Quality Characteristics and Antioxidant Activity of Chrysanthemum indicum L., C. boreale M., and C. zawadskii K. Flowers Candies (감국, 산국 및 구절초꽃 캔디의 항산화활성과 품질특성)

  • In order to investigate the quality characteristics of candies prepared with Compositae species flower extracts, the samples of Chrysanthemum indicum Linne (CI), C. boreale Makino (CB), and C. zawadskii var. latilobum (Maxim.) Kitam (CZ) were extracted with hot water and concentrated, and then added with various concentrations of 5$\sim$20% for candy preparation. L-value of candy color was decreased, but a- and b-value were increased with increasing amount of added extracts. pH was decreased, but total acidity was increased with increasing amount of added extracts. Moisture content of CI, CB, and CZ candies were 2.61$\sim$4.11, 2.46$\sim$4.57, and 2.27$\sim$4.43%, respectively. Hardness of candies were decreased with increasing amount of added extracts. CZ candy of 20% added extract showed the highest total polyphenol, flavonoid content, electron donating ability and anol, flavonaid content at 2.71 mg/g, 0.86 mg/g, 76.24%, and 3.17 mg AA eq/g, respectively. In sensory evaluation, the CI and CZ candies of 10% added extract obtained higher sensory scores.

Effect of NPK Fertilization on the Yields and Effective Components of Chrysanthemum boreale M. (산국의 수량과 유효성분에 대한 NPK의 효과)

  • To establish the fertilization condition to increase the productivity of Chasanthemum boreale M. with high quality, the effects of three nutrients (N, P, K) on the yields and the effective components were investigated in the pot scale. NPK was applied by chemical fertilizers with $(N-P_2O_5,-K_2O=250-160-160\;kg/ha)$ as a main treatment, and NP $(N-P_2O_5,-K_2O=250-160-160\;kg/ha)$, NK $(N-K_2O =250-160\;kg/ha)$, and PK $(P_2O_5K_2O=160-160\;kg/ha)$ treatments were settled as comparison. Dry yields of C. boreale M. was increased significantly to 4 fold higher by nitrogen. Nitrogen increased apparently plant growth and inorganic nutrient uptake. In the flower, which is most useful and edible part as a herbal medicine, main amino acids were glutamic acid and aspartic acids, and the total content was increased significantly by three elements of application. In addition, the content of cumambrin A, which is known to have the effect of blood-pressure reduction, was increased source to 6.2 times by nitrogen higher than that in PK treatment. Potassium was more effective in biosynthesis of cumambrin A than phosphorus, but the biological pathway was not clear, still.

Effect of Hot-air Drying Temperature on Volatile Compounds in Chrysanthemum boreale M. Flowers (열풍 건조온도에 따른 산국의 휘발성 성분 변화)

  • This study examined the effect of drying temperature on the qualitative properties of Chrysanthemum boreale M. flowers. The flower samples were dried in a hot air dryer at $40^{\circ}C$, $50^{\circ}C$, and $60^{\circ}C$, respectively, to attain a $23{\pm}1%$ moisture content. The time required to reach the target moisture content was 8.5-69 hr, and there was a very high negative correlation between log (spending time) and temperature. The Hunter color L- and b-values of the flowers were decreased with increasing drying temperature, whereas the a-value was increased. The volatile compounds contained in the dried flowers were determined by a solid-phase microextraction method. Twelve primary volatile compounds were detected and then quantified based on the GC chromatograms of the samples. The total contents of volatile compounds were increased with increasing drying temperature, and germacrene D and camphor were the main compounds in all samples.

Isolation of Cytotoxic Substances from Chysanthemum Boreale M. (산국으로부터 항암활성 성분의 분리)

  • Sixty kinds of medicinal plants were examined upon the cytotoxicity against L1210 mouse leukemia cells. Isolation and purification of effective antitumor substances from Chysanthemum boreale had been performed which appeared strong cytotoxicity, In cytotoxicity test of the each solvent fractions, $ED_{50}$ values of chloroform fraction against L1210, K562 and A549 cells were shown as 3.98, 4.28, 3.84 (${\mu}g/ml$), respectively. Compound I and II were purified from the chloroform fraction. Between the purified compounds, $ED_{50}$ values of Compound I against L1210, K562 and A549 cells were shown as 0.55, 0.0003, 0.001 (${\mu}g/ml$), respectivety. Whereas Compound II was shu as 4.79 against K562.

Effects of Nitrogen Fertilization on the Yield and Effective Components of Chrysanthemum boreale M. (질소시비가 산국의 수량과 유효성분에 미치는 영향)

  • Chrysanthemum boreale M. (hereafter, C. boreale M.), a perennial flower, has been historically used as a natural medicine in Korea. With increasing concerns for health-improving foods, the demand for C. boreale M. has become higher than ever. Howevr, the amount of wild C. boreale M. collected from mountainous areas is not enough to cover all demands. The cultivation system and fertilization strategy are required to meet increasing demand on C. boreale M. with a good quality. We investigated the effects of nitrogen application on plant growth and effective components of C. boreale M. to suggest optimum rate of nitrogen fertilization. C. boreale M. was cultivated in a pot scale (1/2000a scale), and nitrogen applied with rate of 0(N0), 50(N50), 100(N100), 150(N150), 200(N200), and $250(N250)kg\;ha^{-1}$. Phosphate and potassium were applied at the same level ($P_2O_5-K_2O=80-80kg\;ha^{-1}$) in all treatments. Maximum yield achieved in 246 and $226kg\;ha^{-1}$ N treatment on the whole plant and the flower part, a valuable part as a herbal medicine, respectively. Proline was the most abundant amino acid in the flower of C boreal M. and the contents of amino acids increased with increasing nitrogen application rate in flower. Nitrogen recovery efficiency was high more than 41% in all nitrogen treatments and increased to 61.8% in nitrogen N100 treatment. From the nitrogen content, the high nitrogen uptake, the low residue of mineral N and the reasonably good apparent fertilizer recovery, it can be inferred that C. boreale M. made efficient use of the available nitrogen. In flower, contents of Cumambrin A. which is a sesquiterpene compound and has the effect of blood-pressure reduction, decreased with increasing nitrogen application. However, the amount of Cumambrin A in flower increased as nitrogen rate increased, because of increasing flower yield. Conclusively, nitrogen fertilization could increase yields and enhance quality. The optimum nitrogen application rate might be on the range of $225{\sim}250kg\;ha^{-1}$ in a mountainous soil.

Variation of Concentration of Terpenes in Chrysanthemum boreale (산국에서의 Terpenes 함량의 변이)

  • The monoterpenes and sesquiterpenes are analysed in the leaf and stem of Chrysanthemum boreale using gas chromaltography-mass spectrometry (GC-MS). The total amount of sesquiterpenes are always higher than monoterpenes in both leaf (2.0-3.4 times) and stem (1.6-8.3 times). The mono- and sesquiterpenes yields of the leaf are higher than the stem. There was no significant difference among the leaf developmental stages, while those of stem were varied. Seventeen monoterpenes and 9 sesquiterpenes compound in this plants comprised more than 5% of the mean total monoterpenes and the total sesquiterpenes in each dates. Among leaf monoterpenes, the concentration of (+)-Limonene and unknown compound no. 13 (Retention time, R.T.=17.28) varied significantly during leaf growing season, and the concentrafion of unknown compound no. 7 (R.T.=35.04) and no. 9(R.T.=35.71) varied in the leaf sesquiterpenes. Similarly the results from the leaf, the concentration of five monoterpenes in stem also varied significantly during maturing period, and much varied in seven compounds of stem sesquiterpene. The major sesquiterpenes of leaf and stem were ${\alpha}-Humulene$ and compound no. 2(R.T.=26.19).

Germacranolides from Flowers of Chrysanthemum boreale Makino (산국 꽃의 Germacranolides)

  • Two sesquiterpene lactones were isolated from the flowers of Chrysanthemum boreale Makino by the silica gel column chromatography and recrystallization. On the basis of spectrometric studies including $^1H-NMR,\;^{13}C-NMR,\;DEPT,\;^1H-^1H\;COSY,\;{13}C-^1H\;COSY$, IR and Mass, compounds 1 and 2 were identified as germacranolide, tulipinolide and costunolide, respectively. And they showed antibacterial activity against Vibrio parahaemolyticus, Bacillus subtilis, Bacillus cereus and Staphylococcus aureus. This is the first report that Chrysanthemum boreale contained tulipinolide and costunolide.

Optimization of Extraction Conditions for Physicochemical Properties of Ethanol Extracts from Chrysanthemum boreale (산국(山菊) 에탄올 추출물의 이화학적 특성에 대한 추출조건의 최적화)

  • To renew interest in Chrysanthemum boreale as a traditional food material, response surface methodology was used for optimizing extraction conditions and monitoring physicochemical properties of ethanol extracts from Chrysanthemum petal. The phenolic compounds of ethanol extracts were minimized in 87ml/g(solvent per sample), 21%(ethanol concentration), and 15 hours(extraction time). The physicochemical properties of ethanol extracts were maximized in the conditions of 115ml/g, 98% and 16 hours on yellow color intensity, 143ml/g, 75% and 19 hours on carotenoid content, and 148ml/g, 53% and 18 hours on soluble solid content. Optimum ranges of extraction condition for physicochemical properties of Chrysanthemum boreale were 130~150ml/g, 70~85% and 20~28 hours, respectively. Predicted values at the optimum extraction condition were in good agreement with experimental values.
