Evolutionary Genetic Studv on the Eight Species of the Drosophila melonogaster Group from Korea: Reproductive Isolation and Protein Analysis (한국산 노랑초파리 군(Drosophila melanogaster group) 8종의 진화유전학적 연구 :생식적 격리 및 단백질 분석)

  • 한국산 노랑초파리 군에 속하는 8종의 유전적 유연관계를 밝히고자 생식적 격리 그리고 수용성 단백질을 전기영동법으로 분석하였다. 생리적 격리 실험에서 교배전격리 실험결과를 Watanane와 Kawanishi model에 근거하여 보면 D. aurario complex 3종 중 D. triauror물가 원시종이며 남 auraria는 팍생종으로 나타났다. 교배후 격리 실험에서 나. melonogaster와 D. simuions의 교배시, D. melonogoster를 수컷으로 하였을 때는 불임의 수컷만이, 또 D. melonogaster를 암컷으로 곯였을 때는 불임의 암컷만이 출현하였다. 그리고 남 ouraria complex 3종간의 교배에서는 수정 능력이 있는 수컷과 암컷이 출현하였는데, 이는 아직 남 auraria Complex가 semispecies단계에 있음을 나타내는 것이라 할 수 있다. SDS-PAGE로 분석한 노랑초파리 군 8종의 band pattern을 densitometer로 scanning한 결과 남 susu상가 가장 특이하였으며, TDE에 의한 유전적 거리(Aquadro and Avise's)는 남 auror지와 봉 triauror지사이가 0.155로 가장 낮았고, D. melonogaster와 고. mfa 사이가 0.422로 가장 높았다. 본 연구의 결과를 UPGMA법 뜨로 분석하면, 한국산 노랑초파리 군 8종은 4개의 아군으로 나뉘어지며 이들은 2개의 다른 큰무리로 구분되었는데, D. suzu가기 아군, D. lutescens의 아군, D. melanogoster와 봉 simulons의 아군이 속한 큰무리와, D. mfa,0. ouroria, D. biauroria 그리고 남 triourario가 속한 다른 큰무리로 나눌 수 있다.

Analysis of Reproduction and CO I Gene Sequence between Two Earthworm Populations Used in Vermicomposting Organic Wastes in Different Localities (서로 다른 지역에서 유기성 폐기물 처리에 이용되는 두 지렁이 집단의 생식 및 CO I 유전자 분석)

  • Two domestic earthworm populations used in vermicomposting in different lacalities were collected. The one was from Hongcheon (Kangwon province) and the other was from Youngdong (Chungcheong province). Reproductive capacities and the degree of reproductive isolation of two population were investigated. CO I gene sequences were also compared. There was no difference in their reproductive capacities. And there was no reproductive isolation between two populations. Two populations were identified as Eisenia andrei or Eisenia fetida by CO I gene biomarkers. Phylogenetic tree formulated by CO I gene sequence strongly suggested that two populations were just the same species.

  • A syrnpatnc sample of Rana nigrnmaculata sand R plancyi (Anura) from Kwangu, Korea was investigated to clarify their taxonornic status and to estimate the degree of genic variation, genetic relatedness, and reproductive isolation between them, based on electrophoretic analysis, marphometric data, mating call, mating season, and habitat. All the characters measured including morphometric, genetic, behavioral, and ecological suggest that they are distinct species and R nigrnmaculata might have originated from R plancyi (tropical form about three and a half million years ago. An adult female was found to be Fl hybrid between two species The hybrid female possessed fettile eggs and morphologically intermediate to the parental species in head parts, hind limb, and web characters. The differences in mating call, mating behavior, breeding season, and habitat strongly suggest that postmating isolating mechanism is operating, even though very rare hybrid is found.

  • The geographic distribution and variation for Rhinogobius brunneus were surveyed by means of allozymic and morphological analyses and it was revealed that the Korean populations of R. brunneus comprise three distinct types, Type-A, Type-B, and Type-C, which show considerable differentiation to a degree of interspecific level(Rogers' S(1972): $S_{A-B}$=0.631, $S_{A-C}$=0.628, $S_{B-C}$=0.661). In addition, no evidence of gene flow among the types was found at sympatric area and it is assumed that reproductive isolation is completed. Moreover there is microhabitat segregation according to the distance from river mouth among each types and which segregation was regarded as a factor to facilitate reproductive isolating mechanism. Therefore, based on the evidence presented above, these three types of R. brunneus are considered as typical discrete species.

Interspecific Hybridization between Matsumuraeses phaseoli and M. falcana (Lepidoptera: Tortricidae) and Postzygotic Reproductive Isolation (팥나방(Matsumuraeses phaseoli)과 어리팥나방(M. falcana)의 종간 교잡과 접합후 생식격리)

  • Artificial interspecific matings between Matsumuraeses phaseoli and M. falcana (Lepidoptera: Tortricidae) were conducted to know the possibility of hybridization of the two sympatric species. Reciprocal crossings successfully produced F1 hybrids. Most of F2 crosses yielded progenies except all mating trials with females of F1 hybrid obtained from M. phaseoli female. Inbreedings of the F2 hybrids produced F3 progenies. In backcrossings between F1 hybrids and parent lines, all the two mating trials with females of F1 hybrid obtained from M. phaseoli females did not produce any progeny, while other 6 backcrosses produced the next generations. Inbreedings of the backcross lines also produced subsequent progenies. These results indicated that F1 females produced from hybridization between M. phaseoli females and M. falcana males were sterile. Conclusively, it suggested that a partial reproductive isolation at a postzygotic stage can occur between the two sympatric species.

Genetic Differentiation and Reproductive Isolation among Three Tvpes of the floating Gobv (Chuenogobius annufuris) in Korea (한국산 꾹저구(Chaenogobius annularis) 3형의 유전적 분화 및 생식적 격리)

  • Since the floating goby, Choenogobius onnuloris, has intricate and diverse morphological variations, allozymic analysis at 25 loci was carried out for their populations in Korea to clarify its taxonomic status. A genetic assay carried out revealed that the floating gobies were clearly divided into three genetic groups (Tvpe-A, Tvpe-B and Tvpe-C) in Korea. Alternative alleles were fated at six loci (Aco, 6pgd, Ldh-1, Got-1, Gpf, Gp3l. Some loci had considerable heterogeneitv among three types and no evidence of gene exchange in slunpatric populations was found from statistical analyses. The genetic similarity (Rogers'S) among three types was lower than 0.80 and divergent time estimate indicates that they were speciated during 1.2-1.8 million year before present (WBP). ASso, these three types of C. unnuluris were distinguished morpholosicallv from each other by several characters such as band ornamentations. In conclusion, the evidences presented here support recognition of three types of Chuenogobius annularis as typical discrete species.

한국산 검정망둑(Tridentiger obscurus)과 민물검정망둑(T.brevispinis)의 분류학적 고찰

  • Morphological and isozyme analyses were used to examine closely related species of Tridentiger obscurus and \ulcorner: brevispinis, a pair of cryptic species which has intricate problem with regard to species boundary, and to clarify the taxonomic position. In this study, it was revealed from their 17 allopatric streams that there are differences of band ornamentation on the lateral side and three genetic markers between two species. However, genetic relationship between them (S = 0.813, D = 0.192) was within the intraspecific level of similarity. However, at the sympatry of two species no hybrid was found except at Bangjuk stream where the electrophoretic hybrids with slight deficiency of heterozygosity were observed. It is shown that at sympatry there is a finer microenvironmental subdivision between two species at all populations including Bangjuk stream. To manifest the extent of assortative mating, four syrnpatric subpopulations at Bangjuk stream were analyzed for departure from Hardy-Weinberg equillibrium by F-statistics. All subpopulations were deviated significantly from the expectation under random mating. The evidences presented here supports the recognition of Tridentiger obscurus and T. brevispinis as semispecies in the late stage of reproductive isolation

  • Physiological and morphological differences in larvae and female adults of Thecodiplosis Japonensis from Haenam, Chullanamdo, and Chunsung, Kangweondo, were studied by means of electrophoretic technique and scanning electron microscope (SEM). On zymograms of whole body, third instar larvae of T. japonensis showed geographic differences in the band patterns of esterase and MDH iszymes, but patterns and the staining density of general proteins were similar in tow populations. In female adults, the populations revealed geographic differences in general proteins and esterase isozymens. In external ultrastructures, especially in genital segments, each population had distinctive structures in the 2nd segment of ovipositor.

  • The three self-compatible, terrestrial orchids Cephalanthera erecta, C. falcata, and C. longibracteata flower synchronously in sympatric populations. Cephalanthera erecta and C. longibracteata, which have white flowers that do not fully open, are predominantly autogamous, whereas the food-deceptive C. falcata, whose bright yellow flowers open completely, is predominantly outcrossing. The formation of hybrids rarely occurs between species owing to strong prepollination barriers (floral isolation). If these three species have evolved toward the prevention of interspecific hybridization, we can expect that the spatial distribution patterns of the three species would be characterized as spatial segregation (i.e., 'spatial repulsion') from each other. To test this prediction, we studied the three Cephalanthera species in sympatric populations showing coincident flowering within Yeonwhasan Provincial Park (YPP, Gyeongsangnam Province, South Korea). We found strong spatial aggregation in each population and spatial independence in the interspecific spatial distribution, differing from previous studies. We further hypothesize that Cephalanthera species in sympatry within YPP are distributed somewhat randomly in space, perhaps due to the sharing of similar mycorrhizal fungi.

  • Importation pathway of rice cultivar into Korea was re-established with considering ancient geo-ecological characteristics of the Sororibyeo excavated from Cheongwon. It is assumed that Sororibyeo settled down in Korea by the importation pathway along the southern seashore of China through old downstream of Geumgang by a southern Korean human race when China and Korea were not yet separated each other by the Yellow Sea. This importation pathway was designated as "Old Geumgang-Sorori Rice Road", in this study. It is further inferred that Korean Peninsula was geographically isolated by ocean after the Ice Age. In consequence, Gawajibyeo, an ancient rice with little genetic variation, was evolved from Sororibyeo, which is estimated to evolve into rice cultivar in Korean Peninsula.
