Geological Occurrence and Mineralogy of Pyrophyllite Deposits in the Jinhae Area (진해 납석광상의 산상과 광물학적 특성)

  • The pyrophyllite deposits located in Jinhae area have been studied through field observations and laboratory works including the X-ray diffraction (XRD), X-ray fluorescence (XRF), Electron probe microanalyzer (EPMA) and Inductively Coupled Plasma (ICP). The pyrophyllite deposits consist of mainly illite, dickite, pyrophyllite, diaspore, chlorite, pyrite and copiapite. According to the mineral assemblages, geological occurrences and alteration modes, the altered rocks can be classified into four types: Type A; quartz with silicifictaion, Type B; quartz + illite with illitization, Type C; quartz + dickite + illite with kaolin alteration, Type D; pyrophyllite + illite + dickite + diaspore with pyrophyllite alteraion. Rocks in Type A, which is generated by silicifictaion, have high $SiO_2$ contents more than 90 wt% and distinctive equigranular textures with microcrtstalline quartz. The pyrophyllites from the study area belong to 2M polytype. The host rocks of the pyrophyllite ore in this mine are rhyolitic rock, andecitic tuff and volcanic breccia. The alteration products seem to be controlled by the different lithology of the host rocks. The hydrothermal solution formed the deposits would be inferred to the acidic and have relatively high ionic activity of hydrogen and silica judging from alteration mineral assemblage. Pyrophyllite alteraion zone is generated by highest temperature condition of all alteration zone.

Isolation and Identification of Halotolerant Bacillus sp. SJ-10 and Characterization of Its Extracellular Protease (세포외 Protease를 생산하는 내염성 Bacillus sp. SJ-10 균주의 분리 동정 및 효소 특성)

  • A bacterium producing the halotolerant extracellular protease was isolated from squid jeotgal, and was identified as Bacillus sp. SJ-10 based on morphological, physiological and biochemical characteristics, as well as phylogenetic analysis using 16S rRNA gene sequence. The strain grew at $20^{\circ}C\sim55^{\circ}C$, pH 5~8, and 0%~14% NaCl and optimal growth conditions were $35{\pm}5^{\circ}C$, pH 7, and 5% NaCl. The major cellular fatty acids were anteiso-$C_{15:0}$, anteiso-$C_{17:0}$, and $C_{16:0}$ DNA G+C content was 50.58 mol% and menaquinone consisted of MK-7 Phylogenic analysis based on the 16S rRNA gene sequence indicated that SJ-10T belongs to the genus Bacillus. About 40 kDa of the salt-tolerant protease was purified by 40% ammonium sulfate saturation and Mono Q column chromatography. The optimal activity of the protease was pH 8 and stable at pH 5~10. The optimum temperature and NaCl concentration were $35{\pm}5^{\circ}C$ and $5{\pm}1%$, respectively.

Characterization of a Small Cryptic Plasmid from Pseudomonas nitroreducens Strain TX1 (Pseudomonas nitroreducens TX1에 존재하는 작은 플라스미드의 특성 규명)

  • Pseudomonas nitroreducens TX1 was isolated from a rice field drainage in Taiwan. The bacterium is of special interest because of its capability to use a group of nonionic surfactants such as alkylphenol polyethoxylates even at high concentrations as a sole carbon source. In this study, a small cryptic circular plasmid, pTX1, was characterized from P. nitroreducens TX1. It is 2,286 bp in length with a GC content of 63.3% and harbors three open reading frames, $Rep_{pTX1}$ and functionally unidentified ORF1 and ORF2. The predicted $rep_{pTX1}$ gene product is homologous to Rep proteins of plasmids belonging to the pC194/pUB110 family, which is predominantly found in Gram-positive bacteria and is known to replicate by the rolling-circle mechanism. The copy number of pTX1 was estimated to be about 150 in each cell. Based on the genetic fingerprints and comparison with other plasmids, it is concluded that pTX1 replicates by a rolling circle mechanism which is rarely found for Pseudomonas plasmids.

Effect of the Beet Addition on the Quality of American Preferred Kimchi during Fermentation (비트 첨가가 미국인 선호 김치의 숙성 중 품질에 미치는 영향)

  • To improve the Quality of American preferred kimchi (APK), the APK added different ratios of beets were prepared and the Qualities of those kimchi were investigated by measuring the changes of physicochemical, microbiological and sensory characteristics during fermentation at $5^{\circ}C$. The pH of APK added beet (APKB) decreased as the amount of beet increased while total acidity gradually increased. The reducing sugar content was the highest in APKB added $3\%$ beet at early stage of fermentation. The number of Lactobacillus sp. and Leuconostoc sp. were the highest in $3\%$ beet added group during fermentation. In a texture experiment, the hardness of Korean standard kimchi used as control group showed rapidly decrease while the hardness of APKB were decreased slowly as fermentation proceeded. In Hunter's color values of APKB, lightness and redness increased as the amount of beet increased while yellowness decreased. Sensory scores of overall acceptance, taste, texture and appearance evaluated by Americans as sensory panels were the highest in APKB added $1\%$ beet. Americans also preferred fresh kimchi to optimum ripened kimchi. Therefore it was suggested that the American preferred kimchi added $1\%$ beet was the best group among American preferred kimchi groups.

Effects of Electron Beam Irradiation on the Quality of Kochujang Powder (Electron Beam 조사가 고추장 분말의 품질에 미치는 영향)

  • Electron beam (EB) irradiation was applied to study microbial decontamination effects for kochujang powder by determining their microbiological and physicochemical qualities over gamma ray (GR) irradiation. The samples showed a high microbial population, such as 105~106 CFU/g of total aerobic bacteria, negative of yeasts & molds and coliforms. Total bacterial counts were decreased by 1~2 log cycles with EB irradiation at 5~7.5 kGy, and 10 kGy irradiation was enough to improve the microbiological quality by reducing populations to below 104CFU/g, which was similar to gamma energy. Such doses were effective for controlling the microbial growth in stored samples during storage for 4 months at room temperature. Decimal reduction doses (D10 value) on initial bacterial populations were 2.88~3.02 kGy in EB and 3.57~3.59 kGy in GR, which were influenced by initial populations and energy types applied. Based upon the above results, 7.5~10 kGy irradiation caused negligible changes in Hunter's color, capsaicin, fatty acid composition and organoleptic qualities. Considering the quality changes resulting from subsequent storage, such as a decrease in capsanthin content and an increase in TBA value in the samples, it is recommendable to irradiate kochujang powder at 7.5~10 kGy when used for food processing.

Formation of Brushite (CaHPO4 · 2H2O) in the Oyster Extracts (굴 엑기스에서 결정물질 Brushite(CaHPO4 · 2H2O)의 생성)

  • Small yellowish brown crystals were found in some concentrated oyster extracts which prepared by heating with the drip and washed water of boiled oyster in oyster processing factories. We collected those crystals by filtering the oyster extracts and analysed X ray diffraction (XRD), scanning electron microscopy (SEM) and minerals by ICP. Those were composed of moisture (8.1%), organic materials (21.6%) and inorganic materials (70.2%). Those major inorganic materials were determined as Ca (53.8%) and P (43.8%), from the ICP and elucidated those crystal as brushite $(CaHPO_4{\cdot}2H_2O)$ by XRD and SEM. Organic materials contained crude protein (33.7%) consisting of 7 amino acids. It is presumed that brushite may be formed by the reaction of excess Ca with P in the oyster extracts during concentrating process under higher temperature.

Development of Yeast Leavened Pan Bread Using Commercial Doenjangs (Korean Soybean Pastes): 1. Physicochemical Properties of Doenjang and Physical Properties of Bread Added with Doenjang (시판 된장을 이용한 식빵 제조: 1. 된장의 이화학적 특성 및 된장을 첨가한 식빵의 물리적 특성)

  • This study was carried out to develope yeast leavened pan bread using the commercial Doenjangs (Korean soybean pastes). Physicochemical properties of the Doenjang products were measured such as aminonitrogen, pH, titratable acidity and salinity, reducing sugar, total free sugar, total organic acid, PDI (protein dispersibility index) and color. Seven products of Doenjang were freeze-dried and powdered to be used in bread formula at the levels of 2.5, 5.0, 7.5 and 10.0%. When the Doenjang powder was added up to 5.0%, the ovenspring during baking remarkably increased resulting in increased loaf volume. However, with Doenjang powder more than 7.5%, the loaf volume became smaller than the control as the dough expansion and ovenspring decreased. Thus when Doenjang was added to bread dough, the loaf volume was highly correlated with ovenspring (r=0.92) but it was not with dough expansion during 1st fermentation (r=0.56). The browning color of bread crust and crumb became deeper with decrease in L value as the addition of Doenjang powder increased. From the mechanical texture measurements of bread crumb, hardness, gumminess and chewiness decreased with addition of Doenjang powders. Regardless of the kinds of Doenjang, the Doenjang powders could be added into the bread dough up to 5.0%, improving the loaf volume and texture of yeast leavened pan bread and demonstrating the possibility of producing a functional bread using the Doenjang powders.

A Study on the Quality Characteristic of Power Bio-Fuel Oil for Alternative Fuel oil (중유 대체연료로서 발전용 바이오중유의 품질특성 연구)

  • In these days, many countries carry out many renewable energy policies to increase the renewable energy portion and to reduce the GHG(Green House Gas). In Korea, RPS(Renewable Portfolio Standards) focused on over 500MW power producers is conducting. And they are using the bio-fuel oil to meet their RPS quota. The oil is a mixture of animal and vegetable fat or fatty acid ester of them and should satisfy some specifications to use the power generation such as viscosity, pour point, acid number. Depends on the raw materials, quality characteristics of power bio-fuel oil are changed. By adding the power bio-fuel oil, pour point, density, sulfur content and viscosity are decreased and acid number, iodine number, oxygen content are increased. In this study, we test the quality characteristic of power bio-fuel oil and the property changes by the blending ratio of power bio-fuel oil & conventional fuel oil.

A Study on Paralytic Shellfish Poison of Sea Mussel, Mytilus edulis -Food Poisoning Accident in Gamchun Bay, Pusan, Korea, 1986- (진주담치의 마비성독에 관한 연구 -1986년 부산 감천만 중독사고를 중심으로-)

  • At various times and places all over the world men have become ill and some have died after eating shellfish that were intoxicated with paralytic shellfish poison(PSP) caused by Protogonyaulax spp. In late March, 1986, two persons were dead by ingesting wild sea mussels, Mytilur edulis, grown at bottom of an anchored waste ship to be dismantled at Gamchun Bay, Pusan, Korea. The samples were collected from the bottom of the ship during April $1\~April$ 8 of the year to find the cause of the food poisoning accident. The toxicity was estimated by bioassay with ICR male mouse, while the toxins were extracted and characterized. The toxins were extracted with acidified $80\%$ ethanol. The extract was defatted three times with dichloromethane, treated with activated charcoal, and then purified by chromatography on Bio-Gel P-2 and Bio-Rex 70. The toxic fractions obtained were analysed by cellulose acetate membrane electrophoresis, thin layer chromatography and high performance liquid chromatogaphy. The range and the average of PSP-toxicity of the samples were $132\~295\;MU/g$, 203 MU/g respectively. The amount of PSP was $26.4\~58.9{\mu}g/g$ of whole meat in range and $40.6{\mu}g/g$ in average. The toxicity of the digestive gland of the samples was 9 times higher than that of edible meat (except digestive gland) as $439\~979MU/g$, and it was about $70\%$ in total toxin. The compositional analytical results of the paralytic shellfish toxin, Gonyautoxin $1\~4$ were the major part of the PSP and Saxitoxin and neosaxitoxin were detected as the minor component. It was concluded that the food poisoning accident was caused not by Saxitoxins but by Gonyautoxins.

The Cycling Performance of Graphite Electrode Coated with Tin Oxide for Lithium Ion Battery (리튬이온전지용 주석산화물이 도포된 흑연전극의 싸이클 성능)

  • Tin oxide was coated on graphite particle by sol-gel method and an electrode with this material having microcrystalline structure for lithium ion battery was obtained by heat treatment in the range $400-600^{\circ}C$. The content of tin oxide was controlled within the range of $2.25wt\%\~11.1wt\%$. The discharge capacity increased with the content of tin oxide and also initial irreversible capacity increased. The discharge capacity of tin oxide electrode showed more than 350 mAh/g at the initial cycle and 300 mAh/g after the 30th cycle in propylene carbonate(PC) based electrolyte whereas graphite electrode without surface modification showed 140 mAh/g. When the charge and discharge rate was changed from C/5 to C/2, The discharge capacity of tin oxide and graphite electrode showed $92\%\;and\;77\%$ of initial capacity, respectively. It has been considered that such an enhancement of electrode characteristics was caused because lithium $oxide(Li_2O)$ passive film formed from the reaction between tin oxide and lithium ion prevented the exfoliation of graphite electrode and also reduced tin enhanced the electrical conduction between graphite particles to improve the current distribution of electrode.
