Yields of oyster mushroom(Pleurotus ostreatus) on addition rate of Juglans mandshurica sawdust (가래나무(Juglans mandshurica) 톱밥의 첨가량이 느타리버섯의 수량에 미치는 영향)

  • This study was carried out to investigated optimum mixing ratio of Korean natural Juglans mandshurica using as functional plants. Total nitrogen and carbon source of J. mandshurica was 0.21% and 46.0%, respectively and C/N ratio was 219. Total nitrogen source and pH of substrate mixed with J. mandshurica was 2.3~2.9 and 5.0, respectively. The contents of CaO, MgO and $Na_2O$ at J. mandshurica media were higher at harvest media than inoculation media. Mycelial growth was the fastest at J. mandshurica 20%, and slower by increase of J. mandshurica substrate. Yields of fruiting body show the highest to 163 g/850 mL of medium which are addition 10% of J. mandshurica sawdust, and diameter and thick of pileus were the highest, too. The L value of pileus and stipes were increased by increase of J. mandshurica substrate, but there was no significant difference in the a-value and the b-value. The contents of $P_2O_5$ and $K_2O$ of fruiting body were increased at J. mandshurica substrate, but there was no significant difference in contents of CaO, MgO and $Na_2O$. The contents of Cu of fruiting body was decreased by increase of J. mandshurica substrate, but Fe, Mn and Zn were increased.

Quality Characteristics of Soy Ice Cream Prepared with Fermented Soybean Powder Base and Oligosaccharide and Its Blood Glucose Lowering Effect (대두분말 발효 베이스와 올리고당으로 제조한 콩아이스크림의 품질특성과 혈당개선능)

  • The purpose of this study was to improve the quality characteristics of soy ice cream supplemented with oligosaccharide, and to test its blood glucose lowering effect. Boiled soybean powder was compared to parched soybean powder and to milk, as an ingredient. The soybean powder base was prepared by incubating with fructooligosaccharide (FOS) and apple juice, along with Lactobacillus acidophilus and L. bulgaricus at $30-40^{circ}C$ for 24 hr. With the fermentation process, the fishy smell of the soybean was removed and the taste improved. The overrun and melt-down values of the boiled soybean ice cream were significantly higher than those of the parched soybean ice cream, although they were significantly lower than those of the milk ice cream. The sensory characteristics of the soy ice cream prepared with the fermented base of boiled soybeans were significantly improved, as compared to those of the ice cream made using parched soybeans, but they were not significantly different from those of the milk ice cream. The blood glucose level at 120 min after ingestion of the ice cream prepared with FOS and the fermented base of boiled soybean powder was significantly lower than that occurring with the milk ice cream made with sugar.

Immune Cell Activation and Co-X-irradiation Effect of Eleutherococcus senticosus Maxim Root (가시오갈피 뿌리의 면역세포 활성 및 방사선 병용효과)

  • Purpose: This study was performed to investigate the effects of immune cell activation and the antitumor effect for the combination of treatment with X-irradiation and E/eutherococcus senticosus Maxim Root (ESMR) on mouse tumor cells. Materials and Methods: ESMR (250g) was extracted with 80% methanol, concentrated under decompression and lyophilized. To determine whether ESMR is able to activate the immune cells or not, the proliferation of splenocytes in vitro and the number of B cells and T cells in splenic lymphocytes in ESMR-pretreated mice were evaluated. X-irradiation was given to the mouse fibrosarcoma tumor cells (FSa II) by 250 kv X-irradiation machine. The cytotoxicity of ESMR was evaluated from its ability to reduce the clonogenecity of FSa II cells. In X-irradiation alone group, each 2, 4, 6 and 8 Gy was given to FSa II cells. In X-irradiation with ESMR group, 0.2 mg/ml of ESMR was exposed to FSa II cells for 1 hour before X-irradiation. Results: The proliferation of cultured mouse splenocytes and thymocytes were enhanced by the addition of ESMR in vitro. The number of B cells and T cells in mouse splenic lymphocytes was significantly increased in ESMR pretreated mice in vivo. In FSa II cells that received a combination of 0.2 mg/ml of ESMR with X-irradiation exposure, the survival fraction with a dose of 2, 4 and 6 Gy was $0.39{\pm}0.005$, $0.22{\pm}0.005$ and $0.06{\pm}0.007$, respectively. For FSa II cells treated with X-irradiation alone, the survival fraction with a dose of 2, 4 and 6 Gy was $0.76{\pm}0.02$, $0.47{\pm}0.008$ and $0.37{\pm}0.01$. The difference in the survival fraction of the mouse FSa II cells treated with and without ESMR was statistically significant (p<0.05). Conclusion: Treatment with ESMR increased cell viability of mouse splenocytes in vitro and especially the subpopulation of B cells and T cells in splenocytes in ESMR-pretreated mice. However, treatment with ESMR did not increase the level of Th and Tc subpopulations in the thymocytes. Treatment with the combination of ESMR and X-irradiation was more cytotoxic to mouse tumor cells than treatment with X-irradiation alone; this finding was statistically significant.

Isolations of Orchid Mycorrhizal Fungi from the Korean Native Orchid Plants (한국의 자생 난과식물에서 난균근균(蘭菌根菌)(Orchid Mycorrhizal Fungi) 분리)

  • This study was to identify the orchid mycorrhizal fungi and to test whether the orchid plants antificially inoculated with this fungus showed better growth them uninoculated plants. Symbioses in the root cells of the native plants of Cymbidium goeringii collected were observed and the digestive forms of peletons were also observed in various native roots. Two types of hyphae, thick $(7{\sim}10\;{\mu}m)$ and thin $(2{\sim}4\;{\mu}m)$ in thickness, were conclusively found to be from various native orchid roots. The symbiotic fungus was isolated by several agars and identified as a Rhizoctonia repens or a R. endophytica var. endophytica. Symbioses on the plantlets of C. karnan and Cymbidium hybrid 'Onomoron' were evaluated as the isolates inoculated on oatmeal agars. The growth of plantlets were measured with the formations of mycorrhizae in the roots. R. repens was shown to be the better isolate than the other in growth stimulation of plantlets on oatmeal agars when grown for two months. The two types of hyphae in the root cells under nature were speculated from the different fungal isolates of Rhizoctonia. Further isolates would be needed for application works for the orchid industries.

The Effect of Root Exudate and Chemotaxis on Host Recognition in Soybean-Bradyrhizobium Symbiosis (대두(大豆)-근류균(根瘤菌) 공생(共生)에서 뿌리분비물(分泌物)과 화학주성(化學走性)이 숙주인식(宿主認識)에 미치는 영향(影響))

  • This study was carried out to research the effect of the chemotaxis of Bradyrhizobium japonicum KCTC 2422 and its mutant toward soybean root exudate and to elucidate the effect of the lectin of host specificity (Host Recognition) in soybean-Bradyrhizobium symbiosis. The results obtained were as follows: The homogeneities of the purified lectins from soybean and pea seed was ascertained chromatographically and electrophoretically. Gel electrophoresis of soybean lectin in the presence of sodium dodecyl sulfate appeared a single protein band, whereas pea lectin appeared two protein bands. Soybean lectin from 2 cultivars formed immunoprecipitin arcs at same position with anti-soybean lectin rabbit IgG, but pea lectin did not form immunoprecipitin lines with anti-soybean lectin rabbit IgG. Chemotactic responses of KCTC 2422, LPN-100 and LCR-101 toward proline in capillary assays were 3.1, 1.3 and 1.0-fold above background, respectively. The chemotactic responses of KCTC 2422, LPN-100, and LCR-101 toward Paldal crude root exudate in capillary assays were 3.5, 1.4 and 1.4-fold above background, respectively. The present work shows that B. japonicum and its mutants are capable of very different responses toward root exudate fraction. The chemotactic responses of KCTC 2422 was most with neutral fraction, least with anionic fraction and intermediate with cationic fraction. The nitrogenase activity of soybean nodule was shown in 15days after inoculation with LCR-101. However, we couldn't find out the nodules when soybean was inoculated with LPN-100. From these result we can suppose that the chemotaxis of Bradyrhizobium plays inportant the role of forming the nodule (host recognition) in the soybean-B. japonicum symbiosis.

Fermentation Process for Odor Removal of Oyster (Crassostrea gigas) Hydrolysate and Its Properties (이취 제거를 위한 굴 가수분해물의 발효공정과 제품의 특성)

  • This study was carried out to investigate the optimal processing conditions for odor removal and maximal antioxidant effects of oyster (Crassostrea gigas) hydrolysate. The optimal hydrolysis conditions were 3.3% neutrase as the protease, $50^{\circ}C$ as the hydrolysis temperature, and 8.3 h as the hydrolysis time. Fish odor of enzymatic oyster hydrolysate was greatly reduced during Saccharomyces cerevisiae fermentation at $24^{\circ}C$ with 0.5% glucose. The protein content of the fermentation product from oyster hydrolysate powder was 25.7%, which contained the major amino acids Glu, Asp, Lys, Arg, Gly, and Ala, whereas Leu, Ala, Phe, Val, and Tau were abundant free amino acids. The important minor minerals were Zn and Fe. Toxicity against Chang cells was not observed in the fermentation product from the oyster hydrolysate up to $200{\mu}g/mL$. The results suggest that fermentation with S. cerevisiae could reduce the fish odor of enzymatic oyster hydrolysate. The hydrolysate has potential application as a food ingredients and nutraceutical.

Rapid Detection for Salmonella spp. by Ultrafast Real-time PCR Assay (Ultrafast Real-time PCR법을 이용한 살모넬라의 신속 검출)

  • Salmonella continue to be a major cause of food poisoning worldwide. The rapid detection method of food-borne Salmonella is an important food safety tool. A real-time polymerase chain reaction (PCR) has been used as a rapid method for the detection of pathogens. It has been recently reported that NBS LabChip real-time PCR is a novel, ultrafast, and chip-type-convenient real-time PCR system. We developed the assay method based on NBS LabChip real-time PCR for the rapid detection of Salmonella, which its reaction time was within 20 minutes. Two target genes (invA and stn) were selected to design target specific primers and probes. The new method was validated by checking specificity and sensitivity (limit of detection). This study included forty-two target and twenty-one non-target strains to assess the specificity. This assay was able to identify the 42 Salmonella strains correctly. The limit of detection (LOD) was $10^1copies/{\mu}L$ in Salmonella genomes DNA, while LOD incubated for 4 hr in the inoculated sausage sample ranged from $10^1CFU/g$ to $10^2CFU/g$ as an inoculated cell count. The assay developed in this study could be applied for the investigation of food poisoning pathogens.

Physiological Characteristics and Angiotensin Converting Enzyme Inhibitory Activity of Lactobacillus brevis HLJ59 Isolated from Salted Shrimp (국내 새우젓에서 분리한 Lactobacillus brevis HLJ59의 Angiotensin Converting Enzyme 저해활성 및 생리적 특성)

  • In this study, lactic acid bacteria with high angiotensin converting enzyme inhibitor activity were isolated from Korean fermented food, such as kimchi and salted seafood. The strain HLJ59, isolated from salted shrimp showed the highest angiotensin converting enzyme inhibitor activity in DeMan Rogosa Sharpe broth. Optimum growth temperature of Lactobacillus brevis HLJ59 was at $34^{\circ}C$. Acid treatment at pH 3.0 for 1.5 h decreased cell viability from $9.9{\times}10^8$ CFU/ml to $3.11{\times}10^4$ CFU/ml. The bile extract concentration of 0.3%, 0.5%, and 1.0% in MRS broth did not inhibit the growth of HLJ59. Isolated strain HLJ59 showed more sensibility to amikacin, gentamycin, neomycin, streptomycin, kanamycin, cefmetazole, cephalothin, ampicillin, ticarcillin, sulbactam+ampicillin, amoxicillin+clavulanic acid (AMC), tetracycline, and sulfamethoxazole+trime thoprim (SXT) as compare to other 7 different antibiotics. However, it showed more resistance to cefoxatin, ceftnaxone, penicillin, ciprofloxacin, nalidixic acid, lincomycin, and chloramphenicol.

Production of Korean Traditional Rice-wines Made from Cultures of the Single Fungal Isolates under Laboratory Conditions (각종 누룩 균을 이용한 실험실조건에서의 막걸리 제조)

  • The objectives of this study were to find the nuruk fungal isolate for Korean traditional rice wine and to compare the tastes of rice wines made with its single inoculum. The fungal isolates including the species of Absidia, Mucor, Rhizopus and Aspergillus were used for Korean traditional rice-wine production. The four species were cultured and measured for productions of amylase under various water contents. The four different species tested were different in their growth under the rice cereals with different moisture contents; the species of Absidia and Rhizopus grew better compared to the other species under the lower moisture conditions. The tastes and alcohol concentrations of the rice-wines made with each different inoculum were compared with those of commercial rice wines made with traditional method. As a result, it was speculated that the traditional rice wines were made with nuruk, which is mainly composed of Zygomycetous fungi, such as Absidia, Mucor, and Rhzopus. Also, it was strongly suggested that the traditional nuruk might not be a source of amylase, but a source of fungal inoculum.

Quality Characteristics of Seasoned Skewered Chicken Added with Fermented Carrot Juice (당근발효액 첨가에 따른 닭꼬치의 품질 특성)

  • This study was conducted to evaluate the quality characteristics of seasoned skewered chicken added with fermented carrot juice (FCJ) during storage at $4^{\circ}C$ for 21 days. The carrot juice was fermented using 2% Leuconostoc mesenteroides SM at $25^{\circ}C$ for 18$\sim$20 hr. Seasoned skewered chicken was produced containing 0, 5% and 10% FCJ. The pH and color values of seasoned skewered chicken were decreased with longer storage periods, while the VBN content was increased during storage. Bacteria was not detected in seasoned skewered chicken containing FCJ. The saturated fatty acid in seasoned skewered chicken decreased with increasing additions of FCJ, whereas the unsaturated fatty acid was increased compared to the control. The texture, resilience, springiness, cohesiveness and chewiness of seasoned skewered chicken were not significantly different, while the hardness of seasoned skewered chicken added with 10% FCJ was significantly the lowest (p<0.05). With regard to sensory evaluation, seasoned skewered chicken added with 10% FCJ resulted in higher overall acceptability than other skewered chickens. These results suggest that manufacture of seasoned skewered chicken added with FCJ can help to improve quality of skewered chicken.
