농산물의 효소적 갈변의 저해제 개발 및 그 응용

  • 농산물의 수확 후 취급이나 가공중에 일어나는 효소적갈변 현상은 외관의 변색에 의한 소비자의 기호성을 저하시킬 뿐만 아니라 상품의 가치를 하락시키며, 경제적인 손실을 초래함으로서 현재 식품에 종사하는 기술자나 가공업자에게 가장 관심이 있는 분야이다. 효소적갈변의 주역을 담당하고 있는 polyphenol oxidase(PPO)(EC1.14, 18.1)은 대부분의 식품에 존재하고 있으며 monophenol의 hydroxylation에 의한 o-diphenol화합물의 형성반응(cresolase)과 o-diphenol화합물의 o-quinone으로의 산화반응(catecholase)을 촉배한다. 그리고 여기서 일단 생성된 o-quinone은 다시 다른 o-quinone이나 그밖의 단백질, 아미노산, 환원당 등의 여러 성분들과 상호 반응하여 갈변식품의 특유의 색소인 melanine을 형성한다. 효소적갈변에 의한 품질의 저하는 식품가공산업에서 해결되어야 할 하나의 큰 과제이다. 현재까지 식품산업에서 가장 널리 쓰이고 있는 효소적갈변 저해제는 sylfite류 이다. 그들은 새우의melanosis(blackspot)를 억제하며, 감자, 버섯, 사과와 다른 과채류의 갈변을 방지할 뿐 아니라 쥬우스와 포도주의 off flavors를 일으키는 폴리페놀 플리머의 형성을 감소 시킨다. 그러나 최근 sulfite가 사람의 건강에 좋지않는 부작용을 일으키며, 특히 천식을(asthma)을 지니고 있는 사람이 sulfite가 함유된 식품을 먹었을 경우 사망을 초래할 수 있어 식품첨가물로서 그들의 사용이 미국 FDA에 의해 재평가되고 있다. 최근에 FDA는 셀러드바에서의 sulfite사용을 금지 시켰으며 점차 감자가공품에의 사용을 금지하는 방향으로 나아가고 있다. 따라서 현재 실질적이고 기능적인 sulfite의 대체 화합물의 개발이 절실히 필요한 실정이다. 이러한 배경으로 본 강연에서는 효소적갈변 저해제의 개발과 그들의 식품가공에의 적용 현환 및 화장품, 의약품으로의 응용에 대해 설명하고자 한다.

다양한 동결처리에 따른 과채류의 내부압력 변화 비교

  • 본 연구에서는 과채류의 냉동냉장시 내부압력에 따른 조직파괴를 방지하기 위한 기초연구로 동결방법에 따른 내부압력 변화에 대하여 조사하였다. 과채류의 동결에 의한 중량감소율은 침지식 동결방식에서 0.44∼l.38% 수준으로 가장 적게 나타났으며, 동결방식에 따른 중량감소율 차이는 수박에서 가장 심하여 수분함량에 클수록 동결속도에 따른 효과가 비교적 크다는 것을 알 수 있었다. 동결속도에 따른 내부압력은 배, 사과 및 메론에 있어 침지식 방법에 의한 동결시 체적팽창 및 수축에 따른 내부압력 차가 가장 적게 나타났으며, 송풍식 동결에서 가장 크게 나타나 동결속도가 빠를수록 내압의 크기는 적다는 것을 알 수 있었으나 딸기 및 수박의 경우는 오히려 침지식에서 내압이 가장 크게 나타났다. 균온 처리하지 않은 동결에 있어 과일의 내부압력 변화는 약 2 psig수준인 반면에 균온 처리한 수박의 내부압력 변화는 균온 처리하지 않은 수박의 내부압력 변화 경향과 유사하지만 일정한 수준의 내압을 발생시킴으로써 내부압력의 크기도 약 1.3 psig수준으로 상당히 적게 나타났다. 또한, 해동시에 있어서도 균온처리한 시료의 내부압력 크기가 균온처리하지 않은 내부압력 크기보다 상대적으로 적게 나타났다. 균온처리 동결시, 다단계 처리보다는 3∼4회 수준의 일정한 균온 처리가 내부압력의 증감 폭을 줄일 수 있었다.

Effect of Ethanol on the Production of Cellulose and Acetic Acid by Gluconacetobacter persimmonensis KJ145 (Gluconacetobacter persimmonensis KJ145를 이용한 Bacterial Cellulose 및 초산발효에 미치는 Ethanol의 영향)

  • We investigated the effect of ethanol on the production of cellulose and acetic acid fermentation by Gluconacetobacter persimmonensis KJ145. Results showed that bacterial cellulose productivity was highest when 2% ethyl alcohol was added to apple-juice medium. For acetic acid production, 7% ethyl alcohol was needed. Optimal concentration of ethyl alcohol was 5% for simultaneous production of bacterial cellulose and acetic acid. For simultaneous production of bacterial cellulose and acetic acid, optimal nitrogen source and optimal concentration were corn steep liquor and 15% (w/v), respectively Optimal culture time for simultaneous production of bacterial cellulose and acetic acid was 14 days. At the optimal condition, Cluconacetobacter persimmonenis KJ145 produced 7.55 g/L of bacterial cellulose (dry weight).

Antioxidant Activation of Plum (Prunus mume Sieb. et Zucc.) Flesh, Pericarp and Stone Extracts (매실 과피, 과육과 핵의 품종별 추출액의 항산화 활성)

  • 매실에는 구연산, 사과산, 주석산, 피루브산 등의 유기산 등이 함유되어 있어서 체내의 대사작용을 돕고 세포와 근육에 필요물질인 젖산을 분해하여 피로를 감소시켜주며 신진대사를 촉진하고 있다. 또한 피루브산은 간의 피로, 해독작용을 활발하게 도와주며 카테킨산은 장내의 유해균의 번식을 억제하여 식중독을 예방하고 있다. 또한 이러한 작용은 설사와 변비를 치료하는데도 효과가 있으며 다량의 비타민C의 함유로 인하여 체내의 피로회복을 돕고 있다. 이러한 효능을 가진 매실은 전라남도 해안가에서 주로 재배하고 있다. 특히 순천과 광양지역에서 다양한 품종들을 재배하고 있다. 따라서 본 실험에서는 순천과 광양지역에 재배되고 있는 남고, 천매, 고성, 앵숙, 풍후, 옥영, 임주, 청축, 백가하 등의 잎과 과육, 핵의 추출물로부터 폴리페놀 성분은 핵, 과육보다 잎에서 양호한 반응을 나타내었다. 이러한 결과들을 중심으로 매실의 각 부위별 항산화 활성을 조사하였던 바, 매실의 대부분의 품종에서 항산화 활성이 양호하게 나타났으며 부위별에서는 핵보다는 잎과 과육에서 양호한 항산화 활성을 보였다. 품종별로는 앵숙, 남고, 청축에서 다소 양호한 항산화 활성을 나타내었다. 이러한 결과들로 미루어 볼 때 매실의 항산화 활성은 품종별, 부위별로 상이한 반응을 나타내었다.

Fruit Characteristics of Highbush Blueberry (Vaccininum corymbosum L.) Cultivars (하이부시 블루베리의 품종별 과실 특성)

  • This study was conducted to compare the internal and external fruit quality of highbush blueberry cultivars introduced in Korea. The fruit of blueberry showed a double S-shaped growth curve, and the fruit weight increased significantly per seed increased by 0.02 g. The sugar content ranged from 9.5 to 14.2°Bx, with a large difference between cultivars. And the free sugar was composed of glucose and fructose as a reducing sugar, and mannitol as a sugar alcohol. The organic acid content was 0.7~1.13%, and citric acid was higher than that of malic acid. Fruit hardness showed a berry characteristic that rapidly declined as the coloring progressed.

Comparison of Virus Elimination Methods for Disease-free Seedlings of the Apple Dwarfing Rootstock (사과 왜성대목 무독묘 생산을 위한 바이러스 제거 방법 비교)

  • Apple (Malus domestica) is one of the most economically important fruits in Korea. But virus infection has decreased sustainable production of apple and caused the serious problems such as yield loss and poor fruit quality. Virus or viroid infection including Apple chlorotic leaf spot virus (ACLSV), Apple stem pitting virus (ASPV), Apple stem grooving virus (ASGV), Apple mosaic virus (ApMV) and Apple scar skin viroid (ASSVd) has been also reported in Korea. In many cases, apple is infected with virus and viroid with no specific symptoms, the damage caused by the virus are unaware significantly. In our research, we tried to eliminate viruses in the rootstock for the disease-free seedlings of the apple dwarfing rootstock M.9 and M.26. The method of virus elimination was meristem culture, heat($37^{\circ}C$, 6weeks) treatment and chemistry($Ribavirin^{(R)}$) treatment. The analytical methods commonly used for the detection of virus is Enzyme-linked Immuno-Sorbent Assay(ELlSA) and Reverse Transcription-polymerase Chain Reaction(RT-PCR). RT-PCR method was more 30% sensitive than ELISA method. Efficiency of method eliminate virus appeared meristem method > heat treatment > chemistry treatment. The higher acquisition rate of disease-free seedlings is 30~40% on meristem treatment. In meristem treatment, the apple dwarfing rootstock M.9 gained infection ratio of ACLSV, ASPV and ASGV were 45%, 60% and 50% respectively. In the apple dwarfing rootstock M.26, infection ratio of ACLSV, ASPV and ASGV were 40%, 55%, 55%, respectively. Based on our results, it was found that most effective method of disease-free seedlings apple dwarfing rootstocks was by meristem treatment than heat method and chemistry treatment.

Study on Pesticide Runoff from Soil Surface-I, Runoff of Captafol by Natural Rainfall in Field (농약의 토양 표면유출에 관한 연구-I 포장에서 자연강우에 의한 Captafol의 유출특성)

  • As a first step of pesticide runoff studies, runoff losses of captafol were measured under natural rainfall conditions in apple orchard area. The maximum concentration of captafol was 180 ppb at 5 th sampling period when the rainfall occurred within 24 hours after captafol was applied, and the concentration of samples from other periods was below than 20 ppb. Total runoff loss of captafol was below 0.1%. About 10 fold of dilution factor was observed at the merging point with stream near outlet from orchard and about 50 fold was observed at the next merging point which is located further down. Therefore, captafol will not harm the aquatic organisms due to dilution factor$(10{\sim}50\;fold)$ and rapid hydrolytic degradation rate even when it was run off into a stream nearby.

On-site Output Survey and Feed Value Evaluation on Agro- industrial By-products (농산업부산물들에 대한 배출 현장 조사 및 사료적 가치 평가)

  • This study was conducted to make on-site survey on the output pattern and utilization situation of 19 by-products selected, to evaluate their nutritional characteristics, to find out a reliable index with which digestion of by-products can be predicted on the basis of chemical compositions analyzed and to diagnose the risk of using book values in the absence of the actual values analyzed for diet formulation. Production and utilization situations of by-products were quite various. Nutritionally, fruit processing by-products such as apple pomace (AP), pear pomace (PP), grape pomace (GP), and persimmon peel (PSP), and bakery by-products (BB) were classified as energy feeds. Soybean curd meal (SCM), animal by- products such as blood (BD), feather meal (FM) and poultry by-products (PB), and activated milk processing sludge (AMS) were classified as protein feeds. Soy hulls (SH), spent mushroom compost (SMC), barley malt hulls (BMH), waste paper (WP) and broiler litter (BL) were classified as roughage. Rumen contents (RC) and restaurant food waste (FW) were nutritionally analogous to complete diets for cattle and swine, respectively. Compared to soybean meal (SBM), BD and FM contained high (P<0.05) levels of amino acids and barley malt sprouts (BMS), AMS and FW contained low (P<0.05) levels of amino acids. Enzymatic (pepsin) digestibilities of proteinaceous feeds ranged between 99 and 66%. In vitro DM digestibility was high (P<0.05) in the order of FW, BB, AP, SH, PP, PSP, BMH, BMS, SCM, GP, RC, PB, BL, WP, SMC, AMS, FM and BD. In vitro DM digestibility had the highest correlation (r=0.68) with nonfibrous carbohydrate among chemical components. Differences between analyzed values of chemical components and book values were considerable. Caution is required in using book values when large amount of by-products are used in diets.

Growth and crop residue of soybean and barley grown at high paraquat level of the orchard soil (고농도 paraquat 잔류 과원토양에서의 콩과 보리 생육 및 작물 잔류)

  • Effect of soil residue paraquat (1,1-dimethyl-4,4-dipyridinium dichloride) on growth of barley (Hordeum vulgare L. cv. Sacheon No.6 and cv. Tapgolbori) and soybean [Glycine max (L.) Merr. cv. Alcheon and Danyeop] was investigated. Changes in soil residue paraquat during the cultivation period and residue amount in the p1ants at harvest were also determined. Experiments were conducted at two paraquat residue conditions; the first was done in an apple orchard soil where paraquat residue recorded 30.2 ppm in 1996, but decreased to about 9 to 9.8 ppm at the time of crop seeding and the second was conducted in the soil fortified to about 27 to 32 ppm paraquat residue. In both conditions, no crop injury due to the residue paraquat was observed and number of emerged seedlings and plant height of the two crops were not affected by soil residue paraquat. Residue amount of paraquat in the plants occurred less than 0.5 ppm detection limit. At the first condition, soil residue paraquat was further slightly decreased for 90 days after seeding, while no great change in the residue level was found at the second condition for 30 days after seeding. The results suggest that no carry-over effect occurs at about 30 ppm of soil residue paraquat and at present crop cultivation in Korean orchard soils are safe with respect to crop growth and paraquat residue in the plants.

Amplified mitochondiral DNA identify four species of Tetranychus mites (Acarina: Tetranychidae) in Korea (미토콘드리아 DNA증폭을 이용한 한국의 잎응애속(Tetranychus;Acarina: Tetranychidae) 4종의 동정방법)

  • Except for a cosmopolitan and major pest of apples, Tetranychus urticae Koch, Tetranychus mites in Korea such as T. viennensis Zaher, T. kanzawai Kishida, and T. truncams Ehara have been considered as quarantine pests by Canada and United States. Even though these mites are not feeders on apples, they are suspected to attach accidentally on apple h i t s in autumn as females enter the diapause. The characters used to identify Tetranychus mites have been confined to the shape of aedeagus in adult male. To develope a fast and accurate alternative identification protocol applied to hibernating female mites on apples, their mitochondrial DNA (mtDNA) were examined to find out any polymorphisms to discriminate each species from the other ones. Three pairs of primers for polymerase chain reaction (PCR) were used to amplify cytochrome oxidase subunit I (CO-I) coding region in mitochondrial DNA5 of four species of Tetranycus mites. The longest amplified product was estimated its size as about 680 bp. Digestion with restriction enzymes, AluI, Ddel, and Sau3A, showed length polymorphisms, which will he useful as diagnostic markers to identify Tetranychus mites. Schematic restriction maps in amplified region were shown for each species.
