The Karyotype of Payamphistomum cervi(Zeder, 1790) from Korean Cattle (한국산 사슴쌍구흡충의 핵형분석)

  • As a series of systematic classification of paramphistomes, the worms in the rumen and reticulum were collected on 214 Korean cattle slaughtered at Jeonju abattoir from January, 1986 to April, 1987 and were classified by means of morphology. Afterwards, the karyotype of Paramphistomum cervi(Zeder, 1790) was detected by means of modified air.drying method from germ cells of the worms. The results were summarized as follows: 1. In the chromosome number of 254 p. cervi, the haploid cell was n:9 and the diploid 2n=18. The meiotic divisions were observed frequently; 1,924 haploid and 32 diploid cells were reliable. Nine pairs of mitotic chromosomes were homologous in the metaphase stage, and the chromosomes were composed of five medium-sized metacentrics (m) , subtelocentrics(st) or submetacentrics(sm) and four small-siRed subtelocentrics(st) or submetacentrics(sm). Meiotic metaphase was composed of five medium and four small chromosomes in size. 2. As a series of C-banding method, C-band was showed in centromeric region from all of the haploid germ cells. Whereas chromosome No. 3 and 5 included heterochromatin on the tip region, chromosome No. 4 on the distal region and No. 6 proximal region. And chromosomes No. 2 and 8 showed a remarkable C-band distinguished from other chromosomes.

Effect of Recombinant Bovine Somatotropin Injections on Deer Stag Plasma Insulin-like Growth Factor-I Profiles (rBST 투여가 숫사슴의 혈액 내 IGF-I 함량에 미치는 영향)

  • This study was conducted to elucidate the influence of recombinant bovine somatotropin (rBST) on the plasma IGF-I profiles in deer stags. Slow release rBST (250mg each) preparations were injected subcutaneously during the antler's non-growing period and growing period. It was observed that significant increases in the plasma IGF-I concentrations were occurred right after injections of the rBST irrespective of antler's non-growing period or growing period. When the rBST preparations were injected two times with one week interval during the antler's non-growing period, the mean level of IGF-I was found to be 2,920.7ng I mL, indicating that the IGF-I concentration increased up to 8.4-folds compared with a control. After two weeks from second shot, however, the IGF-I levels decreased to the level of the control. Similar results were also obtained with both a Sika and a Red deer during the antler's growing period. Increases in the IGF-I level of up to 5.6-folds with the Sika deer and 4.3-folds with the Red deer were found. Based on the present study, it is likely that the rBST may be used as a possible candidate for an antler's growth stimulating agent.

Effect of Lectin Isolated from Serrognathus platymelus castanicolor Larvae on the Various Cytokine Expressions (넓적사슴벌레(Serrognathus platymelus castanicolor) 유충으로부터 분리한 렉틴의 사이토카인 발현)

  • A lectin was purified from Serrognathus platymelus castanicolor larvae and named as SPL. The purification was carried out by ion-exchange chromatography on DEAE Sephadex A-50 and gel filtration chromatography on Sephadex G-200. The purity of the protein was verified by polyacrylamide gel electrophoresis and the purified lectin agglutinated erythrocytes of rabbit and human A, B, O, AB. SPL was tested it's ability to enhance the expressions of cytokines, $IL-1\alpha$, IL-2, IL-6, $TNF\alpha$ and $IFN\gamma$ by human peripheral blood mononuclear cells (PBMC) obtained from healthy donors. mRNA analyses were performed by RT-PCR at the moment of 1, 4, 8, 24, 48, 72 and 96 h after stimulation of PBMC with purified SPL. The patterns of IL-2 band were slightly expressed from 24 h and the strongest band was appeared at 96 h. The expressions of $IL-1\alpha$ and IL-6 mRNA were strong from 1 to 8 h and those of $TNF\alpha$ were from 48 to 96 h. The mRNA encoding $IFN\gamma$ were not detected. The addition of SPL for macrophage cultures induced production of nitric oxide (NO) by cells in a dose-dependent manner. NO release was partially inhibited by $TNF\alpha$ antibodies. These results suggest that SPL has the ability to enhance cytokine expressions in PBMC and to induce the NO release by TNFa in macrophage cultures from PBMC cultures.

  • Efficacy of praBiquantel (CesocideB injection) by intramuscular (1.M.) route against cestode infections was evaluated. Total 93 domestic or laboratory animals such as dogs, cats, rats, mice, goats, deers and chickens were used. Animals were infected with Dipylidium caninum, Spirometra sp. , Taenia pisiformis, Taenia taeniaeformis, Hymenolepis nana, Moniegia expanse, Moniexia sp. or Raillietina sp. A single dose of prasiquantel, 6 mg/kg of body weight, was highly effective (97.9%) against cestodes of various kinds disregarding the host species or their intensity of infection. At higher dose above 6 mg/kg, the cure rate was 100%. All the cestodes treated were expelled from the host within 48 hours. The discharged proglottides were damaged severely except Hymenolepis nana and Moniegia expanse. Intramuscular injection of this drug evoked a brief pain response in a dog, but no other side reactions were observed.

A Morphological Study of Intrachondral Articulations in the Korean Water Deer (한국고라니 갈비연골속관절에 관한 형태학적 연구)

  • Intrachondral articulations are the synovial joints that occur within costal cartilage in artiodactyls. The anatomical structure of Korean water deer differs from that of other deer. However, there have been no reports on the occurrence and shape of intrachondral articulations in the deer. To provide information on these articulations, we examined the occurrence and shape of intrachondral articulations in the Korean water deer by gross findings, radiography, computed tomography (CT), and histological observation. These joints often occur in the second to the tenth ribs. Morphologically, they are spheroidal joints, especially from the third to the eighth ribs, and their configuration is discernable in gross findings, radiography, CT, and histological sections. These basic results would be helpful in the diagnosis and treatment of diseases in the lateral thoracic wall of the Korean water deer.

First Report of Red Thread of Kentucky Bluegrass (Poa pratensis L.) Caused by Laetisaria fuciformis on Golf Course in Korea (국내 골프코스의 켄터키블루그래스에 Laetisaria fuciformis에 의한 붉은뿔마름병(Red Thread) 발생보고)

  • In the middle of May 2018, typical red thread disease symptoms were observed on Kentucky bluegrass (Poa pratensis L.) on a golf course, which locates at Yangsan, Gyeongnam province in Korea. Irregular-shaped patched symptoms were observed in fairway of golf course. The foliar symptom was dried out and faded to straw color and tip of the grass leaves were tangled like thread. Early morning, infected and tangled leaves were covered with the pinkish gelatinous antler-like structure (sclerotinia) as a typical red thread disease symptom. Causal fungal pathogens were isolated from the symptom in Kentucky bluegrass. The fungal culture characteristic on potato dextrose agar color of colony was pale pink and conjugated hyphae, sclerotium of irregular shape was pale pink and 3~5mm diameter in size. The pathogen was identified as Laetisaria fuciformis based on morphological and culture characteristics as well as molecular characteristics. Pathogenicity test was verified on the Kentucky bluegrass by Koch's postulates. This is the first report of red thread disease occurrence in Kentucky bluegrass by L. fuciformis in Korea.

Effect of Seasoning Condition and Fruit Extracts Level on the Quality Characteristics of Venison Jerky (조미배합 조건과 과일농축물 첨가에 의한 노령 사슴육포의 품질특성)

  • The principal objectives of the experiments in this study were to assess the quality of venison jerky made from Elk deer (female, weight; 380 kg, age; 15 old years) in accordance with various seasoning sauce and mixture fruit extract level. Salinity, saccharinity and acidity values of the seasoning sauces were all high(VJ-1; 3.72%), (VJ-3; 3.95%), (VJ-1; 0.35%). The salinity range of the venison jerky products was 5.65~5.92%, the saccharinity range was 3.75~4.18%, and the seasoning sauce samples resulted in a significant difference in acidity values(p<0.05). Crude protein, fat and ash contents of the venison jerky products satisfied the Korean jerky standard value, but the moisture content range was 36.18~38.59%, and this was also in excess of the jerky standard level. The manufacturing yield was 42.24~44.48%, with VJ-1 at lowest levels followed by VJ-2 and VJ-3. The pH value of VJ-1 was high, but not significantly higher than the other samples(p>0.05). The water activity value range was measured at 0.68~0.71, and the lightness(L) and redness(a) value evidenced marked increases with increased amounts of added sodium nitrite(p<0.05), and the yellowness(b) values did not significantly differ(p>0.05). The overall sensory acceptance scores ranged between 5.53~7.49, in the order VJ-3, VJ-2, VJ-1(p<0.05).

A study on the food habits of Sika Deer (Saanen) fed with roughage sources (조사료원에 따른 꽃사슴(Servus nippon)의 채식습성에 관한 연구)

  • The object of this experiment was to investigate the food habits of sika deer fed with various roughage sources. The experimental trials were conducted at Unbong Animal Genetic Resources Station in 2008. The experimental roughages include five sources and 25 species in all; grasses and legumes: 5 species (mixed grasses, orchardgrass, tall fescue, alfalfa, white clover), native grasses and weeds: 5 species (mixed native grasses, Miscanthus sinensis Anderss, Arundinella hirta (Thunb.) Tanaka, barnyard grass, short awn, forage crops (hay, silages and straw): 5 species (barley + hairy vetch, Wheat + hairy vetch, rye silage, barley silage, baled rice straw), browse and fallen leaves: 5 species (mixed browse, oriental white oak, Quercus serrta Thunb., oriental cherry fallen leaves, Japanese chestnut fallen leaves), and imported hays and straws: 5 species (timothy hay, tall fescue straw, annual ryegrass straw, klinegrass hay, alfalfa hay). Five sika deer were used as experimental animals and the averaged body weight was 95+5.4kg. The chemical composition and dry matter digestibility of each roughage source and species were significantly different at the sampling area, plant species, growth stages and cutting period(p<0.05). The sika deer ate more roughages which had low fibrous contents, but high dry matter digestibility. Among all the 25 species of roughages, the favorite intake roughage sources ranking by sika deer was observed like this: browse and fallen leaves (32.2%), grass and legumes (27.0%), native grasses and weeds (22.0%), imported hays (12.9%) and forages crops (5.5%) respectively. Although, the sika deer ate more browse leaves, but ate more roughage which had low fibrous contents (NDF and ADF), but high drymatter digestibility. On the other hand, compared to each roughage source, total intake amount by sika deer was showed as browse and fallen leaves (32.2%). Based on the result, the food habit of sika deer seems to be closer to the typical browser.

Serologic survey of the ruminant bacterial infectious diseases in farmed deer and wild water deer in Jeonbuk province (사육사슴 및 야생고라니에서 소 세균성 전염병에 대한 혈청학적 연구)

  • Deer can be one of the susceptible animals to bovine infectious diseases, and thus, may play a role either as a reservoir or amplifier host for spreading the diseases to other species such as cattle and goat. This study was conducted to determine the serum antibodies to bacterial infectious diseases for brucellosis, tuberculosis (TB), paratuberculosis (Johne's disease) in deer. Serum samples were randomly collected from 78 deer from 31 farms at Jeonbuk province, and 7 wild water deer from Jeonbuk wild animal treatment center during 2005 to 2007, respectively. Four farm deer (5.1%) showed antibodies to tuberculosis using Antigen Rapid Bovine TB Ab Test Kit. One elk (1.3%) and one wild water deer had antibodies for paratuberculosis. Antibody against Brucellosis was not detected in tube agglutination test (TAT) and enzyme-linked immunosorbent assay (ELISA). These data suggest that caution should be applied to inspection of velvet, deer blood and meat for human consumption from deer because of zoonotic bacterial diseases in deer. In addition, farmed deer can be a transmissible host for zoonotic disease to diary or raising farm.

Serosurvey for antibodies against Neospora caninum in farmed deer and wild water deer in Jeonbuk province (사육사슴 및 야생고라니의 Neospora caninum에 대한 항체가 조사)

  • With deer farming now becoming recognized as one of the alternative livestock farming practices, there has been much attention on infectious diseases of cattle, but less awareness of these diseases in deer. Serum samples were randomly collected from 78 deer from 31 farms and 7 wild water deer from wild animal medical center during 2005 to 2007, respectively. A total 85 sera were tested for Neospora caninum antibodies by ELISA. Fourteen farmed deer(17.9%) were positive to N. caninum but no antibodies were found in sera from wild water deer. The sera from all of the area for these study showed positive in the range from 6.7 to 30.7%. On the basis of these study, farmed deer in Jeonbuk province were exposed extensively and seriously to N. caninum which might present a risk to other species included cattle and dog.
