Antioxidant and Antiobesity Activity of Solvent Fractions from Red Garlic (홍마늘 용매 분획물의 항산화 및 항비만 활성)

  • Development of garlic processing products by thermal treatment and researched biological activity of a new product, red garlic. Red garlic MeOH extract was graduated by solvents, such as hexane, chloroform, ethyl acetate, butanol, and water, in order. Each solvent fraction was dried by a rotary evaporator and then resolved in water for analysis of its antioxidant and antiobesity activity. Browning compounds of red garlic fractions were the highest in the chloroform fraction. Total phenolic compounds and flavonoid content were highest in the hexane fraction. The chloroform fraction showed significantly higher activity in DPPH, ABTS radical scavenging activity, and antioxidant activity by FRAP. We propose that the antioxidant activity of the solvent fractions from red garlic was revealed interaction of browning compounds, total phenolic compounds, and flavonoids. Due to the higher activity of the shown fraction, the hexane and the chloroform fractions were have high contents of these compounds. Below $500{\mu}g/ml$, pancreatic lipase inhibition activity was significantly increased by sample concentration. And chloroform fraction, have the highest inhibition activity was shown over the 40%. In 3T3-L1 cells, the lipid accumulation inhibition activity was lower in the hexane and in the chloroform fraction than in the other fractions.

Production of Bioactive Components and Anti-Oxidative Activity of Soybean Grit Fermented with Bacillus subtilis HA according to Fermentation Time (Bacillus subtilis HA를 이용한 soybean grit의 고체발효 기간에 따른 생리활성물질 생산 및 항산화 효과)

  • Soybean grits, fortified with various bioactive components, were produced by solid-state fermentation using Bacillus subtilis HA. ${\alpha}$-Amylase activity gradually increased during fermentation over 5 days. Fibrinolytic and protease activities were highest in the soybean grits fermented for 7 days. The grits fermented for 5 days also showed the highest tyrosine content, indicating a higher peptide content. Peptides of low molecular weight (below 3,000 daltons) and browning pigments increased with increasing fermentation time. The fermented soybean grits showed higher contents of total phenolic compounds, to approximately 18 mg/g. DPPH free radical scavenging effects were higher in the soybean grits fermented for 3 days. Also, ABTS radical scavenging effects were greater in the fermented grits compared to the unfermented grits. Overall, the soybean grits fermented by solid-state fermentation for 5 days showed enhanced production of bioactive compounds and greater antioxidant properties.

Identification of Anti-Microbial Material Originated from Opuntia ficus-indica var. saboten Makino (손바닥 선인장 분말로부터 추출된 항균물질의 특성)

  • In order to discover physiologically active substance, we investigated a powder obtained by processing of Opuntia ficus-indica var. saboten Makino trunk. The powder was treated by sulfuric acid and then extracted by several solvents such as methanol, methylene chloride, ethanol etc. Among them, the best antimicrobial activity was showed by methylene chloride extract. To identify materials exhibiting physiological activation, the acid hydrolyzed extract was separated by 7 fractions through preparative silica gel TLC. The effective fraction exhibiting the best broad antimicrobial activity was identified, named as MBT-01108. From structural analysis of the products extracted to acid hydrolysis, a compound exhibiting the antimicrobial activities is identified to levulinic acid. Levulinic acid isolated from Opuntia ficus-indica var. saboten Makino may be applicable as a natural preservative of food or cosmetic and for prevention of bacterial diseases, an ingredient of acne, ageing and whitening cosmetics and an antimicrobial agent.

Antioxidant activity and comparative analysis of major functional compounds in liqueur using coffee and coffee-ground (커피와 커피박 침출주의 항산화 활성 및 주요 생리활성 물질의 비교 분석)

  • Sensory evaluation, in vitro antioxidant activities and main compounds of coffee water-extract, coffee liqueur (CL) and coffee-ground liqueur (CGL) were investigated to consider their industrialization. Sensory evaluation showed that all groups of CGL without 25% CGL (3 month) were relatively higher than CL groups. Total phenolic compounds and in vitro antioxidant activities such as 1,1-diphenyl-2picryl-hydrazyl (DPPH) radical scavenging activity and ferric reducing/antioxidant power (FRAP) were also performed. The group of 35% CGL had higher total phenolic compounds than others, and the result of DPPH radical scavenging activity was similar to that of total phenolic compounds. In addition, 35% CGL is comparable to the FRAP of coffee water extract (CE). Qualitative and quantitative analysis using high-performance liquid chromatography (HPLC) were performed, and chlorogenic acid as a ployphenolic compound and caffeine as a nonpolyphenolic compound were detected in all samples. Moreover, the HPLC analysis showed that CGLs contain a larger amounts of chlorogenic acid (difference of 0.3~10.5%) and also greater amounts of caffeine (difference of 10.0~18.2%) more then CE. Consequently, these results suggest that coffee-ground as coffee by-products could be used as commercially available food substances because of its physiological molecules remained.

  • 방선균은 생리활성물질의 탐색원으로서 많은 주목을 받고 있는 미생물이지만 현재까지 이용되어온 균주의 대부분은 육상의 토양에서 분리된 것들이고, 해양의 방선균에 관하여서는 아직까지 거의 연구되지 않은 분야이다. 미생물의 서식지로서 해양의 환경조건은 육상과 현저히 다르기 때문에 해양에 있어서 방선균의 microflora는 육상과는 상당히 다를 것으로 생각되어지며, 또한 연안해역에 존재하는 방선균 중에는 육상에서 유입된 담수 등으로 인하여 육상의 토양으로부터 유래된 방선균도 많은 것으로 알려지고 있다. (중략)

  • This study was carried out to investigate the anthocyanin pigments in the fruits of mulberry and grape. The anthocyanin pigments in the fruits of mulberry and grape were extracted with l% methanolic hydrochloric acid. The cyanidin-3-glu-coside (C3G) was separated and quantified by HPLC system using a Nova-Pack C$\sub$18/ column. The cyanidin-3-glucoside (C3G) contents of mulberry fruits were higher than that of grapes. Especially, anthocyanin pigments of mulberry fruits showed only C3G peak, but anthocyanin pigments of grapes showed some species peaks.

  • This study was conducted to purify and characterize further a potent cholesterol lowering material found in Sogi. The ethanol extract of Sogi was purified with acetone and ethanol, and a white powder was obtained. This material yielded t재 spots on a Silicagel thin layer chromatogram. The major material identified by M.P. measurement, elementary analysis and spectrography was considered to be gyrophoric acid. Beside this, minor components (orsellic and leccanolic acids ) were detected.
