A CLINICAL STUDY ON THE MAINTENANCE OF LIGHT INTENSITY OF VISIBLE-LIGHT CURING MACHINES FOR THE POLYMERIZATION OF COMPOSITE RESINS (복합레진 중합용 가시광선 광중합기의 적정 광강도 유지를 위한 임상적 고찰)

  • It is well known that numerous factors influence the light output of curing units, but many dentists are un aware that the output of their curing lights are inadequate. This study was conducted to evaluate the light in tensity of visible-light curing units in some private dental clinics and hospital dental clinics. In order to determine the maximum light intensity of the curing units, lamps, filters and fiber optic bundles, they were replaced with new ones and light intensity was remeasured. Light intensity was measured by employing a digital radiometer (EFOS model #8000, USA). Light intensity ranged in $29\sim866mW/cm^2$ (below $150mW/cm^2$ ; 17.8%, $150\sim300mW/cm^2$ : 46.6%, above $300mW/cm^2$ ; 35.6%). The replacement of the components increased the light intensity, with maximum increases of 94.8% for lamps, 82.3% for filters, 200.8% for fiber optics and 361.5% for all three parts. According to the manufacturer of radiometer, curing light is considered as unsuitable for use with a reading of above $300mW/cm^2$ by the radiometer. Applying these criteria to the present study, 64.4% of the curing units required repair or replacement. The results of this study indicated that the light intensities of the curing units used in dental practice were lower than optimum level.

Microleakage and Anticariogenic Effect of S-PRG Filler-containing Pit and Fissure Sealant (S-PRG filler를 함유한 치면열구전색제의 미세누출 및 항우식효과)

  • Pit and fissure sealant prevents biofilm accumulation, plays a role in forming a barrier to acidic substance made by the bacteria. The Surface reaction-type pre-reacted glass ionomerI(S-PRG) filler was developed in 1999. S-PRG filler releases fluoride continuously and does not decompose under wet conditions. The aim of this study was to test the microleakage and anticariogenic effect to adjacent enamel of S-PRG filler-containing pit and fissure sealant. Sound premolars and molars were used in this study. A S-PRG filler-containing pit and fissure sealant, Beautisealant$^{(R)}$(Shofu, Japan) was used for this experiment, the composite resin sealant Concise$^{(R)}$(3M ESPE, USA) was used as control. For the microleakage test, all teeth surface were double coated with finger nail varnish, with the exception of a 1.0 mm window around the restoration margins. The teeth were immersed in 2% methylene blue solution for 24 hours and then rinsed in tap water. For the anticariogenic effect evaluation, all tooth were immersed in artificial carious solution for 9 days and rinsed with tap water. Each tooth was embedded in orthodontic acrylic rein and subsequently sectioned longitudinally in a bucco-lingual direction with a low-speed diamond saw. The cut sections were examined using a stereomicroscope. Differences in microleakage between the two groups were not different significantly. But the S-PRG filler-containing pit and fissure sealant showed higher anticariogenic effect than that of flowable resin sealant.

Varietal characteristics of new Cordyceps militaris 'Dowonhongcho 2ho' improved by mating type molecular markers (교배형 분자마커를 이용한 신품종 밀리타리스 동충하초 '도원홍초 2호'의 품종 특성)

  • The mushroom species Cordyceps militaris has been studied and cultivated as a medicinal mushroom due to its multiple valuable biological and pharmaceutical activities. For breeding new strains of C. militaris, multiplex PCR assays were performed using primers specific for its mating type genes, MAT1-1 and MAT1-2. Mating types and mating status were confirmed, as evidenced by DNA bands at 233-bp and 191-bp for MAT1-1 and MAT1-2 respectively. The novel 'Dowonhongcho 2ho' was developed through mating; they were found to possess high-quality fruiting bodies when grown in artificial media. The stromata of the new strain were club-shaped, with a bright orange-red color, and measured 7.1 cm in length. They had an average cordycepin content of 0.33%. Compared to 'Dowonhongcho,' the new strain had a 7% higher yield, as well as firm fruiting bodies. The optimum temperature for mycelial growth was $20{\sim}25^{\circ}C$, and the optimum temperature for stroma development was $18{\sim}22^{\circ}C$. The fruiting bodies developed after 49.1 days from inoculation. The use of mating type molecular markers improved the breeding efficiency of the new strain 'Dowonhongcho 2ho.' Thus, they may be valuable for artificial cultivation and industrial-scale production of C. militaris with excellent characteristics.

Sclerotinia Rot on Basil Caused by Sclerotinia sclerotiorum in Korea (Sclerotinia sclerotiorum에 의한 바질 균핵병)

  • During growing season of 2011 to 2013, Sclerotinia rot symptoms consistently have been observed on basil in Yesan-gun, Chungcheongnam-do in Korea. The typical symptom formed initially brownish spot on leaf and stem, and then advancing margins, wilting the whole plant and blighting, eventually died. On the surface of diseased lesions was observed cottony, white, dense mat of mycelial growth, and sclerotia ($30-100{\mu}m$ diameter) formed on stem and leaf. Morphological and cultural characteristic on potato dextrose agar, color of colony was white and colorless chocolate, sclerotium of irregular shape of the oval was black and $5-50{\mu}m$ diameter in size. In pathogenicity test, necrosis and wilt of the inoculated stem were observed in all plants and the pathogen was reisolated from stems. On the basis of mycological characteristics, pathogenicity, and internal transcribed spacer rDNA sequence analysis, this fungus was identified as Sclerotinia sclerotiorum. This is the first report of Sclerotinia rot on basil caused by S. sclerotiorum in Korea.

Factors Influencing Somatic Embryo Induction and Plant Regeneration in Aralia elata Seem. (두릅(Aralia elata)의 체세포배 유도, 발아 및 식물체 재분화에 미치는 요인)

  • In order to find optimum conditions for somatic embryogenesis from different individual (2-year-old) in Aralia elata were cultured on MS medium supplemented with 1.0 mg/L 2,4-D, 3% sucrose, and 0.3% gelrite. We also investigated the effect of MS medium salt concentration, BA and ABA on the embryo germination and plant regeneration. While noticeable difference was observed on somatic embryo induction among different individual tree, no apparent difference was seen in both germination and regeneration frequencies. Compared with nonembryogenic calli, embryogenic calli tended to look yellow and/or pale brown in color, slowly growing and soft in their texture. Regardless of BA or ABA treatment, half-strength MS salt medium proved to be better than full strength MS medium in both embryo germination and plant regeneration. Both hypocotyl and cotyledon developments were slightly promoted by adding 0.1 mg/L BA. However, its effect on germination and regeneration seemed inferior to control. ABA treatment on somatic embryos at their torpedo and early cotyledonary stages resulted in poor response in germination and regeneration. Although most regenerated plantlets varied greatly in cotyledon number and shape, they could be developed into normal plants after 4 weeks in culture. More than 95% plantlets were acclimatized in an artificial soil mixture, successfully transplanted to nursery bed and grew normally without any phenotypic abnormalty.

Calretinin-Containing Neurons in the Deeper Layers of the Hamster Superior Colliculus (햄스터 상구의 deeper layers에서 calretinin이 함유 신경세포)

  • Calcium-binding protein calretinin is thought to play important roles in calcium buffering. Recently, we reported on the distribution, morphology of calretinin-immunoreactive (IR) neurons and the effects of eye enucleation on the immunoreactivity of calretinin in the superficial layers of the hamster superior colliculus (SC). In the present study, we describe the distributions and types of labeled cells and effects of enucleation in the deeper layers by immunocytochemistry. We also compare this labeling to that of GABA, the major inhibitory neurotransmitter in the central nervous system. In contrast to the superficial layers, the deeper layers contained many calretinin-IR neurons which formed two tiers. The first tier, which was very distinctive, was found within the intermediate gray layer. The second tier was found in the deep gray layer. Labeled neurons varied dramatically in morphology and included vertical fusiform, stellate, round/oval, and horizontal neurons. In contrast to the superficial layers, enucleation appeared to have no effect on the distribution of calretinin immunoreactivity in the deeper layers. Two-color immunofluorescence revealed that none of calretinin-IR neurons were labeled with an antibody to GABA. The present results demonstrate that calretinin identifies unique neuronal sublaminar organizations in the hamster SC. The present results also demonstrate that none of the calretinin-IR neurons in the hamster SC is GABAergic interneurons. As many calretinin-IR cells are GABAergic interneurons in most other brain areas, this phenomenon in hamster SC is exceptional.

A Study on the Preparation and Rheological Properties of Chik Mook (칡전분을 이용한 칡묵의 조리과학적 연구)

  • Arrowroot Mook was prepared as a trial to develop new food items from natural resources, and its rheological properties were measured. 1. The starting point of gelatinization of 7% arrowroot starch solution was $67.5^{\circ}C$ as measured by amylograph. The viscosity of arrowroot starch reached the maximum 375 B.U. at $95^{\circ}C$ and decreased to 315 B.U. while holding at $95^{\circ}C$ for 15 min. When the temperature dropped to $50^{\circ}C$, the viscosity increased again to 530 B.U. 2. The gelatinization enthalpy of arrowroot starch was 15.603 J/g. 3. For the color, there were significant differences in b value for the arrowroot Mook made of arrowroot starch and in L value for the Mook made by the addition of arrowroot juice. 4. In descriptive analysis, $S_3$(14% arrowstarch) was most favored in color, and $S_1$(10% arrowstarch)was most favored in transparency, brittleness, fineness, and shininess. There were no significant differences in hardness, adhesiveness, and astringency. 5. On the contrary to mechanical measurement, there were no significant differences in hardness of samples as the addition of arrowroot juice increased, although there appeared a tendency to be softer in descriptive analysis. 6. The Mook prepared with 12% arrowroot starch was most favored by sensory evaluation. In case of adding arrowroot juice, the Mook containing 3% of arrowroot juice was most favored. Overall, arrowroot Mook showed a potential as a new food item.

Occurrence and Pathogenicity of Verticillium Wilt on Chrysanthemum Caused by Verticillium dahliae (Verticillum dahliae에 의한 국화 반쪽시들음병 발생과 병원성)

  • A wilt disease on the greenhouse-grown chrysanthemum occurred at Kumi, Masan and Busan, Korea in $2003{\sim}2005$. Symptoms of wilting, yellowing, and marginal burning started from lower leaves and progressed upward. Early symptoms often appeared on one side, involving only one part of the plant or one side of individual leaves or stems without a vascular discoloration. Vascular tissues of the infected leaf-base's discolored to brown. Fungal isolates obtained from discolored tissues were identified as Verticillium dahliae based on its cultural and morphological characteristics. The fungus showed whitish to creamy colony pattern with abundant dark brown to black elongated microsclerotia on PDA. Conidiophores were verticillately branched and conidia were ellipsoidal to sub-cylindrical shape, and measured as $2.5{\sim}8.8{\times}2.0{\sim}3.8{\mu}m$. Pathogenicity tests by root dipping resulted in similar symptoms observed in the naturally infected chrysanthemum in fields.

  • Cordyceps militaris is being studied and cultivated as a medicinal mushroom having many valuable biological and pharmaceutical activities. In the breeding of new C. militaris mushroom, single ascospores were isolated and examined their mycelial growth, mycelial density, and production of stroma and perithecia. Among them selected isolates were crossed and hybrids were produced showing high quality fruiting bodies on artificial media. Mycelial growth rate of new strain 'Dowonhongcho' was higher than that of better on 'Yedang 3' on SDAY at $10-25^{\circ}C$. The stromata of new strain were club-shaped and bright orange-red. Its height was 6.1 cm and the cordycepin content was 0.34% on average. The new strain showed 9% higher yield than 'Yedang 3' with producing firmer fruit bodies. The optimum temperature for mycelial growth was $22{\sim}25^{\circ}C$ and the optimum temperature for stroma development was $18{\sim}22^{\circ}C$. Fruiting bodies were began to produce 45 days later after inoculation. This new cultivar may serve as a valuable one for artificial cultivation and industrial-scale production of C. militaris.

  • 최근 화학, 물리, 생명과학, 전기, 전자등의 다양한 분야에 활발하게 연구가 이루어지고 있는 초분자 화학은 선택적 분자인지를 위한 효율적인 골격구조와 나아가 다양한 계에 응용할 수 있다. 초분자 화학의 분자인지 과정의 특징은 일반적으로 수용체 (receptor 혹은 host)가 목표가 되는 기질 (substrate, analyte, 혹은 guest)에 대하여 선택적으로 식별하고 반응하는 것이다. 비공유 결합성 상호작용에 의하여 이루어지는 초분자 화학의 분자 인지 과정의 특징은 일반적으로 수용체 (receptor 혹은 host)가 목표가 되는 기질 (substrate, analyte, 혹은 guest)에 대하여 선택적으로 식별하고 반응하는 것이다. 이는 공유결합을 이용하는 분자화학과는 차별화 된 것이다. 수용체는 간단한 구조의 화합물 및 금속 이온들과 같은 기질과 가역적으로 상호 작용할 수 있는 착물을 형성한다. 최근들어 급격한 산업화가 진행되어 환경문제가 심각하게 대두 되어져 왔고, 그 중에서 특히 수은이나 카드뮴에 의한 질병, 납에 의한 중독 등 중금속에 의한 오염이 크게 나타남에도 불구하고, 현재 그러한 중금속을 검출함에 있어 많은 비용과 시간이 드는 문제점이 있다. 또한 우리에게 이로운 금속은 효율적 분석을 통해 환경계와 의료계에 많은 도움을 줄 것으로 사례되므로 화학센서 기술의 개발은 절실히 요구되어지고 있다. 이에 새로운 1,3-bisdicyanovinylindane 을 통해 $Hg^{2+}$ 금속의 감지 여부 알아보고, 그 특성을 파악하고자 한다. 1,3-indandion (2.18g, 14.9mmol), malononitrile (2.95g, 44.7mmol), ethanol 50ml를 20분간 상온에서 용해시킨다. 후에 sodium trihydrate acetate(3.05g)을 첨가한 후 5시간 동안 환류반응 시킨다. 이 과정에서 얻어진 용액을 필터과정을 통하여 에서 합성 반응 중에 생성된 불순물(1,3-dicyanovinylindane-1-one, monocondensation)을 제거한다. 필터과정을 통해 걸러진 미 반응 물질을 제�G 용반응욕액을 증류수(100ml)를 이용하여 희석시키고 난 후 염산을 이용, 산성화 시켜 고체 생성물을 얻어낸다. 이렇게 생성된 고체 생성물은 다시 필터 및 건조를 통하여 회색의 고체 화합물을 얻어낸다. 1,3-bisdicyanovinylindane과 금속이온에 대한 감응도를 확인하기 위하여 metanol/water(1:2)을 용매로 하여 금속이온의 농도를 변화시켜 발색특성을 살펴보았다. 본 색소화합물과 Hg2+에 대한 UV 흡광도 변화 적정결과와 그 화합물의 상태 살펴본 결과 금속이온이 0.2ml씩 더 참가되면서 색의 변화를 뚜렷하게 나타내었다. 반면 그 밖에 이온($Fe^{3+}$, $Ag^{2+}$, $Pd^{2+}$, $Zn^2$, $Fe^{2+}$, $Cu^{2+}$, $Pb^{2+}$)은 UV 흡광도 변화가 적거나 변화 자체가 없었다. 하지만 과량의 $Fe^{3+}$, $Ag^{2+}$, $Pd^{2+}$는 색상 변화를 나타내었으며,이와 같은 흡광도 변화는 금속에 따라 약간의 차이가 있지만, 420nm를 등흡수점으로 하여, 580nm의 파장 영역에 있는 흡수 밴드의 세기는 감소하는 반면 400nm 파장 영역에 있는 흡수 밴드의 세기가 증가하였다. 1,3-bisdicyanovinylindane 화합물은 다양한 생물계 및 환경계에서 요구되는 micro mol에서 milli mol 농도 영역의 $Hg^{2+}$ 이온의 선택적이고 민감한 검출과 정량적인 분석에 유용하게 사용될 수 있을 것이다.
