Kinetics of Cultivating Large Quantities of Mammalian Cells (tPA생산을 위한 동물 세포 배양에 관한 동력학적 연구)

  • Growth kinetic parameters for mass cultivation of Chinese Hamster Ovary (CHO) cells are estimated by measuring oxygen uptake rates. It Is found that there is strong correlation between cell growth and oxygen consumption, showing that correlation factor is 0.83. Derived linear model predicts actual cell density very well. It tells that oxygen uptake rate can play important role in indirectly measuring cell density when conventional method of estimating cell density is no longer meaningful due to heavy cell clumpings. Cell yield per oxygen consumption, $Y_{\chi}o$ and mass transfer coefficient for oxygen, Ka are also estimated as 1.26$\times$10$^4$cells/mmole $O_2$ consumed and 1.01/h, respectively. Average specific growth rate over all runs is 2.891/day for CHO cells with producting 2 grams of tPA per day under continuous perfusion operations.

  • In this study, a refined beam analysis model has been developed for multi-celled composite blades with elliptic cross-sections. Reissner's semi-complimentary energy functional is introduced to describe the beam theory and also to deal with the mixed-nature of the formulation. The wail of elliptic sections is discretized into finite number of elements along the contour line and Gauss integration is applied to obtain the section properties. For each cell of the section, a total of four continuity conditions are used to impose proper constraints for the section. The theory is applied to single- and double-celled composite blades with elliptic cross-sections and is validated with detailed finite element analysis results.

Effect of 2-D DBP/PLGA Hybrid Films on Attachment and Proliferation of Intervertebral Disc Cells (2차원적 DBP/PLGA 하이브리드 필름이 디스크 세포의 부착과 증식에 미치는 영향)

  • Because demineralized bone particle (DBP) contains various bioactive molecules such as cytokines, it is widely used biomaterials in the field of tissue engineering. In this study, we investigated the effect of 2-dimensional DBP/PLGA hybrid films on adhesion, proliferation and phenotype maintenance of intervertebral disc cells. PLGA films incorporated with different amount (0, 10, 20, 40 and 80 wt%) of DBP were prepared by the solvent evaporation method and characterized by scanning election microscopy (SEM). PLGA film has a flat and smooth surface. According to the increase of content of DBP, the surface of DBP/PLGA film exhibited few agglomerates and increased the roughness of the surface. Annulus fibrosus (AF) and nucleus pulposus (NP) cells were cultured on PLGA and DBP/PLGA film surface, and then examined the cell adhesion and proliferation by the cell count and SEM observation. The result of cell count and SEM observation revealed that 10 and 20% DBP in DBP/PLGA films were superior to adhesion and proliferation of both AF and NP cells. We confirmed that specific gene expression of disc cells on DBP/PLGA film based on the cell count result. Disc cells seeded on 20% DBP/PLGA film expressed the gene of type I and II collagen continuously. Therefore, pertinent content of biomaterials could provide more appropriate condition on adhesion and proliferation of cell. And this results may be used as a basic data for the intervertebral disc regeneration using tissue engineering.

A Novel Cell Count Method Using Micro Lattice Engraved On A Culture Dish (미세 격자가 형성된 배양 접시를 이용한 새로운 세포 계수 방법)

  • A novel cell count method, which can improve the count efficiency and reduce contamination problem, was presented using micro lattice engraved on culture dish. The micro lattice has feature of $50{\mu}m{\times}50{\mu}m$ rectangular shape, $2{\mu}m$ line width, and $2{\mu}m$ depth in $3mm{\times}3mm$ area. In this paper, nickel mold was fabricated with thickness of 3mm and diameter of 80 mm, and transcription of the micro lattice on a polystyrene cell culture dish was performed by hot embossing at $200\;^{\circ}C$. The tedious and error-prone harvest/load processes of conventional cell counts with a hemocytometer could be omitted, and these advantages became magnified during periodical counts involving long-term cultures. SupT1 cells and HeLa cells were cultivated with the dish for 7 days in $CO_2$ incubator and counted as $371.84/mm^2$ and $123.36/mm^2$, respectively, during the cultivation without harmful effects on the cells.

Application of qDVC Method for Measuring Viable Cells in Lakes (호수 생태계에서 살아있는 세균을 측정하기 위한 qDVC 방법의 적용)

  • For measuring the viable cells in lakes, quantitative direct viable count (qDVC) method is applied. In the qDVC process, the final concentration of glycine is fixed as 2%. For confirming the effectiveness of qDVC for enumerating the viable cells, the viable bacterial numbers were measured by plate count, CTC reduction method and qDVC method at 5 different lakes. Among these 3 methods, the bacterial numbers by qDVC is $2.4{\sim}6.0$ times higher than those by the other 2 methods. And by the qDVC method, the viable cells were easily discriminated from dead or dormant cells.

  • River and stream are the important water supply source in our lives. Eutrophication causes excessive green algae growth including microcystis, which makes harmful to ecosystem and human health. Therefore, the water purification process to remove green algae is essential. In Korea, green algae alarm system exists depending on the concentration of green algae cells in river or stream. To maintain the growth amount under control, green algae monitoring system is being used. However, the unmanned, small and automatic monitoring system would be preferable. In this study, we developed the 3D printed device to measure the concentration of green algae cell using microfluidic droplet generator and deep learning. Deep learning network was trained by using transfer learning through pre-trained deep learning network. This newly developed microfluidic cell counter has sufficient accuracy to be possibly applicable to green algae alarm system.

Mast Cell Distribution at Predilection Sites of Atopic Dermatitis in Normal Canine Skin (개의 아토피성 피부염의 피부증상 호발부위의 비만세포분포조사)

  • Mast cell distribution was quantified in acidified toluidine blue sections of normal skin from 8 different sites in 10 dogs and compared to the predilection sites of canine atopic dermatitis. Mast cell counts varied significantly from site to site (p<0.0001) and counts in the superficial dennis were significantly higher than the deeper dennis (p<0.05). The highest mast cells distribution sites were the concave surface of the ear (mean $74.88{\pm}17.93\;per\;mm^{2}$) and the interdigital skin of the forefeet (mean $28.326{\pm}6.24\;per\;mm^{2}$). Counts in these sites were $280\%$ higher than all the other sites. Our results may provide some evidence that cutaneous mast cell distribution may be a factor in the frequent occurrence of ear and foot pruritus in atopic dermatitis. However, the low mast cell count in the predilection sites of atopic dermatitis did not explain the common occurrence of atopic lesions. Therefore, other factors or more complicated pathogenesis may be correlated with these predilection sites.

Correlation between Phytoplankton Dynamics and Water Quality in Paldang Reservoir (팔당호에서 식물플랑크톤 군집 동태와 수질과의 상관성)

  • This study was aimed to analyze the long-term fluctuation of water quality and phytoplankton dynamics of Paldang reservoir in Korea and to assess the relationship between algal bloom patterns and hydrological, limnological data. Diatoms in Paldang reservoir occurred continuously through the year. Blue- green algae occurred during the summer season (from June to Sept.), and the highest count was observed in July. Occurrence pattern of green algae was similar to that of blue-green algae. The rest of algae contained a lot of Cryptomonas spp. whose concentration was high from May to Aug. Dominant algal genera (>>7,000 cells $mL^{-1}$) in Paldang reservoir were Aulacoseira, Cyclotella, Microcystis, and Cryptomonas spp. Microcystis and Anabaena occurred during the summer season. Many different green algal genera were found in Paldang reservoir but their abundances were very low. There were some significant correlations (r>0.3, p<0.05) between algal taxa and water quality; diatoms and water temperature, TP:blue-green algae and water temperature, pH, DO saturation, COD, TP; green algae and water temperature, pH, DO saturation, COD, SS, TP. Furthermore, algal genera and water quality was significantly correlated (r>0.3, p<0.05) ; Aulacoseira and TN, TP; Anabaena and water temperature, DO saturation, COD, TP : Microcystisand water temperature, pH, DO saturation, TP; Coelastrum and COD, SS; Scenedesmus and water temperature, COD, TN, TP; Cryptomonas and DO saturation, TN. In Paldang reservoir, the water temperature had relatively big effect on blue-green algal bloom that was also dependant upon its hydrologic condition.

  • The value of the cell enzyme-linked immunosorbent assay as a possible replacement for the indirect immunofluorescence antibody test for the estimation of antibodies against BD virus was assessed in four animal models. The serum antibody response was measured by both assay systems;the variability of both tests was less than one diluent step, and correlation of the two tests was assessed using regression analysis. The study showed that the all four animal models gave satisfactory correlation of CELISA and IFA. There, CELISA is acceptable for use in mouse, rabbit, chicken and rat models.

Relationship between Radiation Induced Activation of DNA Repair Genes and Radiation Induced Apoptosis in Human Cell Line A431 (인체세포주 A431에서 방사선 조사 후 DNA수선 유전자 발현과 세포고사와의 관계에 관한 연구)

  • Purpose: The purpose of this study was to evaluate the relationship between radiation-induced activation of DNA repair genes and radiation induced apoptosis in A431 cell line. Materials and Methods: Five and 25 Gys of gamma radiation were given to A431 cells by a Cs-137 cell irradiator. Apoptosis was evaluated by flow cytometry using annexin V-fluorescein isothiocyanate and propidium iodide staining. The expression of DNA repair genes was evaluated by both Northern and Western blot analyses. Results: The number of apoptotic cells increased with the increased radiation dose. It increased most significantly at 12 hours after irradiation. Expression of p53, p21, and hRAD50 reached the highest level at 12 hours after 5 Gy irradiation. In response to 25 Gy irradiation, hRAD50 and p21 were expressed maximally at 12 hours, but p53 and GADD45 genes showed the highest expression level after 12 hours. Conclusion: Induction of apoptosis and DNA repair by ionizing radiation were closely correlated. The peak time of inducing apoptosis and DNA repair was 12 hours in this study model. hRAD50, a recently discovered DNA repair gene, was also associated with radiation-induced apoptosis.
