Stability of Pigment Produced by Monascus pilosus (Monascus pilosus가 생성하는 색소의 안정성)

  • Stability of Monascus pilosus pigment was investigated under various conditions. The concentration of the pigments stored under air and sub-atmosphere (250 mmHg) for 30 days at room temperature decreased by 77.9% and 48.4%, respectively. The pigment solution was stable under temperature ranges of $20-80^{\circ}C$, pH 4-8, darkness and presence of KCl, NaCl, $CaCl_2$, $MgCl_2$, and $ZnCl_2$. In contrast, the pigment solution was relatively unstable, decreasing in concentration by 6.0 and 11.6% at 100 and $121^{\circ}C$, 15.5 and 13.7% at pH 3 and 9, 22.9 and 66.8% under fluorescence and sun light, respectively, and 20.2% in the presence of $AlCl_3$.

  • Antioxidant activity of monascus pigment of Monascus purpureus P-57 mutant was studied. Methanol extract from monascus pigment was separated into five organic solvent fractions; hexane, chloroform, ethyl acetate, butanol and water fractions. Hexane fraction showed the highest free radical scavenging effect on 1,l-diphenyl-2-picryl hydrazyl(DPPH), and the strongest inhibitory effect against xanthine oxidase, followed by chloroform fraction. But butanol and water fraction did not show inhibitory effect against the enzyme. Lineweaver-Burk plot showed that hexane fraction inhibited xanthine oxidase by non-competitive mode.

  • The optimum cultural conditions for production of red pigment from Monascus rubber KCTC 6122 is liquid culture were studied. Monascus robber KCTC 6122 was shown to give the maximum production of red pigment in the medium containing 4% rice powder, 0.2% NaNO$_3$, 0.3% Na$_2$HP0$_4$ and 0.15% MgSO$_4$. The optimum culture conditions, temperature, initial pH and shaking speed were 30$^{\circ}C$, 6.5 and 150 rpm, respectively. The red pigment production reached a maximum level at 8days of cultivation.

  • 물김치는 발효 중 국물에 생성된 젖산을 비롯한 각종 유기산과 이산화탄소가 주는 독특한 신선미와 상쾌한 탄산미 그리고 배추나 무의 아삭아삭한 조직감 때문에 즐겨 먹는 김치의 종류이다. 일반 배추로 담근 물김치가 무색인데 비해, 안토시아닌 색소를 함유한 자색 양배추를 주재료로 물김치를 담그면 발효됨에 따라 유기산이 생성되어 pH가 낮아짐에 따라 색소가 안정화되어 시각적으로 아름다운 색을 띌 것으로 생각된다. 따라서 본 실험에서는 배추, 흰양배추, 적색양배추로 물김치를 담그어 숙성되는 동안의 이화학적, 관능적 특성을 분석하였다.   • Conidia and cultural characteristics of isolates of chromogenic and teleomorphic strains of Colletotrichum gloeosporioides from apple and red pepper were compared. The mycelial growth of teleomorphic strains was faster than that of chromogenic strains in potato dextrose agar and V-8 agar. The chromogenic isolates from apple and red pepper developed. white gray to gray green mycelial rings interspersed with salmon to apricot colored conidial masses in colonies on potato dextrose agar and V-8 agar and none formed on ascigerous stage in cultures. The chromogenic isolates from red pepper produced conidia, most with one apex attenuated on apple and potato dextrose agar whereas fusiform and smaller conidia were produced in V-8 agar and water agar leaf medium. The chromogenic isolates from apple produced fusiform conidia in the media tested. The teleomorphic isolates from apple and red pepper produced cylindrical conidia, most with both apices rounded, developed white gray to dark olive green in a zonate pattern with small dark spots throughout colonies and formed the ascigerous stage in cultures.

  • The optimal culture conditions of Monascus purpureus MMK2 for production of red pigment were investigated in submerged culture. Monascus purpureus MMK2 showed a maximal production of red pigment in the medium containing of 3.0% wheat flour, $0.15%\;NaNO_3,\;0.25%\;Na_2HPO_4\;12H_2O$ and $0.15%\;MgSO_4\;7H_2O$. The optimal culture conditions of temperature and initial pH were $30^{\circ}C$ and 6.5, respectively. The red pigment production reached to a maximal level at 7th day of cultivation.

Pigment and Monacolin K Content of Beni-koji Fermented with Soybean Curd Residue (비지홍국의 색소 및 Monacolin K 함량)

  • This study was conducted to investigate the preparation possibility of Beni-koji by Monascus pilosus using dried soybean curd residue(Biji). The additional effect of water(0-50%), glucose(0-10%, w/w), monosodium glutamate(0-0.l%, w/w) and citrus peel water extracts (0-0.5%, v/w) on the pigment and monacolin K content of the Biji Beni-koji were examined. Optimal added amounts of water was 20% of dried Biji. The highest pigment content(OD at 500 nm) of Biji Beni-koji was 1.06 in 10% glucose, 2.26 in 0.01% monosodium glutamate and 2.61 in 0.4% citrus peel water extracts. The content of monacolin K in the Biji Beni-koji added with 10% glucose, 0.01% monosodium glutamate and 0.4% citrus peel water extracts showed 96.38 mg%(w/w), 118.25 mg%(w/w) and 104.50 mg%(w/w), respectively.

  • The optimum cultural conditions for production of red pigment from Monascus anka KCTC 6121 on solid culture were studied. The optimal conditions were found that the strain was cultivated on polished rice with 25% initial moisture content, at 3$0^{\circ}C$, 90% humidity for 12 days. It was also found that the maximum red pigment was extracted when the final culture was left in 80% ethanol for 2 days. The light stability of the extracted red pigment was relative stable since the discoloration rate was less than 8% in 30 days under the indirect light.

Effect of Exogenous Hormones on Anthocyanin Accumulation and Phenylalaine Ammonia-lyase and Chalcone-synthase Activity in the Hairy Root Cultures of Raphanus sativus cv. Chungpihongsim (청피홍심무우의 모상근 배양에서 안토시아닌의 생성 및 PAL, CHS의 활성에 미치는 호르몬의 영향)

  • When effects of exogenous hormone on hairy root cultures of Raphanus sativs cv. Chungpihongsim examined, the highest anthocyanin synthesis and disorganization were observed when 2, 4-D was supplemented to the culture medium Cytokinins showed early weak induction after transfer and ABA showed inhibitory effect and GA3 showed no effects in anthocyanin synthesis. Hormones except for 2, 4-D in 1 mg/$\ell$ concentration did not induce disorganization of hairy root and retarded growth of hairy root. Time-course changes in anthocyanin synthesis, phenylalanine ammonia-lyase activity and chalcone synthase activity were examined in culture condition contalning 2, 4-D and kinetin. In a medium containing 2, 4-D, anthocyanin synthesis began to increase on the 9th day and reaching maxima on the 18th day after transfer. Maximum peak of PAL activity appeared on the 3-9th day and another minor peak appeared on the 18th day. CHS activity increased from 9th day, reaching maximum on the 18th day and remained at a relatively high level for culture period. In a medium containing kinetin, anthocyanin synthesis increased temporarily on the 6-9th days, early days after transfer and maintained at a low level for remaining culture period. Peak of PAL activity appeared on the 6th day and CHS activity increased from the 6th days, reaching maxima about 18th day and remained at a relatively high level. In particular, addition of kinetin after preculture in hormone free medium for 2 weeks which was thought of wound healing period showed no effects in anthocyanin synthesis. This results showed that stimulation of anthocyanin synthesis by 2, 4-D and kinetin was meaningfully connected with changes of PAL, CHS activity, and then suggested rate-limiting role of CHS on anthocyanin synthesis in that there is close correlation between anthocyanin synthesis and changes of CHS activity in time-course. Besides, it is considered that cytoklnins involving kinetin stimulated anthocyanin synthesis be due to "wound response" by cutting of young roots, and that difference in time-course peak and PAL, CHS activities expressed by 2, 4-D and kinetin result from occurrence of isozyme which have different regulatory mechanism.mechanism.

Isolation of a Pigment Producing Marine Strptomyces sp. from Shallow-sea Muds around in Korea (한국 근해 연안저토에서 분리한 해양 방선균이 생성하는 색소의 분리)

  • A marine Streptomyces sp., which produce water-soluble blue pigment was isolated from shallow-sea muds. The effect of various nutritional conditions on growth of isolated strain were investigated to facilite the potential use of this organism in industry. The effect of carbon source was characterized as a 10g/1 of soluble starch. Growth was optimum at pH 7.0 and slightly affected with more alkaline range but was shown to decrease dramatically in acidic range. Under the optimum growth conditions, isolated strain produced substantial amounts of blue pigment and biomass.
