The Risk Assessment of Butachlor for the Freshwater Aquatic Organisms (Butachlor의 수서생물에 대한 위해성 평가)

  • To assess the effect of butachlor on freshwater aquatic organisms, acute toxicity studies for algae, invertebrate and fishes were conducted. The algae grow inhibition studies were carried out to determine the growth inhibition effects of butachlor (Tech. 93.4%) in Pseudokirchneriella subcapitata (formerly knows as Selenastrum capriconutum), Desmodesmus subspicatus (formerly known as Scendusmus subspicatus), and Chlorella vulgaris during the exposure period of 72 hours. The toxicological responses of P. subcapitata, D. subspicatus, and C. vulgaris to butachlor, expressed in individual $ErC_{50}$ values were 0.002, 0.019, and $10.4mgL^{-1}$, respectively and NOEC values were 0.0008, 0.0016, and $5.34mg\;L^{-1}$, respectively. P. subcapitata was more sensitive than any other algae species. Butachlor has very high toxicity to the algae, such as P. subcapitata and D. subspicatu. In the acute immobilisation test for Daphnia magna, the 24 and $48h-EC_{50}$ values were 2.55 and $1.50mg\;L^{-1}$, respectively. As the results of the acute toxicity test on Cyprinus carpio, Oryzias latipes and Misgurnus anguillicaudatus, the $96h-LC_{50}s$ were 0.62, 0.41 and $0.24mg\;L^{-1}$, respectively. The following ecological risk assessment of butachlor was performed on the basis of the toxicological data of algae, invertebrate and fish and exposure concentrations in rice paddy, drain and river. When a butachlor formulation is applied in rice paddy field according to label recommendation, the measured concentration of butachlor in paddy water was $0.41mg\;L^{-1}$ and the predicted environmental concentration (PEC) of butachlor in drain water was $0.03 mg\;L^{-1}$. Residues of butachlor detected in major rivers between 1997 and 1998 were ranged from $0.0004mg\;L^{-1}$ to $0.0029mg\;L^{-1}$. Toxicity exposure ratios (TERs) of algae in rice paddy, drain and river were 0.004, 0.05 and 0.36, respectively and indicated that butachlor has a risk to algae in rice paddy, drain and river. On the other hand, TERs of invertebrate in rice paddy, drain and river were 3.6, 50 and 357, respectively, well above 2, indicating no risk to invertebrate. TERs of fish in rice paddy, drain and river were 0.58, 8 and 57, respectively. The TERs for fish indicated that butachlor poses a risk to fish in rice paddy but has no risk to fish in agricultural drain and river. In conclusion, butachlor has a minimal risk to algae in agricultural drain and river exposed from rice drainage but has no risk to invertebrate and fish.

Butachlor and Silicate Fertilizer Interaction to Rice (Butachlor와 규산질비료(珪酸質肥料)의 수도(水稻)에 대(對)한 상호작용(相互作用))

  • Pot and laboratory tests were undertaken to investigate the influence of silicate fertilization on butachlor phytotoxicity to rice. Growth of rice seedlings at 150 ppm of $SiO_2$ was stimulated, while adverse effect was observed over 300 ppm of $SiO_2$ and growth reduction was enhanced with combination of butachlor and $SiO_2$ Rice growth in pot trial at 150kg/10a of silicate fertilization was not influenced by recommended amounts of butachlor and nitrofen, however, the growth of Seokwang byeo at 300kg/10a of silicate was markedly retarded by butachlor in the initial stage of growth. Growth reduction of Seokwang byeo caused by combined application of silicate and butachlor was recovered 50 days after herbicide application. Growth reduction from butachlor was not influenced by pH level and also degradation behaviors of butachlor in submerged soil was not altered by silicate fertilization. Adsorbed amount of butachlor on rice root was increased with addition of $SiO_2$ and its amount in Seokwang byeo was higher than that of Jinju byeo. Butachlor absorption by Seokwang byeo was accelerated by 150 ppm of $SiO_2$ applied simultaneously, but those effect was not encountered in Jinju byeo. Butachlor absorption of rice seedlings was also increased by 150 ppm of $K_2O$, while CaO hindered the absorption and $Na_2O$ had no effect on the absorption. Residual level of butachlor in Seokwang byeo treated with combined solution of butachlor and $SiO_2$ was continued higher than that with butachlor alone during 10 days after transplantation to culture solution.

Bioactivities and Isolation of Functional Compounds from Decay-Resistant Hardwood Species (고내후성 활엽수종의 추출성분을 이용한 신기능성 물질의 분리 및 생리활성)

  • Wood of Robinia pseudoacacia and bark of Populus alba$\times$P. glandulosa, Fraxinus rhynchophylla and Ulmus davidiana var. japonica were collected and extracted with acetone-water(7:3, v/v) in glass jar to examine whether its bioactive compounds exist. The concentrated extracts were fractionated with hexane, chloroform, ethylacetate and water, and then freeze-dried for column chromatography and bioactive tests. The isolated compounds were sakuranetin-5-O-$\beta$-D-glucopyranoside from Populus alba $\times$Pl glandulosa, 4--ethyoxy-(+)-leucorobinetinidin frm R. pseudoacacia and fraxetion from F. rhynchophylla and were characterized by $^1H$ and$^{13}C $ NMR and positive FAB-MS. Decay-resistant activity was expressed by weight loss ratio and hyphae growth inhibition in the wood dust agar medium inoculated wood rot fungi. R. pseudoacacia showed best anti-decaying property in both test and its methanol untreated samples, indicating higher activity than methanol treated samples in hyphae grwoth test. In antioxidative test, $\alpha$-tocopherol, one of natural antioxidants, and BHT, one of synthetic antioxidants, were used as references to cmpare with the antioxidant activities of the extacted fractions. Ethylacetate fraction of F. rhynchophylla bark indicated the hightest activity in this test and all fractions of R. pseudiacacia extractives also indicated higher activities compared with the other fractions. In the isolated compounds, aesculetin isolated from F. rhynchophylla bark showed best activity and followed by robonetinidin from R. pseudoacaica.

In vitro Effect of Ecklonia stolonifera Okamura Extract on the Cell Growth in CCD-986sk Human Fibroblast and Melanin Formation Inhibition in Clone M-3 Mouse Melanocyte Cell Line (청정해역 곰피추출물의 세포생리활성 연구)

  • In order to investigate whether or not CCD-986sk cell line can be affected by Korean Ecklonia stolonifera Okamura, we examined the MTT assay when we treated Korean Ecklonia stolonifera Okamura extract in CCD-986sk human fibroblast cell line. The sample were tested for cell proliferation activity by means of a modification of the MTT assay. Ecklonia stolonifera Okamura extract showed significantly strong cell proliferation activity at the range of from 6.25 mg $mL^{-1}$ to 1.56 mg $mL^{-1}$ compared with control group. And in order to search for inhibition agents of skin melanin formation, we tested for inhibition effect of melanin pigmentation of Korean Ecklonia stolonifera Okamura using Clone M-3 mouse melanocyte cell lines. when we treated the extracts of Ecklonia stolonifera Okamura to the mouse melanocyte cell lines, the sample showed a significantly little formation of melanin pigments compared with control group at the only range of 200 mg $mL^{-1}$. These results suggest that extract of Korean Ecklonia stolonifera Okamura may represents an excellent candidate for inhibition of melanin pigmentation and for protection of human skin aging at in vitro level.

Effect of Sodium Hypochlorite(NaOCl) Treatment On Bacterial Yellow Blotch in Oyster Mushroom, Pleurotus ostreatus (Sodium Hypochlorite(NaOCl) 처리가 느타리버섯의 세균성 갈반병에 미치는 효과)

  • Studies were conducted to determine the potential of sodium hypochlorite(SHC) on the control of bacterial yellow blotch in cultivated oyster mushroom, Pleurotus ostreatus. SHC at the concentration of 80 ppm was effective on the control of Pseudomonas agarici causing yellow blotch in oyster mushroom except number 916 isolate. In vitro the mycelial growth was slightly inhibited at the concentration higher than 100 ppm of sodium hypochlorite, but retardation of the mycelial growth was soon recovered. Spray of SHC solution at the concentration of 40-50 ppm per day significantly reduced the incidence of the yellow blotch without impairing the growth of oyster mushroom in field culture. However, the higher concentration of SHC(67 ppm) induced yellow brown or dark gray in color and deformed cap and elongated stripe in morphology of fruiting body. Results indicate that periodical spray of sodium hypochlorite seems to be the recommendable method for protection against bacterial yellow blotch disease in oyster mushroom without reducing food quality.

Antibacterial Action against Food-Borne Pathogens by the Volatile Flavor of Essential Oil from Chrysanthemum morifolium Flower (국화 꽃 휘발성 향기성분의 식중독균에 대한 항균 작용)

  • The aim of this study was to investigate antibacterial activities of essential oil from C. morifolium against four Grampositive bacteria and six Gram-negative bacteria. The antibacterial activity of the oils was determined by agar-well diffusion assay, minimum inhibitory concentration(MIC), and minimum bactericidal concentration(MBC). Essential oil of C. morifolium had a large inhibition zones especially against Salmonella enterica(21 mm) and Bacillus cereus(19 mm). Essential oil of C. morifolium generally showed higher antibacterial activity against Gram-positive bacteria than Gram-negative bacteria. MIC of essential oil from C. morifolium was 5 ${\mu}g/m{\ell}$ against ten food-borne pathogens. MBC values were determined to be from 5 to 20 ${\mu}g/m{\ell}$ against eight bacteria except Salmonella choleraesuis and Listeria monocytogenes. Therefore, the essential oil of C. morifolium and its components have a potent antibacterial activity against food-borne pathogens, and is expected to be used as a novel food preservative.

Physiological Characteristics and ACE Inhibitory Activity of Lactobacillus zeae RMK354 Isolated from Raw Milk (원유에서 분리한 Lactobacillus zeae RMK354의 생리적 특성 및 ACE 억제능)

  • In order to develop a new starter for fermented milk, 1037 bacterial strains were isolated from raw milk. The strain that showed excellent acid producing and angiotensin converting enzyme (ACE) inhibitory activity (88.6%) was selected and identified as a Lactobacillus zeae based on the result of API carbohydrate fermentation pattern and 16S rDNA sequence. Lactobacillus zeae RMK354 was investigated further to study its physiological characteristics. It showed strong ACE inhibitory activity compared with commercial LAB starters tested. The optimum growth temperature of L. zeae RMK354 was $40^{\circ}C$ and it took 10 hr to reach pH 4.3 under this condition. L. zeae RMK354 showed more sensitive to penicillin-G, bacitracin, novobiocin, in a comparison of 14 different antibiotics, and showed most resistance to polymyxin B and vancomycin. It showed higher esterase and leucine arylamidase activities compared with 16 other enzymes. It was comparatively tolerant to bile juice and able to survive at pH 2 for 3 hr. It showed inhibitory activity against Salmonella Typhimurium with the rate of 60%. Based on these and previous results, L. zeae RMK354 could be an excellent starter culture for fermented milk with high level of ACE inhibitory activity.

Growth, Yield and Photosynthesis of Introduced Kenaf Cultivars in Korea (신도입 케나프 품종의 파종시기에 빠른 생육 및 수량 변동과 광합성 특성)

  • Kenaf (Hibiscus cannabinus L.), an annual plant of the family Malvaceae, is considered to be the most promising for alternative plants with potential use as a non-wood fiber source. The objectives of this study were to select the optimum seeding date and adaptable cultivar among newly introduced kenaf cultivars (Everglades-41, Tainung-2 and Chingpi-3) in Jeju island, and to clarify the photosynthetic characteristics of those cultivars. Among the three cultivars, the fresh shoot weight per unit area of Chingpi-3 at harvest season showed highest through all seeding dates, that of while Everglades-41 was the lowest. The difference of shoot yield at harvest mainly due to seedling stand rate and plant number per unit area among the cultivars. The Chingpi-3 showed the highest shoot fresh weight in the seeding date of May 11 and decreased trend in the late seeding dates. Net photosynthesis rate was higher on Everglades-41 with entire type leaf than other two cultivars with palmate type leaf. The activity of ribulose-1,5-bisphosphate carboxylase/foxygenase (rubisco) estimated from the A-Ci curve showed highest in Chingpi-3 among three cultivars. These results suggest that Chingpi-3 might be adaptable cultivar with seeding date of around May 10 for kenaf production in Jeiu island.

The Isolation and Characterization of the Antagonistic Microorganisms, Serratia marcescens-YJK1, for Major Pathogens on Paprika (파프리카에 발생하는 주요 병원균에 대한 길항미생물, Serratia marcescens-YJK1, 분리와 특성)

  • Synthetic agro-chemicals have been widely used to control diseases on paprika but these days negative attention has been increasing to use of them because of several adverse effects. This research was conducted to isolate and to characterize the antagonistic microorganism to control major paprika diseases, gray mold rot, fruit and stem rot, phytophthora blight, sclerotium rot, and wilt disease. Analysis of the fatty acid and analysis of the 16S rDNA gene sequence revealed that YKJ1 isolated in this research belongs to a group of Serratia marcescens. Specially, 16S rDNA gene sequence of YKJ1 showed 99% of sequence similarity with S. marcescens. Observation through the optical microscope revealed that YKJ1 suppressed the spore germination and the hyphal growth of pathogens. YKJ1 treatment on pathogens induced marked morphological changes like hyphal swelling and degradation of cell wall. In the case of phytophthora blight, the zoosporangium formation was restrained. S. marcescens found in this study call as S. marcescens-YKJ1 and it may be valuable as one of biological control agents against major diseases of paprika in the future even though it is require to be tested with more study on field test.

Studies on the Synthesis of Mannich Bases of Hexachlorophene and their Antimicrobial Activities (Hexachlorophene의 Mannich Bases 합성 및 항미생물작용에 관한 연구)

  • Thirty-three Mannich bases of 2,2'-methylene bis (3,4, 6-trichlorophenoxy-acetic acid) were synthesized hexachlorophene as potential antimicrobial agents and tested against Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Trichophyton rubrum, Microsporum gypseum, Epidermophyton floccosum, Aspergillus niger and Aspergillus oryzae in vitro.
