Effect of the Ethanol Extract from Citrus Peels on Oxidative Damage in Alloxan-induced HIT-T15 Cell (진피 에탄올 추출물이 Alloxan에 의해 유도된 HIT-T15 세포의 산화적 손상에 미치는 영향)

  • This study was carried out to investigate the effect of ethanol extract from citrus peels (CP-Et) against the alloxan-induced oxidative damage on HIT-T15, Hamster pancreatic $\beta$-cell. Total polyphenol and flavonoid contents in CP-Et were $57.00{\pm}2.91\;mg/g$ and $8.11{\pm}2.83\;mg/g$, respectively. Cell toxicity on HIT-T15 by CP-Et (0.125~0.75 mg/mL) was not observed. CP-Et (0.125 mg/mL) increased cell proliferation rate of HIT-T15, which was treated alloxan ($IC_{50}=11.58\;mM$) (cell viability=$80.52{\pm}3.29%$ of normal cell, p<0.05). In comparison with insulin secretion of oxidative damaged HIT-T15, 1.5 fold ($116.93{\pm}2.11\;{\mu}g/mg$ protein) was increased by treatment CP-Et treatment (0.125 mg/mL) in HIT-T15 (p<0.05). These results showed that CP-Et contribute to repairing cells and improvement of insulin expression on oxidative stress pancreatic $\beta$-cell, and also suggested application of CP-Et as a functional food material for type 2 diabetes.

Effects of carbendazim on DNA, gene and chromosome (살균제 carbendazim이 DNA, 유전자 및 염색체에 미치는 영향)

  • Benzimidazole pesticide carbendazim that is effective against a wide range of fungal plant pathogens is a protective, eradicant, and systemic fungicide. For genetic toxicity evaluation of carbendazim on DNA, genes and chromosome, were investigated with chromosome aberration, bacterial reverse mutation, micronucleus test in mouse born marrow and DNA damage assay by single cell microgel electrophoresis. Substitution and frameshift mutation were not induce at variable concentration of carbendazim on Ames test with or without rat liver microsomal activation. For the result of chromosome aberration test, numerical changes of chromosome were detected at the concentrations higher than $4.0{\mu}g/m{\ell}$, but structural aberration was not induced. Positive control, Mitomycin-C and captafol made a structural aberration, but numerical change of chromosome did not appear. In the micronucleus test for mouse born marrow, carbendazim was negative, but was weak positive in DNA damage assay by single cell microgel electrophoresis because of increased DNA moving length of 20% to control.

  • Background: Hyperoxemic cardiopulmonary bypass (CPB) has been recognized as a safe technique and is widely used in cardiac surgery. However, hyperoxemic CPB may produce higher toxic oxygen species and cause more severe oxidative stress and ischemia/reperfusion injury than normoxemic CPB. This study was undertaken to compare inflammatory responses and myocardial injury between normoxemic and hyperoxemic CPB and to examine the beneficial effect of normoxemic CPB. Material and method: Thirty adult patients scheduled for elective cardiac surgery were randomly divided into normoxic group (n=15), who received normoxemic CPB (about Pa $O_{2}$ 120 mmHg), and hyperoxic group (n=15), who received hyperoxemic CPB (about Pa $O_{2}$ 400 mmHg). Myeloperoxidase (MPO), malondialdehyde (MDA), adenosine monophosphate (AMP), and troponin-T (TnT) concentrations in coronary sinus blood were determined at pre- and post-CPB. Total leukocyte and neutrophil counts in arterial blood were measured at the before, during, and after CPB.   • This review describes the neurophathological mechanisms that are implicated in perinatal brain injury. Perinatal brain injury is the most important cause of morbidity and mortality to infants, often leading to spastic motor deficits, mental retardation, seizures, and learning impairments. The immature brain injury is usually caused by cerebral hypoxia-ischemia, hemorrhage, or infection. The important form of perinatal brain injury is the hypoxic-ischemic injury and the cerebral hemorrhage. The pathology of hypoxic-ischemic injury include delayed energy failure by mitochondrial dysfunction, neuronal excitotoxicity and vulnerability of white matter in developing brain. The immature brain has the fragile vascular bed of germinal matrix and can not effectively centralize their circulation. Therefore, the cerebral hemorrhage process is considered to be involved in the periventricular leukomalacia.

  • Background: Predicting the important role of intercellular adhesion molecule-1 expression on the acute ischemia-reperfusion injury, we set out to demonstrate it by assessing the degree of expression of ICAM-1 after warm ischemia-reperfusion in canine unilateral lung ischemia model. Material and Method: Left unilateral lung ischemia was induced by clamping the left hilum for 100 minutes in seven adult mongrel dogs. After reperfusion, various hemodynamic pararmeters and blood gases were analyzed for 4 hours, while intermittently clamping the right hilum in order to allow observation of the injured Ieft lung function. The pulmonary venous blood was collected serially to measure TNF- and cICAM-1 level. After 4 hours of reperfusion, the lung tissue was biopsied to assess cICAM-1 expression, and to measure tissue malondialdehyde(MDA) and ATP level. Result: The parameters including arterial oxygen partial pressure, pulmonary vascular resistance and tissue MDA and ATP level suggested severe lung damage. Serum TNF-$\alpha$ level was 8.76$\pm$2.37 ng/ml at 60 minutes after reperfusion and decreased thereafter. The cICAM-1 level showed no change after the reperfusion during the experiment. The tissue cICAM-1 expression was confirmed in 5 dogs. Conclusion: The increase of TNF-$\alpha$ Ievel and expression of tissue ICAM-1 were demonstrated after ischemia reperfusion injury in canine lung model.

Curcumin Induces Recovery from Indomethacin-Induced Gastric Mucosal Lesions in Rats (커규민의 인도메타신 유도 위점막 손상에 대한 치료 효과)

  • In the present study, the curative effect of curcumin on indomethacin-induced gastric mucosal lesions in rats was investigated. Indomethacin is a nonsteroidal anti-inflammatory drug (NSAID), with serious side effects, including erosion, ulcerative lesions, and petechial bleeding in the mucosa of the stomach. Gastric mucosal lesions were caused by oral administration of 25 mg/kg of indomethacin. Various doses (10, 50, and 100 mg/kg) of curcumin were treated for 3 days by oral gavage. Indomethacin clearly increased the gastric ulcer area in the stomach, and curcumin significantly decreased the gastric ulcer area in a dose-dependent manner. Curcumin also markedly inhibited lipid peroxidation in the gastric mucosa and activated radical scavenging enzymes, such as superoxide dismutase (SOD), catalase, and glutathione peroxidase, in a dose-dependent manner. These results suggest that curcumin can induce recovery from indomethacin-induced gastric mucosal lesions through inhibition of lipid peroxidation and activation of radical scavenging enzymes, such as SOD, catalase, and glutathione peroxidase. Curcumin appears to be a powerful free radical quencher, and it may offer an attractive strategy for healing gastric mucosal lesions in humans.

  • 정상적인 뉴런의 활성전위는 외부에서 일정한 자극이 인가되었을 때 세포막을 기준으로 하여 각 이온간의 농도 차에 의해 발생한다. 최근에 관심이 되어지고 있는 쇼크에 의한 세포가 손상이 발생할 경우, 즉 신호를 받아들이고 전달하는 뉴런 중에서 축색에 이온채널이 이상증세를 발생하면 신경 전달 흐름을 흐트러지게 하여 이웃한 정상세포에게 커다란 영향을 미치게 된다. 이것은 병리학적인 중요한 역할을 하는 축색 내에 이상이 발생하였다고 가정을 하지만 이 가정을 뒷받침 해 주는 증거는 매우 적다고 보고되고 있다. 최근 연구에서 손상된 축색의 모델은 쇼크이후에 이온의 칼륨 채널에 blocking 현상이 발생하여 나트륨 이온이 다수 유입됨을 고려하고있다. 이에 본 연구에서는 쇼크나 충격에 의해 축색의 손상을 입을 경우 운동신경의 변형으로부터 병리학적인 중요한 이상결과를 일으킬 수 있는 상태를 고려하여 신경모델을 설계해 시뮬레이션 해 보았다.

  • 암석이 화학적으로 매우 불균일하기 때문에 암석에 있어서 자연적인 풍화와 인위적인 오염에서 기인한 손상을 구별짓는다는 것은 간단하지 않다. 석재의 화학적인 풍화는 스며든 빗물이나 오염먼지 등에 의해 생성된 물질의 농도변화로 표현되어 진다. 특히 벽면 표면의 두터운 검은 외각과 얇은 검은 막은 미관상으로 뿐만 아니라 암석 자체에도 큰 손상을 끼친다. 일반적으로 이런 검은 물질들은 비 등의 수분과 직접적인 접촉이 없고, 농축된 오염물질들이 쉽게 쌓일 수 있는 곳에서 찾아볼 수 있다. 천연 암석과 마찬가지로 검게 손상된 층 또한 화학적으로 매우 복잡한 체계를 갖고 있어 그 생성 원인과 메커니즘을 규명하는 것이 어려운 일이다. 이 흑색 층은 일반적으로 공기오염물질, 유기물, 철과 망간등의 유색광물의 이동과 침착의 현상에서 생성될 수 있다. 건물들의 외벽에 사용된 여러 종류의 사암과 석회암, 인조석의 표면에는 여러 풍화 손상 형태가 나타나고 있다. 특히 표면에 있는 검은 막의 성질을 알아보기 위해 화학성분을 주성분과 미량성분으로 나누어 측정하였고, 화학적인 특징을 예측하기 위해서 분석자료를 여러 통계적인 방법으로 처리하였다.

Photoprotective Effect of Grape Pruning Stem Extracts on the UVB Induced HR-1 Mice Skin (포도전정가지 추출물이 UVB에 유도된 HR-1 mice의 피부손상에 대한 광보호 효과)

  • This study intends to analyze the contents of rutin, procyanidin B3, quercetin, kaempferol, known to have antioxidant, anti-inflammatory and anti-carcinogenic effects, among the polyphenol type contained in the grape pruning stem extracts (GPSE), utilizing grape stems being discarded after harvest, measure the effects on the skin moisture, inhibition of skin cell proliferation, anti-inflammatory on the damaged skin of a HR-1 mice induced with UVB, and verify the applicability as a material for functional food and functional cosmetics. The results of verifying the photoprotection effects through the skin proliferation control through of GPSE showed similar result to suncream was achieved at the GPSE concentration of 2,000 mg/kg on the epidermis (p<0.05). The results showed anti-inflammatory effects on all groups applied with GPSE as compared to the control group irradiated with UVB, but at the GPSE concentration of 1,000 mg/kg, a lower COX-2 protein expression at 8%, lower than the 22% of suncream, was observed to achieve an excellent anti-inflammatory effect (p<0.05). The results of this study confirmed the existence of active polyphenol type, such as rutin, kaempferol, querocetin and procyanidin B3, within the GPSE, and GPSE has improvement effects on moisturizing effects, skin proliferation control effect, inflammatory control effect and improvement effects on the skin barrier function through UV ray damage. GPSE is a functional ingredient with a potential for skin protection effects, and has high utilization as an ingredient for functional food and functional cosmetics.

Differences in Urine Cadmium Associations with Renal Damage Markers According to the Adjustment with Specific Gravity or Urinary Creatinine (요비중 또는 크레아티닌 보정에 따른 요중 카드뮴과 신장손상지표와의 관련성 비교)

  • In general, specific gravity (SG) and urinary creatinine (CR) have been used to adjust urinary cadmium (Cd) concentrations. However, the validity of correction methods has been controversial. We compared the two adjustments to evaluate associations between urinary Cd and various renal damage markers and to evaluate the relationship between urinary Cd concentration and renal disease markers, such as estimated glomerular filtration rate (eGFR), in a relatively large general population sample. Among the 1,086 volunteers who were enrolled in this study, 862 healthy volunteers who did not have kidney disease were included in the final analysis. Urinary Cd, malondialdehyde (MDA), and N-acetyl-${\beta}$-D-glucosaminidase (NAG) concentrations were measured, the creatinine-based eGFR was calculated, and the relationships between these markers were subsequently analyzed. This study showed the use of urinary Cd concentration adjusted with SG rather than with urinary creatinine may be appropriate in studies evaluating renal function based on Cd exposure. Urinary Cd concentration adjusted with SG had a positive correlation with urinary MDA levels and a negative correlation with eGFR. This relationship was relatively stronger in women than in men. This study showed that urinary Cd level was associated with decreased eGFR in the general population, and oxidative stress was likely to act as an intermediator in this process. These results suggest that eGFR can be a very good indicator of kidney damage caused by Cd exposure in the general population.
