Rapid Identification of Cow and Goat Milk in Milk Products Using a Duplex PCR Technique (Duplex PCR을 이용한 유제품 안에 있는 산양유와 우유의 신속한 동정에 대한 연구)

  • A duplex PCR technique was applied for specific identification of cow and goat milk in milk products by using primers targeting the mitochondrial 12S rRNA gene. Duplex PCR using primers specific for cow and goat generated specific fragments of 223bp and 326bp from cow and goat milk DNA, respectively. Duplex PCR was applied to 15 milk products purchased from the market to verify label statements. The labeling statements of four market milk products, three yoghurt products, and one whole milk powder product were confirmed in the duplex PCR. The labeling statements of five of seven infant milk powder products were also confirmed by duplex PCR but the other two products were shown to be contaminated with either cow or goat milk. The proposed duplex PCR provides a rapid and sensitive approach to detection of as little as 0.1% cow milk in goat milk and one-step detection of cow or goat milk in milk products.

Re-Cloning by Somatic Cell Nuclear Transfer from a Cloned Korean Native Goat (복제 산양(진순이)의 체세포 핵이식에 의한 Re-Cloning에 관한 연구)

  • The present study was conducted to examine some factors affecting in vitro development and fecundity of embryos recloned with somatic cell nuclear transfer (SCNT). Fibroblast cells retrieved from the ear of a 3-week-old, cloned Korean goat (Jinsoonny) were used as karyoplast donors and serum-starvation was conducted in tissue culture medium (TCM)-199 supplemented with 0.5% FBS. Recipient oocytes were surgically collected by flushing the oviducts 35 h after hCG injection following FSH priming. The zonae pellucidae of the oocytes were partially perforated with a laser drill and a donor cell was transferred into an enucleated oocyte. The couplets were electrically fused and activated by ionomycin (5 min) and 6-DMAP (4 h). The reconstructed embryos were cultured in mSOF medium containing 0.8% BSA at $39^{\circ}C$ in an atmosphere of 5% $CO_2$, 5% $%O_2$, 90% $N_2$ for 12 to 15 h. Re-cloned embryos (2- to 4-cell stages) were surgically transferred into the oviducts of the recipients and pregnancy was subsequently diagnosed by progesterone assay and ultrasound on Days 21 and 63 of pregnancy. The fusion rate following 1st fusion pulse was higher (p<0.05) in 2nd cloning (65.9%) compared to 1st cloning (51.0%), but it was not different in the other groups. The rate of cleavage after fusion was significantly higher (p<0.05) in 1st (77.7%) than in 2nd cloning (56.0%). A total of 175 re-cloned embryos were transferred into 28 recipients. On day 21 and 60 after transfer, 11 (39.3%) and 4 recipients (17.4%) were pregnancy, respectively. In comparison of pregnancy rate by estrous synchronization, a total of 66 and 109 re-cloned embryos were transferred into 11 recipients in natural estrus and 17 recipients in induced estrus, respectively. Five (45.4%) and 2 recipients (18.2%) in natural estrus were pregnant on days 21 and 63 while 6 (35.3%) and 2 (11.8%) recipients in induced estrus were pregnant, respectively. These results show that recloning of goat can be achieved by SCNT and estrous synchronization between donor and recipient animals may be one of the major factors affecting success rate.

Current status of population size and habitat selection of the long-tailed goral(Naemorhedus caudatus) in Seoraksan National Park (설악산국립공원 멸종위기 산양(Naemorhedus caudatus) 개체군 크기와 서식지 이용 현황)

  • This study was conducted investigate population size and habitat use for the conservation and management of the endangered long-tailed goral in the Seoraksan National Park using feces and camera trap during 2010 to 2014 (track survey, camera trap). As a result of feces tracking and camera trap, its population size was estimated as 160 (camera trap)~251 (feces) individuals in the Seoraksan National Park. The goral prefer $35^{\circ}{\sim}60^{\circ}$ (slope), 600~700m (elevation), NE (aspect), 0~50m (distance to stream), 300~600m (distance to road) and bread-leaved forest (forest type) according to field tracking of fecal. Based on field camera trap, we estimated the age classes of goral populations and activity of gorals during day-time (07-18 time, 56.5%) and night-time (18-07 time, 43.5%). Such analyses of population size and habitat use of the goral could be applied as important fundamental data for conservation of gorals and management of their habitats.

Optimization of Goat Milk Yoghurt Preparation Conditions by Response Surface Methodology (반응표면 분석에 의한 산양유 요구르트의 제조조건 최적화)

  • This study was carried out to establish the optimal preparation conditions of yoghurt made from goat milk with reduced goaty flavor by adding citrus concentrate and flavor. A central composite design was applied to investigate the effects of citrus concentrate ratio (0.5, 1.0, 1.5, 2.0, 2.5%), citrus flavor ratio (0.01, 0.02, 0.03, 0.04, 0.05%) and fructose ratio (3, 4, 5, 6, 7%). The physico-chemical and sensory characteristics of the sixteen yoghurt samples were compared. The addition of citrus concentrate had a significant (p<0.01) effect on the pH, $a^*,\;and\;b^*$ values. Regarding organoleptic properties, the addition of citrus concentrate had a significant (p<0.01) effect on color, and fructose had an effect on overall palatability. The maximum value of organoleptic goaty flavor was 2.35, more than double the minimum value. The optimum conditions predicted for minimizing goaty flavor of the yoghurt were 1.44% citrus concentrate, 0.0357% citrus flavor, and 6.91% fructose.

Characteristics of Exopolysaccharide Produced in Goat Milk Yogurt Cultured with Streptococcus thermophilus LFG Isolated from Kefir (Kefir에서 분리한 Streptococcus thermophilus LFG를 배양한 산양유 발효물에서 분리된 다당체의 특성)

  • This study was carried out to investigate the properties of crude exopolysaccaride (CEPS) produced by Streptococcus thermophilus LFG in goat milk. The yields of CEPS from yogurt cultured with Str. thermophilus LFG were greater at higher temperatures $(40-45^{\circ}C)$ than at lower temperatures $(30-35^{\circ}C)$. Goat milk yogurt had lower viscosity values than cow milk yogurt. However, the CEPS yield was higher in goat milk yogurt than in cow milk yogurt. The yields of CEPS from yogurt were also higher in cultured milk containing 3% glucose (14-21%), and 3% sucrose (4-16%) relative to the control yogurt. Antioxidant activities were higher in goat milk yogurt supernatant (21%) and its CEPS (28%) than cow milk yogurt supernatant (11%) and its CEPS (24%). The amino acid contents of CEPS were higher in yogurt using goat milk than that using cow milk. The CEPS extracted from goat milk yogurt produced by Str. thermophilus LFG consists of carbohydrate (37% w/w) and protein (63% w/w). The CEPS consisted of monosaccharides such as glucose 56.45% (w/w), galactose 42.35% (w/w), galactosamine 1.37% (w/w), glucosamine 1.09% (w/w) and fucose 0.27% (w/w).

  • 산양에서 수혈후 적혈구과다증을 지속시킬 수 있는지 또 해당중간생산물들이 변하는 가에 관하여 연구한바 아래의 결론을 얻었다. 1. 산양의 상동적혈구는 순환혈액에서 곧 사라졌다. 이와 같은 현상은 수혈적혈구가 지속적으로 적혈구과다증을 일으킬 수 없다는 것을 시사한다. 2. 적혈구용적과 혈색소농도는 수혈후 유의성 있게 증가되었으나 해당중간생산물인 GSH 2,3-DPG 및 ATP는 유의성 있게 변하지 아니하였다.

  • 한국 재래산양에 대한 L. monocytogenes균의 감염원을 조사하기 위하여 총 459개의 재래산양 분변, 비즙, 장기, 사료 및 토양재료로부터 균분리를 시도하였으며, 분리균의 항균성 물질에 대한 감수성 및 인공 감염시의 L. monocytogenes의 침입장기를 조사하였던 결과는 다음과 같다. L. monocytogenes는 분변에서만 분리되었으며 (분리율 8.2%) 분변이 본균의 주요 감염원으로 인정되었다. 가검재료로부터 L. monocytogenes의 분리방법으로는 유제화 재료를 혈액배지로써 37에서 24~48시간 동안 호기배양함이 우수하였다. 분리된 L. monocytogenes 8주 중 100%의 균주가 amikacin, colistin, tanamycin, neomycin에 내성을 나타내었으며, clinmycin, gentamicin, lincomycin, methicillin 및 refampin에 87.5%, ampicillin및 oles-ndomycin에 75.0%, Chloramphenico에 62.5%, tetracycline에 50.0% 및 cephalot hin에 37.5%의 균주가 내성이었다. Nitrofurantoin에는 전 균주가 감수성을 나타내었다. L. monocytogenes를 재래산양에 경구 및 정맥내로 접종하였을때 패혈증이 인정되었으며 대부분의 실질장기가 이 균의 침입부위인 것으로 관찰되었다.

Ultrasonography of the Kidney and Urinary Bladder in Male Korean Native Goat (웅성 한국재래산양에서 신장 및 방광의 초음파검사법)

  • The purpose of this study was to make fundamental ultrasonographic observations of kidney and urinary bladder in Korean native goat. The position, dimensions, and structure of the kidneys and bladders In 7 male Korean native goats were determinded by use of ultrasonography. A 3.5%.MHz linear transducer was used. All examinations were performed on goats in lateral recumbency under sedation. The left kidney was 4.2 to 5.9 cm long, 2.5 to 3.6 cm wide, and 2.4 to 3.2 cm deep. Diameter of the parenchyma and renal sinus of the left kidney ranged between 0.7 and 1.3 cm and 0.7 and 1.4 cm, respectively. Circumferences of the medullary Pyramids varied between 1.3 and 1.9 cm. Similar ultrasonic measurements were obtained for the right kidney. The diameter of the urinary bladder varied between 2.1 and 5.4 cm in the goats. It was concluded that the ultrasonographic findings described in this study can be used as references for diagnosis of morphologic changes in the kidney and urinary bladder of Korean native goat.

The Study of Mountain Ginseng-added High Fat Diet on Anti-Apoptosis of Skeletal Muscle (산양삼 첨가 고지방식이가 골격근의 항세포사멸에 대한 작용 연구)

  • The purpose of this study was to investigate the anti-apoptosis-related protein expression in skeletal muscle of rats with different amount of mountain ginseng (MG) added high-fat diet fed. Twenty-four Sprague Dawley male rats were divided into the high-fat diet control group (CON), 0.5% of MG added diet group (MG1), and 1.0% of MG added diet group (MG2) with eight rats each. The P53, anti-apoptotic protein, was significantly lower in MG2 than CON and MG1. The bcl-2 and bcl-xl, however, were not significantly different from MG1 but from CON. The caspase-9 and -3, were significantly lower in MG groups than CON. In addition, it was dramatically lower in MG2. These results suggested that MG addition to the high-fat diet suppressed p53 protein expression and enhanced anti-apoptototic protein expression. MG may be a positive effects on health as a medicinal plant.
