Effects of Red Ginseng Saponins on Growth of and Aflatoxin Production by Aspergillus flavus (홍삼(紅蔘) 사포닌이 Aspergillus flavus의 발육(發育)과 Aflatoxin 생산(生産)에 미치는 효과(效果))

  • The study was carried out to find effects of the saponins that were extracted from red ginseng on the growth of, aflatoxins production by, and protein patterns of Aspergillus flavus NRRL 3357. A. flavus with $10^6$ conidia as grown at $30^{\circ}C$ for seven days on the enriched medium. Mycelial growth and pH changes of medium which cultured the mold, were similar to those of the control group. However, aflatoxin which produced by the mold was less than that of the control in all concentration of the saponin. To be more specific, 0.3 % of the saponin inhibited production of aflatoxin $B_1$ and $G_1$ to the extent of 31.6 and 21% of the control. The protein peaks of A. flavus at the fourth day of the culture were shown high intensity near the level of 14,300 daltons. However, the mold which cultured in the medium containing the saponin showed low intensity of protein than that of the control group on all molecular weight.

Effect of Microwave Treated-Wild Ginseng on the Quality of Korean Traditional Yakju (마이크로 웨이브로 처리한 산양삼 첨가가 전통 약주의 품질에 미치는 영향)

  • To increase the quality of Korean traditional yakju, we prepared seed cultures by fermentation at $20^{\circ}C$ for 2 days after addition of 140% water, 3% nuruk and 1.5% yeast into cooked rice. After the 200% cooked rice, 120% water and 0.08% commercial saccharifying enzyme were added to seed cultures and fermented for 2 days at $20^{\circ}C$, wild ginseng was added and then further fermented for 5 days. Physicochemical properties of traditional yakju were investigated. Ethanol was produced (18.5%) by the addition of 1.2% wild ginseng. However, ethanol content was not increased by addition of microwave treated-wild ginseng and rice (either cooked rice or raw). The traditional yakju obtained by fermentation at $20^{\circ}C$ for 5 days, after 90 sec of microwave treated-wild ginseng was added into main fermentation broth, showed good total acceptability and also contained 791 ppm saponin.

Increase of Functional Saponin by Acidic Treatemnt and Temperature of Red Ginseng Extract (홍삼엑기스의 산(pH) 및 온도처리에 의한 기능성 사포닌 함량증대)

  • To increase the contents of functional ginsenosides by conversion, especially ginsenoside-$Rg_3$ and $Rh_2$, the extracts of red ginseng were treated with high temperature and citric acid or apricot extract. When the extracts were subject to $120^{\circ}C$ for 2 hours, the content of ginsenoside-$Rg_3$ was increased 2 times than in control. When the extracts were subject to $120^{\circ}C$ and acidic conditions adjusted with citric acid, the ginsenoside-$Rg_3$, was detected 2.8 times, but other ginsenoside were decreased heavily to 65%. When the extract were treated with for 12 hours at $80^{\circ}C$, the content of ginsenoside-$Rg_3$ was increased to 3.3 times, Also, when the red ginseng extracts were treated with apricot extract, the ginsenoside-$Rg_3$ was detected to 4 times than in control, but other ginsenoside were decreased lightly to 35%, not same as at the $120^{\circ}C$ treatment.

  • Ginsenosides have been regarded as the principal components responsible for the pharmacological and biological activities of ginseng. Absorption of major ginsenosides from the gastrointestinal tract is extremely low, when ginseng is orally administered. In order to improve absorption and its bioavailability, conversion of major ginsenosides into more active minor ginsenoside is very much required. Here, we isolated lactic acid bacterium (Lactobacillus brevis LH8) having ${\beta}-glucosidase$ activity from Kimchi. Bioconversion ginsenoside Rd by this bacterium in different temperatures was investigated. The maximum activities of crude enzymes precipitated by ethanol were shown in $30^{\circ}C$ and then gradually decreased. In order to compare the effect of pH, the crude enzymes of L. brevis LH8 were mixed in 20mM sodium phosphate buffer (pH 3.5 to pH 8.0) and reacted ginsenoside Rd. Ginsenoside Rd was almost hydrolyzed between pH 6.0 and pH 12.0, but not hydrolyzed under pH 5.0 and above pH 13.0. Ginsenoside Rd was hydrolyzed after 48 h incubation, whereas ginsenoside F2 appeared from 48 h to 72 h, and ginsenoside Rd was almost converted into compound K after 72 h.

Study on the Additional Effect of Fat-Soluble Vitamins to Antioxidative Action of Ginseng Saponin (인삼(人蔘)사포닌의 항산화작용(抗酸化作用)에 대한 지용성(脂溶性) 비타민의 첨가효과(添加效果))

  • This study was to demonstrate the additional effect of fat-soluble vitamins on inhibitory action of Korean ginseng (Panax ginseng C. A. Meyer) saponin to lipoperoxide formation in vitro and in vivo. The ginseng saponin and vitamins were added to the substrate of linoleic acid and incubated on a shaking water-bath at $60^{\circ}C$, and the inhibitory action on lipoperoxide formation was examined by measuring the TBA value (532 nm), POV (500 nm) and EDA (electorn donating ability to ${\alpha}$, ${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl at 525nm) for in vitro. The ginseng saponin and vitamins were administered to Sprague-Dawley rats (♂, 100~120g) orally, and the inhibitory effect on lipoperoxide formation was examined by measuring the TBA value in vivo. EDA to DPPH of ginseng saponin added with vitamin A and D were higher than that of ginseng saponin only. Ginseng saponin added with vitamin A and D inhibited strongly lipoperoxide formation of initial step by extension of induction period in vitro. Fat-soluble vitamins such as vitamin E and A were approved the additional effect on inhibitory action of lipoperoxide formation in vitro. The additional effect of fat-soluble vitamins to ginseng saponin for inhibitory action of lipoperoxide formation was effective vitamin E and D for blood, vitamin A and E for liver in vivo.

The Efficacy of Shampoo Containing Ginseng Radix on Preventing Hair Loss and Promotion Hair Growth (홍삼사포닌 Rg3가 함유된 샴푸의 탈모방지와 양모개선 효과)

  • Recently, hair growth effect of red ginseng was proved by many in vitro experiments, but there are not enough clinical studies. In the present study, we evaluated the efficacy of Rg3 hair shampoo (Somang Co., Korea) containing Rg3 0.003% extracted from ginseng radix on preventing hair loss and promoting hair growth by clinical study. 42 volunteers were enrolled present study and devided into two groups; one group used the Rg3 hair shampoo and another group used shampoo not containg Rg3. Before and after 8 and 16 weeks use of Rg3 hair shampoo, we measured the density, thickness and growth rate of hair. At the same time, an assessment of improvement of hair loss and subjective satisfaction of patients were conducted by clinicians and patients. Results obtained from the group used Rg3 hair shampoo showed statistically significant improvement in the density, thickness and growth rate of hair. Results from the clinicians, patients and subjective satisfaction of patients also showed better responses for the group used Rg3 hair shampoo. In conclusion, Rg3 hair shampoo is an effective product on preventing hair loss and promoting hair growth.

Effect of the Extruded Ginseng on Antioxidant Activity (압출 성형 인삼의 항산화 활성 증강 효과)

  • This study was conducted in order to evaluate the antioxidant activity of extruded ginseng in different extracted fractions. Each of the fractions obtained from extruded ginseng and ginseng (control) were extracted with 80% ethanol, and then the lipophilic components were removed with ether while the hydrophilic components were separated with water-saturated butanol. Each of the 80% ethanol/butanol/water layers were collected and evaporated to acquire samples for tests of saponin content and antioxidant activity. The antioxidant activity of extruded ginseng fractions and ginseng fractions were determined via the oxygen radical absorbance capacity (ORAC) assay. Overall, the extruded ginseng samples harbored saponin contents of 2.2 (Rg1), 2.3 (Re), 1.2 (Rc), 1.3 (Rb2), and 2.2 (Rd) times that measured in the ginseng prior to extrusion. Antioxidant capacity was also higher, not only in the 80% ethanol/butanol which harbor a significant quantity of saponin, but also in the water fractions, which harbor relatively low quantities of saponin as compared to the control samples.   • To study the effect of Korea red ginseng (KCG) on the skin and cerebral blood flow, we evaluated the change of skin perfusion rate and cerebral perfusion rate after the intravenous, intraperitoneal, and oral administration of crude saponin (CS) and saponin-free fraction (SFF) of KRG in the rats. The change of skin perfusion rate and cerebral perfusion rate was measured laser doppler flowmetry. The intravenous injections of CS or SFF of KRG and intraperitoneal injection of SFF of KRG did not change the relative skin and cerebral blood flow in the rats. When the rats were treated by the intraperitoneal injection of CS of KRG, relative cerebral blood flow was significantly increased with a time-dependent manner, however, relative skin blood flow was not influenced by the them. Oral administration of CS of KRG slightly increased skin blood flow in the rats. Also, the change of cerebral blood flow by transient bilateral carotid arterial clamp in the CS-treated rats was significantly decreased, compared with control groups. From the above results, it was suggested that Korea red ginseng have a increasing property of cerebral blood flow in the rats.

Effects of Red Ginseng Total Saponin on the Polycystic Ovaries in Rats (홍삼 사포닌이 랫드의 다낭성난소에 미치는 영향)

  • Polycystic ovarian syndrome (PCOS) is a very common endocrine disorder in women of reproductive age. There are some evidences that nerve growth factor (NGF) is involved in the pathogenesis of PCOS. In this study, we investigated the effect of Korean red ginseng total saponin (GTS) on the ovarian morphology and NGF expressions in the ovaries, pituitary and hippocampus. The oil control animals were injected with 0.2 ml oil/rat. Animals in estradiol valerate (EV) control group were injected i.m. with 4 mg EV in 0.2 ml oil/rat. The GTS was administered (100 mg/kg) i.p. every other day for 60 days, beginning 1 day after the EV injection. PCO was induced by a single injection of EV (4 mg, i.m.). At day 60, the expressions of NGF were examined by immunohistochemistry. The main findings were as follows; PCO was fully developed with a single i.m. injection of EV, and PCO showed the increased expression of NGF, and GTS administration decreased NGF expressions in the ovaries without affecting pituitary and hippocampus significantly. The present results demonstrate that GTS attenuates PCOS by the stimulation of NGF expression.

Determination the optimum extraction method for saponin lancemasides in Codonopsis lanceolata (더덕 사포닌인 lancemasides의 최적 추출 방법 구명)

  • This study was conducted to select the optimal extraction method of codonopsis lanceolata saponin. To investigate the lancemasides content depending on each extraction method, various extractions were performed: reflux (methanol and butanol), hot water, as well as ultrasonic bath (40 kHz; continuous irradiation/interval irradiation) and ultrasonicator (20 kHz) extractions. From the result, the overall lancemasides content were the highest in ultrasonic bath (MeOH; continuous irradiation) extraction, followed by ultrasonic bath (water; continuous irradiation)>ultrasonic bath (MeOH; interval irradiation)>ultrasonicator (MeOH)>hot water>MeOH reflux>BuOH reflux extractions in that order. Sample drying method prior to ultrasonic bath extraction was more effective shade drying than freeze drying. Effective duration and temperature of extraction was 2 hr at $64^{\circ}C$. And ingredient change diverted from aster saponin Hb to lancemasides was identified by extraction condition such as extraction time and temperature.
