The effects of antibodies and complement in macrophage-mediated cytotoxicity on metacercariae of the lung fluke, Paragonimus westeymani (폐흡충(Paragonimus Tuestermani) 피낭유충에 대한 대식세포의 세포독성에 있어서 항체 및 보체가 미치는 영향)

  • Paragonimus westermani is a tissue migrating parasite in the early stage until arriving at lung, and most of the parasites spend their life spans there. Considerable immune responses including activation of macrophages are taken place during the residence of parasites in the host. However, concerning the immunologic defense mechanisms of the host against this parasite, only a few document is available so far. In this study, the cytotoxic effect of peritoneal macrophages under the presence of antibody and/or complement against metacercariae of F. westermani was investigated in vitro. Metacercarlae were collected from the crayfish, Cambaroides similis and hatched out in Tyrode solution (pH 7.4). Plastic adherent cells from normal or infected rat (Wistar) peritoneal exudates were used as experimental macrophages. Polyclonal antibodies were obtained from infected rats and a cat. Cat IgG was fractioned with ion exchange chromatography. Fresh rabbit complement was used according to experimental scheme. Various combinations of peritoneal macrophages, normal or infected rat serum, complement and cat IgG were incubated at $36^{\circ}C$ in 5% $CO_2$ incubator for 6, 14, 24 and 48 hours. The results obtained were as follows: 1. P. westermani infection activated peritoneal macrophages non-specifically and this activation induced increases of cell adherence and cytotoxicity on metacercariae. 2. In the presence of infected rat serum the antibody.dependent cell-mediated cytotoxicity of peritoneal macrophages on metacercariae was significantly increased and showed a peak at 6-hour incubation. But the cytotoxic effect was markedly reduced after inactivation of complement and heat.labile IgE antibody by the heating of infected serum at 56$^{\circ}C$ for 30 minutes. 3. The highest cytotoxic effect (100%) of concomitant incubation with IgG and complement showed 24 hours after incubation, although cell adherence was relatively low at 6-hour incubation and 0% at 24-hour incubation. 4. Coordinative functions of complement with serum and IgG were effective in cell adherence and in cytotoxicity, but it is not clear the independent role of complement on the macrophage- mediated cytotoxicity in this study- With these results it is assumed that P. westermani infection can induce the non-specific activation of peritoneal macrophages, and strum antibodies including IgE antibody might enhance the cytotoxicity by macrophages,

Gnawing and Escaping Behaviors of Monochamus alternatus (Coleoptera: Cerambycidae) in a Confined Environment: Suggesting a Bioassay Method of Netting for Adult Escape Prevention (인위적 구속환경에서 솔수염하늘소의 쏠기와 탈출행동: 성충탈출 방지용 그물망의 생물검정법 제안)

  • The Japanese pine sawyer, Monochamus alternatus Hope, is a representative vector of the pine wood nematode, Bursaphelenchus xylophilus, which causes wilting symptoms in pine trees. A control method using a net has been introduced, which is an alternative method to the fumigation for the control of dead pine trees by pine wilt disease. This study was carried out to investigate the factors that induce gnawing and escaping behaviors of M. alternatus. The behaviors were examined after M. alternatus adult was placed in a confined space at different temperatures. M. alternatus adults could escape through mesh net torn by gnawing when they were confined in a space of 30 mm or less in diameter. The success rate of escape was high at 20 to $30^{\circ}C$, and no adults escaped at $15^{\circ}C$. The enticement of M. alternatus adults by food didn't affect the success rate of escape. In the case of not being confined in a narrow space, the escaping hole could not be formed because the gnawing was not concentrated on one part. M. alternatus moved its body in a narrow space using the tarsus of middle and hind legs, and made an escape hole by concentrically gnawing the obstacle on the front side with mandible, and showed a behavior of getting out while supporting the body by supporting the front legs. The present results will be able to use as an important basic information for evaluating the performance of mesh net which confines M. alternatus adults and suggested by alternative method to fumigation technology.

Control of Food Pests by $CO_{2}$ Modified Atmosphere: Effects of Packing Materials and Exposure Time on the Mortality of Tribolium castaneum and Plodia interpunctella (고농도 탄산가스 충전에 의한 식품 해충 방제: 거짓쌀도둑거저리와 화랑곡나방의 살충효과에 미치는 노출시간과 식품포장 재질의 영향)

  • Effects of three packing materials and exposure time on the mortality of Plodia interpunctella and Tribolium castaneum were studied using $CO_{2}-modified$ atmosphere control. Materials used were triple layered craft paper (KKK), triple layered craft paper with one laminate coating layer (KLK), and triple layered craft paper with one HD film coating layer (KHK). In the test balls (${\phi}1.2\;m$) containing 85% $CO_{2}-modified$ atmosphere, concentration of $CO_{2}$ in small packing envelopes ($9.8{\times}9.8cm$) made of the materials after one day was higher in KKK ($26.67{\pm}0.58%$) than KLK ($23.33{\pm}0.58%$) or KHK ($20.67{\pm}0.58%$). Both P. interpunctella and T. castaneum showed higher mortality in KKK than in either KLK or KHK Similar results were obtained by larger space ($9{\times}4{\times}3m$) and packing volume (20 kg) studies. The effect of the packing materials was clearer on T. castaneum more tolerant to $CO_{2}$ than P. interpunctella. Regression of the insect mortality on cumulative concentration time (${\int}_{0}^{t}c{\times}tdc{\approx}{\sum}concentration{\times}time$) was highly significant. Control of P. interpunctella and T. castaneum by $CO_{2}-modified$ atmosphere were discussed in relation to packing materials, cumulative concentration time of $CO_{2}$ and food products.

  • The susceptibility of the western flower thrips, Frankliniella occidentalis and garden thrips, Frankliniella intonsa was evaluated using 51 commercial insecticides. 15 kinds of insecticides which showed more than 90% mortality against both thrips, F. occidentalis and F. intonsa was selected. Many active ingredients were misused and abused in commercial mixture formulation insecticides. Since the F. intonsa was more susceptible than F. occidentalis, it was considered that both thrips can be controlled by insecticides that showed insecticidal activity on the F. occidentalis. Lethal time ($LT_{50}$ and $LT_{95}$), systemic toxicity and residual toxicity of selected insecticides were compared. Both chlorpyrifos WP and chlorpyrifos + diflubenzuron WP revealed the fastest toxicity within 2 h ($LT_{95}$), while spinetoram WG revealed the slowest toxicity as 62.3 h ($LT_{95}$). Chlorfenapyr SC showed toxicity at foliar and drenching application while spinetoram WG was toxic only in foliar application. Chlorfenapyr SC showed residual effect at 3, 5, 7, 10, 15 days after treatment and both benfuracarb WG and chlorpyrifos WP showed residual effect at 3 days after treatment. As a result of treatment of selected insecticides for field population of F. occidentalis, the population collected from horticultural crops showed lower susceptibility than the population collected from vegetable crops.

Development of a Fennel (Foeniculum vulgare) Oil-based Anti-insect Sachet to Prevent the Indian Meal Moth (Plodia interpunctella) (화랑곡나방 유충 방제를 위한 회향오일 기반 방충향낭 개발)

  • The stored-product insects have been a serious problem during the entire process of distribution on the food industry. Especially, the Indian meal moth (Plodia interpunctella) is one of stored-product insects which causes harm through penetrating into the food packaging. The objective of this study was to develop the anti-insect packaging material with fennel (Foeniculum vulgare) oil (FO). The FO has been selected for insecticidal substance against P. interpunctella, which was tested by fumigant toxicity assay. An anti-insect sachet was prepared by FO and filterpaper placed in a small paper bag. Repellent test was performed to evaluate the repellent activity of anti-insect sachet. In addition, the controlled release of FO from the anti-insect sachet was determined at $28^{\circ}C$ by gas chromatography (GC). It was demonstrated that FO was an effective substance against P. interpunctella. The mortality of FO was 56% at 800 mg/0.5 L in 120 h. In repellent test, the FO sachet showed effective repellency against P. interpunctella. The developed anti-insect sachet could be a promising source for insect repellent materials in food packaging.

Isolation and Activity of Insect Pathogenic Bacillus thuringiensis Strain from Soil (국내 토양으로부터 곤충병원성 세균인 Bacillus thuringiensis 균주의 분리 및 생물검정)

  • Bacillus thuringiensis strains were isolated from the domestic soil and a strain was selected that had a new host range and high toxicity against agriculture insect pest. The 142 samples of soil were sampled from the mountains, paddy fields and patches, in Daejon, Chungnam, Chungbuk and Jeonbuk and used for the investigation. Sixteen B. t strains were isolated from 12 samples among collected samples. There were 11 strains that showed toxical activity on Plutella xylostella (Lepidoptera: Yponomeutidae), 7 steins on Spodoptera litura (Lepidoptera: Noctuidae), 5 strains on Arete coerulea (Lepidoptera: Noctuidae), 5 strains on Culex pipiens pallens (Diptera: Culicidae) among the 16 isolated B. t strains. But there were not any strains that showed activity against Hyphanria cunea (Lepidoptera: Arctiidae) and Sitophilus oryzae (Coleoptera: Rhynchophoridae). And also some of B. thuringiensis strains showed insecticidal activity with 2, 3 or 4 kinds of insects. But there were also 3 strains that did not show any activities to the 6 insects which were used in the experiment. When examined with a phase-contrast microscope, the insecticidal crystal protein produced from 16 selected strains had 13 bipyramidal and 3 spherical shapes. The insecticidal bioactivity of the S. litura showed 100% mortality when there were $1.3{\times}10^{7}\;(cfu/ml)$ of CAB109 isolates.

An Integrated Biological Control Using an Endoparasitoid Wasp (Cotesia plutellae) and a Microbial Insecticide (Bacillus thuringiensis) against the Diamondback Moth, Plutella xylostella (배추좀나방에 대한 프루텔고치벌과 미생물농약의 통합생물방제)

  • All tested Korean populations of the diamondback moth, Plutella xylostella, are known to be resistant especially against pyrethroid insecticides by mutation in its molecular target, para-sodium channel. Moreover, P. xylostella is able to develop resistance against most commercial insecticides. This study was performed to develop an efficient control technique against P. xylostella by a combined treatment of an endoparasitoid wasp, Cotesia plutellae, and a microbial insecticide, Bacillus thuringiensis. To investigate any parasitism preference of C. plutellae against susceptible and resistant P. xylostella, five different populations of P. xylostella were compared in insecticide susceptibilities and parasitism by C. plutellae. These five P. xylostella populations showed a significant variation against three commercial insecticides including pyrethroid, organophosphate, neonicotinoid, and insect growth regulator. However, there were no significant differences among five P. xylostella populations in their parasitic rates by C. plutellae. Moreover, parasitized larvae of P. xylostella showed significantly higher susceptibility to B. thuringiensis. As an immunosuppressive agent, viral ankyrin genes (vankyrins) encoded in C. plutellae were transiently expressed in nonparasitized larvae. Expression of vankyrins significantly enhanced the efficacy of B. thuringiensis against the third instar larvae of P. xylostella. Thus an immunosuppression induced by C. plutellae enhanced the insecticidal efficacy of B. thuringiensis. These results suggest that a combined treatment of C. plutellae and B. thuringiensis may effectively control the insecticide-resistant populations of P. xylostella.

Cultivation Condition of Transformant Alcaligenes eutrophus Harboring Cloned phbC Gene for Production of P(3-hydroxybutyrate-3-hydroxyvalernte) Containing High Molar Fraction of 3-Hydroxyvalerate. (P(3-hydroxybutyrate-3-hydroxyvalerate)의 생산을 위한 재조합 phbC 유전자를 형질전환시킨 Alcaligenes eutrophus의 배양조건 검토)

  • The cultivation conditions of transformant Alcaligenes eutrophus AER5 harboring cloned phbC gene for mass production of poly (3-hydroxybutyrate-3-hydroxyvalerate)[P(3HB-3HV)] containing high molar fraction of 3-hydroxyvalerate (3-HV) were investigated. In two-stage batch cultivation, transformant accumulated P(3HB-3HV) containing 52.2 mol% of 3HV compared to 30 mol% of parent strain A. eutrophus H16. The increased 3-HV molar fraction was due to the amplified activity of PHB synthase participating in condensation of 3-HB and 3-HV. To increase efficiency of P(3HB-3HV) accumulation, fructose was added along with precursor compound valerate, and total cell mass and P(3HB-3HV) concentrations remarkably increased, but not 3-HV molar fraction. The effect of magnesium ion showed that P(3HB-3HV) concentration and 3-HV molar fraction were significantly increased upto 6.1 g/L and 71.3 mol% at 0.01 g/L of MgSO$_4$, respectively. The efficiency of several pH adjuster, NaOH, NaOH and (NH$_4$)$_2$SO$_4$, and NH$_4$OH, on total cell mass, p(3HB-3HV) concentration, and 3-HV molar fraction was also compared. To overcome the disadvantage of two-stage cultivation, one-stage intermittent fed-batch cultivation was attempted, such a way 10.0 g/L of fructose was supplied for cell growth at initial 36 hr and then 10.0 g/L of valerate and 5.0 g/L of fructose were applied to induce the accumulation of P(3HB-3HV), consequently, 10.4 g/L of P(3HB-3HV) with 38 mol% of 3-HV fraction could be obtained after 72 hr. These results can be used for elucidating cultivation strategy for mass production of P(3HB-3HV) containing high 3-HV molar fraction using transformant A. eutrophus AER5 harboring cloned phbC gene.

Survey the Occurrences and Establishment of Environment-friendly Control System of Ricania shanthungensis in Jeonnam Province (전남지역 갈색날개매미충 발생현황과 친환경 방제)

  • This study was carried out to establish of environment-friendly control system and to survey the occurrence status of Ricania shanthungensis in Jeonnam province from 2016 to 2017. R. shanthungensis occurred at 1,344 ha in 61 towns of 15 cities in Gwangju and Jeonnam province. At four sites in the stationary area, the densities of eggmass and pupae were lower in 2017 than in 2016, the hatching rate decreased, and the first hatching time, emergence of adults, and the start of spawning tended to be slower. Telenomus sp., an egg parasitoid of R. shanthungensis, was found in most areas. Sunchon had the highest rate of 46.1%, followed by Muan 40.6, Goksung 29.2, Gurye 25.8, and Gwangju 17, respectively. The optimum spraying time for the control of R. shanthungensis was early March. The spraying material and the dilution multiples are 10 times of the machine oil at environmental friendly cultivation and 500 times of the chlorpriphosphate wp. at normal cultivation. When sprayed these materials, 95% of egg prohibited to hatch. There was more than 80% of the insecticidal effects on the organic materials of the machine oil emulsion, the Sophora root extrect + microbial extract agent, and the castor emulsion. For adults, Sophora root extrect + microbial extract, neem extract, and machine oil were better. Four kinds of chemical pesticides such as dinotefuran wp were effective for nymph and adult control. We have developed an adult catching device using the most preferred daylight color and behavioral habits of R. shanthungensis. The capture device consisted of two daylight compact lamps (30W and 20W), a yellow plate, and a catcher using water, and caught about 700 individuals a day. Based on the above results, we have established a system for controlling and life cycle of R. shanthungensis in Jeonnam province.

Toxicological Effects of Some Insecticides against Welsh Onion Beet Armyworm (Spodoptera exigua) (Metaflumizone을 포함한 몇 가지 약제의 파밤나방(Spodoptera exigua)에 대한 독성검정)

  • The Welsh onion beet armyworm, Spodoptera exigua (Hubner), has attacked Welsh onion and is now the most important pest of Welsh onion in southwestern Korea. The beet armyworm has a wide host range, occurring as a serious pest of vegetable and fields. The relatively high abundance of beet armyworm has stimulated frequent application of insecticides to foliage. Insecticide resistance is a major problem in management of this insect. Accordingly, pesticide application for the control of beet armyworm was tried in both the open field and in laboratory, using 4 synthetic compounds such as metaflumizone and chlorfenapyr, indoxacarb, flufenoxuron, emamectin benzoate. In the laboratory, each developmental stages from eggs, larva to pupa was tested against 4 insecticides. Against the eggs of welsh onion beet armyworm, there was no significantly different with each other. These tested chemicals no killing effect to eggs. However, the population of 1st larva hatched from eggs were reduced because they eaten the egg shell with residual insecticides. The tested insecticides were taken very high mortalities to 1st to 3rd larva of Welsh onion beet armyworm. Otherwise, there were decreased the death rate from 4th to 6th larva. On the other hand, their value of control effects were relatively good against Welsh onion beet armyworms in the field between 87.2 and 90.5% on 10 days after insecticide application.
