Detection of single-nucleotide polymorphism in RPB2 of Wolfiporia hoelen strains and assessment of its applicability for strain breeding (복령 균주의 RPB2 유전자 내 단일염기다형성 및 육종 활용성 분석)

  • The demand for novel strains has been rising in the domestic market to increase the production of sclerotia from Wolfiporia hoelen. To improve strain breeding efficiency, we investigated whether single-nucleotide polymorphisms (SNPs) in the RNA polymerase II subunit (RPB2) gene, which may be linked to the mating type locus, are useful for distinguishing monokaryons from dikaryons in Korean W. hoelen strains. We designed a specific primer set to efficiently amplify a region of RPB2 using PCR with the genomic DNA of 12 cultivated strains and 31 wild strains of W. hoelen collected from Korea. Nucleotide sequences of the PCR-amplified RPB2 genes were determined and analyzed for the presence of SNPs among the 43 W. hoelen strains. Previously reported SNP loci were detected in the RPB2 gene of all W. hoelen strains tested. However, these previously reported SNP loci could not be applied to differentiate monokaryons from dikaryons in approximately one-third of Korean wild strains with homozygous genotypes. Three additional SNPs in the RPB2 gene, which may improve the ability to distinguish monokaryons from dikaryons, were identified by searching through the multiple sequence alignments of the 43 W. hoelen strains. The applicability of these three novel SNPs, together with the previously known SNPs, in the RPB2 gene to W. hoelen strain breeding was verified by examining the hybrid strains and their parental strains.

  • This study is a precedent study for deriving transfer function model between growing stock and forest management policies. Its goal is to solve the multicollinearity between forest works inducing growing stock changes through principal component analysis using annual time series data from 1997 to 2008. As the results, the total explanatory power showed 91.4% on the summarized 3 principal components. They were renamed 'good forest management' 'pest & insets management' 'forest fires' for conceptualization on the derived each component.

Extraction of the Tree Regions in Forest Areas Using LIDAR Data and Ortho-image (라이다 자료와 정사영상을 이용한 산림지역의 수목영역추출)

  • Due to the increased interest in global warming, interest in forest resources aimed towards reducing greenhouse gases have subsequently increased. Thus far, data related to forest resources have been obtained, through the employment of aerial photographs or satellite images, by means of plotting. However, the use of imaging data is disadvantageous; merely, due to the fact that recorded measurements such as the height of trees, in dense forest areas, lack accuracy. Within such context, the authors of this study have presented a method of data processing in which an individual tree is isolated within forested areas through the use of LIDAR data and ortho-images. Such isolation resulted in the provision of more efficient and accurate data in regards to the height of trees. As for the data processing of LIDAR, the authors have generated a normalized digital surface model to extract tree points via local maxima filtering, and have additionally, with motives to extract forest areas, applied object oriented image classifications to the processing of data using ortho-images. The final tree point was then given a figure derived from the combination of LIDAR and ortho-images results. Based from an experiment conducted in the Yongin area, the authors have analyzed the merits and demerits of methods that either employ LIDAR data or ortho-images and have thereby obtained information of individual trees within forested areas by combining the two data; thus verifying the efficiency of the above presented method.

  • 전 세계적으로 산불로 인한 산림 자원의 손실로 인한 피해는 막대하다. 산불로 인한 인명 및 재산 피해는 증가하는 추세이다. 또한 산불로 인한 산림 자원의 손실은 생태계에 회복되기 힘든 상처를 남긴다. 이런 산불을 분석하고 예방하기 위해 다양한 연구가 진행되고 있으나, 산불의 발생을 예측 할 수 있는 연구는 부족한 실정이다. 본 논문은 미래 예측 연구에 많이 사용되는 마코프 체인을 이용하여 산불을 예측 할 수 있는 산불 예측 모델링을 제안 하고 그 기대 효과에 대해 논의한다.

Desiccation Tolerance and Storage Behavior of Sarcandra glabra Seeds for Use in ex-situ Conservation (현지외 보존을 위한 죽절초 종자의 건조 내성과 저장성 평가)

  • This study sought to determine the desiccation tolerance and storage behavior of Sarcandra glabra seeds to assist in the establishment of an ex-situ conservation strategy for the species. The basic germination characteristics of S. glabra seeds were investigated. Subsequently, the seeds were dried to various desiccation levels to allow for analysis of their germination characteristics and seed integrity. In addition, the seeds were subjected to germination tests after being stored for different durations. The initial seed moisture content following the removal of the fleshy fruit was 32.8%, and the fresh seeds maintained their seed viability even after a decrease in the seed moisture content to 4.8%, while the germination percentage was 81.0%. The germination percentage of the seeds decreased to 55.0% and 30.0% when they were desiccated to moisture contents of 2.3% and 2.2%, respectively. By contrast, when the stored seeds were desiccated, they maintained their seed viability and vigor at a moisture content of 18.6%; however, when the moisture content decreased below 5.0%, both the seed viability and vigor significantly declined and the germination percentage fell below 20.0%. Furthermore, analysis of the seed leachate revealed that the decrease in the seed viability and vigor was associated with both increased electrical conductivity and increased concentrations of inorganic compounds such as potassium and calcium. In conclusion, the findings of this study show S. glabra seeds to possess desiccation tolerance that falls somewhere between the recalcitrant and intermediate seed types, with the observed variations depending on the degree of deterioration.

  • Forest lodge are divided into forest resources, auxiliary facilities, and users, and are constantly being used along with well-being culture. In addition, attention to the safety of users is also required, and this study aims to study how users evacuate within a short time (golden time) in situations of natural disasters that may occur in forests. In order to search for the current location of the user and find the best evacuation route, 3D scans of the entire forest lodge(forest resources, auxiliary facilities, etc.) are performed, and the optimal trajectory to the evacuation site is found through recognition of the current location. It is believed that it is possible to provide a quick evacuation guide through a mobile device with gps.

Change Analysis of Aboveground Forest Carbon Stocks According to the Land Cover Change Using Multi-Temporal Landsat TM Images and Machine Learning Algorithms (다시기 Landsat TM 영상과 기계학습을 이용한 토지피복변화에 따른 산림탄소저장량 변화 분석)

  • The acceleration of global warming has required better understanding of carbon cycles over local and regional areas such as the Korean peninsula. Since forests serve as a carbon sink, which stores a large amount of terrestrial carbon, there has been a demand to accurately estimate such forest carbon sequestration. In Korea, the National Forest Inventory(NFI) has been used to estimate the forest carbon stocks based on the amount of growing stocks per hectare measured at sampled location. However, as such data are based on point(i.e., plot) measurements, it is difficult to identify spatial distribution of forest carbon stocks. This study focuses on urban areas, which have limited number of NFI samples and have shown rapid land cover change, to estimate grid-based forest carbon stocks based on UNFCCC Approach 3 and Tier 3. Land cover change and forest carbon stocks were estimated using Landsat 5 TM data acquired in 1991, 1992, 2010, and 2011, high resolution airborne images, and the 3rd, 5th~6th NFI data. Machine learning techniques(i.e., random forest and support vector machines/regression) were used for land cover change classification and forest carbon stock estimation. Forest carbon stocks were estimated using reflectance, band ratios, vegetation indices, and topographical indices. Results showed that 33.23tonC/ha of carbon was sequestrated on the unchanged forest areas between 1991 and 2010, while 36.83 tonC/ha of carbon was sequestrated on the areas changed from other land-use types to forests. A total of 7.35 tonC/ha of carbon was released on the areas changed from forests to other land-use types. This study was a good chance to understand the quantitative forest carbon stock change according to the land cover change. Moreover the result of this study can contribute to the effective forest management.

Vegetation Structure and Soil Condition of Acer okamotoanum Communities in Ulleung Island (울릉도 우산고로쇠나무 군락의 식생구조와 토양환경)

  • Vegetation structure and soil condition were analyzed to provide information for effective management of Acer okamotoanum community. Importance value of A. okamotoanum in upper layer was highest as 120.7, and that of Camellia japonica in middle and lower layer was highest as 61.8 and 15.7, while those of A. okamotoanum were 37.5 and 2.6, respectively. Taxus cuspidata var. latifolia which was designated as vulnerable species by Korea National Arboretum began its existence in lower layer. Species diversity and evenness were 0.674 and 0.706 in upper layer, 0.947 and 0.805 in middle layer, and 1.312 and 0.938 in lower layer, respectively. Soil pH of A. okamotoanum community was 5.79. The contents of organic matter, total N, and available $P_2O_5$ were 7.2%, 0.33%, and 51.1ppm, respectively.

Spreading Pattern of Evergreen Broad-leaved Trees in Forest Community adjacent to the Camellia japonica Stands (동백나무림 주변 산림군집에서 상록활엽수의 확산패턴)

  • In this study, the amount of seedlings and seed dispersal of evergreen broad-leaved trees in forest community adjacent to Camellia japonica forest were studied to provide basic information for effective management of evergreen broad-leaved forest. Evergreen broad-leaved trees including C. japonica, Neolitsea sericea, Machilus thunbergii, Ligustrum japonicum, Cinnamomum japonicum, Litsea japonica, Pittosporum tobira showed high density and ratio of seedlings in community adjacent to C. japonica forest. Although individual densities of N. sericea, M. thunbergii, L. japonicum were low, their seedlings were distributed up to Pinus thunbergii and coniferous broad-leaved forest at a distance of 100m and 200m from C. japonica forest. Distribution of DBH class of C. japonica suggested a continuous spread from higher frequency of young individuals, N. sericea, M. thunbergii and L. japonicum did not showed an obvious trend. Seed of C. japonica mainly dispersed within 50m from mother tree.

Methodological Consideration for Estimating Growing Stock of Young Forests based on Early Growth Characteristics of Standing Trees in Korea (우리나라 입목의 초기 생장 특성에 따른 유령림의 임목축적 산출방안 고찰)

  • The growing stocks of young forests that are less than10 years of age have been excluded from the Korean forest resource statistics, despite the existence of standing trees; however, sustainable forest management and carbon removals in the forestry section require complete information regarding forest resources. This study developed a method to estimate the growing stocks for young forests from National Forest Inventory (NFI) data. After reviewing previous research on growth characteristics for young forests, we conducted stem analysis of major species, and examined stand characteristics by site index, based on real yield tables. Our statistical analysis results showed that there were few standing trees with diameters at breast height (DBH) above 6 cm in young stands, and that it would have taken 12 years, on average, to reach 6 cm DBH. This suggests that mean tree height by diameter should be assessed at the root, in order to assess growing stocks for young stands through the NFI. Moreover, the database system should be improved to differentiate tree species, since diverse shrubs, including trees, have been surveyed.
