A Study on Trace-metals in Korean Yeongdeok Crab and Russian Snow Crab (영덕대게와 러시아산대게의 체내 미량금속 함량 연구)

  • This study is to measure the differences of the trace-metals resulted from the different ecologies, such as nickel(Ni), copper(Cu), zinc(Zn), cadmium(Cd), plumbum(Pb), arsenic(As) and chrome(Cr) remaining in the parts of Korean Yeongdeok Crab (KYC) and Russian Snow Crab (RSC) based on ICP-MS. The recovery rate of each metal certified the reference materials (CRM) was in the average of 81~99%, which corresponded with the level required in Codex. The level of metals in the parts of KYC was in the order of Ni > As > Zn > Cu > Cr > Cd in the both male and female shell; the order of Zn > As > Cu > Cr > Ni > Cd in the leg flesh; the order of Zn > As > Cu > Cr > Cd > Ni in the body flesh; the order of Cu > Zn > As > Cd > Cr > Ni in the gill; the order of Cu > As > Zn > Cd > Ni > Cr in the male hepatopancreas; the order of Cu > Zn > As > Cd > Cr > Ni in the female hepatopancreas, thereby showing some differences. It was revealed that the levels of most metals (nickel, copper, zinc, arsenic and chrome) were similar between KYC and RSC except cadmium which was somewhat lower than that of KYC. However, the cadmium in RSC was discovered in high level in most of the parts, two times higher in the hepatopancreas, and four times in the gill. It was also revealed that the trace metal contents were changing according to the size of KYC; the metals with the highest level of Ni in shell, Zn in leg and body flesh, Cu in gill tended to decrease as growing, whereas the cadmium contents tended to increase overall and accumulated the most in hepatopancreas. The results showed there was a possibility that the phenomenon of bioaccumulation within hepatopancreas would increase as growing.

Estimation of Terminal Sire Effect on Swine Growth and Meat Quality Traits (돼지 성장 및 육질 형질에 영향하는 종료웅돈의 효과)

  • A submerged biofilm sequencing batch reactor (SBSBR) process, which liquor was internally circulated through sandfilter, was designed, and performances in swine wastewater treatment was evaluated under a condition of no external carbon source addition. Denitrification of NOx-N with loading rate in vertical and slope type of sandfilter was 19% and 3.8%, respectively, showing approximately 5 times difference, and so vertical type sandfilter was chosen for the combination with SBSBR. When the process was operated under 15 days HRT, 105L/hr.m3 of internal circulation rate and 54g/m3.d of NH4-N loading rate, treatment efficiencies of STOC, NH4-N and TN (as NH4-N plus NOx-N) was 75%, 97% and 85%, respectively. By conducting internal circulation through sandfilter, removal performances of TN were enhanced by 14%, and the elevation of nitrogen removal was mainly attributed to occurrence of denitrification in sandfilter. Also, approximately 57% of phosphorus was removed with the conduction of internal circulation through sandfilter, meanwhile phosphorus concentration in final effluent rather increased when the internal circulation was not performed. Therefore, It was quite sure that the continuous internal circulation of liquor through sandfilter could contribute to enhancement of biological nutrient removal. Under 60g/m3.d of NH4-N loading rate, the NH4-N level in final effluent was relatively low and constant(below 20mg/L) and over 80% of nitrogen removal was maintained in spite of loading rate increase up to 100g/m3.d. However, the treatment efficiency of nitrogen was deteriorated with further increase of loading rate. Based on this result, an optimum loading rate of nitrogen for the process would be 100g/m3.d.

Uronic Acid Composition, Block Structure and Some Related Properties of Alginic Acid (4) On Alginic Acid from Myagropsis myagroides Fensholt and Sargassum horneri C. Agardh (알긴산의 화학적 조성 및 그 물성에 관한 연구 (4) 외톨개모자반 및 괭생이모자반의 알긴산)

  • In the previous papers (Kim and Park, 1984 a, b; 1985 a), we have reported on alginic acid from Ecklonia cava and Sargassum ringgoldianum. The seasonal variation in the composition of uronic acids and their block structures of alginic acid from Myagropsis myagroides Fensholt and Sargassum horneri C. Agardh (collected from Iee Chun village on the coast of Ilgwang-myon, Yansan-gun, Kyungnam, Korea, in the period of January to December in 1982) are investigated, and their relationship between the chemical composition and some related properties are discussed in this study. One year average contents of alginic acid were $25.2\%$ in the M. myagroides and $26.5\%$ in the S. horneri, and one year average values of M/G ratios were 1.97 in the M. myagroides and 1.38 in the S. horneri. The value of M. myagroides was largest in the period of December to April, and smallest in May to June and October to November. The value of S. horneri was largest in January and smallest in March to April. The proportion of alternating, M and G block in M. myagroides were $18.4\%,\;40.4\%$, and $41.2\%$, and those in S. horneri $9.8\%,\;33.3\%$ and $56.9\%$, respectively. The higher viscosity showed the value of 45.3 cP in M. myagroides (in November), and 26.0 cP in S. horneri(in January), respectively. Furthermore, the dependence on temperature of M. myagroides alginic acid was also larger in November, that of S. horneri alginic acid in June. Ion exchange ability of M. myagroides alginic acid was highest in November and the exchange amounts were $Pb^{2+}\;4.4,\;Cu^{2+}\;1.8,\;Zn^{2+}\;2.5$ and $Co^{2+}\;2.0\;meq/g$. Na-Alg., and the ability of S. horneri alginic acid was highest in June and the amounts were $Pb^{2+}\;4.5,\;Cu^{2+}\;2.2,\;Zn^{2+}\;2.4$ and $Co^{2+}\;2.1\;meq/g.$ Na-Alg. The affinity with metallic ions appeared higher in order of $Pb^{2+}>Cu^{2+}>Zn^{2+}>Co^{2+}$, and the exchange ability assumed to relate with the block ratio of uronic acid.

Lowering Effects in Plasma Cholesterol and Body Weight by Mycelial Extracts of Two Mushrooms: Agaricus blazai and Lentinus edodes. (아가리쿠스와 표고버섯 균사체 추출물의 혈중롤레스테롤 및 체중감소 효과)

  • The effects of protein-bound polysaccharides (A-PBP and L-PBP) that were extracted from the mycelia of two edible mushrooms, namely Agaricus blazai and Lentinus edodes, on serum cholesterol and body weight were investigated in mice and female volunteers. Six groups of Male Balb/c mice were fed six kinds of diet supplement- solutions composed of L-PBP, A-PBP, chitosan, and other fiber constituents, for 30 days under the normal diet. Ninety female volunteers were also supplemented for 8 weeks with six kinds of capsules including control and five test groups as the same manners (two times a day, 4 capsules). From 12 days after feeding of L-PBP (Group I) and A-PBP (Group II), the weight of mice began to reduce as compared with control, whereas that of Group III fed chitosan was decreased 15 days after feeding. Group W and Group V which were fed mixture of L-PBP, A-PBP, chitosan, and other dietary fiber, were more significant in lowering weight. After 4 weeks of the supplementation in women, their serum LDL-cholesterol level and body weights in Group I and II were reduced, but Croup 111 taken with chitosan capsule showed weaker effect than Group I and II. After 8 weeks, LDL-cholesterol content in the sera of Group I (132.5 mg/dL) and II(131.5 mg/dL) was decreased to ideal level (125.4 and 122.8 mg/dL) for healthy blood vessel. In the case of Group W supplemented with mixture of L-PBP, A-PBP, and chitosan, the weight-reduction effect (11.8%) and hypocholesterolemic effect (11.0%) was most significant, indicating their synergistic action. These data suggested that the weight-controlling and hypolipidemic effect of L-PBP and A-PBP was involved, at least in part, in absorption of cholesterol as their role of dietary fiber, as well as cholesterol metabolism.

Environmental characteristics on habitats of Viola diamantiaca Nakai and its RAPD analysis (금강제비꽃(Viola diamantiaca Nakai) 자생지의 환경특성과 RAPD 분석)

  • This study investigated the environmental factors and conducted a RAPD analysis for a better understanding of the environmental characteristics and regional genetic variation in samples from 18 different areas of Viola diamantiaca. The habitats are mostly located on the slopes of mountains facing north at an altitude ranging from 614 m to 1,462 m above sea level with angles of inclination ranging from 3 degrees to 30 degrees. A total of 268 vascular plant taxa are identified in 35 quadrates of 18 habitats. The importance value of V. diamantiaca is 11.58%, and four highly ranked species, Sasa borealis (5.61%), Meehania urticifolia (5.21%), Ainsliaea acerifolia (3.62%), Pseudostellaria palibiniana (3.60%) are considered to have an affinity with V. diamantiaca in their habitats. The degree of their average species diversity is 1.36, while this metric for their evenness and dominance are 0.89 and 0.07, respectively. The average field capacity of the soil is 25.99%, with organic matter at 17.47%, and the pH is 5.19. The soil texture was confirmed as sandy loam of eleven and loam of seven. The result of the RAPD analysis, among 78 bands amplified with a primer, 64 (84.6%) showed polymorphism. Eighteen populations could be classified into five groups with similarity coefficient values ranging from 0.53 to 0.86. The Mt. Jiri population, which is geographically segregated, shows basal branching within the 18 populations. Five populations, including two in the southern district in Gangwon-do and three in Chungcheongbuk-do, form a distinct clade. Four populations in the central district of Gangwon-do and Mt. Bohyeon in the Gyeongsangbuk-do clade form a sister to the clade containing two populations in Gyeonggi-do and five populations of the northern district in Gangwon-do. The Mt. Gariwang population is placed between the southern district and the central district in the Gangwon-do clades.

Effect of Dietary Fish Oil on Lipid Peroxidation and Antiperoxidative System in Rat Liver and Brain -Sex-related Differences- (어유(魚油)섭취가 흰쥐의 간과 뇌조직의 지질과산화물 형성과 항산화계에 미치는 영향 -성(性)의 차이를 중심으로-)

  • In order to examine the effect of dietary fish oil on lipid peroxide formation and antiperoxidative efficiency in liver and brain, a group of male and female rats weighing about 70 grams were fed for three months, diet containing mackerel oil(MO) at the level of 10% (w/w). Results were compared, according to sex and source of dietary fat, i.e., in addition to MO, perilla oil(PO), soybean oil(SO), rapeseed oil(RO) or beef tallow(BT). Liver lipid peroxide level was significantly higher and levels of ${\alpha}-tocopherol$ and reduced glutathione(GSH) were lower in MO group than in other groups. This phenomenon was less clear in male than in female. Liver GSH level was lower in male, compared to female, but oxidized glutathione (GSSG) level did not vary, depending on either sex or dietary fat source. Brain lipid peroxide and ${\alpha}-tocopherol$ levels were not different among five experimental groups. Activities of liver and brain glutathione peroxidase and superoxide dismutase were not changed by dietary fat source, but glutathione peroxidase activity was higher in female than in male. The present study shows (a) that there is sex-related difference in antiperoxidatiye activity and (b) that fish oil containing $C_{20-22}({\omega}3)$ fatty acids, increases body lipid peroxide level and consumes more of cellular antioxidant, although it has lower total PUFA content than perilla or soybean oils.

Physicochemical Characteristics of Seafood-Added Kimchi during Fermentation and Its Sensory Properties (수산물 첨가 김치의 이화학적 특성 변화 및 관능성)

  • This study was conducted to investigate the physicochemical characteristics of seafood added kimchi (SAK) during fermentation and its sensory properties. Korean cabbage kimchi (KCK) and four different SAKs were prepared and stored at $5^{\circ}C$ for eight weeks. The SAKs contained pre-treated octopus, squid, abalone, and webfoot octopus added at 12% (w/w) to the brined Korean cabbage. The fermentation patterns of SAKs were similar to those of KCK, indicating that the SAKs followed a typical fermentation process. Comparison of the physicochemical characteristics of SAKs with KCK revealed that the pH and acidity of SAKs was higher. The maximum concentrations of Lactobacillus spp. and Leuconostoc spp. for SAKs ranged from 8.31~8.85 and 7.60~8.14 log CFU/mL, respectively, which were higher than those for KCK. Therefore, the production of organic acids by microorganisms was greater in SAKs, which explained the higher acidities of the SAKs. Nitrogenous compounds hydrolyzed during fermentation, as well as reducing sugars and other nutritious compounds in SAKs might provide a good medium for lactic acid bacterial growth. Sensory evaluation was carried out using optimally ripened kimchi (pH $4.3{\pm}0.1$, acidity $0.7{\pm}0.1$), and the scores for sour taste, sour smell, and carbonated taste were significantly lower for SAKs than KCK. In the preference test, texture and overall acceptability were significantly higher for SAKs than KCK. Significant differences were not observed among SAKs upon subjective and preference evaluations. In conclusion, the fermentation patterns of SAKs were normal, regardless of seafood sources, and their sensory characteristics were comparable to or superior than those of KCK due to free amino acids, nitrogenous compounds produced during the fermentation, and reducing sugar present in the seafood.

THE EFFECT OF FERMENTED MILK ON VIABLE CELL COUNT AND BIOFILM FORMATION OF STREPTOCOCCUS MUTANS (유산균 발효유가 Streptococcus mutans의 생균수 및 biofilm 형성에 미치는 영향)

  • Lactic acid bacteria worked positively on gastrointestinal tract and oral environment. So I selected commercial five fermented milks and milk, and then I evaluated their effect of growth inhibition and biofilm formation of cariogenic bacteria, Streptococcus mutans. And also calculated the acidity, buffering capacity, concentration of Ca and P ion and pH change of those drinks. After adding S. mutans to fermented milks viable cell count of S. mutans in milk was not statistically different but those in all fermented milks were decreased as concentration of fermented milk increased. When I measured the amount of formed biofilm in 10% fermented milks and milk with S. mutans and compared with those without S. mutans, the amount was decreased in Active GG and Bulgaris while being increased in Tootee, Ace and milk(P<0.05). The fermented milk with the lowest pH value was E5(3.48${\pm}$0.01), and the highest was Bulgaris(4.19${\pm}$0.02). pH change of the fermented milks and milk with S. mutans was measured. The highest acid producing fermented milk was Bulgaris, and followed by Active GG, Ace, Tootee, E5, Milk. These results indicated that fermented milks had caries activity due to the value of initial acidity and acid producing capacity. But, concentrated fermented milks had the inhibitory effect against S. mutans, and also had high volume of Ca and P ion that protected teeth. So I suggest that they have positive effect on teeth.

Processing Conditions and Quality Stability of Frozen Seasoned Sardine Meat during Frozen Storage (냉동 정어리 조미육의 가공 및 저장중의 품질안정성)

  • Seasoned sardine meat was prepared to extend the use of sardine for human consumption, and processing conditions and storage stability of frozen seasoned sardine meat were studied during storage at $-20^{\circ}C$. The fish was beheaded, gutted and cleaned in a washing tank. The washed fish was then put through a belt-drum type meat separator which separates the flesh iron the bone and skin. Mechanically deboned fish meat was mixed with $20.6\%$ emulsion curd, $0.5\%$ table salt, $2.0\%$ sugar, $0.4\%$ sodium bicarbonate, $0.2\%$ polyphosphate, $0.1\%$ monosodium glutamate, $0.3\%$ onion powder, $0.1\%$ garlic powder, $0.1\%$ ginger powder, $3.0\%$ soybean protein and $0.1\%$. In sodium erythorbate. This seasoned sardine meat was frozen with contact freezer, packed in a carton box and then stored at $-20^{\circ}C$. The pH, volatile basic nitrogen, viable cell counts, peroxide value, carbonyl value, thiobarbituric acid value, taste compounds, fatty acid composition, salt extractable nitrogen, drip, texture, and color values of the products were determined during frozen storage. The results showed that lipid content in products could be controlled by using emulsion curd, and flavor and texture could be improved by adding spices and soybean protein, and lipid oxidation could be retarded by $0.1\%$ sodium erythorbate. Judging from the results of chemical experiments and sensory evaluation, the products can be preserved in a good quality for 120 days during frozen storage.

Phylogentic Position, Pigment Content and Optimal Growth Condition of the Unicellular Hydrogen-Producing Cyanobacterial Strains from Korean Coasts (한국 연안산 단세포성 수소생산 남세균 종주들의 분류계통, 색소함량 및 최적성장 환경)

  • To set up unicellular cyanobacterial strains with photo-biological $H_2$ production potential, live samples were repeatedly collected from 68 stations in the coastal zone of Korea for the four years since 2005. Among 77 cyanobacterial strains established six (KNU strains, CB-MAL002, 026, 031, 054, 055 and 058) were finally chosen as the excellent strains for $H_2$ production with $H_2$ accumulation over 0.15 mL $H_2\;mL^{-1}$ under general basic $H_2$ production conditions as well as positive $H_2$ production for more than 60 hr. To explore optimum procedures for higher $H_2$ production efficiency of the six cyanobacterial strains, the inter-strain differences in the growth rate under the gradients of water temperature and salinity were investigated. The maximum daily growth rates of the six strains ranged from 1.78 to 2.08, and all of them exhibited $N_2-fixation$ ability. Based on the similarity of the 16S rRNA sequences, all the test strains were quite close to Cyanothece sp. ATCC51142 (99%). The six strains, however, were grouped into separate clades from strain ATCC51142 in the molecular phylogeny diagram. Chlorophyll- a content was 3.4~7.8% of the total dried weight, and the phycoerythrin and phycocyanin contents were half of those in the Atlantic strain, Synechococcus sp. Miami BG03511. The growth of the six strains was significantly suppressed at temperatures above the optimal range, $30{\sim}35^{\circ}C$, to be nearly stopped at $40^{\circ}C$. The growth was not inhibited by high salinities of 30 psu salinity in all the strains while strain CB055 maintained its high growth rate at low salinities down to 15 psu. The euryhaline strains like CB055 might support massive biotechnological cultivation systems using natural basal seawater in temperate latitudes. base seawater. The biological and ecophysiological characteristics of the test strains may contribute to designing the optimal procedures for photo-biological $H_2$ production by unicellular cyanobacteria.
