Green algae are important primary producers in aquatic ecosystem, and they are sensitive test species in bioassay. Green algae are broadly used to assess the adverse effects of various chemicals by measuring the inhibition of metabolism, reproduction and survival. In this study, we extensively gathered domestic and foreign toxicity test methods conducted using green algae, which are distributed in Korean water environment. Selected eight domestic green algae were Chlamydomonas reinhardtii, Desmodesmus subspicatus (=Scenedesmus subspicatus), Scenedesmus abundans, Scenedesmus acutus, Scenedesmus quadricauda, Podohedriella falcata (=Ankistrodesmus falcatus), Pseudokirchneriella subcapitata (=Selenastrum capricornutum), and Chlorella vulgaris. Forty four test methods were collected from the standard test ones, government reports, SCI papers and Korean research papers. P. subcapitata and D. subspicatus are the most common test species recommended by the standard test methods. Initial cell density and dilution water were the main differences among the test methods we collected. We proposed the suitable ecotoxicity test methods based on domestic green algae in Korea. This study could be a fundamental basis to establish the ecotoxicity test methods by green algae distributed in Korea.
The Ministry of Environment plans to introduce the Whole Effluent Toxicity (WET) system in Korea. The WET test is well established in developed countries with recognizing of the limitations of physicochemical analysis method and potential risk of chemicals in water medium. Therefore, it is essential to build the ecotoxicity infrastructure for the induction of WET test. In this study, we extensively collected the domestic and foreign toxicity test methods which employ native test species to Korea. And we suggested that the domestic ecotoxicity test methods with domestic test species in Korea through extracting the range of test conditions. Five domestic fish species selected were Carassiu auratus (Crucian carp), Cyprinus carpio(Common carp), Gasterosteus aculeatus (Three spine stickleback), Misgurnus anguillicaudatus (Oriental weather fish) and Oryzias latipes (Japanese medaka), The toxicity test methods with native test species to Korea were collected from the standard methods (OECD, U.S. EPA, ASTM), government reports, SCI papers and domestic papers. We collected the 32 test methods, and suggested the suitable aquatic toxicity test methods for fish. It is expected that this study could prove a useful information to establish the ecotoxicity test methods with domestic aquatic organisms in Korea henceforth.
The use of aquatic species in ecotoxicity research is well established in developed countries. However, there are limitations of using the species that are not native to Korea, and the toxicity data produced by domestic test species are significantly needed to reflect the domestic situation. The purpose of this study was to investigate the domestic species that can be applicable for the aquatic toxicity assessment in Korea. Aquatic toxicity data were collected in the framework of the project 'Development of integrated methodology for evaluation of water environment' to obtain a range of test species used for aquatic toxicity assessment internationally. The test species collected were evaluated in terms of domestic distribution based on the reliable references and the advices of experts. We figured out the 71 test species native to Korea. They included 7 fish, 26 invertebrates (2 annelids, 2 bryozoa, 13 crustaceans, 3 insects, 4 mollusc, 1 platyhelminth, and 1 protozoan), 26 plants (9 diatoms, 14 green algae, 3 macrophytes), and 12 others (2 amphibians, 3 bacteria, 6 blue-green algae, and 1 fungus). The result of this study should be a very useful information for ecotoxicity assessment in aquatic ecosystem, especially in choosing the test species applicable for the ecotoxicity in Korea hereafter.
Water fleas are very important organisms in aquatic ecosystem because they are key constituents of food webs as either glazers or foods for predators. Daphnia magna is a representative test species and it has been extensively used in ecotoxicity evaluation. However, Daphnia magna has not been found yet in Korean water environment. There are limitations of using the foreign species such as Daphnia magna to reflect domestic situations in Korea. Consequently it is mandatory to find domestic species suitable for bioassay, and to develop corresponding toxicity test methods using the domestic species. In this study, we widely collected the domestic and foreign toxicity test methods performed by using domestic water fleas in Korea. The 58 test methods were collected from the standard methods (OECD, US EPA, ASTM), government reports, SCI papers and domestic papers. Ten domestic water fleas selected were Bosmina longirostris, Ceriodaphnia dubia, Ceriodaphnia reticulata, Daphnia obtusa, Daphnia puex, Moina macrocopa, Moina micrura, Simocephalus mixtus, Simocephalus serrulatus, and Simocephalus vetulus. We suggested the domestic ecotoxicity test methods using domestic water fleas in Korea through providing a range of test conditions, and future directions for toxicity test using water flea. This study could be a useful basis for establishing the aquatic toxicity test methods with domestic organisms in Korea.

This study suggests ways to improve the standard test method and judgment criterion for the "Eco-toxicity Test" based on the rules and regulations provided in 'performance and qualifying test standards for marine pollution prevention materials and chemicals' in the Republic of Korea. Compared with the relevant references of other countries, this study attempted to find the limitations in the existing standards. As for the growth inhibition test of algae using Skeletonema costatum as an indicator, applying comparative analysis to measure specific growth rates, together with statistical analysis, instead of applying current methods, judged by the appearance of colors from the culture medium was suggested. Considering the exponential growth phase of the test species, the test duration was suggested to be reduced to less than four days. For the test with fish as an indicator, resetting the appropriate body size was suggested to, show consistent sensitivity against chenicals applied during testing. Furthermore, it is necessary to consider the criteria needs, which should be established in reasonable and objective ways. For the testing species, marine rotifer, Brachionus plicatilis could be a replacement for Artemia sp., and a bivalve for fish in the test. To improve the performance effectiveness of the "Eco-toxicity test", it is worth considering the designation of accredited testing institutes, by placing it in the same loop. Thus it is also expected to have a reliable management system in place, having the capacity of cost calculation properly and adjusting testing items if required.

Marine ecotoxicological standard method was applicated using marine rotifer and benthic copepod as primary consumer of marine ecosystem. Marine rotifer, Brachionus plicatilis and benthic copepod, Tigriopus japonicus were designated as standard test species with the endpoints of 24hr neonate mortality (24hr $LC_{50}$) and 48hr population growth (48hr $EC_{50}$) for rotifer, and 48hr nauplius mortality (48hr $LC_{50}$) for benthic copepod. Tests method was referred to those of ASTM (American Society for Testing and Materials) with the replacement of test species which are widely distributed in Korean waters. The two species showed a wide tolerance on salinity ($5{\sim}35\;psu$) and can be easily cultured in small space even they were not as sensitive as in the mortality test using the nauplius of marine invertebrates. However, these species revealed the significant "concentrationresponse relationship" tested with ocean disposal wastes, and reproducibility using cadmium as standard reference material between laboratories. Accordingly, these two species have good potential as test species for marine ecotoxicological test species. Also, we strongly recommend that at least one of these species be included in the test species of "battery test" in marine bioassay.

Marine ecotoxicological standard method using fish larvae was established with the standard test species of Oryzias latipes(Japanese Medaka) and Paralichthys olivaceus(flounder) and with the 7 day $LC_{50}$ as endpoint. Test method referred to the USEPA(1994) with the replacement of test species found in the Korean water. Standard test species were selected in terms of the species supply and ecological importance in Korean waters. Japanese medaka can be reared with small tanks in the lab and has wide tolerance on salinity, and flounder eggs can be easily obtained from commercial fish hatcheries. General conditions for larval fish toxicity test are as follows. The possible salinity ranges for toxicity test were $0{\sim}35\;psu$ for medaka and >20 psu for flounder. Test type was designated as static non-renewal test if the dissolved oxygen in the test chamber does not fall below 4.0 mg/L. Ages of test species were selected as 7 days after hatched for medaka(about 5 mm TL) and 25 days for flounder(about 10 mm TL) because of the low natural mortality after these periods. Test can be accepted when the survival rates are over 80% in control. Also, species sensitivity on standard reference materials(copper, cadmium or zinc) must be provided with the toxicity test results.
The toxic responses of flagellate Euglena agilis Carter to 8 heavy metals (Ag, Cd, Cr (VI), Cu, Hg, Ni, Pb, Zn) were measured using E. agilis system (E-Tox), an automated biomonitoring system. The E-Tox measures cell movement parameters, such as velocity, motility, and forms of the cells, as biological endpoints. $EC_{50}$ values from the E. agilis biomonitoring test were compared with the literature data from the tests with Daphnia magna, Vibrio fischeri and Euglena gracilis. Measurement of the E. agilis movement behavior and D. magna acute toxicity test were also conducted for the wastewater samples. E. agilis is less sensitive than D. magna but is comparable to or more sensitive than V. fischeri and E. gracilis for the heavy metals tested in this study. E. agilis shows prompt changes of these parameters for the toxic metal plating wastewater. Major advantages of the E-tox are automatic, easy to handle and fast ecotoxicity monitoring system compared to other biological test systems. These results imply that E. agilis biomonitoring test using E-Tox can be a putative ecotoxicity test as a good early warning tool for the monitoring of toxic wastewater.

Water flea is a widely used test species in the aquatic ecotoxicity test. In Korea, D. magna is currently used as a standard test species, however that do not inhabit in the Korean aquatic ecosystem. In this study, Daphnia galeata, which is a dominant species in the Korean aquatic ecosystem, was collected from domestic lake and investigated to suggest the D. galeata for ecotoxicity assessment in Korea. We investigated the characteristics, life span, and toxicity sensitivity compared with D. magna. The life span test of D. galeata was performed in this study, and then the results were compared to the sensitivity with D. magna to confirm the applicability for ecotoxicity assessment. The 48h-L(E)C50 values for seven heavy metals (As, Cd, Cr, Cu, Hg, Ni, and Zn) of D. galeata and D. magna were collected and analyzed. As a results, shorter lifetime, less reproduction, smaller body size of D. galeata were observed compared with D. magna. D. galeata was similar or more sensitive than D. magna for seven heavy metals. Therefore, we propose that D. galeata is a suitable test species for ecotoxicity testing in Korea.
