Comparison of Growth Characteristics of Tricholoma matsutake Mycelium Among the Types of Air Bubble Bioreactor (공기부양식 생물반응기의 형태별 송이균사의 생장특성 비교)

  • In order to select suitable bioreactor type inhibiting cell stress during submerged culture of Tricholoma matsutake mycelium, the growth characteristics and ergosterol contents were investigated using the external-loop type of air-lift bioreactor (ETAB), balloon type of air bubble bioreactor (BTBB) and column type of air bubble bioreactor (CTBB). Dry weights of the T. matsutake in the BTBB, ETAB and CTBB were 12 g, 11.4 g, and 9.5 g per 1 litter, respectively. BTBB, ETAB and CTBB reached stagnant phases 16, 20, and 24 days after cultivation, respectively, The BTBB was more suitable for liquid culture of T. matsutake mycelium compared to other bioreactors owing to much mycelia product and short culture period. The ergosterol contents produced by the mycelium in the bioreactors were in sequence of BTBB, CTBB, and ETAB at every growth phase. BTBB might affect the mycelium on producing the smallest size of pellets. BTBB and CTBB got the mycelium precipitated and coagulated under operation of bioreactor sparser, whereas ETAB shown no effect of above phenomenon. A renovated bioreactor combined between a balloon shape of BTBB and an external-loop of ETAB was developed to enhance the efficiency of culture technique.

The Analysis of Pre-Service Biology Teachers' Natural Selection Conceptions in Multiple-Choice and Open-Response Instruments (생물 예비 교사의 선택형과 개방형 문항에서 나타난 자연선택 설명 분석)

  • Teachers use explanations to communicate important scientific ideas to students. Consequently, all biology teachers should be evaluated to determine how effective they are at constructing and communicating biological explanations. Open response questions are required to detect pre-service biology teachers' abilities to communicate robust and accurate scientific explanations. Nevertheless, multiple-choice questions are typically preferred by educators because of the common drawbacks of using open-response instruments, such as scoring time, inter-rater scoring disagreements, and delayed feedback to test takers. This study aims to measure pre-service biology teachers' competence in building scientific explanations and to investigate how accurately multiple-choice questions predict the results of open-response questions. One hundred twenty four pre-service biology teachers participated in the study and were administered 20 multiple-choice items and three open-response items designed to measure the accuracy and quality of their explanations of evolutionary change. The results demonstrated that pre-service teachers displayed higher competence when tested with multiple choice items than when tested with open response items. Moreover, scores derived from multiple-choice items poorly predicted the scores derived from open-response items. Multiple-choice items were also found to be poor measures of the consistency, purity and abundance of conceptual elements in teachers' evolutionary explanations. Additionally, many teachers held mixed-models composed of both scientific and naive ideas, which were difficult to detect using multiple-choice formats. Overall, the study indicates that multiple-choice formats are poorly suited to measuring several aspects of biology teachers' knowledge of evolution, including their ability to generate scientific explanations. This study suggests that open-response items should be used in teacher education programs to assess pre-service teachers' explanatory competency prior to being permitted to teach science to children.

DISTRIBUTION OF MUTANS STREPTOCOCCI IN DENTAL PLAQUE OF CHILDREN (소아의 치면세균막에 존재하는 mutans streptococci의 분포)

  • The aim of this study is to survey the frequency of mutans streptococci species and biotypes isolated from dental plaque in Korean children and the relationship between species and biotypes of mutans streptococci and dft index. Dental plaques were collected from the anterior and molar teeth of upper and lower jaws in the subjects, aged below 12 years old. A dental examination was performed for dft (decayed, filled, total) with the WHO caries diagnostic criteria. The mutans streptococci from the sample were cultured selectively on mitis salivarius-bacitracine (MSB) agar plate. For biotyping of mutans streptococci, biochemical test was performed. From the culture, bacterial genomic DNA was prepared for using of PCR template for the identification of mutans streptococci at the species-level. Forty strains of mutans streptococci were isolated from dental plagues of 40 patients. The biotype I (45%) and biotype IV (32.5%) were most frequently detected. The prevalence of S. mutans and S. sobrinus was 69% and 31%, respectively. There was no positive relationship between species and biotypes of mutans streptococci and dft index. Our results revealed that biotype I and S. mutans were frequently detected in Korean children and support that dental caries incidents by many causative factors not only bacterial factor.

Production of Mycelia and Water Soluble Polysaccharides from Submerged Culture of Ganoderma applanatum Using Different Types of Bioreactor (생물반응기 유형에 따른 잔나비불로초(Ganoderma applanatum)의 균사체 및 수용성 다당체 생산 특성)

  • In order to select a suitable bioreactor type for the submerged cultivation of Ganoderma applanatum, both growth characteristics and polysaccharides production were compared among four different types of bioreactor. These include an external-loop type air-lift bioreactor (ETAB), a balloon type air bubble bioreactor (BTBB), a column type air bubble bioreactor (CTBB) and a stirrer type bioreactor (STB). The mycelial biomass produced from the reactors were in decreasing order: ETAB ($7\;g/{\ell}$) > BTBB ($6.2\;g/{\ell}$) > STB ($6\;g/{\ell}$) > CTBB ($5\;g/{\ell}$). Maximal soluble exopolysaccharides ($1\;g/{\ell}$) and endopolysaccharides (2.7%) were also obtained from ETAB. Thus, the ETAB was most suitable for submerged culture of G applanatum mycelium. Based on the results, ETAB was chosen for further detailed study. The most effective aeration rate for the mycelial growth in ETAB ranged from 0.05 to 0.1 vvm. For the maximal production, the mycelium at the initial growth stage needed low aeration rate to reduce cell damages by fluid flow. However, as the mycelia grew, the culture became viscous and thus needed higher aeration. The molecular weight of exopolysaccharides obtained from the culture grown in ETAB was higher than that from the culture grown in other bioreactors.

Time-course Analysis of Biofilm Formation in Quorum Sensing-deficient Bacteria (Quorum sensing 결핍 세균에서 생물막 형성의 시간적 추이 분석)

  • Pseudomonas aeruginosa and Vibrio vulnificus are Gram-negative human pathogens, which exert their virulence through quorum sensing (QS) regulation. The infection of these pathogens have been known to be mediated by biofilm formation in many cases and this study carried out the time-course analysis of biofilm formation depending on the QS regulation in P. aeruginosa and V. vulnificus. In P. aeruginosa, our results demonstrated that QS-deficient mutant better attached to surface at initial stage of biofilm formation, but poorly proceeded to the maturation of the biofilm structure, while wild type less attached at initial stage but developed highly structured biofilm at late stage. Because of this, the quantitative comparison of biofilm formation between wild type and the QS mutant showed the reversion; the QS mutant formed more biofilm until 10 h after inoculation than wild type, but wild type formed much more biofilm after 10 h than QS mutant. V. vulnificus has been reported to form more biofilm with the mutation on QS system. When we performed the same time-course analysis of the V. vulnificus biofilm formation, the reversion was not detected even with prolonged culture for 108 h and the QS mutant always forms more biofilm than wild type. These results indicate that the QS regulation negatively affects the attachment at early stage but positively facilitates the biofilm maturation at late stage in P. aeruginosa, while the QS regulation has a negative effect on the biofilm formation throughout the biofilm development in V. vulnificus. Based on our results, we suggest that the developmental stage of biofilm and bacterial species should be considered when the QS system is targeted for biofilm control.

Identification of mutans streptococci isolated from dental plaque between the bracket and tooth surface in orthodontic patients (교정환자의 브라켓과 치아 경계부에 존재하는 치면세균막내 mutans streptococci 종 및 생물형의 식별)

  • The aim of this study was to compare the species and biotypes of mutans streptococci isolated from dental plaques sampled from the interfaces between the bracket aid tooth surface and smooth tooth surfaces In orthodontic patients. Dental plaque was collected from the interfaces between brackets aid teeth (test group), and from smooth tooth surfaces distant from brackets by more that 2mm (control group). The dental plaque collected by a sterilized curette was transferred into a vial of 1 X PBS. The sample in the vial was vigorously vortexed for 1 min and plated ou mitis-salivarius bacitracin (MSB) agar plate using cotton tips. The agar plates were incubated at $37^[\circ}C$ in a candle jar for 2 days, and again incubated for 1 more day at anambient temperature Individual colonies were cultured in TH broth at $37^[\circ}C\;CO_2$ incubator. The PCR-RFLP based on dextranase gene was performed for the identification of mutans streptococci at the species-level For biotyping of mutans streptococci, biochemical tests were performed There was no significant difference of the species of mutans streptococci isolated from both test and control groups However, the biotypes of the mutans streptococci isolated from test and control groups were different. These results may offer the basic data to verify the relationship between the mutans streptococci biotype and enamel decalcification or dental caries in orthodontic patients with fixed appliances.

  • 생물정보원은 이질성이 높고 사용자의 요구사항이 다양하다. 본 논문은 이러한 이질성을 해결하고 사용자의 다양한 요구사항에 쉽게 대처할 수 있는 XML 기반의 생물정보원 통합시스템의 설계개념과 구조 및 구현결과를 제시한다. 제시하는 통합시스템은 관계형테이블, 객체, XML, 플랫파일 등 다양한 자료형을 지원하면서, 관계형, 객체관계형, 웹자원, 응용프로그램 등 데이터 관리모델에 무관한 뷰 정의 및 질의처리모델이다. 그리고 사용자정의 XML 뷰 기반의 뷰 관리 및 질의처리를 통하여 사용자의 다양한 요구사항에 쉽게 적응할 수 있는 미디에이터 질의처리 기반의 생물정보원 통합시스템을 제시한다.
