Preparation and Characterization of p-Styrenesulfonates of Isopropylidene Dicyclohexanol as Acid Amplifiers to Enhance the Photosensitivity of Positive-Working Photoresists (포지티브 포토레지스트의 감도 증진을 위한 산 증식제로 이소프로필리덴 디시클로헥산올의 p-스티렌술폰산 에스테르의 합성 및 특성연구)

  • The photosensitivity enhancement of photoresist achieved by the addition of acid amplifiers stems from the autocatalytic decomposition of the acid amplifiers triggered by acidic species generated from a photoacid gen-erator. In this research we synthesized and evaluated 4-hydroxy-4'-p-styrenesulfonyloxy isopropylidene dicyclohex-ane(1), 4,4'-di-p-styrenesulfonyloxy isopropylidene dicyclohexane(2) and 4-p-styrenesulfonyloxy-4'-tosyloxy isopropylidene dicyclohexane(3) as novel acid amplifiers. These acid amplifiers(1-3) showed reasonable thermal stability for resist pro-cessing temperature. As estimated by the sensitivity curve, 1-3 were 2X-12X sensitive than poly(tert-butyl meth-acrylate) film in the presence of a photoacid generator and, therefore, provides practical applicability for photoimaging.

A Novel Photoresist based on Polymeric Acid Amplifier (고분자형 산 증식제에 기초한 새로운 포토레지스트의 연구)

  • Acid amplifying copolymers are synthesized by the copolymerization of tert-butyl methacrylate(tBMA) with acid-sensitive functional group and 4-hydroxy-4'p-styrenesufonyloxyisopropylidene dicyclohexane(HSI) or 4-p-styrenesulfonyloxy-4'-tosyloxyisopropylidenedicyclohexane(STI) with acid-amplifying group as novel polymeric acid amplifying photoresists. Poly(HSI-co-tBMA) film and Poly(STI-co-tBMA) film as acid amplifying photoresists show reasonable thermal stability in the absence of an acid species. Poly(STI-co-tBMA) film exhibits 2X higher photosensitivity, whereas Poly(HSI-co-tBMA) film show 2X lower photosensitivity compared with ptBMA homopolymer. The attachment of acid-amplifying units to polymer backbones could provide a novel way to enhance the photosensitivity of acid-sensitive polymers depending on the structure of acid-amplifying units.

Effects of Ascorbic Acid on the Proliferation of Subcultured Canine Chondocytes in Monolayer and Alginate Beads Culture (단순배양과 알긴산배양에서 개 연골세포의 증식에 있어 계대배양에 따른 아스코빅산의 영향)

  • Ascorbic acid has been used widely as a medium supplement to stimulate cell proliferation, but its effects on cell proliferation have not yet been elucidated, and no reports have analyzed effects on subcultured chondrocytes. Subcultured canine chondrocytes of passage one, two and four were cultured in monolayer and alginate beads with and without ascorbic acid. Cell proliferation was examined by 2,3-Bis(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide inner salt) (XTT) colormetric assay. Ascorbic acid stimulated cell proliferation significantly in both culture methods (p<0.05). The increased cell numbers by stimulation with ascorbic acid were significantly high in passage one cells compared to that of other passages. Differences in cell proliferative capacity by subculturing were not determined. These results suggest that ascorbic acid stimulated the proliferation of subcultured canine chondrocytes and enhanced it more in low-passage cells than in the other cells tested.

Change of amino acids contents of Gastrodia elata Blume with harvest times and seed tuber (수확시기와 자마의 특성별 천마의 아미노산 함량 변화)

  • Gastrodia elata Blume, an achlorophyllous orchid plant, has been used in traditional medicine and harvests in spring and fall. Here we investigated the quantitative changes of amino acids in G. elata by harvest times and seed tubers. In the results, we found that there was not big difference in contents of total amino acids but the distribution of amino acids differed depending on harvest times. 19% of total amino acids were asparagin and valine, an essential amino acid, occupied 10% within total amino acids in the tubers harvested in October. Among amino acids in the tubers harvested in April, serine occupied 23% and arginine, an essential amino acid, occupied 10% within total amino acids in tuber. Interestingly, the use of sexually propagated seed tubers produced high concentration of total amino acids compared to vegetatively propagated seed tubers. As for sexually propagated seed tubers, essential amino acids contents similar to the tubes harvested in October and nonessential amino acids contents similar to the tubers harvested in April. In this study, we found that amino acids contents in G. elata tubers altered depending on various cultivation practices. Therefore if these results can be applied to food industry, the value of G. elata as a natural food resource will be enhanced to a great extent.

효모에 의한 과실주중의 감산 효과에 관한 연구 제1보 균주의 분리 및 동정

  • 미생물학적 감산현상에 의하여 과실주중의 산미를 조절하기 위한 방법으로 효모에 의하여 사과산을 알코올로 분해하는 Maloalcohol 발효를 유도하고자 하여 본 연구를 하였다. 이미 발표된 Schizosaccharomyces pombe 0-77보다 단 시간에 강력히 사과산을 소비하는 효모균을 딸기의 과피로부터 분리, 동정하였다. 본 분리균을 J. Lodder의 “The Yeasts”에 준하여 동정한 바 분열법에 의하여 증식하며 포자를 형성하고 galactose 를 발효못하므로 Schizosacoha-romyces 속으로 분류하였다. 위균사를 잘 형성하며 melibiose를 발효하므로 Schizosaccharomyces japonicus와 일치 하였다. 그러나 변종의 동정은 어려우나 위균사의 형태로 보아 Schizosaccharomyces japonicus var. japonicus로 동정하였다.

홍화자 추출물의 치주 경조직 재생 촉진 효과

  • 최근 치의학 영역에서 사용되는 천연물 특히 생약제에 대한 연구가 활발히 진행되고 있다. Scutellariae Radix, Centella asiatica 등이 치주인대세포의 활성을 증가시키고 홍삼사포닌이 배양한 치주인대세포의 성장, 분화에 관계된다는 보고 등이 이에 해당된다. 본 연구에서는 홍화자 메탄을 추출물과 키토산의 치주인대 세포의 증식, 분화, 석회물 결정 생성 촉진작용을 검토하여 치주경조직 재생 약물로서의 사용여부를 보고자 하였다. 치주인대 세포는 교정치료목적으로 경희의료원에 내원한 환자의 제1 소구치를 발거하고 치근시작점에서 중앙으로 1/3되는 지점에서 치주인대 조직을 절취하여 1차 배양하였다. 실험에는 계대배양하여 5-7 세대의 세포를 사용하였다.

A Study on Synergisitic Effect of Chitosan and Sorbic Acid on Growth Inhibition of Escherichia coli O517:H7 and Staphylococcus aureus (E. coli O517:H7 과 Staphylococcus aureus의 증식억제에 대한 키토산과 소르빈산의 상승효과에 관한 연구)

  • This study was performed to investigate the synergistic effect of chitosan and sorbic acid as a new food preservative. So it was performed to investigate inhibitory effect on growh of E. coli 0157:H7, gram negative pathogenic food borne disease bacteria and of S. aureus, gram positive food borne disease bacteria in chitosan, sorbic acid and combination of chitosan and sorbic acid. Minimun Inhibitory Concentration (MIC) of chitosan in E. coli 0157:H7 was 500 ppm at pH 5.0, 250 ppm at pH 5.5, 500 ppm at pH 6.0, and 2000 ppm at pH 6.5, while in Staph. aureus 31.25 ppm at pH 5.0 and 62. 5 ppm at more than pH 5.5. also, MIC of sorbic acid in E. coli 0157:H7 was 500 ppm at pH 5.0, 1500 ppm at pH 5.5, and 2000 ppm at more than pH 6.0, while in Staph. aureus 1500 ppm at pH 5.0 and more than 2000 ppm at more than pH 5.5. Due to the effect of pH in E. coli 0157:H7, MIC of combined chitosan and sorbic acid was 500 ppm of chitosan with 500 ppm of sorbic acid at pH 6.5, but 250 ppm of chitosan with 31.3 ppm of sorbic acid at pH 5.0. In Staph. aureus, there was great effect of chitosan, but neither effect of pH nor sorbic acid. When E. coli 0157:H7 were treated with 500 ppm of chitosan with 500 ppm of sorbic acid and 250 ppm of chitosan with 250 ppm of sorbic acid at pH 6.5, they were inhibited. But, they were increased at the initial concentration of bacteria at 1000 ppm of chitosan in 18 hours, at 500 ppm of chitosan in 36 hours. There was no effect of growth inhibition with sorbic acid but great effect with chitosan on Staph. aureus. The correl~tions between MICs of chitosan and sorbic acid in E. coli 0157:H7 accoding to pH were higher than those in Staph. aureus. R values in E. coli 0157:H7 were 0.95 (p<0.01), 0.99 (p<0.01), 0.97 (p<0.01), and 0.99 (p<0.01) at pH 6.5, 6.0, 5.5, and 5.0 respectively. The synergistic effect of chitosan and sorbic acid in E. coli 0157:H7 could be confirmed from the result of this experiment. Therefore, it was expected that the food preservation would increase or maintain by using sorble acid together with chitosan, natural food additive that did no harm to human body.

Effect of Ursolic Acid on the Development of Mouse Embryonic Stem Cells under Hypoxia (저산소 상태에서 우르솔산이 배아줄기세포 성장에 미치는 효과)

  • Ursolic acid (UA) a bio-active ingredient found in a variety of fruits and vegetables, and it has potent antioxidant activity. However, the role of UA in mouse embryonic stem (ES) cells is poorly understood. This study investigated the functional role of UA in regulating the development of mouse ES cells under hypoxia. Hypoxia did not exert a significant effect on the undifferentiated state of mouse ES cells. However, it induced reactive oxygen species (ROS) generation and increased the level of lactate dehydrogenase (LDH) production at 48 h of hypoxic exposure. Conversely, oxidative stress induced by hypoxia was significantly inhibited by UA ($30{\mu}M$) pretreatment. Hypoxia significantly decreased cell survival and the level of [$^3H$] thymidine incorporation, both of which recovered following pretreatment of UA. In addition, UA decreased the apoptotic effect of hypoxia by attenuating caspase-3 cleavage or by recovering cellular inhibition of the apoptotic protein (cIAP)-2 and Bcl-2 expression. We further found that UA decreased senescence-associated beta-galactosidase activity. We suggest that UA is a natural antioxidant and one of the functional modulators of hypoxia-induced survival, apoptosis, proliferation, and aging in mouse ES cells.

Convergence Study on Natural Preservatives from Various Native Plant Species in Jiri Mountain Area (지리산 지역 자생식물 활용 천연보존제 융합 연구)

  • The purpose of the study aimed to explore the possibility for natural preservative convergence materials by examining the antioxidant activity and antimicrobial effect of 19 wild plants in Jiri Mountain. Total polyphenols were higher in the order of CS(Camellia sinensis L., 87.9 mg GAE/ g), MP(Mentha piperascens Holmes., 85.1 mg), NN(Nelumbo nucifera Gaertn., 65.0 mg) and PD(Pinus densiflora Siebold & Zucc., 52.8 mg). Total flavonoids were high in NN(25.7 mg QUE / g) and MP (25.4 mg QUE / g). CS(58.1%), NN(47.9%), and MP(40.6%) showed high ABTS radical scavenging ability and the result was similar in DPPH radical scavenging ability. The extracts of HC(Hemerocallis coreana Nakai.), PD, and CO(Cornus officinalis Siebold et Zucc.) showed the highest inhibitory effect on the growth of E. coli. The extracts of PK(Pulsatilla koreana Nakai ex Nakai.), SC(Saururus chinensis Baill.), and MC(Smilax china L.) completely inhibited the proliferation of S. aureus, showing the possibility to be developed as natural preservatives and disinfectants.

Biological Activity of Polyphenol Group Fraction from Korean Pear Peel (한국산 배과피 폴리페놀 분획군의 생리활성효과)

  • Biological activities and anticarcinogenicity of Korean Pear peel were investigated, Electron donating activity and superoxide dismutase (SOD)-like activity of fraction II, III were up to 80% and 50-60% at 50 ppm, respectively, Inhibitory effects on xanthine oxidase were about 80% at 50 ppm, breast adenocarcinoma was about 60% at 2,000 ppm, higher III than II Inhibitory effect on prostate adenocarcinoma was about 23% at 500 ppm. In conclusion, Korean pear peel was expected to use as a functional material.
