride. In addition, the observations on the change of hydroxyapatite crystals also have been scarcely seen. In this study, enamel caries in intact premolars or molars was induced by using lactic acidulated buffering solutions over 2 days. Then decalcified specimens were remineralized by seven groups of solutions using different degree of saturation(0.212, 0.239, 0.301, 0.355) and different pH(5.0, 5.5, 6.0) over 10 days. A qualitative comparison to changes of hydroxyapatite crystals after fracturing teeth was made under SEM(scanning electron microscopy) and AFM(atomic force microscopy). The results were as follows: 1. The size of hydroxyapatite crystals in demineralized area was smaller than the normal ones. While the space among crystals was expanded, it was observed that crystals are arranged irregularly. 2. In remineralized enamel area, the enlarged crystals with various shape were observed when the crystals were fused and new small crystals in intercrystalline spaces were deposited. 3. Group 3 and 4 with higher degree of saturation at same pH showed the formation of large clusters by aggregation of small crystals from the surface layer to the lesion body than group 1 and 2 with relatively low degree of saturation at same pH did. Especially group 4 showed complete remineralization to the body of lesions. Group 5 and 6 with lower pH at similar degree of saturation showed remineralization to the body of lesions while group 7 didn't show it. Unlike in Group 3 and 4, Group 5 and 6 showed that each particle was densely distributed with clear appearance rather than crystals form clusters together.
