Nitric oxide production and inducible nitric oxide synthase expression induced by Porphyromonas gingivalis lipopolysaccharide (Porphyromonas gingivalis의 세균내독소가 RAW264.7세포에서의 nitric oxide의 생성과 inducible nitric oxide synthase의 발현에 미치는 영향 및 기전)

  • 본 연구는 치주질환 주요 병인균주 중의 하나인 Porphyromonas gingivalis의 세균내독소가 마우스 대식 세포주인 RAW264.7 세포에서의 nitric oxide의 생성과 iNOS의 발현에 미치는 영향을 분석하고 그 기전을 규명하기 위해 수행되었다. Butanol추출법과 phenol-water법에 의해 P. gingivalis 381로부터 세균내독소를 추출하였으며, NO의 생성은 배양 상층액 내의 nitrite 농도를 측정하여 결정하였다. 또한, iNOS의 western blot 분석과 reverse transcription (RT)-PCR 산물의 분석을 수행하였다. P. gingivalis의 세균내독소는 부가적인 자극이 없는 상태에서도 iNOS의 발현과 NO 생성을 유발하였으며, NF- ${\kappa}B$, microtubule polymerization, protein tyrosine kinase, 그리고 protein kinase C 등이 P. gingivalis 세균내독소에 의한 NO 생성에 간여하는 것으로 여겨진다. 또한, P. gingivalis 세균내독소에 의한 NO 생성에는 L-arginine이 요구되었다. P. gingivalis 세균내독소에 의한 NO 생성은 염증성 치주질환의 발병과 진행에 있어 중요한 역할을 하는 것으로 여겨진다.

  • Cross contamination between a patient and rescuer or CPR trainees can occur when performing mouth to mouth ventilation during cardiopulmonary resuscitation (CPR). On the other hand, there has been a lack of research on the filtration efficacy of face shields that are designed to protect people from cross-contamination. This study aims to secure the safety of rescuers from communicable diseases in pre-hospital emergency settings and CPR trainees by verifying the protective effects of face shields. The FA shield and CM Shield were used to verify the safety. The bacteria collected from filters used by CPR trainees were incubated. These incubated bacteria were smeared onto the new filters, and were then blown out through the filters using a Bag Valve Mask (BVM) and the pathogens at the front and the back of the filters were checked. While the FA shield was effective in preventing the transmission of pathogens, the CM shield did not prevent the transmission of pathogens. Therefore, some of face shields that received national certification are ineffective in preventing cross-contamination. Accordingly, it is necessary to verify the safety of other face shields used domestically.

Effects of the Proliferation of Beneficial and Harmful Enteric Bacteria after Intake of Soybean Fermentation (Zen) Produced by a Mixture of Lactobacilli and Saccharomyces (Lactobacilli와 Saccharomyces 혼합균주의 대두발효액(Zen) 섭취 후 장내 유익세균과 유해세균의 증식에 미친 영향)

  • Purpose: The purpose of this study was to investigate the increase or decrease of important intestinal beneficial bacteria and inhibitory bacteria in 30 stools of clinical subjects after ingesting Zen fermentation broth as a mixed microbial fermentation solution for eight weeks. Methods: Intestinal bacteria were identified by PCR amplification using specific primers. Results: Bifidobacterium genus gi% of test group ingested Zen-fermented broth was 55.15% before and 70.1% after ingestion, so it was a significant difference (p<.009). Lactobacillus genus of the test group was 46.87% before and 60.91% after ingestion, it was a significant difference (p<.01). Clostridium genus of the test group was 85.64% before and 65.99% after ingestion. There was a significant difference (p<.017) as the pre-post-difference decreased to -19.65%. Bacteroides genus of the test group was 17.11% before and 20.22% after ingestion. There was a significant difference (p<.048) as the pre-post-difference increased to 3.11%. Prevotella genus of the test group was 14.01% before and 16.79% after ingestion, so it was not a significant difference. Conclusions: Intestinal bacteria increased the proliferation of beneficial bacteria and suppressed harmful bacteria in the intestines after ingesting the Zen-fermented broth of the mixed microorganism. The Zen fermentation broth evaluated as a beneficial drink for intestinal health.

Comparison of community structure of sulfate reducing bacteria in rice paddy and dry farming soils (논과 밭 토양의 황산염 환원세균 군집 구조 비교)

  • The goal of this study was to identify relationships between the composition of sulfate reducing bacterial assemblages and terminal restriction fragment length polymorphism (T-RFLP) patterns in rice paddy and dry farming soils. Samples of organic farming soils, conventional farming soils, and dry field farming soils were collected in August and November. Analyses of the soil chemical composition revealed similar total nitrogen, total carbon and total inorganic phosphorus levels; however, the moisture content and total carbon were higher than in the other soils in both August and November, respectively. Sulfate reducing bacteria utilizing lactic acid were more widely distributed than those that used acetic acid, and the number of sulfate reducing bacteria in organic farming soil was most abundant. Phylogenetic analysis based on 181 clones revealed that most showed low similarity with cultured sulfate reducing bacteria, but more than 90% similarity with an uncultured sulfate reducing bacteria isolated from the environment. T-RFLP analysis revealed that fragments of 91, 357, 395, and 474 bp were most common, and the community structure of sulfate reducing bacteria changed seasonally.

A Study on the Change of the Corrosion Products by the Activity of Iron Reducing Bacteria for Corrosion Carbon Steel (부식철편에 있어서 철환원능력을 갖춘 세균의 활동에 의한 부식생성물의 변화)

  • Bacteria with ability for iron reduction in the soil can use corrosion products of iron remains as energy source. The activities of this bacteria cause the change of corrosion products. As a result, it can be difficult to identify corrosion products promoting corrosion of iron remains. The purpose of this study, is to investigate the change in corrosion products that bacteria causes and to improve understanding about the corrosion of iron remains. To simulate corroded condition of excavated iron remains, carbon steel corroded by solution of NaCl and $Na_2SO_4$ was prepared. Then the prepared carbon steel was immersed in a liquid medium with bacteria. The incubation period was 42days. After experiment, the carbon steel was analyzed by SEM-EDS, X-ray diffraction method. The result is that the carbon was changed to green because of activity of bacteria and that the plate crystal and lozenge crystal were generated on the corrosion specimen. Also, we confirmed that the activities of bacteria differenciated colors and forms of corrosion products.

  • Volatiles exist ubiquitously in nature. Volatile compounds produced by plants and microorganisms confer inter-kingdom and intra-kingdom communications. Autoinducer signaling molecules from contact-based chemical communication, such as bacterial quorum sensing, are relayed through short distances. By contrast, biogenic volatiles derived from plant-microbe interactions generate long-distance (>20 cm) alarm signals for sensing harmful microorganisms. In this review, we discuss prior work on volatile compound-mediated diagnosis of plant diseases, and the use of volatile packaging and dispensing approaches for the biological control of fungi, bacteria, and viruses. In this regard, recent developments on technologies to analyze and detect microbial volatile compounds are introduced. Furthermore, we survey the chemical encapsulation, slow-release, and bio-nano techniques for volatile formulation and delivery that are expected to overcome limitations in the application of biogenic volatiles to modern agriculture. Collectively, technological advances in volatile compound detection, packaging, and delivery provide great potential for the implementation of ecologically-sound plant disease management strategies. We hope that this review will help farmers and young scientists understand the nature of microbial volatile compounds, and shift paradigms on disease diagnosis and management to aromatic (volatile-based) agriculture.

Protection of Tobacco Plants from Bacterial Wilt with an Avirulent Isolate of Pseudomonas solanacearum (비병원성 Pseudomonas solanacearum을 이용한 담배 세균성마름병의 방제)

  • Microbial antagonism between virulent and avirulent isolates of Pseudomonas solanacearum was studied in relation to the control of bacterial wilt of tobacco. In nutrient broth media or in soil, the avirulent isolate of P. solanacearum grew faster than did the virulent one. Inhibitory effect of avirulent isolate against growth of virulent one was negligible in mixed culture of the two isolates. The disease severity of bacterial wilt was significantly reduced when the roots of cultivar BY 4 susceptible to bacterial wilt was dipped in suspension of an avirulent isolate for 6 hours prior to transplanting to the soil infested with virulent bacteria. When the seedlings of tobacco were poured with the suspension of an avirulent isolate onto the soil in pre-planting pots 24 hours before ransplanting, there was a significant reduction in disease severity in the field. However, the reduction was noticed until early July, but after middle of July, no difference between the avirulent isolate-treated and non-treated plants was found in severity of the bacterial wilt.

Study on Development of Novel Biopesticides Using Entomopathogenic Bacterial Culture Broth of Xenorhabdus and Photorhabdus (Xenorhabdus 및 Photorhabdus 세균 배양액을 이용한 생물농약 개발에 관한 연구)

  • Two groups of entomopathogenic bacteria, Xenorhabdus and Photorhabdus, are known to suppress insect immune responses by inhibiting eicosanoid biosynthesis. This study used these bacterial culture broths to develop novel biochemical insecticides against the diamondback moth, Plutella xylostella. Though the bacterial culture broths alone showed little insecticidal activity, they significantly enhanced pathogenicity of Bacillus thuringiensis against the fourth instar larvae of P. xylostella. Sterilization of the bacterial culture broth by autoclaving or $0.2\;{\mu}m$ membrane filtering did not influence the synergistic effect on the pathogenicity of B. thuringiensis. Three metablites identified in the culture broth of X. nematophila also showed similar synergistic effects. In field test, both entomopathogenic bacterial culture broth also enhanced the control efficacy of B. thuringiensis against P. xylostella.

Identification of Bacterial Strains Adhered to Dog Hair and Antibacterial Effects of Scutellaria baicalensis Gorgi (애완견의 털에 부착되어 있는 세균의 분리 및 황금의 살균효과)

  • This study was carried out to identify bacterial strains adhered to domestic pet dog hair and to identify antibacterial extracts from natural compounds. A total of 76 strains were isolated from dog hair. The most common species isolated was Staphylococcus spp. (41 isolates), followed by Micrococcus spp. (21 isolates), Enterococcus spp. (8 isolates), Bacillus sup. (3 isolates), Exiguobacterium sup. (2 isolates), Shigella spp. (1 isolate) and Zoogloea spp. (1 isolate). These results suggested that dog hair could be a source of bacterial contamination to human. The susceptibility of isolates to antibiotics and antibacterial activities of the natural compounds were examined by disk diffusion method. Water and ethanol extract from Scutellaria baicalensis revealed high antibacterial activities against Staphylococcus, Micrococcus, Enterococcus and Shigella. Our results suggest that the extract of Scutellaria baicalensis can be used a antibacterial agent against the antibiotic-resistant microorganisms.

Bacterial Biomass and Secondary Productivity in Naktong River Estuary (낙동강 하구생태계의 세균 생물량과 이차생산성)

  • To investigate the bacterial potentials for utilizing dissolved organic matter in highly eutrophic estuary, the annual fluctuations of microbiological and physicochemical environmental parameters were analyzed in Naktong River Estuary. Total bacterial number ranged from 0.33 to $2.09{\times}10^7$ cells/ml, and correlated with the heterotrophic bacterial numbers in more eutrophic sites, especially. Bacterial biovolume and biomass varied between 0.064 and 0.156 2.09${\mu}m^3$/cell, 0.163 and 1.036 ${\mu}g$-C/ml, respectively. Bacterial secondary productivity ranged from 0.24 to 60.86 ${\mu}g$-C/l/h, and showed high correlations with the environmental parameters of pollution indicator. The seasonal variation pattern of bacterial productivity in freshwater sites was high in winter and low in summer, which was interpreted as the results of pollution loads varied with the amount of rainfall. In seawater site, the pattern was different from those of freshwater sites; high in summer and low in winter. In this site, the values of bacterial productivity showed positive correaltions with chlorophyll a, heterotrophic bacterial number, and temperature (r>0.5, p<0.05). These results suggested that the main source of organic matter which influences the bacterial productivity may be allochthonous materials in the upper freshwater zone of Naktong River Barrage, and autochthonous algal excretory products in the lower seawater zone of Naktong River Barrage.
