Extraction of Intercellular Material from Rice Leaf Tissue (수도의 세포간극 물질 추출)

  • The intercellular material was extracted from rice leaf tissue. The quantitative tests of protein and soluble carbohydrate, and activity of peroxidase, showed differences between tissue extract and intercellular material. Also electrophoretic patterns of peroxidase and esterase isozymes were not similar between them. It indicated that the intercellular material which was extracted, was not the mixture of cellular materials.

Changes of the Cell Wall Structures during Maturation of Jujube Fruits (성숙 중 대추의 세포벽 조직 변화)

  • This study was investigated to the structure of cell wall during maturation for the research of softening of jujube fruits. Cell was hardly combined with each other untill turning stage, but middle lamella of cell wall was splited at mature stage and was observed splited cell. The middle lamella of cell wall was not observed at green mature stage, but was observed at turning stage. Cell wall was degraded at mature stage. It was observed mitochondria, endoplasmic reticulum et. at in jujube fruit of green mature stage, but cytoplasm and organelle was attached on cell wall as vacuole was grown up after turning stage.

Mechanism for Gating of Gap Junction Channel. (간극결합채널의 개폐기전)

  • Gap junction is a membrane structure facilitating the direct transmission of several ions and small molecules between two cells. It is also called an 'intercellular channel' to distinguish it from other well-known cellular channels (e.g. sodium and potassium channels). Gap junction channels are not passive conduits, rather the ion channels modulated by several stimuli including pH, calcium ion, voltage, and a chemical modification (mainly known as phosphorylation). Among them, the effects of voltage on the gating of gap junction channels have been well studied. Gap junction channels are more sensitive to the transjunctional potential ($V_j$) between two cells rather than the membrane potential($V_m$) between inside and outside the cell. In this review, I will summarize the general properties of gap junction channel and discuss the gating mechanism for the gap channels.

Effect of Light Quality on Growth of Cucumber Plug Seedlings (광질이 오이 플러그 묘의 생육에 미치는 영향)

  • 광파장 및 조사시기에 따른 오이묘의 생육 반응을 검토하였다. 광원에 따른 오이 묘의 생육상태를 조사해본 결과 청색광, 적색광을 처리한 것이 생체중, 하배축 무게, 근중이 증가하면서 하배축의 신장이 억제되었고, 청색광과 적색광을 혼합하여 야간 12시간씩 조사하여 30일간 육묘해 본 결과 뿌리의 발육도 우수하고, 줄기와 잎의 생육 정도에서도 우량묘의 소질을 보였다. 청색광과 적색광을 혼합하여 야간 6시간과 12시간씩을 조사하여 30일간 육묘한 결과 하배축의 길이는 각각 60.0mm와 44.9mm였고, 하배축의 무게는 0.59g, 0.62g으로 나타나 12시간 조사구가 하배축의 신장억제효과가 큰 것으로 나타났다. 하배축의 엽록소 함량은 청색광과 적색광의 조사시간이 길어질수록 뚜렷이 증가하였고, 식물체도 진한 녹색을 나타냈다. 광처리구와 무처리구의 오이묘 하배축의 횡단면과 종단면을 검경하여 세포구조를 비교해 본 결과 광처리구의 횡단면 표피조직은 단층의 밀착된 세포로 되어 있으며, 바깥쪽 세포는 현저하게 두꺼웠다. 후각조직의 세포는 작고 부정형이며, 세포간극이 조밀하였으며, 세포구조도 치밀한 특성을 보였다. 종단면은 광처리구에서 세포가 짧고 세포간에 간극이 없는 반면 무처리구는 세포가 길고 뚜렷이 구분되지 않은 것이 관찰되어 대체로 광처리구는 무처리구에 비해 세포신장이 억제되는 대신 세포구조가 치밀하여 식물체의 도장이 억제된다는 것을 알 수 있었다.

Anatomical Observation of Vitrified and Glaucous Leaf from Rehmannia glutinosa Plant Produced in Vitro (지황 기내배양시 투명화된 잎과 정상잎간의 조직학적 관찰)

  • Addition of growth inhibitors such as ancymidol, ABA, chloromequat, and pachlobutrazol into MS medium had no effect to preventing vitrification in cultures of Rehmania glutinosa. Anatomical investigation revealed that vitrified thick leaf tissue in vitro had larger intercellular space with poor development of sponge and pallisade tissue compared to those of in vitro grown glaucous and field grown plants. In vitro grown glaucous leaf had smaller and round type stomata showing distinguishable guard and subsidiary cell than those of reestablished plantlets into soil whereas abnormal stomata and poor development of epicuticular wax on the surface of leaf was observed in verified plantlet.

Analysis of CMTX Mutants Using Connexin Membrane Channels (커넥신 세포막채널을 이용한 씨엠티엑스 돌연변이체의 분석)

  • Mutations in the human connexin 32 (Cx32) gene are responsible for X-linked Charcot-Marie-Tooth (CMTX) disease. Although over 300 different mutations have been identified the detailed molecular etiology of CMTX disease is poorly understood. Several studies reported that connexin membrane channels share most biophysical properties with their parental gap junction channels. In this study, two connexin mutant membrane channels (one mutant channel called the M34T channel in which the methionine residue at the $34^{th}$ position of the Cx32 protein is replaced with threonine residue and the other mutant channel called the T86C channel in which the threonine residue at the $86^{th}$ position is replaced with cysteine residue) associated with CMTX mutations were characterized at the single-channel level instead of using mutant gap junction channels. The biophysical properties of the M34T channel were very similar to those of the gap junction channel formed by M34T mutation. In addition, the mutant membrane channel study revealed the reversal of the gating polarity, the loss of fast gating and the gain of slow gating. The T86C channel also behaves like its parental wild type Cx32 membrane channel. Taken together, these results suggest that a study using connexin membrane channels is useful to characterize CMTX mutants.

Induction of intercellular chitinase isozymes in bean and soybean leaves under stress (강낭콩과 대두 잎에서 세포간액 키틴분해효소의 분포와 유도)

  • Using the enzyme activity staining, we studied the induction and distribution of chitinase isozymes, pathogenesis-related proteins, in intercellular fluids of bean and soybean leaves under stress conditions. The chitinase in intercellular fluids was barely detected in healthy plant leaves. By treatment of ethylene, pathogen (Fusarium oxysporum), or wounding, only 34 kD intercellular endochitinase was induced in bean leaves, while 30 kD and 36 kD intercellular endochitinases were induced in soybean leaves.

  • When a piece of the dorsal skin(cat fish) was observed under a light microscope, melanophores were horizontally expanded and had a few processes filled with melanosomes. They were isolated from one another. Some melanophores looked black, while others were d&k brown. On observation with a transmission electron microscope, the epidermal melanophore of cat fish was highly branched and small segments of processes were found frequendy m the vidnity of the interecellular spaces. The cross section of the processes of melanophore was almost circular, and often invested by a thin layer of epidermal cells. Some processes, however, occurred free m the wide interecellular spaces or at the cytoplasm of the superilcial layers. In the mature melanophore, the cell organelles including melanosomes, mitochondria and free ribosomes were prominent in the perinuclear portions. Many melanosomes were spherical or elipsoidal in shape. Each melanosomes was surrounded by a limiting membrane. The processes of the mature melanophore were well developed, but the processes of the immature melanophores were incompletely developed.

  • In the present work, production of organophosphorus hydrolase (OPH) that is secreted in periplasmic space of recombinant Escherichia coli was performed for degradation of environmental toxic organophosphate compounds, paraoxon. The optimal conditions for enhancement of OPH production were 1.0 mM isopropyl-${\beta}$-D-thiogalactopytanoside (IPTG), 0.25 mM $Co^{2+}$, and 0.1 mM ethylenediamine tetraacetate (EDTA). Under these culture conditions, the maximum OPH production was $174Unit/L{\cdot}OD$. In addition, 1 mM of paraoxon was completely degraded by OPH. These results can be used as a bioremediation tool for removal of environmental toxic organophosphate compounds remaining in soil and aquatic environment.

  • At 5 DAS/T, leaf primordia of rice stems that were grown under dry condition in transverse sections were strongly stained while those under water condition had many aerenchyma cells well developed. On the other hand, leaf primordia and large air spaces in stem of transplanted rice were well developed. Rice in leaf anatomy had small and fine epidermal cells, chlorophyllous mesophylls, and bulliform cells but had no chlorophyllous vascular bundle sheath cells, while barnyardgrass leaf had large, rough and irregularly arranged epidermal cells, chlorophyllous vascular bundle sheath cells, and non-bulliform cells but had no chlorophyllous mesophylls. Epidermal cells of transplanted rice, however, were well developed, differentiated and sclerified. Cross sections of rice root under dry condition showed cell contents, regularly arranged cells, non-intercellular spaces and non-aerenchyma while under water condition had well-developed intercellular spaces, aerenchyma cells, small and densely arranged epidermis, sclerified exodermis and sclerenchyma cells. But root anatomy of transplanted rice consisted of finely, regularly arranged epidermis, well-developed intercellular spaces and nucleous cells.
