Phylogenetic relationships of genera Trametes on the basis of ITS region sequences (rDNA의 ITS 부위 염기서열 분석에 의한 구름버섯 균주의 유전적인 유연관계 분석)

  • This study was carried to identify a correct species and asses genetic diversity within the same species of Trametes spp. preserved in Division of applied Microbiology The morphological and cultural characteristics of preserved strains were observed through microscope and investigated on PDA, respectively. Contaminated isolates showed different growth rates, morphology and color of hyphae. We have reconstructed the phylogenetic tree of a select group of Trametes spp. using nucleotide sequences of the internal transcribed spacer region(ITS) region. The phylogenetic tree was constructed by using the neighbor-joining method. PELF primers of 20-mer were used to assess genetic diversity of preserved isolates. Sequence analysis showed that five strains were different species and six strains were identified completely different nomenclature. According to the analysis of ITS sequences, the genus Trametes clustered into four distinct group, most of which correlated with species-groups identified by RAPD method. Seven isolates included TM 01 strain showed high similarity with Trametes versicolr, TM 07 and TM 10 high similarity with Trametes gibbosa, and TM 05 high similarity with Trametes elegans. But isolates collected in the United States was identified as T. junipericola. T. gibbosa and T. versicolor by RAPD analysis of genetic polymorphisms showed a very different band patterns and these strains showed different band patterns on areas. As the result of RAPD and ITS region sequences analysis for preserved isolates, it seems likely that 11 isolates of Trametes spp. may be need to reclassify or eliminate from preserved catalogue.

Isolates were collected in the United States(PM10, PM11) was identified as P. alveolarius and P. arcularius. RAPD analysis of genetic polymorphisms of genus Polyporus showed a very different band patterns. As the result of RAPD and ITS region sequences analysis for preserved isolates, it seems likely that 6 isolates of Polyporus spp. may be need to reclassify or eliminate from preserved catalogue.

A Study on Detecting Moving Objects using Multiple Fisheye Cameras (다중 어안 카메라를 이용한 움직이는 물체 검출 연구)

  • Since vision-based surveillance system uses a conventional camera which has a narrow field of view, it is difficult to apply it into the environment whose the ceiling is low and the monitoring area is wide. To overcome this problem, the method of increasing the number of camera causes the increase of the cost and the difficulties of camera set-up For these problems, we propose a new surveillance system based on multiple fisheye cameras which have 180 degree field of view. The proposed method handles occlusions using the homography relation between the multiple fisheye cameras. In the experiment, four fisheye cameras were set up within the area of $17{\times}14m$ at height of 2.5 m and five people wandered and crossed with one another within this area. The detection rates of the proposed system was 83.0% while that of a single camera was 46.1%.

Morphological Variation and Partial Mitochondrial Sequence Analysis of Echinoid Species from the Coasts of the East Sea (동해 연안에 서식하는 성게의 형태변이와 미토콘드리아 유전자 분석)

  • Morphological classification of echinoid species has many difficulties because of their phenotypic variations. In the present study, we analyzed morphotypes and partial mitochondrial 12S rDNA sequences of four sea urchin species classified as Pseudocentrotus depressus, Anthocidaris crassispina, Hemicentrotus pulcherrimus and Strongylocentrotus nudus, and unidentified four species collected from the coasts of the East sea. Their genomic DNAs were extracted from gonads and mitochondrial 12S rDNA sequences were amplified by the polymerase chain reaction (PCR) method. The sequence identities among the known four sea urchin species were 87.4-95.6%. The sequence identities among the unidentified four species were 99.4-99.6% and showed the highest homology to S. intermedius(99.8%). Thus, our phylogenetic tree indicates that the unidentified four species belong to S. intermedius.

Significance of Ages of Tungsten Mineralization (중석(重石) 광화작용(鑛化作用) 시기(時期)의 의의(意義))

  • It is understood that many big tungsten deposits such as the Sangdong in Korea, Fugigatami in Japan, Yukon in Canada, Pine Creek in U.S.A and Vostok in Russia were formed at late Cretaceous ages. However, most of tungsten mineralization in China where half the total world tungsten ores is reserved took place in late Jurassic to early Cretaceous ages. While the close association of molybdenum with tungsten mineralization is observed in the deposits related with Cretaceous magma, tungsten deposits in China related with late Jurassic to early Cretaceous show a close association of tin as well as molybdenum mineralization. It is characteristic that tungsten mineralization in China was followed by tin mineralization. The mode of occurrence of tungsten ore deposits in China is various and may represent the origin of tungsten in general, since the larger half of total amount of tungsten ores in the world are reserved in China. In case of Korea, more than 90% of total production of tungsten was occupied by the Sangdong tungsten deposit, which produced molybdenite as a byproduct Even if tin is detected in ppm unit content, no cassiterite is found in the Sangdong tungsten orebody. A similar type of two tungsten deposits is comparatively studied in order to confirm the published data; one is the Moping tungsten deposit in China and the other is the Dehwa tungsten deposit in Korea. Mineral assemblages occurring in quartz veins of both deposits are more or less same except that zinnwaldite and cassiterite occur only in the former deposit Ages of zinnwaldite and muscovite closely with molybdenite in the former deposit are 181.1 Ma and 167.8 Ma respectively, while muscovites associated with molybdenite in the latter deposit show ages of 80.9 Ma and 80.2 Ma. These results may represent deficient supply of tin from the source granitoid from which tungsten was derived in Korean peninsula during Cretaceous period, while tin supplied during tungsten mineralization tended to increase and the active tin mineralization followed the Jurassic tungsten mineralization in China.

Genetic Analysis of 5 Mountain Cultivated Ginseng and Wild Ginseng in Korea (국내 5개 지역의 장뇌삼과 산삼의 유전 분석)

  • ISSR PCR technique was applied to investigate genetic relationship among 5 Mountain cultivated ginseng populations (Jinan, Hongcheon, Punggi, Andong and Yeongju) and cDNA libraries of wild ginseng roots were constructed and analyzed functional genes related to morphogenesis via EST. Twenty four ISSR markers tested produced 127 polymorphic loci from 5 regional Mountain cultivated ginseng. Among the regional samples, Yeongju was made 18 polymorphic loci that were the highest level of variations among the cultivated regions. The range of similarity coefficient was 0.46~0.58 and the regional samples of Punggi and Hongcheon, Jinan and Andong were classified to similar groups respectively, whereas Yeongju was shown to be separate group with high level of genetic variation in UPGMA cluster analysis. As a result, there was no relationship according to geographical distance and genetic similarity. Eleven cDNA clones were consisted of 9 known genes and 2 unknown genes analyzed by BLAST program of NCBI. To recognize expression pattern of Homeodomain transcription factor related genes, Northern Blot analysis was performed for wild ginseng's leaf and root. As a result, the gene was only expressed by Mountain wild ginseng root.

Phylogenetic and Morphological Comparison between Thamnaconus septentrionalis and T. modestus Collected in Southwest Seashore (서남해에서 채집된 말쥐치 (Thamnaconus modestus)와 유사종 (T. septentrionalis)의 형태 및 계통유전학적 비교)

  • Thamnaconus modestus, distributed in the Northwest Pacific, has high economic value and is used in various seafood. In this study, the morphological and genetic characteristics of T. modestus and T. septentrionalis were compared and analyzed. We observed the external and internal morphology of T. modestus, sketched skeletal elements, and analyzed phylogenetic evolutionary relationships using the cytochrome c oxidase subunit I (COI) gene on mitochondrial DNA compared to T. septentrionalis. The T. modestus observed in this study had blackish-brown patterns irregularly scattered on the gray-brown body, and the fins were blue-green. Genetic analysis results based on the COI sequences of T. modestus showed seven types of base sequence variation; however, the homology was more than 98.8%. In addition, as a result of comparison of the COI nucleotide sequences and phylogenetic analysis in Tetraodontiformes, two T. septentrionalis sequences (JN813099, MW485059) were similar to T. modestus with 99% homology, and the other two T. septentrionalis sequences (EF607583, KP267619) were similar to those of species belonging to another genus Thamnaconus with 95% homology with T. modestus. It was not easy to classify the species based on morphological characteristics, and phylogenetic analysis between T. modestus and T. septentrionalis confirmed the difference in classification. These results provide the external and internal morphology of T. modestus and will be used as important information for the taxonomic study of T. modestus and T. septentrionalis.

Effect of Zebularine on Chromosomal Association between Meiotic Homoeologous Chromosomes in Wheat Genetic Background (Triticum aestivum L.) (제부라린이 생식세포분열 동안 동조 염색체 사이의 염색체 접합에 미치는 영향)

  • The objective of this study was to identify the effect of zebularine, a DNA methylation inhibitor, on the chromosomal association between homoeologous chromosomes in the wheat genetic background. Zebularine at a final concentration of 10 µM was used to treat the spikes of the double monosomic wheat addition line (DMA) with one Leymus mollis chromosome and one Leymus racemosus chromosome, both of which were in a homoeologous relationship. In late prophase, zebularine led to chromosome breakage in the Leymus homoeologous chromosomes. Chromosome breakage caused an increase in the frequency of chromosomal associations between the Leymus homoeologous chromosomes. Ordinary DMA showed 65 cells (35.3%) with chromosomal associations and 119 cells (64.7%) with no association, whereas treated DMA showed 102 cells (60.0%) with chromosomal associations and 67 cells (39.4%) with no association. In diakinesis, the Leymus bivalent showed a chromosomal association in the whole euchromatic region. In metaphase, the Leymus bivalent showed association in the whole chromosomal region, unlike other Leymus bivalents with partial chromosomal association. Chromosomal association by chromosome breakage occurred not only between Leymus chromosomes but also between Leymus and wheat chromosomes. The frequency of other chromosomal association (such as fusion and insert) was increased. Chromosome breakage by zebularine treatment is a useful method at the chromosome level as the spores with others are hereditary stable, although the homologous index (h) was not significantly different between ordinary DMA and treated DMA. It is necessary to study how to control zebularine treatment with a more stable concentration for chromosome breakage during meiosis.

Phylogenetic Relationships of Morus Species on the Basis of RAPD (RAPD를 이용한 뽕나무속 식물의 유전적 유연관계 분석)

  • Phylogenetic relationships among mulberry varieties (Morus species) were analyzed on the basis of RAPD in order to identify the possibility of classification for the species. Polymorphisms under RAPD method were compared among 41 mulberry varieties. From the results of RAPD for 41 mulberry varieties by use of 30 different primers, 151 polymorphic bands were formed out of 201 ones. Under the dendrogram based on cluster analysis with the polymorphic bands, the varieties were classified into 7 groups including two large and five small ones on 0.747 value of genetic similarity coefficient. In the large groups 19 and 16 varieties were belong to group I and III, respectively. On the other hand, relatively high genetic similarity was shown among the varieties belonging to the group I, II and III. While, relatively low similarity were done between them in the group IV-Ⅵ and the other groups, and Mohusang in the group Ⅶ showed the lowest phylogenetic relationship with the other varieties.

  • 음절과 글자의 내부구조에 대한 언어학적 심리학적 논의들을 개관하였다. 영어의 음절구조에 대한 연구들은 초두자음/각운 구조를 지지하고 있다. 한편 한국어의 음절과 글자의 내부구조에 대한 최근의 연구는 영어에서 얻어진 결과들과 다른 결과를 얻고 있다. 자모대체 과제를 사용한 이 연구에 의하면, 글자유형에 관계없이 종성자모의 대체시간이 초성자모의 대체시간보다 짧았다. 이러한 결과는 음절에 대응하는 글자의 내부구조로서 초중성자모/종성자모 구조를 지지하고 있다. 선행연구 결과들을 바탕으로 음운단위와 표기단위의 상동성, 그리고 언어특유적 음절구조의 가능성에 대해서 논의하였다.
