Influence of Post-planting Fertilizer Concentrations Supplied through Sub-irrigation in Winter Season Cultivation of Tomato on the Seedling Growth and Changes in the Chemical Properties of Root Media (저면관비 방법으로 동절기 토마토 육묘시 추비 농도가 묘 생장과 상토의 화학성 변화에 미치는 영향)

  • This research was conducted to investigate the influence of post-planting fertilizer concentrations on the growth of seedlings and changes of nutrient concentrations of media in tomato seedling production through sub-irrigation. Two root media such as peat moss (grade of 0 to 6 mm, PM06) plus perlite (grade of 1 to 2 mm (PE2)(7:3, v/v) and peat moss (grade of 5 to 15 mm, PM515) plus PE2 (7:3, v/v) were formulated and filled into 72-cell plug trays. After seeds of 'Dotaerang Dia' tomato were sown and germinated at $28^{\circ}C$, the trays were moved to greenhouse and seedlings were raised 35 days. When the cotyledons were emerged, post-planting fertilizers of 13-2-13, 15-0-15 and 20-9-20 ($N-P_2O_5-K_2O$) were applied in a sequence. The fertilizer concentrations based on N in each plug stage were differed with $25mg{\cdot}L^{-1}$ in three treatments. The fertilizer solutions were supplied when the weight of plug trays decreased to 40 to 50% compared to container capacity. The root media were collected in 1, 2, 4, and 5 weeks after sowing and were divided into top, middle, and bottom parts, then were analysed for pH, EC and macro-nutrient concentrations. The seedling growth was investigated 5 weeks after sowing. The pH and EC in PM06+PE2 was higher than those of PM515+PE2. The bottom and mid-part had higher pH and lower EC compared to upper part in each medium. The differences of EC between upper and bottom parts were around 2 times in each medium. The $NH_4-N$ and K concentrations in program 3 of PM06+PE2 showed the highest concentrations among all treatments. The $NO_3-N$ concentrations in PM06+PE2 increased gradually and this rising tendency become severe as post-planting fertilizer concentrations were elevated. The seedling growth in terms of fresh and dry weights was the highest in the treatment of program 2 in PM06+PE2 among all treatments tested. Comparing Photosynthesis, Growth, and Yield of Paprika (Capsicum annuum L. 'Cupra') under Supplemental Sulfur Plasma and High-Pressure Sodium Lamps in Growth Chambers and Greenhouses (황 플라즈마 및 고압나트륨 램프의 보광에 따른 생육상 및 온실에서의 파프리카 광합성 및 생산성 비교)

  • Supplemental lighting with artificial light sources is a practical method that enables normal growth and enhances the yield and quality of fruit vegetable in greenhouses. The objective of this study was to investigate the effect of sulfur plasma lamp (SP) and high-pressure sodium lamp (HPS) as supplemental lighting sources on the growth and yield of paprika. For investigating the effectiveness of SP and HPS lamps on paprika, the effects of primary lighting on plant growth were compared in growth chambers and those of supplemental lighting were also compared in greenhouses. In the growth chamber, plant height, leaf area, stem diameter, number of leaves, fresh weight, and dry weight were measured weekly at SP and HPS from 2 weeks after transplanting. In the greenhouse, no supplemental lighting (only sunlight) was considered as the control. The supplemental lights were turned on when outside radiation became below $100W{\cdot}m^{-2}$ from 07:00 to 21:00. From 3 weeks after supplemental lighting, the growth was measured weekly, while the number and weight of paprika fruits measured every two weeks. In the growth chamber, the growth of paprika at SP was better than at HPS due to the higher photosynthetic rate. In the greenhouse, the yield was higher under sunlight with either HPS or SP than sunlight only (control). No significant differences were observed in plant height, number of node, leaf length, and fresh and dry weights between SP and HPS. However, at harvest, the number of fruits rather than the weight of fruits were higher at SP due to the enhancement of fruiting numbers and photosynthesis. SP showed a light spectrum similar to sunlight, but higher PAR and photon flux sum of red and far-red wavelengths than HPS, which increased the photosynthesis and yield of paprika.

  • In batch bioreactor fermentations for cyclosporin A (CyA) production, good potential for bioprocess improvement was demonstrated in the immobilized cell system, providing appreciably better utilization of the catalytic activity of the biomass than the freely suspended cells, especially during the exponential phase. When concentrated nutrient medium was added pulsely during the exponential phase of cell growth(at hour 139 of fermentation), reactivation and regermination in both immobilized and suspended cell cultures were observed to contribute to the longevity of CyA production, maintaining maximum CyA titre until 250 hours of fermentation. Contrarily, simple batch fermentations without any supplement of medium in both systems showed repid decrease in CyA concentrations during the late stationary phase. Notably, the CyA yield coefficient $(Y_p/x)$ for the immobilized cell system was maintained quite high even after the pulse addition of the concentrated full medium, reaching almost 80% of the level attained during the exponential phase. This is in sharp contrast when compared with the corresponding value of 58% in the case of parallel-suspended cells. This pattern of CyA production resulted in considerably enhanced CyA production in the immobilized cell system, reaching almost 2 time higher maximum CyA production in comparison with the free cell system.

  • This study was carried out to determine leaf photosynthetic capacity and growth characteristics of Kalopanax picturs Nakai seedlings under the different light condition. The seedlings grown under low light condition showed larger leaf area and petiole, and higher relative growth rate than those grown under high light condition. Chlorophyll contents as chlorophyll a, chlorophyll b, and chlorophyll a + b were high in the seedlings grown under highlight condition compared to those grown in low light conditions. The mean absorption value of shade leaf within photosynthetically active radiation (400-700 nm) was slightly higher than that of sun leaf, Leaf photosynthetic capacity of seedling was variable under the different light conditions. Seedling grown under high light condition had the higher photosynthetic capacity. Leaf photosynthetic rates under forestry and nursery were 700 and $300\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, respectively. However, leaf photosynthetic rates under high and low light conditions were 500 and $300\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, respectively.

Biological Treatment of Processed-Leachate from Landfills by Reed (Phragmites australis)-Bed in a Continuous Flow System (갈대-상(床)을 이용한 쓰레기 매립지 침출수의 생물학적 연속흐름 처리)

  • We investigate the biological treatment of processed-leachate from SUDOKWON landfill site in Korea by the reed (Phragmites australis)-bed with a continuous flow system. The reed individuals showing superior growth in processed-leachate experiment were selected among the reeds of thirteen natural habitats by means of the comparisons between the removal rates of nutrient salts, eco-physiological responses and growth. The reed-beds (combination of the reed individuals showing superior growth with helper microorganisms) were continuously supplied with processed-leachate. We monitored the effluents that passing through the reed-beds during the experiment period. After five weeks, analysis results of effluent from each reed-bed were as follows; chromaticity, total nitrogen, total phosphorus, biological oxygen demand, chemical oxygen demand, total dissolved solid and salinity decreased $29.5{\sim}36.9\%,\;49.4{\sim}67.2\%,\;42.1{\sim}94.6\%,\;74.5{\sim}88.8\%,\;15.6{\sim}20.8\%,\;17.5{\sim}35.4\%\;and\;15.3{\sim}34.7\%$, respectively. These results represented the substantial improvement of water-quality after passing through reed-bed in a continuous flow system.

Effect of Bedsoil on Cutting Propagation of Old Growth and Protected Tree of Salix chaenomeloides Kimura (노거수 및 보호수 왕버들 삽목증식에 미치는 상토의 영향)

  • Old growth and protected-trees are worth history and culturally in Korea. This research carried out to investigate habitat characterization and the vegetative propagation methods of Salix chaenomeloides(100-600 years old trees) in Gyeongsangnamdo Province. It has been preserved in good and grow naturally in area surrounding village(4 trees), riverside(1 tree) and field(1 tree). Cutting from old growth and protected trees was conducted using 10 kinds of bedsoils. It was survived above 93% in all bedsoils. Growth of cuttings is a distinct difference according to bedsoils and tree ages. Growth of cutting was high on CBS, followed mixed bedsoils(VPMP) including vermiculite, peatmoss and perlite. Root collar diameter was difference depend on tree ages and bedsoils. All trees are produced 1 to 4 branch. Root growth of cutting was 20 cm on CBS and VPMP, which is the best shoot growing on cutting. Bedsoil with physico-chemical characteristics of high moisturizing ability, high air permeability, high EC value and slightly acid may be suitable for efficiently cutting propagation of old growth and protected S. chaenomeloides. CBS and VPMP bedsoil are favorable for propagating this species by cuttings.

Cultural Conditions for the Mycelial Growth of Phellinus spp. (진흙버섯속의 배양적 특성)

  • This study was carried out to determine the optimum culture conditions for Phellinus spp. known as white rot fungi showing anti-cancer activity. The optimum solid medium for mycelial growth at $25^{\circ}C$ was potato dextrose agar medium and optimum pH range was $6.0{\sim}8.0$, while all species showed reduced or no growth at pH 4.0. Most species showed good growth at $25{\sim}30^{\circ}C$. Out of 10 species of Phellinus examined, P. biscuspidatus was the best growing fungus in the range of pH $6.0{\sim}7.0$ based on mycelial density. Three species such as P. biscuspidatus, P. johnsonianus and P. lloydii could be grouped in mesophile fungi, showing $30{\sim}35^{\circ}C$ optimum temperature.

  • Basic studies on the cultural characteristics of Pycnoporus coccineus and P. cinnabarinus were performed. They exhibited $30{\sim}40{\circ}C$ optimal temperature ranges and optimal pH ranges of $5{\sim}6$. Among 6 media, they were good at mycelial growths on PDA, LBA and YMA.P. coccineus grew more than P.cinnabarinus on the same medium. Among 10 sawdust media, they were good at mycelial growth on three oak trees and Alnus hirsuta. However, the sawdust of Castanea crenata was bad at mycelial growth. Among 3 coniferous trees, Larix leptolepis showed better growth than the other trees such as Pinus densiflora and P. koraiensis. The fruit body production P. coccineus was about twice better than P. cinnabarinus on Quercus spp. sawdust cultivation.

  • 우리나라에서의 수도작은 후기등숙이 불량해지기 쉬우므로 못자리기간을 앞당기므로서 저온기의 보온육묘가 필연적으로 수행되어야 하는 특성을 지니고 있다. 보온육묘는 자연상태에서 육묘가 불가능한 시기에 인공가온으로 묘의 생장을 시키게 되므로 보온상내 환경이 외기환경과 차이가 크므로 세심한 관리를 해야한다.
