Growth Promotion of Pavlova viridis by Bacteria Isolated from the Microalga (파블로바 비리디스로부터 분리한 세균에 의한 미세조류의 생장 촉진)

  • The marine microalga Pavlova viridis can grow fast and has the ability to accumulate essential nutrients for culturing marine animals, such as EPA and DHA, and it has been used as food for raring larval fish and prawn. The symbiotic relationship between the flagellate microalga Pavlova viridis and its associated bacteria was investigated. An axenic culture of P. viridis was obtained by repeated treatment of the microalga with an antibiotic cocktail. The axenic status was confirmed after sub-culturing three times in a sterile f/2 medium without an antibiotic. The axenic alga was then co-inoculated with five bacteria, arbitrarily designated as I1–I5, isolated from the alga to test the growth promotion of the algae. All bacterial strains promoted the growth of P. viridis, and bacterial isolate I3 was the most effective among the five bacteria tested. The cell number of P. viridis in the co-culture with I3 was significantly higher than that of the control culture. A sequence analysis of the 16S rRNA gene isolated from I3 revealed a 97% nucleotide sequence similarity to that of Citrobacter sp. The growth of strain I3 was also significantly enhanced by co-culturing with P. viridis, indicating a symbiotic relationship between the microalga and its associated bacterium. The association between the microalga and bacterium was confirmed by scanning electron microscopy.

The Potential Probiotic and Functional Health Effects of Lactic Acid Bacteria Isolated from Traditional Korean Fermented Foods (한국 전통발효식품에서 분리한 유산균의 프로바이오틱스 특성 및 건강기능성 연구)

  • This study investigated the probiotic properties and physiological activities of Korean fermented foods such as sikhae, young radish kimchi, and bean-curd dregs. Among the isolated lactic acid bacteria, Pediococcus inopinatus BZ4, Lactobacillus plantarum SH1, Lactobacillus brevis SH14, Pediococcus pentosaceus YMT1, and Leuconostoc mesenteroides YMT6 demonstrated a greater than 60% survival rate at pH 2.5, along with an excellent survival rate even at 0.3% bile acid. These five bacteria showed strong flocculation ability in autoaggregation and coaggregation tests, indirectly clustering useful micro-organisms and inhibiting the attachment of pathogenic bacteria. In a cell surface hydrophobicity test, these bacteria showed adhesion to three solvents (ethyl acetate, chloroform, and xylene) and high hydrophobicity, thereby indicating excellent indirect cell adhesion to intestinal cells. The cell-free supernatants and intracellular extracts of the five lactic acid bacteria showed antioxidative activity in the form of 2,2-Diphenyl-1-picrylhydrazyl and 2,2'-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging ability and lipid peroxidation inhibition. Antimicrobial activities were also observed in four pathogenic bacteria, namely E. coli KCTC 2571, H. pylori HPKCTC B0150, L. monocytogenes KCTC 13064, and S. aureus KCTC 1916. These results demonstrate that these five lactic acid bacteria could be used as probiotics with antioxidant and antimicrobial properties.

A Study on the Performance Development of Sewage Concrete by Application of Antibiotics (항균제 도포에 의한 하수시설 콘크리트의 성능향상에 관한 연구)

  • Recently sewage facilities mainly consisted of concrete structures are being deteriorated seriously by biodeterioration originated from sulfur-oxidizing bacteria. In this study, to prevent biochemical corrosion of the sewer concrete, antibiotics which prevent growth of sulfur-oxidizing bacteria were developed and antimicrobial performance of it was investigated. After that, to consider applicability of antibiotics to concrete, physical properties of concrete covered with antibiotics were investigated. As a results of the study, it was proved that the antimicrobial performance of antibiotics was available. Also compressive strength and bond strength of concrete didn't closely connected with antibiotics, and resistance to abrasion, water absorption, air permeability, carbonation, salt damage and chemical attack of concrete was improved remarkably by covering with it.

  • Aortic valve replacement with aortic allograft has been considered a treatment of choice for aortic valve disease secondary to bacterial endocarditis because of its good homodynamic performance and higher resistance to infection. The aortic root replacement technique might be superior to the subcoronary allograft implantation technique with regard to aortic regurgitation. A 46 yea,rs old male patient had acute aortic regurgitation with progressing heart failure secondary to acute bacterial endocarditis. The patient underwent emergent aortic root replacement using 20 mm aortic allograft. At operation, right coronary cusp perforation and heavy calcification of commissure between right and left coronary cusp were observed. The patient recovered well and postoperative echocardiography demonstrated no aortic regurgitation. Inflammatory signs were subsided after 8 weeks of antibiotics therapy. Medically uncontrolled acute bacterial endocarditis was treated successfully by aortic root replacement using aortic homograft.

Screening of Bifidobacteria for the Development of Probiotics Inhibiting Intestinal Pathogenic Bacteria (장내 유해세균을 억제하는 양돈용 프로바이오틱스 개발을 위한 비피도박테리아 탐색)

  • In order to isolate probiotic lactic acid bacteria possessing high inhibitory activities against porcine and zoonotic pathogens, such as enterotoxigenic E. coli, Salmonella Typhimurium, and Clostridium perfringens, a total of 65 anaerobic strains were initially isolated from a variety of sources including cattle rumen fluids, chicken intestines and swine feces. Four Bifidobacterium strains were selected for their high anti-pathogenic bacterial activities. By using the 16S rDNA sequencing method, three B. boum strains and one B. thermophilum were identified. B. thermophilum demonstrated the best adhesive ability to epithelial cells of swine intestine among the isolates. Indeed, B. thermophilum was seen to have superior characteristics as a probiotic for swine, as judged by their high growth inhibitory activities against various pathogens, and high acid- and bile-tolerance.

  • If you don't regularly go to the dentist to check your teeth, it is difficult to notice cavities or various diseases of your teeth until you have pain or discomfort. Dental plaque is produced by the combination of food or foreign substances and bacteria in the mouth. Starch breaks down from the bacteria that form tartar. The acid that occurs at this time melts the enamel of the teeth and becomes a cavity. So tartar management is important. Poppyrin, the metabolism of bacteria in the mouth, reacts at 405 nm wavelengths and becomes red fluorescent, which can be seen by imaging through certain wavelength filters. By the above method, Frag and tartar are fluorescently detected and photographed with a yellow series of filters that pass wavelengths of 500 nm or more. It uses MATLAB to detect and display red fluorescence through image processing. Using the difference in voltage between normal teeth and tartar through an optical measuring circuit, it was connected to an Arduino and displayed on the LCD. This allows the user to know the presence and location of dental plaque more accurately.

  • Anaplasma phagocytophilum major surface protein-2 (Msp2 or p44) is the immunodominant outer membrane protein of the bacterium. Recently, we disclosed that Msp2 was an A. phagocytophilum adhesin for binding to host neutrophils and HL-60 cells, probably mediated by attachment to platelet selectin glycoprotein ligand-1 (PSGL-1). In this study, we further elucidated that Msp2 bound to PSGL-1/FucT IV-transfected BJAB but not nontransfected BJAB cells. Binding of recombinant Msp2 or cell (lee bacteria to the surface of PSGL-1/FucT IV-transfected BJAB cells was significantly higher than to nontransfected BJAB cells (p<0.01 and p<0.01). Also, Msp2 monoclonal antibody and soluble recombinant Msp2 as antagonist led to concentration-dependent reductions in A. phagocytophilum adhesln (p<0.05 and p<0.01) to transfected BJAB cells. Thus, we conclude that Msp2 of. A. phagocytophilum acts as an adhesin by which the bacterium binds to PSGL-1 on host neutrophils and myeloid cells.

CHANGES IN ADHESION OF STREPTOCOCCUS MUTANS TO NANOCOMPOSITE RESINS AFTER ACIDULATED PHOSPHATE FLUORIDE GEL APPLICATION (나노컴포지트에서 Acidulated Phosphate Fluoride 적용에 따른 Streptococcus mutans 부착량 변화)

  • Topical application of APF gel can increase the surface roughness of resin composites and the roughened surfaces may allow increased bacterial accumulation and surface staining. Resin specimens of two proprietary resin composites, Filtek Z250(FZ) and Filtek Supreme Universal(FS), and experimental resin composites containing 0%, 3%, 6% nanofillers(E0, E3, E6) were fabricated and divided into two groups of the same number; APF treatment group and no treatment group. The amount of S. mutans adhered to specimens and the mean surface roughness(Ra) were measured. The results were as follows; 1. In no treatment group, the amount of S. mutans adhered to FS was the smallest. It was significantly different from those of FZ, E3, E6(p<0.05) although it was not significantly different from that of E0(p>0.05). 2. For all resin composites used, the amount of S. mutans adhesion in APF treatment group was significantly greater than that in no treatment group(p<0.05). 3. In APF treatment group, the amount of S. mutans adhesion was significantly different between materials(p<0.05), and increased in order of FS, FZ, E0, E3 and E6. 4. Difference of the surface roughness(Ra) between materials was not statistically significant in both no treatment group and APF treatment group(p>0.05)
