We examined the effects of selenium-binding peptide from sericin hydrolysates on the bioavailability of selenium-deficient rats. Three-week-old male rats were fed a selenium-deficient diet for 4 weeks while the normal control group was fed a normal diet. The selenium-deficient rats were divided into three groups: no treatment, organic selenium (OS), and inorganic selenium (IS). After selenium supplementation for 4 weeks, the level of serum glutathione reduced form in rats treated with organic selenium was significantly higher than that of inorganic selenium. Selenium retention rate also increased significantly in the organic selenium group compared to the inorganic selenium group [selenium deficient diet (DD)+OS 50.25% vs. DD+IS 17.04%, P<0.05]. In conclusion, binding of selenium to peptides from sericin hydrolysates seems to improve its bioavailability, and can hasten a cure for selenium deficiency in experimental rats.

필수 미량원소인 셀레늄은 인간의 건강에 기본적으로 중요한 역할을 하는 물질이다. 항산화 관여 효소의 작용과 구조역할을 담당하는 셀레늄이 함유된 단백질(selenoproteins)은 항산화 기능이 있을 뿐만 아니라, 갑상선호르몬의 활성화에 관여하는 것으로 밝혀졌다. 또한, 셀레늄은 정상적인 면역체계의 유지에 필요하며, HIV(human immunodeficiency virus)가 진행되어 AIDS로 발전되는 것을 억제하는 작용이 있다는 사실도 밝혀지고 있으며, 정자의 생성과 운동성에 관계하기도 한다. 우울증 환자가 셀레늄을 섭취하게 되면 효과가 있으며, 셀레늄이 순환계질환의 발생을 억제하는 것으로 알려지고 있다. 특히 최근에는 셀레늄의 섭취가 암발생을 예방할 수 있다는 역학조사결과를 토대로, 대규모 셀레늄 임상실험이 다국적 연구의 형태로 2개의 프로젝트(PRECISE, SELECT)가 수행 중이다. 한편, 셀레늄의 항암효과에 대한 세포수준에서의 연구가 활발하게 이루어지고 있으며, 어느 정도 가시적인 결과를 보이고 있다. 따라서, 셀레늄에 대한 재평가가 우리 나라에서도 이루어져야 한다. 그러기 위해서는 지역별로 셀레늄의 정확한 섭취량조차도 파악되어 있지 않을 뿐만 아니라 일일권장량 값조차 정확하지 않은 현실을 볼 때, 셀레늄의 기본적 연구에 대한 관심을 가져야 할 때라고 판단된다.
Human blood selenicum(Se) level depends on the Se content and bioavailablilty of foods. In countries where the soil has low Se content, the differences of Se intake and blood Se concentration are shown according to the type of diet. In this study, Se status of women eating an average German diet(175 German healthy women) and wholesome nutrition group(243 women) were studied. There were significant differences in wholeblood and plasma Se levels between the two groups. In average German diet group, mean wholeblood Se concentration was 84.97ug/l and plasma Se concentration was 75.02ug/l. In wholesome nutrition group mean wholeblood Se concentration and plasma Se concentration with Se levels, correlation analysis was conducted between Se concentration and BMI, age and daily food intake. BMI and age did not correlate significantly with wholeblood and plasma Se levels. In wholesome nutrition group, serum and wholeblood Se levels had significant correlation with fish intake.

This study was conducted to investigate the effects of feeding seleniferous whole crop barley silage (WCBS) on the growth performance, blood and carcass characteristics, and tissue selenium deposition infinishing Hanwoo steers. A total of 20 growing Hanwoo steers were selected and assigned to one of the following feed groups: control (0.1 ppm Se), T1 (0.3 ppm Se), T2 (0.9 ppm Se), and T3 (0.9 ppm of inorganic Se). Five steers were allocated to each treatment group, and the trial lasted for 90 days. All experimental diets combined seleniferous and/or non-seleniferous WCBS up to a ratio of 30%. For the T3 diet, 0.9 ppm Se in the form of sodium selenite was added to the control diet. Dietary level and type of selenium did not affect feed intake and body weight gain. Blood total lipid and triglyceride concentrations were significantly (p<0.05) lower in the T2 group than in control. Blood immunoglobulin G concentration increased significantly (p<0.05) with increasing levels of dietary selenium; concentrations were significantly (p<0.05) higher in T2 and T3 than in control. Blood selenium concentration was the highest (p<0.05) in T2. No significant difference was observed in dressing rate, ribeye area, marbling score, meat color and fat color. Further, no association was found between levels and chemical form of dietary selenium and quality and quantity of meat. To the contrary, tissue selenium content in kidney, liver, and hind leg increased significantly (p<0.05) with increasing levels of selenium; however, feeding inorganic selenium did not introduce a significant increase in tissue selenium content of finishing Hanwoo steers. The results indicated that the selenium present in seleniferous WCBS was an effectively absorbable selenium source, suitable for increasing tissue selenium content in beef. Approximately 100 g of hind leg muscle from steers fed on the T2 diet met approximately 69% of the recommended daily selenium requirements.
This study was carried out to investigate the effect of application concentration and period of sodium selenate ($Na_2SeO_4$) in the soil on the growth of garlic. Fresh weight were decreased with the application of higher concentration and longer period of Se compared with the control. The treatment of higher concentration and longer period of Se increased the content of Se in leaf sheath than bulb of garlic after 3 weeks. But, the Se content of garlic was higher in order of bulb, stem, root after 6 weeks of selenium treatment. Se was also detected also in control, the non-treated plot, but the amount was negligible. The content of selenium in the soil was not affected by the concentration and period of selenium application.
Selenized yeast (Se yeast) containing $0.1{\%}$(w/w) of selenium was obtained when the yeast was incubated at a selenium concentration of 1$1.14{\times}10_-3 M$ in rich medium. After washing several times, the inorganic selenium on the cell wall was confirmed with MBRT. There was no indication of inorganic selenium on the cell wall when the blue color in MBRT was stayed for 15 minutes. The selenized yeast was sonicated, then the selenium contained protein was obtained after salting out by ammonium sulfate at the concentration $80{\%}$ saturation. The seven protein bands were seperated by SDS-PAGE and the selenium concentration in protein was measured by ICP-AES. Analytical data showed that the large expressed protein band contained a relatively large amount of selenium. The proteins of the 47kDa was contained the concentrations of 69.5 ${\mu}$ Se/g of most many content. The protein (47 kDa) was seperated from PVDF membrane by tank-electroblotting. The isolated protein was hydrolyzed under acid condition and reacted with PITC. The derivatives of amino acids were analyzed by HPLC and compared with the data obtained from regular yeast. The resulting selenium-yeast was analyzed with the selenomethionine concentration of $2{\%}$ comparaed with general amino acids. The goal of this study is to analyze the selenium concentration in protein bands and measure the degree of biotransformation of selenomethionine in a specific protein.

This study was conducted to investigate effects of feeding seleniferous whole crop barley silage (WCBS) on growth performance and blood characteristics in growing Hanwoo steers. A total of 20 growing Hanwoo steers, initially weighing on average 208.8 kg of body weight, were used and treatments were consisted of 1) controls : 0.1 mg/kg Se, 2) T1 : 0.3 mg/kg Se, 3) T2 : 0.9 mg/kg Se by combining seleniferous and/or non-seleniferous WCBS, and 4) T3 : 0.9 mg/kg Se of inorganic Se treatment. Five steers were allocated to each treatment, and the trial was lasted for 90 days. All experimental diets were included to 30% of combination of seleniferous and/or non-seleniferous WCBS, and in T3 diet, sodium selenite that corresponds to 0.9 mg/kg Se was added to control diets. Also, the diets were isonitrogenous and isocaloric among treatments. Dietary level and type of selenium did not affect feed intakes and daily gain, and blood glucose concentration was significantly (p<0.05) lower for controls than T1 and T2 treatments. Blood total lipid concentration was significantly (p<0.05) decreased with increasing levels of dietary selenium, and also that of T2 and T3 was significantly (p<0.05) lower than controls. LDL-cholesterol concentration was significantly (p<0.05) lower for treatments including dietary selenium than controls, and also blood triglyceride concentration was significantly (p<0.05) lower for T2 than controls. Overall, it was tended that feeding seleniferous WCBS or inorganic Se increased blood IgG concentration, and in the same dietary selenium levels, T2 treatment was higher for IgG than T3 group (p>0.05). Blood selenium concentration was significantly (p<0.05) increased by feeding increasing levels of seleniferous WCBS, but there was no significant difference between controls and T3 group. These results showed that feeding seleniferous WCBS to growing Hanwoo steers was responsible for saccharide and lipid metabolism, and in particular, it reduced their total lipid and blood LDL-cholesterol concentrations. Furthermore, selenium present in seleniferous WCBS rather than inorganic selenium was better available in intestinal absorption, and it might help to enhance immunity in growing and younger stages of Hanwoo steers.

This study was conducted to clarify the effects of supplying methods of selenium on the growth and Se uptake of hydroponically grown tomato plants. Tomato seeds (Lycopersicum esculentum Mill. cv. Momotaro T-93, Daki Seed Co.) were sown in plug tray with fifty holes, and raised for sixty days. Tomato seedlings transplanted to coco fiber slabs were supplied with the nutrient solutions adjusted to EC $2.3dS{\cdot}m^{-1}$ and pH $5.8\~6.2$ recommended by the Japanese Horticultural Experiment Station. Selenium forms used were inorganic $SeO_2$ (here in after referred to Se) and organic selenium chlenium with sugar fatty acid ester (here in after referred to chelated-Se). 10 ppm selenium solutions were treated to tomato plants with foliar applications, drenching, and foliar application plus drenching. Growth characteristics in terms of plant height, number of leaves, leaf area and chlorophyll content were significantly increased in the plot of foliar application ot Se, and in the plot of foliar application plus drenching of chelated-Se than other plots, respectively. Transported contents of selenium into the tomato fruits were highest as 0.302 ppm in the plot of foliar application plus drenching of chelated-Se. Also, it had tended to be higher in the plot of foliar application plus drenching than in the plots of foliar application or drenching in both of Se and chelated-Se. Foliar application and drenching of organic chelated-Se were effective to produce the functional tomato fruits.
Selenium exists in various forms of chemical species. The activity and bioavailability is strongly dependent on its chemical form and concentration. Consequently the information on each selenium species and its concentration must be exactly determined for the food we take in. In this study, selenium species in seafood were separated and quantified by RP (reversed phase) HPLC (high performance liquid chromatography) coupled with ICP-MS (inductively coupled plasma mass spectrometry) using post-column isotope dilution. $^{79}Br$, which interferes on $^{80}Se$, has mostly been removed by solid phase extraction and then mathematical correction has been applied for the more accurate correction. The experimental result for CRM (certified reference material) DOLT-4 agreed well with the certified value but each selenium species could not be compared. SeCys (selenocysteine) and SeMet (selenomethionine) were the major species detected in seafood such as belt fish, spanish mackerel, and squid that have been serving as Korean diet. The concentrations found in Korean sea food for SeCys and SeMet were in the range of 0-661.6 mg/kg and 137.3-462.7 mg/kg, respectively.
The selenium (Se) contents of 20 herbs, harvested in three areas in Korea, were analyzed. Many herbs had no Se. Angelica ($138.4{\mu}g \;Se{\cdot}kg^{-1}$ FW), caraway ($167.8{\mu}g \;Se{\cdot}kg^{-1}$ FW), chamomile ($116.6{\mu}g \;Se{\cdot}kg^{-1}$ FW), marjoram (158.7 and $132.6{\mu}g\;Se{\cdot}kg^{-1}$ FW), and oregano ($62.5{\mu}g\;Se{\cdot}kg^{-1}$ FW) accumulated Se as higher concentration compared with other plants. The Se content of herbs was dependent on the species, cultivars and areas. The status of Se uptake by herbs was investigated according to Se concentrations in hydroponics. During four weeks, before harvest of several herbs in greenhouse, sodium selenate ($Na_2SeO_4$) was added to the nutrient solution at concentrations of 2, 4, 6, and $8mg{\cdot}L^{-1}$. Field-mint accumulated Se too high, but external symptom of Se toxicity was not found. Essential oil content was generally proportionated to treated-Se concentration. The essential oil contents in basil and balm increased two to three times higher by sodium selenate treatment compared to non-treatment, but they did not show any linear relationship between essential oil contents and treated-Se concentration.
